This paper describes a new method for the preparation of 1,5-bis(4-hydroxy-3-methoxyphenyl)-1,4-pentadien-3-one 1 and its derivatives 2-5. This set of synthetic compounds exhibited high antitumoral activities regarding in vitro screening against several human tumor cell lines as lung carcinoma NCI-460, melanoma UACC-62, breast MCF-7, colon HT-29, renal 786-O, ovarian OVCAR-03 and ovarian expressing the resistance phenotype for adriamycin NCI-ADR/ RES, prostate PC-3, and leukemia K-562. Compounds were also tested against murine tumor cell line B16F10 melanoma and lymphocytic leukemia L1210 as well as to their effect toward normal macrophages. Specific activity against colon cancer cells HT-29 was observed for all tested compounds and suggests further studies with models of colon cancer. Compounds 1, 2, and 4 showed significant cytotoxic activity with IC(50) values <= 2.3 mu M for all human cancer cell lines. Intraperitoneal acute administration of compound 1 and 2 showed very low toxicity rate. (C) 2010 Elsevier Ltd. All rights reserved.

Inhibition of microtubule function is an attractive rational approach to anticancer therapy. Although taxanes are the most prominent among the microtubule-stabilizers, their clinical toxicity, poor pharmacokinetic properties, and resistance have stimulated the search for new antitumor agents having the same mechanism of action. Discodermolide is an example of nontaxane natural product that has the same mechanism of action, demonstrating superior antitumor efficacy and therapeutic index. The extraordinary chemical and biological properties have qualified discodermolide as a lead structure for the design of novel anticancer agents with optimized therapeutic properties. In the present work, we have employed a specialized fragment-based method to develop robust quantitative structure - activity relationship models for a series of synthetic discodermolide analogs. The generated molecular recognition patterns were combined with three-dimensional molecular modeling studies as a fundamental step on the path to understanding the molecular basis of drug-receptor interactions within this important series of potent antitumoral agents.

The nitrocompounds mode of action involves the biotransformation of the nitro group, releasing intermediates in the redox process. Some of those intermediates attack enzymes, membranes and DNA, providing the basis for their biological activity and adverse effects. In this report, some aspects regarding the biological activity, mechanism of action and toxicity of nitrocompounds are explored, purposing the research of new bioactive derivatives having low toxicity.

Phytochemical investigation of hexanic extract of leaves of Rollinia laurifolia afforded three known acetogenins, uvariamicin-I (1), solamin(2) and gonionenin(3), for the first time isolated from Rollinia genus. The structures of 1, 2 and 3 were elucidated by spectroscopic methods.

Muchos esfuerzos se están realizando en el diseño de nuevos métodos para la eliminación de las células tumorales y así inhibir el crecimiento neoplásico. Entre los métodos no convencionales se encuentran la Terapia Fotodinámica.La Terapia Fotodinámica (TFD) es un tratamiento experimental de algunos tipos de cáncer, basado en el efecto citotóxico inducido en el tejido tumoral, por la acción combinada de una droga (fotosensibilizador) y la luz visible. El fotosensibilizador posee la propiedad de absorber la luz y reaccionar con el oxígeno molecular, produciendo una forma activa del oxígeno: el oxígeno singlete (1O2) que oxida diversas moléculas biológicas, induciendo un efecto citotóxico que se traduce en la regresión tumoral. Los nuevos avances en la dosimetría de la luz, así como la búsqueda de una segunda generación de nuevos fotosensibilizadores más eficaces que los actualmente utilizados, han permitido incluir protocolos de Terapia Fotodinámica en numerosos centros hospitalarios principalmente para el tratamiento de cánceres de pulmón, vejiga, esófago y piel. Plantas fototóxicas, sus metabolitos fotosensibilizantes y sus posibles usos; En general, dentro de las especies vegetales tóxicas existen aquellas denominadas plantas alergénicas, que son las que pueden producir sus efectos indeseables por vía dérmica. También existen aquellas que pueden producir efectos tóxicos por vía sistémica. Sin embargo, coexiste en la naturaleza otro grupo de plantas tóxicas que desencadenan sus efectos nocivos bajo la acción de la luz, por lo que son llamadas plantas fototóxicas, cuyos principios activos son comúnmente denominados agentes fotosensibilizantes La apoptosis como blanco terapéutico contra el cáncer: Los conocimientos moleculares sobre la apoptosis adquiridos en los últimos años están siendo aplicados al desarrollo de nuevos fármacos que puedan modular selectivamente las señales involucradas en la muerte de las células. Una de las razones que justifica el interés en el estudio de este tipo de moléculas, es que una de las características más tempranas en la transformación de la células neoplásicas esta relacionada con la incapacidad de responder a los estímulos de muerte. Esto lleva a una desregulación del proceso de apoptosis desencadenando una proliferación descontrolada. Los otros eventos que desencadenan el cáncer son, la invasión vascular y la metástasis a distanciaLa adquisición de resistencia a los efectos citotóxicos de los tratamientos anticancerígenos ha emergido como un significante impedimento para el efectivo tratamiento de la enfermedad. Por ello, en el presente proyecto se investigará si la adquisición de resistencia a TFD inducida en la línea celular estudiada es conferida por el aumento de la proteína MDRP1 a través de la vía de señalización PI3K/Akt. Además, se estudiará la correlación entre la posible resistencia a drogas y la inducción de apoptosis, analizando los mecanismos involucrados. Los resultados obtenidos contribuirán a dilucidar y entender los mecanismos moleculares implicados en la resistencia y sensibilidad tumoral a la TFD, y de esta manera mejorar la eficacia de dicha terapia antitumoral para sensibilizar a las células a la apoptosis. OBJETIVOS Estudiar el efecto de agentes fotosensibilizadores de origen sintético (ftalocianinas), comercialmente ya aprobadas por la FDA (Me-ALA), de origen natural (antraquinonas), y obtenidas en procesos nanotecnologicos (nanofibras) respecto a su capacidad de inducir la muerte celular en sistemas experimentales in vivo, para el desarrollo de nuevas drogas de aplicación en Terapia Fotodinámica (PDT). Estudiar las señales de apoptosis que se desencadenan, combinando la PDT con iRNA (antisurvivina) con la finalidad de aumentar la eficiencia de la muerte tumoral. Estudiar los mecanismos de resistencia a la Terapia Fotodinámica en carcinoma de células escamosas con fotosensibilizadores permitidos (Me-ALA).

Two new complexes of platinum(II) and silver(I) with acesulfame were synthesized. Acesulfame is in the anionic form acesulfamate (ace). The structures of both complexes were determined by X-ray crystallography. For K(2)[PtCl(2)(ace)(2)] the platinum atom is coordinated to two Cl(-) and two N-acesulfamate atoms forming a trans-square planar geometry. Each K(+) ion interacts with two oxygen atoms of the S(=O)(2) group of each acesulfamate. For the polymeric complex [Ag(ace)](n) the water molecule bridges between two crystallographic equivalent Agl atoms which are related each other by a twofold symmetry axis. Two Agl atoms, related to each other by a symmetry centre, make bond contact with two equivalent oxygen atoms. These bonds give rise to infinite chains along the unit cell diagonal in the ac plane. The in vitro cytotoxic analyses for the platinum complex using HeLa (human cervix cancer) cells show its low activity when compared to the vehicle-treated cells. The Ag(I) complex submitted to in vitro antimycobacterial tests, using the Microplate Alamar Blue (MABA) method, showed a good activity against Mycobacterium tuberculosis, responsible for tuberculosis, with a minimal inhibitory concentration (MIC) value of 11.6 mu M. The Ag(I) complex also presented a promising activity against Gram negative (Escherichia colt and Pseudomonas aeruginosa) and Gram positive (Enterococcus faecalis) microorganisms. The complex K(2)[PtCl(2)(ace)(2)] was also evaluated for antiviral properties against dengue virus type 2 (New Guinea C strain) in Vero cells and showed a good inhibition of dengue virus type 2 (New Guinea G strain) replication at 200 mu M, when compared to vehicle-treated cells. This paper reports on the synthesis and characterization of two new ternary copper(II) complexes: [Cu(doxy-cycline)(1,10-phenanthroline)(H(2)O)(ClO(4))](ClO(4)) (1) and [Cu(tetracycline)(1,10-phenanthroline)(H(2)O)(ClO(4))](ClO(4)) (2). These compounds exhibit a distorted tetragonal geometry around copper, which is coordinated to two bidentate ligands, 1,10-phenanthroline and tetracycline or doxycyline, a water molecule, and a perchlorate ion weakly bonded in the axial positions. In both compounds, copper(II) binds to tetracyclines`. via the oxygen of the hydroxyl group and oxygen of the amide group at ring A and to 1,10-phenanthroline via its two heterocyclic nitrogens. We have evaluated the binding of the new complexes to DNA, their capacity to cleave it, their cytotoxic activity, and uptake in tumoral cells. The complexes bind to DNA preferentially by the major groove, and then cleave its strands by an oxidative mechanism involving the generation of ROS. The cleavage of DNA was inhibited by radical inhibitors and/or trappers such as superoxide dismutase, DMSO, and the copper(I) chelator bathocuproine. The enzyme T4 DNA ligase was not able to relegate the products of DNA cleavage, which indicates that the cleavage does not occur via a hydrolytic mechanism. Both complexes present an expressive plasmid DNA cleavage activity generating single- and double-strand breaks, under mild reaction conditions, and even in the absence of any additional oxidant or reducing agent. In the same experimental conditions, [Cu(phen)(2)](2+) is approximately 100-fold less active than our complexes. These complexes are among the most potent DNA cleavage agents reported so far. Both complexes inhibit the growth of K562 cells With the IC(50) values of 1.93 and 2.59 mu mol L(-1) for compounds I and 2, respectively. The complexes are more active than the free ligands, and their cytotoxic activity correlates with intracellular copper concentration and the number of Cu-DNA adducts formed inside cells.
