Cytogenetic analysis were done on specimens from two populations of Lysapsus limellus limellus. three of L. l. bolivianus and of one of Lysapsus caraya. All animals showed a diploid chromosomal number of 2n=24. The karyotypes of the two L. limellus subspecies were very similar, differing only by the larger amount of telomeric heterochromatin and a small pericentromeric C-band on the short arms of pair 2 in L. l. limellus specimens. The karyotype of L. caraya differed from those of the two L. limellus subspecies in terms of chromosomal morphology, C-banding pattern and location of the main NOR on chromosomes 7 and 6. respectively. The karyotype of the L. l. bolivianus population from Guajara-Mirim/RO differed from those of the other populations of the same subspecies in morphology and heterochromatin pattern of chromosomes 7 and 8. Additional NORs were detected by silver staining and confirmed by FISH in one of the homologues of pairs 1 and 8 in L. l. bolivianus and in pair 7 in L. caraya. These results suggest that a reassessment of the taxonomic status of L. limellus subspecies, especially of the L. l. bolivianus populations, may be necessary. (c) 2005 Elsevier Ltd. The speciose Brazilian Elateridae fauna is characterized by high karyotypic diversity, including one species (Chalcolepidius zonatus Eschscholtz, 1829) with the lowest diploid number within any Coleoptera order. Cytogenetic analysis of Conoderus dimidiatus Germar, 1839, C. scalaris (Germar, 1824,) C. ternarius Germar, 1839, and C. stigmosus Germar, 1839 by standard and differential staining was performed with the aim of establishing mechanisms of karyotypic differentiation in these species. Conoderus dimidiatus, C. scalaris, and C. ternarius have diploid numbers of 2n(male) = 17 and 2n(female) = 18, and a X0/XX sex determination system, similar to that encountered in the majority of Conoderini species. The karyotype of C. stigmosus was characterized by a diploid number of 2n=16 and a neoXY/neoXX sex determination system that was highly differentiated from other species of the genus. Some features of the mitotic and meiotic chromosomes suggest an autosome/ancestral X chromosome fusion as the cause of the neoXY system origin in C. stigmosus. C-banding and silver impregnation techniques showed that the four Conoderus species possess similar chromosomal characteristics to those registered in most Polyphaga species, including pericentromeric C band and autosomal NORs. Triple staining techniques including CMA(3)/DA/DAPI also provided useful information for differentiating these Conoderus species. These techniques revealed unique GC-rich heterochromatin associated with NORs in C. scalaris and C. stigmosus and CMA(3)-heteromorphism in C. scalaris and C. ternarius.

Specimens of Hyla nana and Hyla sanborni from a syntopic population were studied cytogenetically. These species are morphologically very similar and are frequently misidentified, confused with each other. Both species had a diploid chromosome number, 2n = 30. However, the karyotypes of H. nana and H. sanborni differed considerably from each other in the number of submetacentric and telocentric chromosomes. The two species also differed in their primary NOR-bearing chromosomes (metacentric pair 13 in H. nana and telocentric pair 12 in H. sanborni). Additional nucleolus organizer regions (NORs) were detected by Ag-NOR staining and FISH in chromosome pairs 1, 5, 6, 12, and 14 in seven specimens of H. nana. Thus, a total of six patterns of NOR were identified. These differences in karyotype and in NOR location allowed the unambiguous identification of syntopic individuals of the two species. However, the chromosomal morphology of both species differed from that reported for populations from other geographic regions, suggesting that a systematic reevaluation of this group of Hyla may be necessary.

The chromosome study of five species of the family Elateridae, belonging to the subfamilies Agrypninae and Elaterinae, and the analysis of the cytogenetic data previously recorded for this family permitted the establishment of the main strategies of karyotypic differentiation that has occurred in the elaterids. In Agrypninae, the three species studied (Conoderus fuscofasciatus, Conoderus rufidens, and Conoderus sp.) showed the male karyotype 2n = 16 + X0. This karyotypic uniformity detected in these Conoderus species has also been shared with other species of the same genus, differing considerably from chromosomal heterogeneity verified in the subfamily Agrypninae. The use of the C-banding technique in C. fuscofasciatus and Conoderus sp. revealed constitutive heterochromatin in the pericentromeric region of the majority of the chromosomes. In C. fuscofasciatus, additional constitutive heterochromatin were also observed in the long arm terminal region of almost all chromosomes. Among the representatives of Elaterinae, the karyotype 2n = 18 + Xy(p) of Pomachilius sp.2 was similar to that verified in the majority of the Coleoptera species, contrasting with the chromosomal formula 2n = 18 + X0 detected in Cardiorhinus rufilateris, which is most common in the species of Elaterinae. In the majority of the elaterids, the chromosomal differentiation has frequently been driven by reduction of the diploid number; but, among the four cytogenetically examined subfamilies, there are some differences in relation to the trends of karyotypic evolution. Drosophila serido is considered to be a superspecies consisting of two species: D. serido, from Brazil and D. koepferae from Argentina and Bolivia. However this probably does not express the entire evolutionary complexity of its populations. Isofemale lines A95F3 (from Brazil) and B20D2 (from Argentina), at present representing, respectively, the first and second species, were analyzed for fertility and fecundity in pair-mating intracrosses and intercrosses, as well as for development time, banding patterns and asynapsis of polytene chromosomes in the isofemale lines and their hybrids.Although variations in experimental conditions resulted in some variability in the results, in general A95F3 fertility and fecundity were lower than in B20D2. Intercrosses of A95F3 females and B20D2 males showed lower fertility and fecundity than the reciprocal crosses, following more closely characteristics of the mother strains. This is in contrast to the results obtained by Fontdevilla et al. (An. Entomol. Soc. Amer. 81: 380-385, 1988) and may be due to the different geographic origin of D. serido strains they used in crosses to B20D2. This difference and others cited in the literature relative to aedeagus morphology, karyotype characteristics, inversion polymorphisms and reproductive isolation strongly indicate that A95F3 and D. serido from the State of Bahia, Brazil are not a single evolutionary entity, reinforcing the idea of greater complexity of the superspecies D. serido than is known today.The reproductive isolation mechanisms found operating between A95F3 and B20D2 were prezygotic and postzygotic, the latter included mortality at the larvae stage in both directions of crosses and sterility of male hybrids in intercrosses involving B20D2 females and A95F3 males. The two isofemale lines differed in egg-adult development time, which was also differently affected by culture medium composition.A95F3 and B20D2 also showed differences in the banding patterns of proximal regions of polytene chromosomes 2, 3 and X, a fixed inversion in chromosome 3 (here named 3t), apparently not described previously, and a high degree of asynapsis in hybrids.These observations, especially those related to reproductive isolation and chromosomal differentiation (including the karyotype, previously described, and the differentiation of banding patterns, described in this paper), as well as the extensive asynapsis observed in hybrids reinforces the distinct species status of A95F3 and B20D2 isofemale lines.

Among the Opiliones, species of the suborders Cyphophthalmi, Eupnoi, Dyspnoi and Laniatores have shown very diverse diploid chromosome numbers. However, only certain Eupnoi species exhibit XY/XX and ZZ/ZW sex chromosome systems. Considering the scarcity of karyotypical information and the absence of structurally identifiable sex chromosomes in the suborder Laniatores, we decided to analyse the chromosomes and bivalents of Discocyrtus pectinifemur (Gonyleptidae) to identify possible sex differences. Testicular cells examined under light microscopy showed it high diploid number, 2n = 88, meta/submetacentric chromosome morphology and a nucleolar organizer region on pair 35. Prophase I microspreading observed in transmission electron microscopy exhibited 44 synaptonemal complexes with similar electron density and thickness. The total and regular synapsis between the chromosomes of the bivalents was also noted in pachytene nuclei. Male mitotic and meiotic chromosomes revealed no distinct characteristic that could be related to the occurrence of heteromorphic sex chromosomes. Evolutionary trends of chromosome differentiation in the four suborders of Opiliones are discussed here.

Foram realizados estudos citogenéticos e morfológicos em nove populações locais de Astyanax scabripinnis paranae. Todas as populações apresentaram 2n = 50 cromossomos e conspícuas diferenças envolvendo a morfologia dos cromossomos, número e posição das NORs e quantidade e localização dos blocos de heterocromatina constitutiva. O estudo quantitativo dos dados citogenéticos, usando a análise das variáveis canônicas (CVA), mostraram que oito populações apresentaram diferentes constituições cariotípicas. Análises morfológicas baseadas em nove caracteres morfométricos e dois merísticos foram eficazes no estabelecimento de uma clara identificação de cinco populações. Análises comparativas dos dados citogenéticos e morfológicos sugerem que as modificações cromossômicas e morfológicas ocorreram em diferentes taxas, sendo as cromossômicas mais rápidas do que as morfológicas. A despeito da similaridade morfológica encontrada entre algumas populações, diferenças cromossômicas foram detectadas entre todas as populações estudadas.

We studied the karyotypes of Hassar cf. orestis and an undescribed Hassar species from the Jarí River and Opsodoras ternetzi, H. orestis and Platydoras cf. costatus from the Xingú River, all with 2n = 58. Constitutive heterochromatin is located in the centromere in most metacentric pairs; in some chromosomes this banding is not present, or it is located on the whole chromosome arm or in the distal regions. The NOR is located on a single biarmed pair at a distal region of the short arm in H. cf. orestis, H. orestis and P. cf. costatus at a distal region of the long arm in O. ternetzi and at a proximal region of the long arm in the Hassar species. In all species (except for Hassar sp.) the CMA3 analysis revealed a rich G-C region coincident with the NOR. Probably inversions occurred in the NOR chromosome during the chromosomal differentiation of the Doradidae species here described.

Os morcegos representam um grupo amplamente distribuído e diversificado. A diversidade de hábitos alimentares faz da ordem Chiroptera uma das mais bem sucedidas entre os mamíferos, desempenhando, em função de seus hábitos, um importante papel no controle de insetos, na polinização e na dispersão de sementes de numerosos vegetais. A família Phyllostomidae constitui a terceira maior família em número de espécies dentro da Ordem Chiroptera. Entre as representantes neotropicais é a mais numerosa, sendo encontrada em florestas tropicais da America do Sul, particularmente, concentrada na Amazônia que é a região com maior diversidade de morcegos do mundo. No presente trabalho foram analisados citogeneticamente exemplares de três espécies da subfamília Phyllostominae: Chrotopterus auritus, Trachops cirrhosus e Vampyrum spectrum coletados no estado do Pará e Amazonas. Os dados cromossômicos obtidos para Chrotopterus auritus (2n = 28 e NF = 52) e Trachops cirrhosus (2n = 30, FN = 56) estão de acordo com os descritos na literatura. Para Vampyrum spectrum (2n=30 NF=56) relatamos os primeiros padrões de bandeamento e FISH (Hibridização in situ Fluorescente). A técnica de bandeamento C demonstrou um padrão pericentromérico de distribuição da heterocromatina constitutiva nas três espécies estudadas. A técnica de FISH com sondas de DNA teloméricas humanas mostrou apenas marcações distais em todos os cromossomos das três espécies e as sondas de rDNA 18S confirmaram a localização das Regiões Organizadoras Nucleares observadas na técnica de Ag-NOR, presentes no braço longo do par 2 de Chrotopterus auritus, no par 11 de Trachops cirrhosus e no braço longo do par 1 de Vampyrum spectrum. A análise comparativa entre elas sugere um extenso grau de diferenciação cromossômica, com poucos cromossomos compartilhados entre os três gêneros. Contudo, cinco pares cromossômicos inteiros se mantiveram conservados sem nenhum tipo de rearranjo após a divergência das três linhagens. A comparação entre as espécies revela que C. auritus e V. spectrum apresentam mais elementos compartilhados entre si do que em relação à T. cirrhosus. Nossos resultados apoiam a proximidade filogenética entre C. auritus e V. spectrum e sugerem a associação de T. cirrhosus com o clado do gênero Phyllostomus.

The present study aimed to cytogenetic analysis and structural and molecular level of four fish species of the genus Trichomycterus: T. diabolus, T. iheringi, T. zonatus and T. cf. mimonha collected in different river basins in Brazil. Techniques were used for classical cytogenetic (Giemsa, Silver nitrate impregnation, C-banding) and molecular with the chromosomal location of genes for 18S and 5S rDNA. All individuals examined had a diploid number 54 chromosomes and karyotype consisting of types of metacentric, submetacentric and subtelocentric. The constitutive heterochromatin identified by C-banding was observed in two small blocks in the karyotype of T. diabolus and large blocks of centromeric several pairs in chromosomal karyotypes of T. iheringi, T. zonatus and T. cf. mimonha. The Silver nitrate impregnation and hybridization with 18S rDNA probe revealed the existence of only a couple carryng nucleolar organizing regions (NORs) on the species T. diabolus, T. iheringi and T. cf mimonha, and two pairs carrying 18S rDNA in T. zonatus. The 5S rDNA was observed in interstitial position 6 of the pair in T. iheringi, the synteny with the two pair in 18S rDNA of T.diabolus in pericentromeric position of and two pairs submetacentric, one being also a case of synteny with the 18S rDNA in T. zonatus and T. cf. mimonha, this rDNA was located in the pairs 3, 18 and 25, and synteny in the 18S rDNA pair 18. Although representatives of these four species Trichomycterus present diploid number and karyotypic formula preserved, three is specific about the distribuition patterns of heterochromatin and location of rDNA sequences, indicating that chromosomal differentiation events in this group of fish are acting directly on these genomic portions
