In the adult olfactory nerve pathway of rodents, each primary olfactory axon forms a terminal arbor in a single glomerulus in the olfactory bulb. During development, axons are believed to project directly to and terminate precisely within a glomerulus without any exuberant growth or mistargeting. To gain insight into mechanisms underlying this process, the trajectories of primary olfactory axons during glomerular formation were studied in the neonatal period. Histochemical staining of mouse olfactory bulb sections with the lectin Dolichos biflorus-agglutinin revealed that many olfactory axons overshoot the glomerular layer and course into the deeper laminae of the bulb in the early postnatal period. Single primary olfactory axons were anterogradely labelled either with the lipophilic carbocyanine dye, 1,1'-dioctodecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI), or with horseradish peroxidase (HRP) by localized microinjections into the nerve fiber layer of the rat olfactory bulb. Five distinct trajectories of primary olfactory axons were observed in DLI-labelled preparations at postnatal day 1.5 (P1.5). Axons either coursed directly to and terminated specifically within a glomerulus, branched before terminating in a glomerulus, bypassed glomeruli and entered the underlying external plexiform layer, passed through the glomerular layer with side branches into glomeruli, or branched into more than one glomerulus. HRP-labelled axon arbors from eight postnatal ages were reconstructed by camera lucida and were used to determine arbor length, arbor area, and arbor branch number. Whereas primary olfactory axons display errors in laminar targeting in the mammalian olfactory bulb, axon arbors typically achieve their adult morphology without exuberant growth. Many olfactory axons appear not to recognize appropriate cues to terminate within the glomerular layer during the early postnatal period. However, primary olfactory axons exhibit precise targeting in the glomerular layer after P5.5, indicating temporal differences in either the presence of guidance cues or the ability of axons to respond to these cues. The plant architecture hypothesis predicts that variation in host plant architecture influences insect herbivore community structure, dynamics and performance. In this study we evaluated the effects of Macairea radula (Melastomataceae) architecture on the abundance of galls induced by a moth (Lepidoptera: Gelechiidae). Plant architecture and gall abundance were directly recorded on 58 arbitrarily chosen M. radula host plants in the rainy season of 2006 in an area of Cerrado vegetation, southeastern Brazil. Plant height, dry biomass, number of branches, number of shoots and leaf abundance were used as predicting variables of gall abundance and larval survival. Gall abundance correlated positively with host plant biomass and branch number. Otherwise, no correlation (p > 0.05) was found between gall abundance with shoot number or with the number of leaves/plant. From a total of 124 galls analyzed, 67.7% survived, 14.5% were attacked by parasitoids, while 17.7% died due to unknown causes. Larvae that survived or were parasitized were not influenced by architectural complexity of the host plant. Our results partially corroborate the plant architecture hypothesis, but since parasitism was not related to plant architecture it is argued that bottom-up effects may be more important than top-down effects in controlling the population dynamics of the galling lepidopteran. Because galling insects often decrease plant fitness, the potential of galling insects in selecting for less architectural complex plants is discussed.

Temperate and temperate x tropical germplasms were introduced into the maize breeding of the Department of Genetics (ESALQ-USP). Twenty populations designated as P-1 to P-20 and a check population were evaluated in a randomized complete block design with two replications at Piracicaba, SP, Brazil. Plots were 2.0 m long, spaced 1.0 m apart, with 10 plants per plot after thinning. The following traits were evaluated: PH, plant height; EH, ear height; TB, tassel branch number; KW, 100 kernels weight; EL, ear length; ED, ear diameter; KR, kernels per row, and RN, number of rows of kernels. The means of the exotic populations showed a variation in performance for all traits. The population means were greater than check means for KW, EL, and KR. Populations P-9, P-10, P-12, and P-13 showed low relative ear placement values (EH/PH index), indicating that these materials could contribute to lower ear placement in local populations. P-1 and P-9 showed a high potential to reduce TB. The phenotypic correlation coefficients among populations were positive for most combinations, except for RN with PH, EH, TB, and KW. Predictions of 19 composite means were obtained for all traits.

Two populations of Chaetophora elegans (Roth) C. Agardh and two of Stigeoclonium helveticum Vischer were investigated for microhabitat characteristics and morphological variation in streams of Sao Paulo State, southeastern Brazil. Different patterns of microhabitat distribution were found between species investigated. Populations of C. elegans were distributed under relatively narrow microhabitat conditions (high irradiance, low depth, moderate to high current velocity, rocky substrata and lower values of niche width) and showing little morphometric variation (colony diameter, main axis cell size, and apical branch number). Stigeoclonium helveticum occurred under more diverse microhabitat conditions, revealed by lack of significant difference between sampling units with and without the alga and wider niche width, but also exhibited relatively narrow morphometric variation (plant length, main axis cell and lateral branch cell sizes). The narrow microhabitat conditions and smaller niche width of C. elegans can explain its low abundance (percentage cover) in streams from the area studied as well as in other regions of Sao Paulo State. In contrast, the wider variation of microhabitat conditions and the higher niche widths of S. helveticum suggest that this green alga is able to grow in a high number of stream ecosystems in the region investigated, ranging from undisturbed to highly disturbed habitats. Thus, the results suggest that S. helveticum is a generalist species.

The present work was conducted with the objective to study the effects of mineral nutrition on Eucalyptus grandis and Brachiaria decumbens (#BRADC) growth, when submitted to inter- and intra-specific competition. The treatments consisted of two plants of Eucalypts/pot, two plants of BRADC/pot, and one plant of each species/pot. The plants were nourished with the Hoagland e Arnon (1950) complete solution; with no K, P or N; or only with either N, P or K. Sixty days after growing side by side, no effect of the inter- and intra-specific competition on eucalypts plant high, branch number and leaf dry mass was observed. When mineral nutrition did not limit plant growth (complete solution or solution with no K), intra-specific competition reduced on average, 23% of eucalypts root length, leaf area, and stem and root dry mass, and inter-specific competition reduced, on average, 75% of BRADC dry mass. When mineral nutrition became a limiting factor, no plant competition effect on the parameters studied was detected.

In order to verify the effect of Panicum maximum plant density on the initial growth of Eucalyptus grandis plants, one assay was conducted under semi-controlled conditions of soil fertility and humidity. Dark red Latossol, collected from the arable layer, was used as substrate in 50 liters amianthus cement boxes. One seedling of Eucalyptus was planted in each box. Fifteen days later, seedlings of P. maximum were transplanted on the same box. The treatments consisted of 0, 4, 8, 12, 16 and 20 plants of P. maximum per m 2. The experimental design was the complete randomized design, with six treatments and eight replicates for P. maximum. One hundred and ten and 190 days after transplanting, Eucalyptus plants grown among P. maximum showed an average reduction of 30.80 and 46.55% stem diameter, 25.10 and 22.50 plant high, 40.18 and 31.29% stem dry weight, 61.32 and 54.06% branch dry weight, 53.72 and 51.82% root dry weight, 44.62 and 38.50% leaf dry weight, 22.51 and 23.16 branch number, 20.72 and 19.97% leaf number, and 33.88 and 17.05% leaf area, respectively.

The aim of this work is to describe the behavior of coffee (Coffea arabica L.) grown for nine years under organic management systems in full sun and shaded by banana trees (Musa sp.) and Erythrina verna Vell., in Valença, RJ. We performed a joint evaluation of vegetative characteristics, nutritional content and yield, with the aid of a principal component analysis. Twelve treatments were arranged in a randomized block design with four replications in a split plot. The plots evaluated farming systems in full sun and shade, and the subplots consisted of the following varieties of coffee: Tupi IAC 1669-33, MG 6851, IAC 3282 Icatu, Catucaí 2SL, Obatã IAC 1669-20; lineage IAC IAC 144. After five years we assessed the following variables, height, stem and canopy diameter, leaf area, number of branches, number of nodes per branch, number of leaves present, the distance between nodes, the percentage of green, ripe and dried fruit, number of dead plants, number of plants with death of the apical bud, coffee yield, and foliar concentrations of N, P, K, Ca and Mg. A multivariate analysis efficiently discriminates the variables in full sun and shaded cropping systems. Shading increases the percentage of green fruit, leaf area, height, diameter, distance between nodes, number of leaves on the branches, number of branches and leaf N content, but does not reduce the level of productivity when the shade is adequate.

The carbohydrates translocation and consequently growth and production of fig tree (Ficuscarica L.) vary according to the different management on cultivation conditions. The aim of this study was to evaluate the changes in the levels and total carbohydrates accumulation together with growth and “Roxo de Valinhos” fig trees production onimplementation of orchards in initial phase, cultivated with and without irrigation. We adopted a factorial arrangement (2 x 7) with four repetitions distributed in installments (with and without irrigation) subdivided in time (collect time). Destructive analyzes were performed at 40, 80, 120, 160, 200, 240 and 280 days after pruning (DAP) and are measured: stem diameter and branch, stem length and branch, number of leaves, internodes and fruit. Subsequently, the plant parts were sectioned to obtain the leaf area, length and roots volume, fresh and dry matter weight. The number, weight and total productivity of fruits were evaluated. The media of all growth attributes and production characteristics were higher in treatments with water irrigation. The total carbohydrate content was higher at 120 and 160 DAP and the carbohydrates accumulation was increasing for most institutions over the plants development, except for the leaves that showed a decrease in the levels at 160 DAP. The fruits showed greater carbohydrates accumulation in relation to the other evaluated organs.

The present study is a comparative functional analysis of three factors controlling glial differentiation in mouse (Fyn Src kinase, hnRNPF/H and NG2) and their homologues in Drosophila (Src42A and 64B, Glorund and Kon-tiki (Kon)). In Drosophila, mutations in any of these genes were not associated with major embryonic neurodevelopmental phenotypes. Src kinases and Glorund were shown to be ubiquitously expressed, whereas kon mRNA showed selective expression in muscles as well as in central and peripheral glia. Kon was also shown to be expressed in L3 larvae with high levels of protein accumulation at the neuromuscular junction (NMJ) and in muscles in the form of speckles. Knockdown of kon in glia resulted in NMJ phenotypes, mainly characterized by a significant increase in bouton number and a reduction in α-Konecto staining intensity at the NMJ. From the three glial layers ensheathing the peripheral nervous system, subperineurial glial showed to be the one contributing the most to kon knockdown dependent NMJ phenotypes, while perineurial glia only had a minor role. The knockdown of kon in glia also showed to affect Glutamate receptor subunit (α-GluRIIA) clustering in the postsynapse, same as microtubule arrangement in the presynapse, as seen by α-Futsch pattern interruptions and alterations. kon knockdown in glia also resulted in impaired axonal transport, as seen by the accumulation of Bruchpilot-positive vesicles along the nerves, abnormal formation of neuronal derived protrusions and swellings, filled with vacuole-like structures. Glia number along the peripheral nerves is also reduced as consequence of kon knockdown. Muscle derived Kon was shown to accumulate at the NMJ and play a role in bouton consolidation and to interfere with phagocytosis of ghost boutons. NMJ bouton and branch number was also significantly increased in Kon overexpression in glia. The overexpression of Kon in glia also resulted in a massive elongation of the ventral nerve cord, which served in a suppressor screen to identify intracellular interaction partners of Kon in glia. It was shown that Kon is processed in glia and preliminary results indicate that the metalloendopeptidase Kuzbanian (the fly homologue of ADAM10) may play a role in the shedding of Konecto. In the present work, Kon is shown as a multifunctional gene with various roles in glia-neuron and glia-neuron-muscle interaction.

Fast-growing tree species of Populus spp.,Salix spp. and Eucalyptus spp. are cultivated to produce wood in a short time. Poplars are cultivated with cycles of 15-18 years to obtain saw timber and peeler logs, but when grown as short -rotation coppice(SRC) to produce biomass, planting density increases and rotation is considerably reduced (3-5 years). In this regard, research efforts are focused in the identification of traits and loci that allow the generation of improved SRC biomass-yielding genotypes. Biomass yield is a highly complex trait as it is the combined outcome of many other complex traits, each under separate polygenic control. Among profitable biomass yield-related traits are the amount of sylleptic branching and the length of winter dormancy. In poplar and in a few other Salicaceae species some lateral buds grow out sylleptically, the same season in which they form without the need of an intervening rest period. Sylleptic branching in poplar increases branch number, leaf area and general growth of the tree in its early years, and is a reasonable predictor of coppice yield. On the other hand, the length of winter dormancy determines the extent of the growth period. Our group has characterized the RAV1 gene of Castanea sativa (CsRAV1), encoding a transcription factor of the subfamily RAV (Related to ABI3/VP1). CsRAV1 expression shows a marked seasonal pattern, being higher in autumn and winter both in stems and buds. We generated transgenic lines of the hybrid clone Populus tremulax P. alba INRA 717 1B4 constitutively expressing CsRAV 1. These CsRAV1-expressing poplars develop sylleptic branches only a few weeks after potting. In addition to the sylleptic branching phenotype, these trees show phenological features that could give rise to an extended growth period. We are currently assessing the phenotype and behavior of these transgenic trees in a field trial, and ultimately, we will evaluate the impact on lignocellulosic biomass quality and production.
