We took a comparative approach utilizing clines to investigate the extent to which natural selection may have shaped population divergence in cuticular hydrocarbons (CHCs) that are also under sexual selection in Drosophila. We detected the presence of CHC clines along a latitudinal gradient on the east coast of Australia in two fly species with independent phylogenetic and population histories, suggesting adaptation to shared abiotic factors. For both species, significant associations were detected between clinal variation in CHCs and temperature variation along the gradient, suggesting temperature maxima as a candidate abiotic factor shaping CHC variation among populations. However, rainfall and humidity correlated with CHC variation to differing extents in the two species, suggesting that response to these abiotic factors may vary in a species-specific manner. Our results suggest that natural selection, in addition to sexual selection, plays a significant role in structuring among-population variation in sexually selected traits in Drosophila.

陆地生态系统的呼吸作用是全球碳循环的一个主要通量和响应全球变化的一个潜在的重要正反馈机制。研究陆地生态系统的呼吸作用特征及其对生物环境因子的响应具有重要意义。本实验利用涡度相关技术对内蒙古库布齐沙漠两个不同土地利用类型的生态系统(人工种植杨树林和天然的油蒿灌丛)2006年生长季(4-10月)的生态系统呼吸特征进行比较研究,并分析了控制生态系统呼吸(Re)的生物与环境因子。结果表明:在这两种生态系统中Re存在着显著的日变化和季节变化,两个生态系统之间Re也存在着显著差异。Re日平均最大值分别2.0 mol CO2 m-2 s-1和1.7 mol CO2 m-2 s-1,都显著低于其他类似生态系统。杨树林和油蒿灌丛的生态系统Re与空气温度都表现出明显的指数相关关系,温度敏感指数Q10分别为1.11和1.12。两个生态系统的Re都与土壤水分含量呈显著的线性正相关关系,表明库布齐沙漠的生态系统的Re受到了土壤水分条件的限制。杨树林和油蒿灌丛生态系统呼吸Re都与叶面积指数的有线性回归关系,说明叶面积指数对生态系统呼吸有很好的指示作用。 本文还选择了两个生态系统内四种常见的土壤覆盖类型(分别是:杨树林生态系统的沙地SL和低洼地BL;油蒿灌丛生态系统的灌丛间BS和灌丛内WS),利用动态密闭气室测定了5-9月土壤呼吸的季节动态以及植株尺度的小尺度空间异质性。结果表明:1)不同土壤覆盖类型的土壤呼吸存在着很大的差异,其中低洼地BL和沙地SL分别有着最大和最小值,灌丛内WS的土壤呼吸要明显高于灌丛外BS。根生物量是导致它们之间差异的主要原因。2)土壤呼吸与土壤含水量之间的线性关系表明,土壤水分是两个生态系统土壤呼吸的限制因子。3)两个生态系统土壤呼吸存在着明显的小尺度差异,在靠近植株(0.5m内)地方的土壤呼吸的值明显高于距植株0.5m外的值,而0.5m外的土壤呼吸没有显著差异。小尺度土壤呼吸与根生物量之间明显的线性关系,说明根生物量是导致小尺度土壤呼吸差异的原因。本实验对沙漠生态系统的土壤呼吸和生态系统呼吸特征及其影响因子的研究,对准确的估计这一地区的碳收支有很大的帮助,为深入的理解干旱半干旱地区的生态系统碳循环提供了有价值的信息。

Global climate change is expected to modify the spatial distribution of marine organisms. However, projections of future changes should be based on robust information on the ecological niche of species. This paper presents a macroecological study of the environmental tolerance and ecological niche (sensu Hutchinson 1957, i.e. the field of tolerance of a species to the principal factors of its environment) of Calanus finmarchicus and C. helgolandicus in the North Atlantic Ocean and adjacent seas. Biological data were collected by the Continuous Plankton Recorder (CPR) Survey, which samples plankton in the North Atlantic and adjacent seas at a standard depth of 7 m. Eleven parameters were chosen including bathymetry, temperature, salinity, nutrients, mixed-layer depth and an index of turbulence compiled from wind data and chlorophyll a concentrations (used herein as an index of available food). The environmental window and the optimum level were determined for both species and for each abiotic factor and chlorophyll concentration. The most important parameters that influenced abundance and spatial distribution were temperature and its correlates such as oxygen and nutrients. Bathymetry and other water-column-related parameters also played an important role. The ecological niche of C. finmarchicus was larger than that of C. helgolandicus and both niches were significantly separated. Our results have important implications in the context of global climate change. As temperature (and to some extent stratification) is predicted to continue to rise in the North Atlantic sector, changes in the spatial distribution of these 2 Calanus species can be expected. Application of this approach to the 1980s North Sea regime shift provides evidence that changes in sea temperature alone could have triggered the substantial and rapid changes identified in the dynamic regimes of these ecosystems. C. finmarchicus appears to be a good indicator of the Atlantic Polar Biome (mainly the Atlantic Subarctic and Arctic provinces) while C. helgolandicus is an indicator of more temperate waters (Atlantic Westerly Winds Biome) in regions characterised by more pronounced spatial changes in bathymetry.

Relief is regarded as the abiotic factor most strongly influencing pedogenic processes at a local scale. The spatial correlations between the composition of the clay fraction (iron - Fe and aluminum - Al oxides, kaolinite and organic matter - OM) and contents of available phosphorus (P) of an Oxisol were evaluated at hillslope scale under sugarcane cultivation. A total of 119 samples were collected at intersection points on a 100. ×. 100. m georeferenced grid of regularly spaced points 10. m apart in the 0.2-0.4. m depth in an area consisting of two landform components namely: component I (an area with a linear hillslope curvature), and component II (one with a concave-convex hillslope curvature). Soil OM and available P contents were subjected to descriptive statistics and geostatistical analyses in order to assess their variability and spatial dependence. All attributes studied were spatially dependent. Available phosphorus had positive spatial correlation with high crystalline goethite, hematite and gibbsite. Identifying small hillslope curvatures is useful with a view to better understanding their relationships with soil organic matter and available phosphorus, as well as kaolinite and Fe and Al oxide attributes. A simple correlation analysis by itself is inadequate to relate attributes, which requires a supplemental, geostatistical technique. © 2012 Elsevier B.V..

Phytochromes are red/far-red light photoreceptors that mediate a variety of photomorphogenic processes in plants, from germination to flowering. In addition, there is evidence that phytochromes are also part of the stress signalling response, especially in response to water deficit stress, which is the major abiotic factor limiting plant growth and crop productivity worldwide. In this study, we used the phyA (far red-insensitive; fri), phyB1 (temporary red-insensitive; tri) and phyB2 mutants of tomato (Solanum lycopersicum L.) to study the roles of these three phytochromes in drought stress responses. Compared to wild type (WT) plants grown under water-deficit stress conditions, the fri, tri, and phyB2 mutants did not exhibit altered dry weights, leaf areas, stomatal densities, or stomatal opening. The stomatal conductance of all three mutants was severely reduced under both fully-hydrated and water-deficit conditions. Although relative water contents did change after drought stress in each mutant, the most significant reduction in water potential during water stress was observed in the fri mutant. However, this mutant returned its water status to WT levels during rehydration. Although the phyB2 mutant lost more water from detached leaves during abscisic acid (ABA) treatment, phyB2 behaved like WT plants, indicating that this mutant was not insensitive to ABA. Overall, these results indicate that the phytochromes phyA, phyB1, and phyB2 modulate drought stress responses in tomato.

Water deficit is one of the factors which most limit agriculture yield and growth. Although sugar cane has moderate tolerance to drought, it presents high yield losses under the influence of this abiotic factor. Based on this fact, selection of genotypes tolerant to water stress may represent an alternative for decreasing the amount of water used for irrigation, while keeping or increasing yield. This study was performed in order to evaluate the performance of four sugarcane cultivars during initial development under water stress conditions, by means of morphological variables to select more tolerant genotypes to drought. The experiment was carried out in a greenhouse at the Department of Rural Engineering, College of Agricultural Sciences - UNESP/Botucatu – SP, from November 26th 2010 to April 5th 2011. A total of four sugar cane cultivars were evaluated (RB855453, RB92579, RB965902 and RB965917) under two treatments as follows: control (100% field capacity) and water stress (50% field capacity). Evaluations were performed at 0, 28 and 63 days after treatment application. The following morphological variables were analyzed: plant height, leaf area, leaf length, leaf width, number of green leaves, shoot and root dry matter. The RB855453 and RB92579 cultivars produced more shoot and root dry matter under water stress treatment, while the RB965902 and RB965917 cultivars had lower shoot and root dry matter production under the same conditions. Therefore, the RB855453 and RB 92579 cultivars can be considered tolerant while the RB965902 and RB965917 cultivars can be considered susceptible.

[EN]Multicellular red algae (Rhodophyta) have some of the most complex life cycles known in living organisms. Economically valuable seaweeds, such as phycocolloid producers, have a triphasic (gametophyte, carposporophyte, and tetrasporophyte) life cycle, not to mention the intricate alternation of generations in the edible “sushi-alga” nori. It is a well-known fact that reproductive processes are controlled by one or more abiotic factor(s), including day length, light quality, temperature, and nutrients. Likewise, endogenous chemical factors such as plant growth regulators have been reported to affect reproductive events in some red seaweeds. Still, in the genomic era and given the high throughput techniques at our disposal, our knowledge about the endogenous molecular machinery lags far behind that of higher plants.

Las masas forestales tienen una importancia colosal para nuestra sociedad y el conjunto de la biosfera. Estudios recientes a escala mundial indican que la sequía es el factor abiótico que más afecta a su crecimiento y supervivencia, seguida por las temperaturas extremas y la salinidad. Aunque comprender los mecanismos con que las especies arbóreas toleran estas formas de estrés tiene un interés aplicado evidente, dichos mecanismos se han estudiado mucho más en especies herbáceas modelo o de interés agronómico. Existen sin embargo diferencias notables entre ellas, como se demuestra en esta tesis y en otros trabajos recientes. Nuestro estudio se centra concretamente en la respuesta molecular del chopo –el sistema modelo forestal más desarrollado– al estrés abiótico, con particular énfasis en la sequía. Utilizando una estrategia proteómica y tratamientos controlados, hemos identificado componentes mayoritarios de dicha respuesta. Su participación en la misma se ha validado mediante análisis transcripcionales detallados utilizando tecnología qRT-PCR (PCR cuantitativa en tiempo real). Hemos identificado proteínas cuyo nexo funcional con mecanismos de tolerancia ya era conocido, como chaperonas moleculares sHSP o enzimas que atenúan el estrés oxidativo, pero también proteínas cuya relación funcional con el estrés es menos clara o incluso novedosa, como polifenol oxidasas (PPO), deshidrogenasas/reductasas de cadena corta (SDR), o bicupinas (BIC), entre otras. El cuerpo central de la tesis consiste en la caracterización detallada de una PPO inusual, cuya inducción por estrés hídrico se describe por vez primera. Estas enzimas están ampliamente distribuidas en plantas, si bien su número es muy variable de unas especies a otras. Algunas, como nogal, tienen un único gen, mientras que Arabidopsis no tiene ninguno. En la última versión del genoma de chopo hemos identificado un total de 12 miembros bona fide, corrigiendo trabajos previos, y hemos caracterizado su expresión individual ante diferentes situaciones de estrés controlado y tratamientos hormonales. La isoforma antedicha es el único miembro de la familia que responde claramente a la deshidratación. También responde a salinidad y a la mayor parte de tratamientos hormonales ensayados, pero no a daño mecánico o tratamientos con metil jasmonato. Esto la diferencia de enzimas homólogas presentes en otras especies de plantas, que se han relacionado experimentalmente con estrés biótico. Los patrones de acumulación de transcritos en árboles adultos son compatibles con un papel protector frente a la sequía. La integración de nuestros estudios funcionales y filogenéticos sugiere que la familia ha sufrido un proceso reciente de diversificación y neofuncionalización, siendo la protección frente a deshidratación su papel primigenio. Aunque se conoce la actividad bioquímica in vitro de este tipo de enzimas, sus sustratos naturales son esencialmente una incógnita. Mediante expresión heteróloga en Escherichia coli BL21(DE3) hemos detectado que la enzima de chopo es capaz de oxidar L-DOPA a dopaquinona, siendo menos activa frente a otros sustratos. Por otra parte, hemos demostrado su localización cloroplástica mediante transformación transitoria de protoplastos con fusiones a la proteína fluorescente YFP. Mediante la obtención de plantas transgénicas de A. thaliana hemos demostrado que la enzima de chopo aumenta considerablemente la tolerancia in vivo frente a la deshidratación y al estrés salino. El análisis fenotípico detallado de las líneas transgénicas, combinando múltiples metodologías, nos ha permitido sustanciar que la tolerancia tiene una base compleja. Esta incluye una mayor protección del sistema fotosintético, una capacidad antioxidante muy incrementada y la acumulación de solutos osmoprotectores como la prolina. Los análisis metabolómicos nos han permitido asociar la expresión de la proteína a la síntesis de un flavano no descrito previamente en A. thaliana, vinculando la enzima de chopo con la síntesis de fenilpropanoides. También hemos observado alteraciones en los niveles hormonales que podrían subyacer a efectos pleiotrópicos con interés aplicado, como un aumento consistente del tamaño de la planta o el acortamiento del ciclo de crecimiento. Además de aportar datos novedosos sobre la funcionalidad in vivo de esta familia de oxidasas, los resultados de esta tesis demuestran que los árboles son sistemas de estudio interesantes para caracterizar nuevas estrategias de tolerancia al estrés abiótico con potencial aplicado. ABSTRACT Forests masses have an extraordinary importance for our society and the biosphere. Recent worldwide studies indicate that drought is the abiotic factor that affects more their growing and survival, followed by extreme temperatures and salinity. The understanding of how the arboreal species tolerate the stress has an evident practical interest, but their mechanisms have been studied much more in herbaceous species or with agronomic interest. However, considerable differences exist between them, as this thesis and recent studies show. Our study is focused on the molecular response of the poplar –the more developed forestry model system- to abiotic stress, specifically focused in the drought. Using a proteomic strategy and controlled treatments, we have identified main components in such response. Its participation has been validated through transcriptional analysis using qRT-PCR technology. We have identified proteins whose functional connection with tolerance mechanisms were already known, as molecular chaperones sHSP or enzymes that attenuate the oxidative stress, but also some proteins whose functional relationship with the stress is less clear or even novel, as polifenol oxidases (PPO), short chain deshidrogenases/reductases (SDR), or bicupines (BIC), among others. The central body of the thesis consists of the detailed characterization of an unsual PPO, whose induction due to drought stress is first described. These enzymes are thoroughly distributed in plants, but their number of members is very variable among species. Some of them, as the walnut tree, have a single gene, while Arabidopsis has none. We have identified a total of 12 members in the last version of the poplar genome, correcting previous works, and have characterized their individual expression against different situations of controlled stress and hormone treatments. The aforementioned isoform is the only member of the family that responds clearly to the drought. It also reacts to salinity and the majority of hormonal treatments tested, but it does not respond to mechanical damage or treatments with methyl jasmonate. This is the difference with homologue enzymes present in other plant species, which have been related experimentally with abiotic stress. The accumulation patterns of transcripts in adult trees are compatible with a protector role against drought. The integration of our functional and phylogenetic studies suggests that the family has suffered a recent process of diversification and neofunctionalization, being the protection against drought their original role. Although the in vitro biochemistry activity of this kind of enzymes is already known, their natural substracts are essentially a mystery. By means of heterologous expression of Escherichia coli BL21(DE3) we have detected that the enzyme of poplar is able to oxidize L-DOPA to dopaquinone, being less active against other substrates. Additionally, we have proven its chloroplastic location with transitory transformation of protoplasts with YFP protein fusion. By means of getting transgenic plants of A. thaliana, we have demonstrated that the poplar enzyme increases notably the in vivo tolerance against the drought and salinity stresses. The phenotypic analysis of the transgenic lines, and the use of multiple methodologies, allowed us to test the complexity of the tolerance. This includes a major protection of the photosynthetic system, a very increased antioxidant capacity and the accumulation of osmoprotectant solutes as the proline. The metabolic analysis has allowed to associate the protein expression with the synthesis of a Flavan non described previously in A. thalaiana, linking the enzyme of poplar with the synthesis of phenylpropanoids. We have observed alterations in the hormonal levels that could underlie pleiotropic effects with applied interest, as a consistent increase of the size of the plant and the reduction of the growth cycle. The results of this thesis, in addition to provide novel data about the in vivo functionality of the oxidase family, demonstrate that the trees are interesting systems of study to characterize new strategies of tolerance against abiotic stress with applied potential.

A utilização de gradientes ambientais no estudo de comunidades vegetais possibilita a eleição de sítios onde há a predominância de um fator abiótico que determina o sucesso ou o fracasso de espécies ao longo de sua extensão. Entre as inúmeras ferramentas utilizadas no estudo de gradientes climáticos, se destaca a anatomia do lenho, pois, é um ramo da ciência que permite analisar, além dos aspectos espaciais, os aspectos temporais dos sítios por meio dos anéis de crescimento. Além disso, a ampla distribuição das plantas lenhosas ao longo do globo possibilita análises em praticamente todos os tipos de biomas e ecossistemas terrestres. Dentro desse contexto estão os bosques andino-patagônicos de Araucaria araucana (Pehuén) ao norte de sua distribuição na Argentina. Esses bosques ocupam territórios caracterizados por um acentuado gradiente de precipitação, que vai de cerca de 3000 a 100 milímetros anuais, entre a cordilheira do Andes e a estepe patagônica, que os define como bosques mésicos e xéricos, com diferenças ecológicas que condicionam a formação vegetal, dinâmica, estrutura, relações com o clima e vulnerabilidade em cenários de mudanças ambientais. O objetivo do projeto foi descrever e analisar comparativamente a estrutura anatômica do lenho de 33 espécies arbóreas e arbustivas ocorrentes ao longo de um gradiente de precipitação, entre a encosta da cordilheira dos Andes e a estepe Patagônica, para verificar possíveis alterações anatômicas que permitam a determinação de tendências e elucidem os limites de distribuição das espécies. Foram coletadas amostras não destrutivas de lenho durante três expedições para a Patagônia entre 2012 e 2014, seguidas de procedimentos laboratoriais de confeccção de lâminas histológicas, preparo de amostras para análises dendrocronológicas, captura de imagens com câmeras acopladas a microscópios óptico e eletrônico de varredura, mensurações, descrições e análises comparativas com o auxílio de softwares. Foram identificados caracteres anatômicos exclusivos de sítios secos e úmidos que permitiram a caracterização, posicionamento ao longo do gradiente de precipitação e grupamento de espécies xerófitas e mesófitas; A ultra-estrutura das pontoações de traqueídeos de Araucaria araucana apresentou diferenças marcantes na frequência e porosidade de suas membranas, sendo maior e menos porosas em sítios xéricos, e menor e mais porosas nos mésicos; a chave dicotômica microscópica permitiu a identificação das 32 espécies arbustivas, as espécies Chuquiraga oppositifolia e Nothofagus antarctica apresentaram potencial dendrocronológico, e foram identificadas tendências anatômicas latitudinais influenciadas pelo clima nos extremos da América do Sul. A anatomia do lenho se mostrou uma ferramenta confiável no estudo de um gradiente de precipitação na Patagônia argentina, e os resultados apontam para riscos de embolismos e morte induzida por falha no sistema hidráulico de Araucaria araucana ao longo de todo o gradiente, em função do atual cenário climático, e suas projeções em médio e longo prazos.

Common bean (Phaseolus vulgaris L.) is the most important legume crop in the world, providing low-cost, high quality protein, minerals and dietary fiber for human nutrition. The crop was originated from diversity centers in America and exhibits adaptation abilities to different environmental conditions, including soil with low pH. Acid soils occupy 30% of the agro ecosystem areas in the world. In Madeira, acid Andosols and unsatured Cambisols are the dominant groups of soils. Generally, under acidic and infertile conditions, besides of H+ toxicity, soluble aluminium (Al) is the most important abiotic factor limiting plant development and crop productivity. In the field, the hidden roots are also affected and the reduction of root growth under Al stress can be clearly observed in early stages. Seedlings of fifty bean accessions from the Archipelago of Madeira were tested under controlled conditions in the presence of 50 mM Al at pH 4.4. In general, the tested germplasm appeared to be sensitive or very sensitive to Al toxicity. However, fifteen traditional cultivars clearly exhibited elevated Al-tolerance, with an average root relative elongation (RRE) exceeding 50%, while top six accessions surpassed the 60% RRE mark. The Madeira bean germplasm is a valuable resource for sustainable crop production in acid soils and it could be used as parental lines in breeding programs aimed for Al tolerance in common beans.
