In many countries, fermentation studies regarding the use of bacteria instead of yeasts to reduce the period of alcoholic fermentation have been carried out. In Brazil, all the industrial alcohol production is carried out by yeasts as fermentation microorganisms and little is known about other microorganisms with potential to produce alcohol industrially. Brazil stands out in the energy sector worldwide and thus some institutions have been selecting microorganisms which are more efficient in the alcohol production process. Alcoholic bacteria from species Zymomonas mobilis present technological characteristics with potential to be used for alcoholic fermentation at industrial scale, since it exhibits promising abilities to transform sugars into alcohol and carbon dioxide, at conditions similar to the ones required by yeasts. Zymomonas mobilis is a unique bacteria among the microbial world, with peculiar growth, energy production and response to culture conditions, causing a great interest in scientific, biotechnological and industrial fields. The bacteria's ability to make possible energy production in favor of product formation, respond to physical and chemical environmental manipulation as well as its limited product formation make it an ideal microorganism for the study and development of microbial processes for ethanol production.

Levan is an exopolysaccharide synthesized by several microorganisms during fermentation of a culture medium containing sucrose, yeast extract and minerals. This biopolymer has applications in the food segment as stabilizers, thickeners, as carriers for flavor and fragrances, as well as in the pharmaceutical segment as a hypocholesterolemic agent and for exhibiting antitumor activity. This work aimed to analyze the kinetic parameters for levan production. The microorganism used was Zymomonas mobilis CCT 4494, incubated in a synthetic medium containing 200.0 g L-1 of sucrose, in a rotary shaker at 200 rpm and 30°C. Samples were taken every 24h, during a period of 96h, in order to determine variations of pH, biomass, reducing sugar, total reducing sugar and levan formation. It was observed that yields of biomass and synthesized biopolymer in 24h were superior to those obtained in 48, 72 and 96h of fermentation.

The production of ethanol using Zymomonas mobilis had been reported to be three to four times larger than with Saccharomyces cereviseae. The influence of pH, temperature and composition of the means of fermentation are parameters that can direct the metabolism for the production of ethanol. The objective of this study was to evaluate the production of ethanol by Zymomonas mobilis CCT 4494, by variations of the initial pH, temperature and concentrations KCl, K 2SO4, MgSO4, CaCl2 and sucrose, by a factorial experimental design of type 27-2, according to the model proposed by Box et al. (1978). For this, the broth of sugar cane was used as sole carbon source, because it is cheap and easily accessible in the region of São José do Rio Preto, São Paulo State. According to the experimental design, the bacteria Zymomonas mobilis CCT 4494 has adapted in the fermentation mean containing high concentrations of sucrose, and supported the change of pH and temperature of fermentation. The highest amount of ethanol produced was 8.89 mg mL-1. This is not similar to the levels of secondary metabolites produced by Zymomonas mobilis CCT 4494.

Zymomonas mobilis was immobilized using a cell suspension fixed to 8.6 x 10(7) CFU mL(-1) by spectrophotometry. This biomass was suspended in sodium alginate solution (3%) that was dropped with a hypodermic syringe into 0.2 M calcium chloride solution. Was test two initial pH of fermentation medium (4 and 5) and different sucrose concentrations 15, 20, 25, 30 and 35% at 30 degrees C, without stirring for 24, 48, 72 and 96 hours. The levan production to pH 4 was high in sucrose 25% for 24 (16.51 g L-1) and 48 (15.31 g L-1) hours. The best values obtained to pH 5 was in sucrose 35% during 48 (22.39 g L-1) and 96 (23.5 g L-1) hours, respectively. The maximum levan yield was 40.8% and 22.47% in sucrose 15% to pH 4 and 5, respectively. Substrate consumption to pH 4 was bigger in sucrose 15 (56.4%) and 20% (59.4%) and to pH 5 was in 25 (68.85%) and 35% (64.64%). In relation to immobilization efficiency, Zymomonas mobilis showed high adhesion and colonization in support, indicated by cell growth increased from 107 to 10(9) CFU mL(-1) during fermentation time.

Sucrose utilization by Zymomonas mobilis: Levan production optimization using submerged fermentation
Resumo:
Levan is an extracellular polysaccharide (EPS), constituted by linked fructose units β (2,6), obtained by transfructosilation reaction during fermentation of microorganisms in a sucrose rich culture medium. The bacterial levan production is a good alternative of fructose source, besides having certain functional characteristics in the human body, such as a hypocholesterolemic and an anticarcinogenic agent. In the food industry, the levan can be used to fix colors and flavors, as well as to thickening and stabilizing agent in foods. This work aimed to analyze the kinetic parameters for levan production by Zymomonas mobilis CCT 4494, using submerged fermentation. The response surface methodology (RSM), was utilized to predict the optimization of medium for exopolymer production and the independent variables studied were: initial pH, incubation temperature, sucrose, KCl, K2SO4, MgSO4 and CaCl2. It was observed that the bacterium Z. mobilis CCT 4494 well adapted in medium containing high concentrations of sucrose.
