TdT-mediated dUTP-biotin nick end labeling (TUNEL) is a sensitive and valid method for detecting DNA cleavage in programmed cell death (PCD). Using this method, DNA cleavage was observed in Laminaria japonica sporophytic tissues, which were infected with alginic acid decomposing bacterium. It was found that DNA cleavage occurred 5 min after the infection, the fragments with 3'-OH groups of cleaved nuclear DNA increased with time of infection and spread from the infection site. Although no typical DNA ladder (200 bp/ 180 bp) was detected by routine agarose gel electrophoresis, the cleavage of nuclear DNA fragments of 97 similar to 48.5 kb could be detected by pulsed field gel electrophoresis (PFGE). By using CaspGLOW(TM) fluorescein active caspase-3 staining method, caspase-3 activity has been detected in response to the infection of alginic acid decomposing bacterium. Our results are similar to the observations in hypersensitive response (HR) of higher plant, suggesting that the rapid cell death of L. japonica infected by alginic acid decomposing bacterium might be involved in PCD, and indicating that the occurrence of PCD is an active defense process against the pathogen's infection.

We collected the diseased blades of Laminaria japonica from Yantai Sea Farm from October to December 2002, and the alginic acid decomposing bacterium on the diseased blade was isolated and purified, and was identified as Alterornonas espejiana. This bacterium was applied as the causative pathogen to infect the blades of L. japonica under laboratory conditions. The aim of the present study was to identify the effects of the bacterium on the growth of L. japonica, and to find the possibly effective mechanism. Results showed that: (1) The blades of L. japonica exhibited symptoms of lesion, bleaching and deterioration when infected by the bacterium, and their growth and photosynthesis were dramatically suppressed. At the same time, the reactive oxygen species (ROS) generation enhanced obviously, and the relative membrane permeability increased significantly. The contents of malonaldehyde (MDA) and free fatty acid in the microsomol membrane greatly elevated, but the phospholipid content decreased. Result suggested an obvious peroxidation and deesterrification in the blades of L. japonica when infected by the bacterium. (2) The simultaneous assay on the antioxidant enzyme activities demonstrated that superoxide dismutase (SOD) and catalase (CAT) increased greatly when infected by the bacterium, but glutathione peroxidase (Gpx) and ascorbate peroxidase (APX) did not exhibit active responses to the bacterium throughout the experiment. (3) The histomorphological observations gave a distinctive evidence of the severity of the lesions as well as the relative abundance in the bacterial population on the blades after infection. The bacterium firstly invaded into the endodermis of L. japonica and gathered around there, and then resulted in the membrane damage, cells corruption and ultimately, the death of L. japonica.

A detailed account of the morphological, staining, penicillin sensitivity and serological peculiarities of five strains of an oxalate-decomposing bacterium including the well-recognized strain Vibrio extorquens, has been given. Inasmuch as all the strains share many of the characteristics of the genus Arthrobacter and not Vibrio the desirability of placing the bacterium in the former genus for the time being has been suggested. The possibility of the strains falling under an altogether new genus which represent a phylogenic link between the pseudomonads and diphtheroids has been speculated.

Sargassum cervicone Greville is one of the species of Sargassum occuring in abundance along the South Western Coast of Ceylon. Durairatnam 1966 carried out a survey from Ambalangoda to Hambantota, and estimated that approximately 130 tons of dried Sargassum can be harvested manually. The bulk of the Sargassum found in this area was Sargassum cervicone Greville.

From the hexane-soluble fraction of an ethanol extract from leaves and stems of Stemodia foliosa (Scrophulariaceae), the new stearic acid 4-[(n-pentoxy)phenethyl] ester (1) was isolated. This compound exhibited antibacterial properties at 10μg/mL concentration by using disc diffusion method against Gram-positive bacteria Bacillus cereus and Bacillus subtilis and fast-acid bacterium Mycobacterium fortuitum. The structure of the new compound was elucidated by spectroscopic methods and by chemical conversion.

É crescente o aumento da preocupação do homem com a contaminação de ambientes aquáticos uma vez que da subsistência dos mesmos depende o bem estar de todos. Ao mesmo tempo em que cresce a preocupação tem aumentado a quantidade de pesquisas em busca de tecnologias alternativas ao tratamento e remediação de efluentes aquosos contaminados pelas mais diversas substâncias incluindo os metais pesados. Nas últimas décadas, tem crescido o número de trabalhos avaliando a capacidade de sorção e a viabilidade da utilização de biossorventes de baixo custo na captação de íons metálicos e um desses materiais são as biomassas de algas pardas. Dentre os vários gêneros existentes no planeta o território brasileiro é rico na macroalga do gênero Sargassum. Muito embora a composição dessas biomassas varie o principal constituinte das mesmas é o ácido algínico e seus sais alcalinos na forma de um copolímero linear, homopolimérico, com unidades do ácido manurônico, (M), (1-4)-β-D- ligado e seu epímero em C-5 nos resíduos α-L-gulurônicos, (G), respectivamente, covalentemente ligados entre si em sequencias diferenciadas ou em blocos. Os monômeros podem aparecer em blocos homopoliméricos consecutivos, resíduos G (unidades G), resíduos consecutivos M (unidades M), pequenas unidades alternadas M e G (blocos MG), ou em blocos aleatoriamente organizados. A proposta dessa dissertação é realizar a modificação do copolímero existente na biomassa com epicloridrina, avaliar e comparar a captação de íons Cu (II) e Hg (II) pela biomassa da alga Sargassum sp., com a biomassa não modificada e resultados publicados na literatura

Pseudomonas aeruginosa and Escherichia coli are the most prevalent Gram-negative biofilm forming medical device associated pathogens, particularly with respect to catheter associated urinary tract infections. In a similar manner to Gram-positive bacteria, Gram-negative biofilm formation is fundamentally determined by a series of steps outlined more fully in this review, namely adhesion, cellular aggregation, and the production of an extracellular polymeric matrix. More specifically this review will explore the biosynthesis and role of pili and flagella in Gram-negative adhesion and accumulation on surfaces in Pseudomonas aeruginosa and Escherichia coli. The process of biofilm maturation is compared and contrasted in both species, namely the production of the exopolysaccharides via the polysaccharide synthesis locus (Psl), pellicle Formation (Pel) and alginic acid synthesis in Pseudomonas aeruginosa, and UDP-4-amino-4-deoxy-l-arabinose and colonic acid synthesis in Escherichia coli. An emphasis is placed on the importance of the LuxR homologue sdiA; the luxS/autoinducer-II; an autoinducer-III/epinephrine/norepinephrine and indole mediated Quorum sensing systems in enabling Gram-negative bacteria to adapt to their environments. The majority of Gram-negative biofilms consist of polysaccharides of a simple sugar structure (either homo- or heteropolysaccharides) that provide an optimum environment for the survival and maturation of bacteria, allowing them to display increased resistance to antibiotics and predation.

The importance of marine algae, often referred to as seaweeds, has been felt over a long time and is appreciated more and more in modern times. The economic value of marine algae is understood both indirectly and directly. The indirect benefit is due to the role of marine phytoplankton as well as the benthic macrophyte biomass along the shore and in the continental shelf, in primary production of the sea. Direct benefit includes the use of marine algae as food, feed, fertilizer and as source of various products of commercial importance such as agar and alginic acid. Hence to understand the potential resources of seaweeds, their distribution, density, standing crop and interrelated environmental parameters, a detailed study (survey and ecological work) was carried out for a period of 20 months from August 1988 to March 1990 in South Andaman, North Andaman, Middle Andaman, Havelock, Neil, Car Nicobar, Terassa, Chowra and Bumpoka islands. However in South Andaman, data were collected from five fixed stations fortnightly during this period for the purpose of modelling and system analysis.

The alginic acid or alginates are acidic polysaccharides found in brown seaweed widely used in food, cosmetic, medical and pharmaceutical industry. This paper proposes the extraction, chemical characterization and verification of the pharmacological activities of brown seaweed variegata Lobophora . The alginate was extracted from the seaweed Lobophora variegata and part was sulphated for comparative purposes. The native extract showed 42% total sugar, 65% uronic acid, 0,36 % protein and 0% of sulfate, while the sulfate showed 39% , 60%, 0.36% and 27,92 % respectively. The presence of a sulfate group may be observed by the metachromasia with toluidine blue in electrophoresis system and characteristic vibration 1262,34 cm-1 in infrared spectroscopy connections assigned to S = O. We observed the formation of films and beads of native alginate, where more concentrated solution 6% resulted in a thicker and more consistent film. Native alginate showed proliferative activity at concentrations (25 and 50 mcg), (50 mg) and (100 mg) in 3T3 cell line in 24h, 48h and 72h, respectively , as the sulfated (100 mg) in 24 . Also showed antiproliferative or cytotoxic activity in HeLa cells of strain, (25 and 100 mg), (25 and 100 mg) and (25, 50 and 100 mg), to native, now for the sulfate concentrations (100 mg) in 24 (25, 50 and 100 mg) in 48 hours, and (50 and 100 mg ) 72h. For their antioxidant activity, the sulfated alginates have better total antioxidant activity reaching 29 % of the native activity while 7.5 % of activity . For the hydroxyl radical AS showed high inhibition ( between 77-83 % ) in concentrations, but the AN surpassed these numbers in the order of 78-92 % inhibition. The reducing power of AN and AS ranged between 39-82 % . In the method of ferric chelation NA reached 100 % chelating while the AS remained at a plateau oscillating 6.5%. However, in this study , we found alginates with promising pharmacological activities, to use in various industries as an antioxidant / anti-tumor compound

Due to an increasing interest, a vast number of biodegradable polymers have been obtained recently. Polymers naturally produced, such as cellulose, starch, chitosan and alginate, represent biodegradable materials, with low toxicity and low cost. Among polysaccharides, chitosan has been of great interest of the industrial and academic research, due to its special qualities of biodegradability and biocompatibility and, on the other hand, to the versatility of its use in several physical forms and products. A significant growth in the development of new dosage forms capable to deliver the drug in a controlled and targeted way has been observed in these last years. Such pharmaceutical forms search, mainly, the reduction of the dose administered and of the administration frequency, the reduction of adverse side effects and, consequently, a better patient compliance. The present paper describes the use of chitosan in pharmaceutical products, especially in drug controlled delivery systems.

The formation of complex coacervates between chitosan and alginate was evaluated during microparticles formation. Mass ratio between polyelectrolytes and calcium chloride concentration were determinated by conductimetric analysis and by calcium ions quantification, respectively. Inert microparticles were prepared using a complex coacervation method in W/O emulsion and morphological analyses of microparticles were carried out. This method enabled the production of spherical particles, with slightly rough surface and narrow size distribution with maximal diameter of 10 μm.

Tuberculosis (TB) is a serious public health problem all over the world, which was recognized by the WHO in 1993 as a global emergency because of its very high incidence in the 22 countries that concentrate 80% of TB cases. One of these countries is Brazil, where TB occurrences are well-documented in major cities, but little is known of its spread in rural areas and small towns. Therefore, an epidemiological study was done on medical records of TB sufferers in the rural district and small town of Américo Brasiliense, São Paulo state, from 1992 to 2002, with the aim of improving TB prevention and treatment. The results showed that the incidence of TB peaked in the years of intense migration of rural workers, largely cane-cutters. Among these, the disease attacks mainly the men, in their productive years (20 to 40 years old). The predominant clinical form observed was pulmonary tuberculosis. The treatment abandonment rate was less than 1.8%, while the cure rate was around 90%. The disease detection rate by examination of sputum for acid-fast bacilli was around 60%.

In this study, the aim was to measure the concentration of fluoride ions in dental alginates and the quantity released from alginate molds immersed in milliQ water, 0.1N hydrochloric acid and artificial saliva. Two separate lots of each of seven brands of alginate commercially available in Brazil were analyzed: Avagel, Deguprint, Hydrogum, Orthoprint, Jeltrate, Jeltrate Plus and Jeltrate Chromatic. Fluoride was measured in each sample by direct potentiometry, using the combined fluoride-ion selective electrode. The brands with the highest fluoride concentrations were Hydrogum (7052.87 μg/g), Jeltrate Plus (6519.68 μg/g) and Orthoprint (6218.18 μg/g). Only in Hydrogum and Jeltrate were different fluoride concentrations found in lots one and two. The various materials showed differences in the amount of fluoride released from the molds into the immersion medium, the mean concentration being highest in Hydrogum and Orthoprint, in all three media. The immersion medium also influenced the release of fluoride, which was lower in saliva than in water and highest in acid. Considering that the concentrations of fluoride found in the alginates tested were high and that various sources of exposure to fluoride exist, there is a need for constant monitoring of alginates intended for dental use.
