Thlaspi caerulescens é espécie hiperacumuladora de metais como Cd2+, Ni2+ e Zn2+, considerada como uma plantamodelo para estudar a acumulação e tolerância a metais pesados. No entanto, a baixa produção de sementes em nossas condições climáticas tornam necessária a determinação de condições que possam maximizar a germinação e o vigor de suas sementes. Para identificar as melhores condições para a germinação, sementes de T. caerulescens foram colocadas na presença (15 mmol.m-2.s-1, 8 h luz/16 h escuro) ou ausência de luz nas temperaturas de 10, 15, 20 e 25 ºC, em papel germitest previamente umedecido com solução de ácido giberélico (GA3) a 0,05% ou água destilada. Foram avaliados a porcentagem de germinação (%G) e o índice de velocidade de germinação (IVG). Maior porcentagem de germinação (66%) foi observada nos tratamentos com GA3 e temperaturas de 15 e 20 ºC, na presença de luz. Maiores valores do IVG foram obtidos com a utilização de GA3 nas temperaturas de 15 e 20 ºC, tanto na presença quanto na ausência de luz. Maiores germinação e IVG de T. caerulescens foram observados com uso de GA3 na presença de luz nas temperaturas de 15 e 20 ºC.

The effects of short- and long-term exposure of cells to elevated cyclic adenosine monophosphate (c-AMP), using dibutyryl-c-AMP, 8-bromo-c-AMP, cholera toxin or forskolin, or cyclic guanosine monophosphate (c-GMP), using dibutyryl-c-GMP or 8-bromo-c-GMP, on the activity and expression of the noradrenaline transporter (NAT) were examined. Short- or long-term c-GMP elevation had no effects on H-3-noradrenaline uptake by rat PC12 phaeochromocytoma cells or human SK-N-SH-SY5Y neuroblastoma cells. Short-term c-AMP elevation (for 17 min experiment duration) caused a decrease in H-3-noradrenaline uptake by PC12 cells, but had no effects on SK-N-SH-SY5Y cells or COS-7 cells transfected with human or rat NAT cDNA. c-AMP did not affect H-3-nisoxetine binding to PC12 cells. Long-term (24 h) exposure to elevated c-AMP levels caused a decrease in H-3-noradrenaline uptake and NAT mRNA in PC12 cells, but had no effects on SK-N-SH-SY5Y cells and caused a small increase in H-3-noradrenaline uptake in COS-7 cells heterologously expressing rat or human NAT. Hence, c-AMP, but not c-GMP, causes a cell type-dependent reduction in NAT activity after short-term exposure and a reduction in NAT expression after long-term exposure. Estudo taxonômico do gênero Micrasterias C. Agardh ex Ralfs realizado em duas Áreas de Proteção Ambiental (APA Rio Capivara e APA Lagoas de Guarajuba), ambas situadas no Município de Camaçari, Estado da Bahia, nordeste do Brasil. Os materiais estudados, de origem do plâncton e do perifíton, provieram de 96 amostras coletadas no verão (dezembro de 2006 a março de 2007) e no inverno (maio a agosto de 2007), em ambientes lóticos e lênticos. Foram identificados 14 táxons, dos quais M.americana (Ehrenb.) Ralfs var. bahiensis I. B. Oliveira, C. E. M. Bicudo & C. W. N. Moura foi descrita como nova para a ciência e 10 constituem citacões pioneiras para a desmidioflórula da Bahia, quais sejam: M. alata G. C. Wall., M. arcuata Bailey var.subpinnatifida West & G. S. West f. subpinnatifida, M. borgei H. Krieg. var. borgei, M. foliacea Bailey ex Ralfs var. foliacea, M. furcata C. Agardh ex Ralfs var. furcata f. furcata, M. laticeps Nordst. var. laticeps, M. laticeps Nordst. var.acuminata H. Krieg., M. mahabuleshwarensis J. Hobson var.ampullacea (W. M. Maskell) Nordst., M. mahabuleshwarensis J. Hobson var. mahabuleshwarensis f. mahabuleshwarensis, M. pinnatifida (Kütz.) Ralfs var. pinnatifida f. pinnatifida, M. radiosa Ralfs var. elegantior (G. S. West) Croasdale, M. rotata (Grev.) Ralfs var. rotata e M. truncata (Corda) Bréb. ex Ralfs var. pusilla G. S. West. As espécies mais bem representadas na área estudada foram M. alata e M. pinnatifida presentes, respectivamente, em 50% e 45,83% das amostras analisadas. Micrasteriaslaticeps var.acuminata e M.americana var. bahiensis, foram as menos representadas, por terem sido registradas apenas em 2,08% e 1,04% respectivamente.

Understanding the drivers of population divergence, speciation and species persistence is of great interest to molecular ecology, especially for species-rich radiations inhabiting the world's biodiversity hotspots. The toolbox of population genomics holds great promise for addressing these key issues, especially if genomic data are analysed within a spatially and ecologically explicit context. We have studied the earliest stages of the divergence continuum in the Restionaceae, a species-rich and ecologically important plant family of the Cape Floristic Region (CFR) of South Africa, using the widespread CFR endemic Restio capensis (L.) H.P. Linder & C.R. Hardy as an example. We studied diverging populations of this morphotaxon for plastid DNA sequences and >14 400 nuclear DNA polymorphisms from Restriction site Associated DNA (RAD) sequencing and analysed the results jointly with spatial, climatic and phytogeographic data, using a Bayesian generalized linear mixed modelling (GLMM) approach. The results indicate that population divergence across the extreme environmental mosaic of the CFR is mostly driven by isolation by environment (IBE) rather than isolation by distance (IBD) for both neutral and non-neutral markers, consistent with genome hitchhiking or coupling effects during early stages of divergence. Mixed modelling of plastid DNA and single divergent outlier loci from a Bayesian genome scan confirmed the predominant role of climate and pointed to additional drivers of divergence, such as drift and ecological agents of selection captured by phytogeographic zones. Our study demonstrates the usefulness of population genomics for disentangling the effects of IBD and IBE along the divergence continuum often found in species radiations across heterogeneous ecological landscapes.

SUMMARY Heavy metal presence in the environment is a serious concern since some of them can be toxic to plants, animals and humans once accumulated along the food chain. Cadmium (Cd) is one of the most toxic heavy metal. It is naturally present in soils at various levels and its concentration can be increased by human activities. Several plants however have naturally developed strategies allowing them to grow on heavy metal enriched soils. One of them consists in the accumulation and sequestration of heavy metals in the above-ground biomass. Some plants present in addition an extreme strategy by which they accumulate a limited number of heavy metals in their shoots in amounts 100 times superior to those expected for a non-accumulating plant in the same conditions. Understanding the genetic basis of the hyperaccumulation trait - particularly for Cd - remains an important challenge which may lead to biotechnological applications in the soil phytoremediation. In this thesis, Thlaspi caerulescens J. & C. Presl (Brassicaceae) was used as a model plant to study the Cd hyperaccumulation trait, owing to its physiological and genetic characteristics. Twenty-four wild populations were sampled in different regions of Switzerland. They were characterized for environmental and soil parameters as well as intrinsic characteristics of plants (i.e. metal concentrations in shoots). They were as well genetically characterized by AFLPs, plastid DNA polymorphism and genes markers (CAPS and microsatellites) mainly developed in this thesis. Some of the investigated genes were putatively linked to the Cd hyperaccumulation trait. Since the study of the Cd hyperaccumulation in the field is important as it allows the identification of patterns of selection, the present work offered a methodology to define the Cd hyperaccumulation capacity of populations from different habitats permitting thus their comparison in the field. We showed that Cd, Zn, Fe and Cu accumulations were linked and that populations with higher Cd hyperaccumulation capacity had higher shoot and reproductive fitness. Using our genetic data, statistical methods (Beaumont & Nichols's procedure, partial Mantel tests) were applied to identify genomic signatures of natural selection related to the Cd hyperaccumulation capacity. A significant genetic difference between populations related to their Cd hyperaccumulation capacity was revealed based on somè specific markers (AFLP and candidate genes). Polymorphism at the gene encoding IRTl (Iron-transporter also participating to the transport of Zn) was suggested as explaining part of the variation in Cd hyperaccumulation capacity of populations supporting previous physiological investigations. RÉSUMÉ La présence de métaux lourds dans l'environnement est un phénomène préoccupant. En effet, certains métaux lourds - comme le cadmium (Cd) -sont toxiques pour les plantes, les animaux et enfin, accumulés le long de la chaîne alimentaire, pour les hommes. Le Cd est naturellement présent dans le sol et sa concentration peut être accrue par différentes activités humaines. Certaines plantes ont cependant développé des stratégies leur permettant de pousser sur des sols contaminés en métaux lourds. Parmi elles, certaines accumulent et séquestrent les métaux lourds dans leurs parties aériennes. D`autres présentent une stratégie encore plus extrême. Elles accumulent un nombre limité de métaux lourds en quantités 100 fois supérieures à celles attendues pour des espèces non-accumulatrices sous de mêmes conditions. La compréhension des bases génétiques de l'hyperaccumulation -particulièrement celle du Cd - représente un défi important avec des applications concrètes en biotechnologies, tout particulièrement dans le but appliqué de la phytoremediation des sols contaminés. Dans cette thèse, Thlaspi caerulescens J. & C. Presl (Brassicaceae) a été utilisé comme modèle pour l'étude de l'hyperaccumulation du Cd de par ses caractéristiques physiologiques et génétiques. Vingt-quatre populations naturelles ont été échantillonnées en Suisse et pour chacune d'elles les paramètres environnementaux, pédologique et les caractéristiques intrinsèques aux plantes (concentrations en métaux lourds) ont été déterminés. Les populations ont été caractérisées génétiquement par des AFLP, des marqueurs chloroplastiques et des marqueurs de gènes spécifiques, particulièrement ceux potentiellement liés à l'hyperaccumulation du Cd (CAPS et microsatellites). La plupart ont été développés au cours de cette thèse. L'étude de l'hyperaccumulation du Cd en conditions naturelles est importante car elle permet d'identifier la marque, éventuelle de sélection naturelle. Ce travail offre ainsi une méthodologie pour définir et comparer la capacité des populations à hyperaccumuler le Cd dans différents habitats. Nous avons montré que les accumulations du Cd, Zn, Fe et Cu sont liées et que les populations ayant une grande capacité d'hyperaccumuler le Cd ont également une meilleure fitness végétative et reproductive. Des méthodes statistiques (l'approche de Beaumont & Nichols, tests de Martel partiels) ont été utilisées sur les données génétiques pour identifier la signature génomique de la sélection naturelle liée à la capacité d'hyperaccumuler le Cd. Une différenciation génétique des populations liée à leur capacité d'hyperaccumuler le Cd a été mise en évidence sur certains marqueurs spécifiques. En accord avec les études physiologiques connues, le polymorphisme au gène codant IRT1 (un transporteur de Fe impliqué dans le transport du Zn) pourrait expliquer une partie de la variance de la capacité des populations à hyperaccumuler le Cd.

Background: Providing analgesia without suppressing motor or sensory function is a challenge for regional anesthesia and postoperative pain management. Resiniferatoxin (RTX), an ultrapotent agonist for transient receptor potential subtype-1 (TRPV1) can produce this selective blockade, as TRPV1 is selectively expressed on nociceptors. Futhermore, after peripheral nerve injury, spontaneous ectopic activity arises from all types of nerve fibers that can affect spinal neurons and glial cells. The goal of the present experiment is to determine whether spontaneous activity generated in C-fibers or in both A&amp;C-fibers is required for microglia activation. Method: We applied RTX (0.01%) or bupivacaine microspheres to the sciatic nerve of rats to block the conduction of C-fibers or A&amp;C-fibers, respectively, before spared nerve injury (SNI). Behavior was tested and all the rats were sacrificed 2 days later; immunohistochemistry was performed on their spinal cord for mitogen-activated protein kinase (MAPK) p38, bromodeoxyuridine (BrdU, marker of proliferation) and Iba1 (microglial marker). Result: At day 2 after SNI robust mechanical allodynia and p38 activation in spinal microglia were documented. There was also a substantial cell proliferation in the spinal cord, all proliferating cells (BrdU+) being microglia (Iba1+). RTX blocked heat sensitivity and produced heat hypoalgesia without affecting mechanical allodynia and motor function. Microglial proliferation and p38 activation in the spinal cord were not affected by RTX (p >0.05). In contrast, a complete sensory and motor blockade was seen with bupivacaine which also significantly inhibited p38 activation and microglial proliferation in the spinal cord (p <0.05). Conclusion: We conclude that (1) RTX can provide a selective nociceptive blockade but that (2) blocking only nociceptive fibers does not impair the development of mechanical allodynia and microglia activation. Therefore (3) if microglia activation is important for chronic pain development then specific nociceptive blockade won't be sufficient to prevent it.

Bipolaris maydis (Y. Nisik. &amp; C. Miyake) Shoemaker foi consistentemente isolado de plantas de Pennisetum purpureum Schum., com sintomas de manchas foliares, no Cerrado, em 2005 e 2006. Testes de patogenicidade em mudas sadias de capim-elefante, em casa de vegetação, e o subseqüente reisolamento do fungo confirmaram que B. maydis era o agente causal das lesões foliares observadas. Os primeiros sintomas apareceram dois dias após a inoculação. Onze outras espécies de gramíneas foram suscetíveis ao fungo.

Introduction: In 2012, a study by K. Chatziioannidou and S-C. Renteria showed that teenagers chose to undergo a surgical termination of pregnancy (TOP) more often than a medical TOP (mifepristone followed by misoprostol) when they decided to terminate a pregnancy. It also showed that the teenagers' choice for a medical versus surgical method is inversely proportional to the adults' choice although the efficiency of the medical method showed even better results for teenagers than for adults. According to the hypothesis made, the reasons for this choice might be influenced by the following facts: (i) the belated call to make an appointment, the medical procedure not being available after 9 weeks of gestation; (ii) the imperative request for confidentiality; (iii) the beliefs and subjective appreciation of the medical staff. Objectives: The aim of this retrospective and qualitative study is to analyse the reasons why, in case of a TOP, teenagers chose the surgical method more often than their adult counterparts. Material: (i) All teenagers who were admitted for an abortive procedure during 2011 in the in- or outpatient ward. (ii) The professional team (midwives and sexual and reproductive counsellors) in charge in the case of a TOP request. Methods: The information about the patient's history and the biopsycho- social data was retrieved from the patient files filled out by midwives and sexual and reproductive health counsellors during the first appointment for a TOP request or during its process. The professionals' appreciation was evaluated by means of a semi-structured questionnaire. Results: Concerning the choice of the method for a pregnancy termination, the results of our research show that: (i) Out of 47 teenagers, 27 chose the surgical method and 17 the medical method. (ii) Three had a second trimester abortion (which includes use of the medical method). (iii) Fifteen teenagers out of the 27 who chose a surgical method consulted between the 9th and 14th weeks of amenorrhoea and therefore did not have any other choice. The reasons for their 'late arrival' will be explained in detail. The 12 teenagers who arrived before the 8th week of amenorrhoea and chose to undertake abortion by suction &amp; curettage under general anaesthesia did it for the following reasons: (i) Four were afraid of bleeding and pain. (ii) Five thought that the organisation of the surgical procedure was easier. (iii) Two did not trust the abortion pill. (iv) One was taken to her mother's gynaecologist where she had a D&amp;C. Confidentiality was requested nine times out of 27 when choosing the surgical method, and six times out of 17 when choosing the medical method. Therefore, although confidentiality concerns a third of the teenagers' pregnancy termination requests, it does not seem to be a significant element for the choice of the method. As for the subjective appreciation of the professionals, the first results of the discussions seem to show that teenagers were reluctant or resistant towards the medical method. Conclusion: This study shows that the reasons why teenagers still prefer the use of the surgical over the medical method compared to adults, seem to include the late request for an appointment, fear of pain and bleeding and organisational issues. Confidentiality does not seem to greatly influence the teenagers' choice. Nonetheless, medical professionals seem to favour the suction curettage procedure performed under anesthesia because they associate young age with vulnerability and psychological frailty and consequently diminished ability to cope with pain and emotional distress during the medical procedures.

Um experimento foi conduzido no NuPAM/FCA/UNESP, Botucatu-SP, objetivando avaliar a dinâmica de retenção de água e o caminhamento de um traçante (simulando um herbicida) em diferentes coberturas mortas. Os tratamentos foram constituídos pelo monitoramento do traçante FD&C-1 pulverizado sobre coberturas mortas de cevada, trigo, aveia-preta colhida, aveia-preta rolada, azevém, milheto e capim-braquiária, nas quantidades de 3.000, 6.000 e 9.000 kg ha-1, antes e após simulação de chuvas. As repetições constituíram-se de oito conjuntos de PVC + funil + béquer com palha, onde, através da chuva lixiviada pelas palhadas e do peso dos suportes de PVC, foram estimadas a retenção e transposição da água, assim como quantificado o traçante extraído, através de procedimentos espectrofotométricos. Os diferentes tipos de resíduos culturais mostraram-se similares quanto à retenção da água da chuva, ocorrendo uniformização entre os primeiros 7,5 e 15 mm de precipitação. A formação de "pontos secos" associados a "canais preferenciais de escorrimento" induziu menor capacidade de embebição e retenção da água das chuvas pelas palhadas. As máximas capacidades médias de retenção da chuva pelas coberturas foram de 1,22, 1,99 e 2,59 mm para 3.000, 6.000 e 9.000 kg de matéria seca ha-1, respectivamente. As precipitações iniciais entre 10 e 20 mm foram fundamentais para o molhamento uniforme das palhadas e carregamento do traçante até o solo, independentemente do tipo e da quantidade de palha. Esse comportamento indica ser viável a utilização de programas similares de controle de plantas daninhas para diferentes tipos e quantidades de palha em sistemas de plantio direto.

O grupo está representado no estado por seis espécies e uma variedade: T. angustifolia (Willd.) Proctor, T. chrysodioides (Fée) C.V. Mortonvar. goyazensis (Maxon &amp; C.V. Morton) C.V. Morton, T. longifolia (Desv.) R.M. Tryon, T. macrophylla (Kunze) C.V. Morton, T. maxoniana A.R. Sm., T. salzmannii (Fée) C.V. Morton e T. serrata (Cav.) Alston. São apresentadas descrições do subgênero e das espécies, chave de identificação para as espécies, bem como ilustrações, distribuição geográfica e comentários das espécies.

Las infecciones de vías urinarias (IVU) son frecuentes en la comunidad, existe un incremento progresivo en los mecanismos de resistencia de las enterobacterias, conocer los factores de riesgo representa un interés en la investigación actual. Objetivo: identificar los factores de riesgo para IVU adquirida en la comunidad por enterobacterias productoras de β-lactamasas de espectro extendido (BLEE) o AmpC positivas. Diseño: estudio observacional analítico tipo casos y controles, 50 casos y 100 controles Medición: análisis de las variables de interés para la determinación de las asociaciones en los respectivos Odd ratios (OR). Resultados: se obtuvo un total de 25 casos y 50 controles, en el análisis univariado el uso previo de antibióticos (OR, 6.68; CI 95%, 2-22.32; P 0.001) y los procedimientos previos de la vía urinaria (OR, 3.45; CI 95%, 1.102-10.83; P 0.028) son factores de riesgo para IVU por BLEE o AmpC, en el análisis multivariado el uso previo de antibiótico represento el principal factor de riesgo (OR, 7.36; CI 95%, 1.76-30.77; P 0.006), las quinolonas fueron el antibiótico de uso previo más frecuente. Conclusiones: El uso previo de antibióticos es el principal factor de riesgo para adquirir IVU por enterobacterias productoras de BLEE o AmpC, es necesario ampliar el tamaño de la muestra actual para determinar el impacto que puedan tener las otras variables a estudio.
