Escherichia coli produit diverses entérotoxines thermolabiles et thermostables. STb est une toxine de faible poids moléculaire résistant à la chaleur chargée de la diarrhée chez les animaux de la ferme. Une étude antérieure a montré que les cellules ayant internalisé la toxine STb provoquent un dysfonctionnement de la barrière épithéliale par des changements dans les protéines des jonctions serrées (TJ). Ces modifications contribuent probablement à la diarrhée observée. Pour mieux comprendre le mécanisme de l'augmentation de la perméabilité intestinale, nous avons traité les cellules du côlon humain (T84) avec la toxine purifiée STb une fois que les cellules ont été récoltées et les protéines extraites. Après l'utilisation d'une solution contenant 1% de Nonidet P-40 (un détergent non dénaturant, non ionique), nous avons étudié la distribution de la claudine -1, une protéine majeure des TJs, responsable de l'imperméabilité de l'épithélium, entre la membrane (NP40-insoluble) et le cytoplasme (NP40-soluble). En utilisant l’immunoblot et la microscopie confocale, nous avons observé que le traitement des monocouches de cellules T84 avec STb induit la redistribution de la claudine-1. Après 24h, les cellules cultivées en milieu faible en Ca+ (5 uM) et traitées par STb, ont montré qu’environ 40 % de plus de la claudine-1 se sont délogées dans le cytoplasme par comparaison au contrôle. En passant d’un milieu faible à un milieu contenant des quantités physiologiques de Ca++ (1,8 mM) nous avons observé une augmentation du taux de claudine- 1 délogé, comme la délocalisation comparable et ce, après 6h. Un milieu supplémenté avec la même concentration de Mg++ ou Zn++ n'a pas affecté le taux de délogement comparé au milieu contenant une faible teneur en Ca++. En utilisant des anticorps anti-phosphosérine et anti-phosphothréonine, nous avons observé que la perte des claudines-1 de la membrane a été accompagnée par une déphosphorylation de cette protéine des TJs. Dans l'ensemble, nos résultats ont montré une importante redistribution de la claudine-1 dans les cellules traitées par la toxine STb. La perte de la claudine-1 phosphorylée de la membrane est susceptible d'être impliquée dans la perméabilité accrue observée. Les mécanismes par lesquels ces changements sont provoqués restent à élucider.

La toxine thermostable d’E.coli (STb) est une cause de diarrhée chez l’homme et l’animal. STb se lie au sulfatide, son récepteur, puis s’internalise. Dans le cytoplasme, par une cascade d’événements, STb déclenche l’ouverture des canaux ioniques permettant la sécrétion des ions et la perte d’eau menant à la diarrhée. Les jonctions serrées forment une barrière physique intercellulaire dans les cellules épithéliales intestinales, contrôlant ainsi le flux paracellulaire des ions et de l’eau. Les jonctions serrées sont affectées par divers pathogènes et par leurs toxines. À ce jour, l’effet de STb sur les jonctions serrées n’a pas été étudié. L’étude entreprise visait à explorer l’effet de STb sur les jonctions serrées et la barrière épithéliale des cellules intestinales. Des cellules épithéliales intestinales du colon humain (T84) ont été traitées pendant 24h soit avec la toxine STb purifiée soit avec une souche d’E.coli exprimant STb. La résistance transépithéliale (TER), le flux de marqueurs paracellulaires et la microscopie confocale ont été utilisés pour analyser les effets de STb sur les jonctions serrées. Les monocouches traitées par la souche E.coli exprimant STb et la toxine STb purifiée ont manifesté une forte réduction de TER (p<0.0001) parallèlement à une augmentation significative de la perméabilité paracellulaire à l’Albumine de Sérum Bovin marqué avec l’IsoThioCyanate Fluoroscéine, BSA-FITC (p<0.0001) comparativement aux cellules non traitées et aux cellules traitées par une souche d’E.coli commensale non-toxinogène. L’augmentation de la perméabilité paracellulaire induite par STb a été associée à une dissolution générale et une condensation des fibres de stress centrales des filaments d’actine. Le réarrangement des filaments d’actine a été accompagné par une redistribution et une fragmentation des protéines des jonctions serrées dont l’occludine, la claudine-1 et la Zonula Occludens-1. Les mêmes modifications on été observées après l’intoxication des cellules T84 avec un octapeptide synthétique retrouvé dans la séquence de STb correspondant à une séquence consensus de la toxine ZOT de Vibrio cholerae, impliquée dans la réorganisation des jonctions serrées. Cet effet n’a pas été observé lorsque les cellules ont été traitées avec un octapeptide synthétique comportant les mêmes acides aminés mais distribués de façon aléatoire ou avec la toxine mutée (D30V). Nos résultats montrent pour la première fois que STb induit le dysfonctionnement de la barrière épithéliale intestinale en modifiant la distribution des protéines des jonctions serrées. Ces résultats ouvrent une nouvelle voie pour la compréhension de la pathogenèse de diarrhée causée par la toxine STb.

Background The epidemiology of dengue in the South Pacific has been characterized by transmission of a single dominant serotype for 3–5 years, with subsequent replacement by another serotype. From 2001 to 2008 only DENV-1 was reported in the Pacific. In 2008, DENV-4 emerged and quickly displaced DENV-1 in the Pacific, except in New Caledonia (NC) where DENV-1 and DENV-4 co-circulated in 2008–2009. During 2012–2013, another DENV-1 outbreak occurred in NC, the third DENV-1 outbreak in a decade. Given that dengue is a serotype-specific immunizing infection, the recurrent outbreaks of a single serotype within a 10-year period was unexpected. Findings This study aimed to inform this phenomenon by examining the phylogenetic characteristics of the DENV-1 viruses in NC and other Pacific islands between 2001 and 2013. As a result, we have demonstrated that NC experienced introductions of viruses from both the Pacific (genotype IV) and South-east Asia (genotype I). Moreover, whereas genotype IV and I were co-circulating at the beginning of 2012, we observed that from the second half of 2012, i.e. during the major DENV-1 outbreak, all analyzed viruses were genotype I suggesting that a genotype switch occurred. Conclusions Repeated outbreaks of the same dengue serotype, as observed in NC, is uncommon in the Pacific islands. Why the earlier DENV-1 outbreaks did not induce sufficient herd immunity is unclear, and likely multifactorial, but the robust vector control program may have played a role by limiting transmission and thus maintaining a large susceptible pool in the population. Monolayers of octadecanethiolate on Au(1 1 1) surface were formed under electrochemical control. The influence of the formation time on the reductive desorption process was studied by cyclic voltammetry and chronoamperometry. When the formation time is increased, the reductive desorption peak observed on the voltammograms is significantly shifted in the negative direction, while the cathodic charge is only slightly affected. This behaviour is attributed to a higher degree of organisation of the monolayers for longer formation times, highlighting the role of defect sites in promoting the dissolution. A good agreement was found between our experimental chronoamperograms and theoretical models describing the dissolution process by a shrinkage mechanism. It is demonstrated that a reorganisation process takes place, consisting in the merging of small condensed domains into larger ones. This annealing phenomenon is time and potential dependent, the largest condensed domains being obtained for the longest formation times and least negative potentials. © 2008 Elsevier B.V. The description of the monolayer formed at Au(1 1 1) by 2-mercaptobenzimidazole (MBI) under potential control has been based on electrochemical data (charge measurements) and spectroscopic information from the subtractively normalized interfacial Fourier transform infrared spectroscopy method (SNIFTIRS). From the quantitative analysis of the SNIFTIR spectra, a surface coverage Γ/Γmax was extracted for each sample potential. The evolution of the coverage with potential was in full agreement with the charge density curve. The shift of the pzc in the presence of MBI indicates that the adsorbed molecules have a nonzero component of the permanent dipole moment in the direction perpendicular to the electrode surface. Thanks to the high quality of the spectra, it was possible to determine the orientation of MBI molecules at the surface in the monolayer and submonolayer range. The angle between the C2-axis of the molecule and the direction normal to the surface is close to 64 ± 4° and its small change (<15°) with potential indicates that the orientation of the molecules is chiefly controlled by the chemical interaction between the sulphur atom and the gold surface. © 2005 Elsevier Ltd. The behaviour of a self-assembled monolayer of 2-mercaptobenzimidazole (MBI) at the Au(111) electrode has been examined using cyclic voltammetry and in situ FTIR spectroscopy. The charge associated with the reductive desorption is pH independent while the oxidative partial redeposition charge increases when the pH is lowered. This is due to differences between the nature and the solubility of the MBI desorption product. In alkaline and neutral media MBI desorbs as the thiolate. In contrast, in acidic solutions the thiol is the desorbed product. Subtractively normalized interfacial reflection Fourier transform absorption spectroscopy (SNIFTIRS) has been applied to investigate the MBI monolayer in contact with aqueous solutions of different pH. The SNIFTIRS data are in agreement with the electrochemical results. Moreover, quantitative analysis of the IR data provided evidence that adsorbed MBI molecules assume a tilted orientation with an angle of 60±5° between the C2 axis of the molecule and the direction normal to the gold surface. © 2003 Elsevier B.V. OBJECTIVE: To determine the effects of cognitive-behavioral stress management (CBSM) training on clinical and psychosocial markers in HIV-infected persons. METHODS: A randomized controlled trial in four HIV outpatient clinics of 104 HIV-infected persons taking combination antiretroviral therapy (cART), measuring HIV-1 surrogate markers, adherence to therapy and well-being 12 months after 12 group sessions of 2 h CBSM training. RESULTS: Intent-to-treat analyses showed no effects on HIV-1 surrogate markers in the CBSM group compared with the control group: HIV-1 RNA < 50 copies/ml in 81.1% [95% confidence interval (CI), 68.0-90.6] and 74.5% (95% CI, 60.4-85.7), respectively (P = 0.34), and mean CD4 cell change from baseline of 53.0 cells/microl (95% CI, 4.1-101.8) and 15.5 cells/microl (95% CI, -34.3 to 65.4), respectively (P = 0.29). Self-reported adherence to therapy did not differ between groups at baseline (P = 0.53) or at 12 month's post-intervention (P = 0.47). Significant benefits of CBSM over no intervention were observed in mean change of quality of life scores: physical health 2.9 (95% CI, 0.7-5.1) and -0.2 (95% CI, -2.1 to 1.8), respectively (P = 0.05); mental health 4.8 (95% CI, 1.8-7.3) and -0.5 (95% CI, -3.3 to 2.2) (P = 0.02); anxiety -2.1 (95% CI, -3.6 to -1.0) and 0.3 (95% CI, -0.7 to 1.4), respectively (P = 0.002); and depression -2.1 (95% CI, -3.2 to -0.9) and 0.02 (95% CI, -1.0 to 1.1), respectively (P = 0.001). Alleviation of depression and anxiety symptoms were most pronounced among participants with high psychological distress at baseline. CONCLUSION: CBSM training of HIV-infected persons taking on cART does not improve clinical outcome but has lasting effects on quality of life and psychological well-being.

We report two patients with microdeletions in chromosomal subdomain 15q26.1 encompassing only two genes, CHD2 and RGMA. Both patients present a distinct phenotype with intellectual disability, epilepsy, behavioral issues, truncal obesity, scoliosis and facial dysmorphism. CHD2 haploinsufficiency is known to cause intellectual disability and epilepsy, RGMA haploinsufficiency might explain truncal obesity with onset around puberty observed in our two patients.

Full text: The title of the book gives us a major clue on the innovative approach developed by Anne Freadman in her analysis of a particular Colette corpus, the one devoted to auto-biographical writing: Les Vrilles de la vigne, Mes apprentissages, La Maison de Claudine, Sido ,L’E ́toile Vesper and Le Fanal bleu. Freadman follows the powerful lure of Rimbaldianvieilles vieilleries and its echoes with Colette’s fondness for collecting objects, people and memories. To this must be added a technical aspect, that of the study of the genre of Colette’s writing. Freadman argues that, by largely avoiding the autobiographical form, the writer achieves a new way of ‘telling time’, collecting anecdotes and detail taken from the quotidian and setting them within an all-encompassing preoccupation with time. This provides the second part of the title.The sonata form directs the sequence of the book, orchestrated into five parts,from ‘exposition’ to ‘first subject’ to‘bridge’ to ‘second subject’ to ‘recapitulation’. This has the advantage of enabling Freadman to move and progress between distinct themes—autobiography first,then alternative forms—with grace,whilst preserving within her own writing what she sees as the essence of Colette’s relationship to time in her ‘Livres-Souvenirs’, the telling of time. This‘telling of time’ is itself therefore cleverly subjected to the time constraints and freedoms of musical composition. Freadman’s ‘Exposition’ takes us through a discussion of the autobiographical genre, analysing the texts against anumber of theorists, from Lejeune to Benjamin and Ricoeur, before launching into ‘Colette and Autobiography’. It argues pertinently that Colette did not write a ‘sustained’ autobiography, even inthe most autobiographical of her writings, Mes apprentissages. Measured against Goodwin’s three sources for autobiography, confession, apologia and memoirs, Colette’s autobiographical writings appear to be at odds with all of them. Freadman then goes on in Part II of her argument, to persuasively uncover a project that rejects self-scrutiny and with no autobiographical strategy. In ‘Collecting Time’, despite claims of continuity, narrative logic and causality areabandoned in favour of a collection offragments, family stories that are built up generation after generation into familylegends. A close and fruitful analysis of Sidoleads us to a study of ‘The Art of Ending’, concentrating on L’E ́toile Vesperandle Fanal Bleu. The closing chapter gives a fascinating reading of La Naissance du jouras an exemplar of the way in which the two subjects developed in Freadman’s volume are cast together:Colette’s own working through the autobiographical genre, and her refusal to write memoirs, in favour of collecting memories, and the strategies she uses for her purpose. In ‘Recapitulation’, her concluding chapter, Freadman adroitlyen capsulates her analysis in a fetching title: ‘Fables of Time’. Indeed, the wholepremise of her book is to move away from autobiographical genre, having acknowledged the links and debt the corpus owes to it, and into a study of the multiple and fruitful ways in which Colette tells time.The rich and varied readings of thematerial, competently informed by theoretical input, together with acute sensitivity to the corpus, mark out this study as incontournable for Colette scholars.
