Se ha determinado el effecto inhibidor sobre la actividad Glucogeno sintetasa (E.C.2.41.11) por parte de cuatro antihelminticos: Albendazol, Mebendazol, Parbendazol y Tiabendazol. Observandose que en todos los casos, es el Parbendazol quien ha demostrado un mayor poder inhibidor sobre la glucógeno sintetasa de Ascaris suum, Fasciola hepatica y Moniezia expansa. El Tiabendazol es el anti-helmintico que menor efecto inhibidor ha presentado sobre la enzima en los tres parasitos objeto de nuestro estudio. Con el presente trabajo y otros previstos en la misma linea, se pretende aportar nuevos datos acerca del aun desconocido locus de acción de estos antihelminticos.

Three superoxide dismutase isoenzymes of different cellular location were detected in an homogenate of Thrichuris ovis. Each of these molecular forms was purified by differential centrifugation and precipitation with ammonium sulphate, followed by chromatography on DEAE-cellulose and Sephadex G-75 columns. The activity levels of the two molecular forms detected in the mitochondrial (one cyanide sensitive Cu-Zn-SOD and the other cyanide intensitive Mn-Sod were higher than that of the superoxide dismutase detected in the cytoplasmic fraction (cyanid sensitive Cu-Zn-SOD). All the mollecular forms present evident differences to the SODs contained in the host liver. Molecular mass and some of the physical and chemical aproperties of the enzyme was determined for all three molecular forms. An inhibitory effect on the SOD of the parasite an the host was detected with a series of compounds, some of wich markedly inhibited parasite ensyme but not host enzyme.

A new convenient method for preparation of 2-substituted benzimidazoles and bis-benzimidazoles is presented. In this method, o-phenylenediamines were condensed with bisulfite adducts of various aldehydes and di-aldehydes under neat conditions by microwave heating. The results were also compared with results of synthesis by conventional heating under reflux. Structures of the products were confirmed by infrared, ¹H- and 13C-NMR spectroscopy. Short reaction times, good yields, easy purification of products, and mild reaction conditions are the main advantages of this method.

El treball experimental que ha permès redactar la present Tesi Doctoral ha estat dividit en dues parts. A la primera part es presenten els resultats referents a la síntesi estereocontrolada de noves heteroarilglicines (quinolil i pirazolilglicines) a partir de cetones acetilèniques, substrats quirals que permeten accedir a l'esquelet de diferents heterocicles (quinolines i pirazoles), la posterior obtenció dels corresponents quinolil i pirazolil--aminoalcohols i les diferents metodologies d'oxidació per tal d'accedir a les corresponents quinolil i priazolilglicines objectiu. A la segona part d'aquesta memòria s'ha estudiat, en dissolució, l'habilitat del grup alquilsulfona com a grup sortint eficaç en reaccions d'ipso-substitució nucleofilica. El desenvolupament d'aquesta reacció ha servit de punt de partida per a la creació de llibreries d'heterocicles amb alta diversitat molecular i potencial interès biològic sobre fase sòlida.

Reaction of Cu(1,2-phenylenediamine)(2)(ClO4)(2) with neat RR'=O (R = methyl and/or ethyl) (lives Cu(2,2-dialkyl-2H-benzimidazole)ClO4. demetallation of which by the action of aqueous ammonia yields Pure 2,2-dialkyl-2H-benzimidazoles. These are characterised by NMR. hi the X-ray crystal Structure, Ag(2,2-methyl-2H-benzimi-dazolc)NO3 is Found to be a spiral 1D coordination polymer where the 2H-benzimidazole acts as an N,N bridge between two Ag(I) centus. Although 2H-benzimidazoles are very unstable in the free state, they are quite stable in their Cu(I)(1) and Ag(I) complexes. The 1,2-tautomerisation in imidazole and benzimidazole have been Studied by means of transition state calculations at B3LYP/6-3 11 +G(2d,p)* level.

A straightforward one-step method for the N-methylthiomethylation of benzimidazoles has been developed employing DMSO as a solvent and as a reagent. This methodology has been applied for the synthesis of diverse N-methylthiomethyl derivatives of benzimidazoles. The products can be chemoselectively oxidized to the corresponding sulfoxides with NaBiO3 in acetic acid. Both the N-methylthiomethyl derivatives of benzimidazoles and their corresponding sulfoxides are important medicinal scaffolds.

Over the past 30 years, benzimidazoles have increasingly been used to treat cystic echinococcosis (CE). The efficacy of benzimidazoles, however, remains unclear. We systematically searched MEDLINE, EMBASE, SIGLE, and CCTR to identify studies on benzimidazole treatment outcome. A large heterogeneity of methods in 23 reports precluded a meta-analysis of published results. Specialist centres were contacted to provide individual patient data. We conducted survival analyses for cyst response defined as inactive (CE4 or CE5 by the ultrasound-based World Health Organisation [WHO] classification scheme) or as disappeared. We collected data from 711 treated patients with 1,308 cysts from six centres (five countries). Analysis was restricted to 1,159 liver and peritoneal cysts. Overall, 1-2 y after initiation of benzimidazole treatment 50%-75% of active C1 cysts were classified as inactive/disappeared compared to 30%-55% of CE2 and CE3 cysts. Further in analyzing the rate of inactivation/disappearance with regard to cyst size, 50%-60% of cysts <6 cm responded to treatment after 1-2 y compared to 25%-50% of cysts >6 cm. However, 25% of cysts reverted to active status within 1.5 to 2 y after having initially responded and multiple relapses were observed; after the second and third treatment 60% of cysts relapsed within 2 y. We estimated that 2 y after treatment initiation 40% of cysts are still active or become active again. The overall efficacy of benzimidazoles has been overstated in the past. There is an urgent need for a pragmatic randomised controlled trial that compares standardized benzimidazole therapy on responsive cyst stages with the other treatment modalities.

This report presents evidence that a reduced pyrrolo[1,2-a]benzimidazole (PBI) cleaves DNA as a result of phosphate alkylation followed by hydrolysis of the resulting phosphate triester. The base-pair specificity of the phosphate alkylation results from Hoogsteen-type hydrogen bonding of the reduced PBI in the major groove at only A.T and G.C base pairs. Alkylated phosphates were detected by 31P NMR and the cleavage products were detected by 1H NMR and HPLC. Evidence is also presented that a reduced PBI interacts with DNA in the major groove rather than in the minor groove or by intercalation.

Two series of benzimidazole derivatives were sythesised. The first one was based on 5,6-dinitrobenzimidazole, the second one comprises 2-thioalkyl- and thioaryl-substituted modified benzimidazoles. Antibacterial and antiprotozoal. activity of the newly obtained compounds was studied. Some thioalkyl derivatives showed remarkable activity against nosocomial strains of Stenotrophomonas malthophilia, and an activity comparable to that of metronidazole against Gram-positive and Gram-negative bacteria. Of the tested compounds, 5,6-dichloro-2-(4-nitrobenzylthio)-benzimidazole showed the most distinct antiprotozoal activity.

INTRODUCTION AND OBJECTIVES:Recently, three novel non-vitamin K antagonist oral anticoagulants received approval for reimbursement in Portugal for patients with non-valvular atrial fibrillation (AF). It is therefore important to evaluate the relative cost-effectiveness of these new oral anticoagulants in Portuguese AF patients. METHODS: A Markov model was used to analyze disease progression over a lifetime horizon. Relative efficacy data for stroke (ischemic and hemorrhagic), bleeding (intracranial, other major bleeding and clinically relevant non-major bleeding), myocardial infarction and treatment discontinuation were obtained by pairwise indirect comparisons between apixaban, dabigatran and rivaroxaban using warfarin as a common comparator. Data on resource use were obtained from the database of diagnosis-related groups and an expert panel. Model outputs included life years gained, quality-adjusted life years (QALYs), direct healthcare costs and incremental cost-effectiveness ratios (ICERs). RESULTS:Apixaban provided the most life years gained and QALYs. The ICERs of apixaban compared to warfarin and dabigatran were €5529/QALY and €9163/QALY, respectively. Apixaban was dominant over rivaroxaban (greater health gains and lower costs). The results were robust over a wide range of inputs in sensitivity analyses. Apixaban had a 70% probability of being cost-effective (at a threshold of €20 000/QALY) compared to all the other therapeutic options. CONCLUSIONS:Apixaban is a cost-effective alternative to warfarin and dabigatran and is dominant over rivaroxaban in AF patients from the perspective of the Portuguese national healthcare system. These conclusions are based on indirect comparisons, but despite this limitation, the information is useful for healthcare decision-makers.

[Excerpt] Purine nucleobases are essential biomolecules in living organisms. Playing several key roles in the cell, they have been a significant inspiration for drug design.1 Benzimidazole nucleus is an important pharmacophore in the development of molecules with pharmaceutical or biological interest. Benzimidazoles have been reported to display significant pharmacological activities such as antiulcer, antifungal, antiparkinson, anticancer and antibiotic.2 Fused structures incorporating these two scaffolds might be important for medicinal chemistry and, to the best of our knowledge, there are no reports of these systems in the literature. In particular, benzo[4,5]imidazo[2,1]purines seem to be novel and must be important target molecules in the heterocyclic synthesis. (...)

Estudios prelimares iniciados en el 2007 (Proyecto Pictor Córdoba 877) están indicando que la resistencia a los antihelmínticos por los nematodes que parasitan a los caprinos es un fenómeno común en el centro-norte de Córdoba. En la región, el género parasitario de mayor prevalencia es Haemonchus spp el cual es considerado mundialmente y desde una perspectiva global, como el problema parasitario de mayor importancia económica para los pequeños rumiantes. Poblaciones de este parásito con resistencia a los antiparasitarios de mayor uso (avermectinas y benzimidazoles) están ampliamente difundidos en la región (incluidos aislamientos con resistencia hacia ambas drogas) y de acuerdo a la información preliminar que se está obteniendo, los productores no están advertidos sobre el problema y en muchos casos están utilizando antiparasitarios totalmente inefectivos. El corolario es que se están produciendo pérdidas productivas (potencialmente de magnitud) y la acumulación de genes resistentes en las majadas y superficies de pastoreo lo cual compromete la eficiencia y sustentabilidad de estos sistemas productivos caprinos. Las técnicas diagnósticas actualmente disponibles se basan en características fenotípicas de las poblaciones parasitarias resistentes las que se expresan cuando las fallas terapéuticas y productivas generalmente ya pueden haber ocurrido. Mientras no se encuentren disponibles técnicas diagnósticas para identificar características genotípicas de estas poblaciones que permitan anticiparnos a estas pérdidas, se hace necesario la implementación continua de evaluaciones para detectar los problemas de eficacia de los antihelmínticos tan pronto como sea posible. La menor capacidad económica relativa de los productores caprinos frente a otras actividades pecuarias exige métodos diagnósticos muy simples si es que se pretende la adopción de esta metodología diagnóstica. En este contexto, el proyecto contempla en este punto específico el estudio de técnicas y alternativas para simplificar y economizar los métodos actuales para el diagnóstico de la resistencia antihelmíntica bajo situaciones de campo. La dispersión de la resistencia surge como una consecuencia inevitable de las actuales prácticas de aplicación de antiparasitarios desarrolladas en la región y exige abandonar en forma urgente la lógica simplista y los tratamientos empíricos sin sustento epidemiológico que están comprometiendo seriamente la sustentabilidad del control de los nematodos gastrointestinales. Se hace necesario contar con información regional no solo sobre la epidemiología de estos parásitos si no también sobre sus efectos sobre los diferentes estados fisiológicos de los hospedadores caprinos. En una aproximación al problema, el proyecto propone el estudio sobre el impacto productivo y sanitario que ocasionan los nematodes en el periparto y lactación de las hembras caprinas y determinar los efectos de tratamientos estratégicos durante este período fisiológico específico.

El desarrollo de resistencia de los antihelmínticos por los nematodes que parasitan a los herbívoros domésticos se está incrementando rápidamente en la Argentina. En el área centro- norte de Argentina se han registrado casos de resistencia a los antihelmínticos por nematodes que parasitan ovinos, caprinos, bovinos y equinos. Existen actualmente tres grupos químicos para el control de los nematodes de los herbívoros domésticos: levamisoles, benzimidazoles y lactonas macrocíclicas. En los ovinos la resistencia ya se ha expresado a los tres grupos químicos y en bovinos a dos (benziamiazoles y lactonas macrocrílicas) existiendo también aislamiento de nematodes con resistencia para ambos grupos. Con respecto a los caprinos, la información disponible en nuestro país es notoriamente escasa habiéndose informado sobre casos puntuales de resistencias a benzimidazoles y a las lactonas macrocrílicas. No obstante los ovinos y caprinos son parasitados por los mismos nematodes y por lo tanto los parásitos que desarrollan resistencia en las ovejas pueden transferirse a las cabras y viceversa. En este contexto es probable que el problema de la resistencia se encuentre también difundido en la especie caprina. Recientemente se ha informado también de los primeros casos de nematodes equinos con resistencia a los benzimidazoles en nuestro país. Actualmente, considerando las especies de herbívoros domésticos, es en los ovinos (por su prevalencia y grupos químicos involucrados) donde el problema tiene mayor magnitud. La prevalencia de este fenómeno en los bovinos parece ir asemejándose a la observada en los ovinos hace una década atrás. Debido a la importancia de la producción bovina y a la fuerte dependencia de estos químicos para el control de nematodes, se requiere de la implementación de diagnósticos continuos para identificar poblaciones resistentes en los diferentes sistemas ganaderos. Los objetivos del trabajo son: a. Evaluar el grado de susceptibilidad / resistencia en rodeos bovinos del área central de Argentina y determinar las posibles implicancias de la resistencias a los antihelmínticos sobre la productividad de los mismos; b. Transferir la información obtenida a productores, profesionales y estudiantes e informar a los organismos oficiales o privados de la salud animal intentando limitar la dispersión de la resistencia de los nematodes a los antihelmínticos.

The scaffold protein Islet-Brain1/c-Jun amino-terminal kinase Interacting Protein-1 (IB1/JIP-1) is a modulator of the c-Jun N-terminal kinase (JNK) activity, which has been implicated in pleiotrophic cellular functions including cell differentiation, division, and death. In this study, we described the presence of IB1/JIP-1 in epithelium of the rat prostate as well as in the human prostatic LNCaP cells. We investigated the functional role of IB1/JIP-1 in LNCaP cells exposed to the proapoptotic agent N-(4-hydroxyphenyl)retinamide (4-HPR) which induced a reduction of IB1/JIP-1 content and a concomittant increase in JNK activity. Conversely, IB1/JIP-1 overexpression using a viral gene transfer prevented the JNK activation and the 4-HPR-induced apoptosis was blunted. In prostatic adenocarcinoma cells, the neuroendocrine (NE) phenotype acquisition is associated with tumor progression and androgen independence. During NE transdifferentiation of LNCaP cells, IB1/JIP-1 levels were increased. This regulated expression of IB1/JIP-1 is secondary to a loss of the neuronal transcriptional repressor neuron restrictive silencing factor (NRSF/REST) function which is known to repress IB1/JIP-1. Together, these results indicated that IB1/JIP-1 participates to the neuronal phenotype of the human LNCaP cells and is a regulator of JNK signaling pathway.

We assessed the blockade of the renin-angiotensin system (RAS) achieved with 2 angiotensin (Ang) antagonists given either alone at different doses or with an ACE inhibitor. First, 20 normotensive subjects were randomly assigned to 100 mg OD losartan (LOS) or 80 mg OD telmisartan (TEL) for 1 week; during another week, the same doses of LOS and TEL were combined with 20 mg OD lisinopril. Then, 10 subjects were randomly assigned to 200 mg OD LOS and 160 mg OD TEL for 1 week and 100 mg BID LOS and 80 mg BID TEL during the second week. Blockade of the RAS was evaluated with the inhibition of the pressor effect of exogenous Ang I, an ex vivo receptor assay, and the changes in plasma Ang II. Trough blood pressure response to Ang I was blocked by 35+/-16% (mean+/-SD) with 100 mg OD LOS and by 36+/-13% with 80 mg OD TEL. When combined with lisinopril, blockade was 76+/-7% with LOS and 79+/-9% with TEL. With 200 mg OD LOS, trough blockade was 54+/-14%, but with 100 mg BID it increased to 77+/-8% (P<0.01). Telmisartan (160 mg OD and 80 mg BID) produced a comparable effect. Thus, at their maximal recommended doses, neither LOS nor TEL blocks the RAS for 24 hours; hence, the addition of an ACE inhibitor provides an additional blockade. A 24-hour blockade can be achieved with an angiotensin antagonist alone, provided higher doses or a BID regimen is used.
