This study was designed to evaluate the effects of different fat sources on the performance, egg quality, and lipid profile of the egg yolks of layers in their second production cycle. The fat sources were cottonseed oil, soybean oil, lard, sunflower oil, or canola oil. Experimental diets were fed to postmolt ISA Brown layers at 70 wk of age and the experimental period was 74 to 86 wk of age. The different fat sources did not influence performance or eggshell quality, but lipid profile of the egg yolk changed as a function of dietary fat sources. In general, the best changes, such as lower level of saturated fatty acids, higher levels of alpha-linolenic acid and DHA, and lower linoleic acid levels, were promoted by the addition of canola oil, but it did not promote enrichment of the eggs with polyunsaturated fatty acids.

Oxycarotenoids concentration in layer hens’ feed may be increased whenever the consumer market demands for more pigmented egg yolks. Oxycarotenoids present in Rubrivivax gelatinosus biomass grown in industrial wastewater have already proved their ability on enhancing yolk color. Moreover, there is a current interest on the antioxidant properties that some carotenoids may provide to food and health. So, in this experiment, we investigated the effects of oxycarotenoids supplementation in hens´ diets on yolk composition and stability to rancidity. Hy line hens aging 19 weeks were individually housed in wire cages equipped with feeders and drinkers. After 15 days receiving a corn basal diet nutritionally balanced, they were assigned to four different treatments, with six replicates, that lasted for 28 days: T1 – basal diet (control), T2 – basal diet + 1.5 mg/kg canthaxanthin, T3 – basal diet + 4.5 g/kg freeze dried R. gelatinosus biomass and T4 - basal diet + 4.5 g/kg spray dried R. gelatinosus biomass. Eggs laid on the last 5 days of rearing were collected; the yolks were separated, analyzed for pH and then freeze dried for the further analyses. Proximate composition was determined after drying and the rancidity (TBRAS method) was investigated at 0, 30 and 60 days of storage at room temperature/dark conditions. Yolks that received the oxycarotenoids had the lowest moisture content and the highest protein contents (P < 0.05). Lipids and pH were the same for all treatments (P > 0.05) and ashes only were higher for T2 (P < 0.05). All treatments that received oxycarotenoids had lower TBARS than control group and, among them, oxycarotenoids from spray dried R. gelatinosus biomass were the most effective to prevent rancidity (P < 0.05). So, we concluded that the use of R. gelatinosus biomass in hens feed brings positive effects to the yolk quality, since protein content is increased and conservation is increased due to decreased water content and lipid oxidation.

Egg yolk color may be controlled both by the concentration and the type of xanthophylls added to diets, with the aim of meeting consumers demand. The objectives of this work were to study how yellow and red xanthophylls present in laying hens` diets influence yolks colors and find the concentrations of these ingredients that meet the regional consumer desire. A factorial design 5 x 3 with 5 concentrations of yellow xanthophylls (lutein + zeaxantin 40%; 1.0, 1.25, 1.5, 1.75 and 2.0 mg/hen/d) and 3 concentrations of red xanthophylls (canthaxantin 10%; 0, 0.35, 0.7 mg/hen/d) was used. After a 30 d period receiving corn basal diets and water ad libitum, 60 White Dekalbe hens were distributed to receive the 15 dietary treatments in 4 replicates. Diets were provided daily at 110 g, during 21 d under 16 h light/8 h dark. Yolks colors were evaluated daily using the CIE L, a, b color space and the Roche color index. After the color stabilization, data were analyzed by ANOVA, regression analysis and Response Surface Methodology (MRS). Global acceptance for the Roche colors was evaluated with a 5 points hedonic scale and data were analyzed by Friedman and Dunn tests. Significance was established at 95% (P < 0.05). Regression analysis showed that the red xanthophylls content was the most important factor that defined L, a and b values for yolk color (P < 0.0001; square function), although its effect was significantly affected by the yellow xanthophylls contents (P = 0.0277; P < 0.0001; P = 0.0002 for L, a, b, respectively), providing evidence for a synergistic effect and not for a saturation effect. MRS showed that the highest redness of yolks was reached with 1.5 mg/hen/d of yellow and 0.5 mg/hen/d of red xanthophylls. So, higher supplementations aiming at increasing yolk color would bring an unnecessary cost to the ration. The most accepted yolk color scored 9, which corresponded to mean color attributes L = 65; a = 16; and b = 64. MRS showed that these values could be reached with combinations of yellow:red xanthophylls like 1.0:0.15 or 1.5:0.1 mg/ hen/d or simply with the yellow xanthophylls at 2.0 mg/hen/d. So, it was concluded that both yellow and red xanthophylls are important to define yolks color; that high amounts of xanthophylls are unnecessary to bring changes to color; and that Brazilian consumer requires yolks color attainable with few amounts of red xanthophylls or only with the yellow ones.

Northern rock sole (Lepidopsetta polyxystra) is a commercially important flatfish in Alaska and was recently classified as a distinct species from southern rock sole (L. bilineata). Taxonomic and vital rate data for northern rock sole are still not fully described, notably at early egg and larval stages. In this study, we provide new taxonomic descriptions of late-stage eggs and newly hatched larvae, as well as temperature-response models of hatching (timing, duration, success), and larval size-at-hatch and posthatch survival at four temperatures (2°, 5°, 9°, and 12°C). Time-to-first-hatch, hatch cycle duration, and overall hatching success showed a negative relationship with temperature. Early hatching larvae within each temperature treatment were smaller and had larger yolk sacs, but larvae incubated at higher temperatures (9° and 12°C) had the largest yolk reserves overall. Despite having smaller yolks, size-at-hatch and the maximum size achieved during the hatching cycle was highest for larvae reared at cold temperatures (2° and 5°C), indicating that endogenous reserves are more efficiently used for growth at these temperatures. In addition, larvae reared at high temperatures died more rapidly in the absence of food despite having more yolk reserves than cold-incubated larvae. Overall, northern rock sole eggs and larvae display early life history traits consistent with coldwater adaptation for winter spawning in the North Pacific.

During a two years research hydrogen peroxide efficacy evaluated for Persian sturgeon, Chinese carps and common carp eggs. These series of the experiments conducted in various conditions different concentration of hydrogen peroxide include 250, 500, 750, 1,000 1,500 2,000 3,000 and 9,000 PPM used as ten and fifteen minutes baths, compared with Malachite green and natural control . In the next phase effect of Levaemisole hydrochloride as an immunostimulator which applied as 5 mg/I in twenty minutes baths from day sixth after hatch evaluated by daily mortality rate and leukocytes counts. The results shown that according fertilization percent and temperature condition hydrogen peroxide at 1,000 and 1,500 PPM concentrations is a effective antifungal agent during incubation periods of Persian sturgeon and even sometimes increasing hatching rates significantly comparing with natural controls and Malachite green. In Chinese carps although hydrogen peroxide controls water molds but it is not recommended in high temperatures because it make shortened incubation time and mold infections will decrease. Also the results shown 750 PPM concentration of hydrogen peroxide in common carp eggs controls water moulds infections and increase hatching rate significantly comparing with Malachite green and natural control. Daily mortality rates accessing of Persian sturgeon fries show that 20 minutes baths of 5mg/1 levamisole hydrochloride decreases daily mortality rate during yolk sac absorption. Nitrogenous compounds: nitrate and ammonium differ significantly between treated tanks with control. Blood leucocytes concentrations as an immune index was different significantly in treated fishes by levamisole hydrochloride comparing with controls. In Chinese carps because yolks sac absorption time is short there is not necessary to use the levamisole hydrochloride. Although treated larvae were more active than controls. As a result our suggestions is to use hydrogen peroxide in Persian sturgeon and common carp artificial propagation and also suggest the use levamisole hydrochloride for Persian sturgeon beside management method in stress and pollution condition

Rabies virus was used as the antigen to immunize laying chickens. Anti-rabies virus immunoglobulin Y(IgY) was isolated from yolks of the eggs laid by these chickens using a two-step salt precipitation and one-step gel filtration protocol. The purified IgY was reduced with dithiothreitol, and heavy chains (HC) and light chains (LC) were obtained. In addition, the purified IgY was digested with pepsin and the fragment with specific antigen binding properties (Fab) was produced. Using matrix-assisted laser desorption/ionization mass spectrometry (MALDI-TOFMS), the average molecular weights of IgY, HC, LC, and Fab were determined as 167 250, 65 105, 18 660, and 45,359 Da, respectively. IgY has two structural differences compared with mammalian IgGs. First, the molecular weight of the heavy chain of IgY is larger than that of its mammalian counterpart, while the molecular weight of the light chain of IgY is smaller. Second, upon pepsin digestion, anti-rabies virus IgY is degraded into Feb, in contrast to mammalian IgG, which has been reported to be degraded into F(ab')(2) under the same conditions. The objective was to identify an appropriate cryoprotectant and protocol for vitrification of red sea bream (Pagrus major) embryos. The toxicity of five single-agent cryoprotectants, dimethyl sulfoxide (DMSO), propylene glycol (PG), ethylene glycol (EG), glycerol (GLY), and methyl alcohol (MeOH), as well as nine cryoprotectant mixtures, were investigated by comparing post-thaw hatching rates. Two vitrifying protocols, a straw method and a solid surface vitrification method (copper floating over liquid nitrogen), were evaluated on the basis of post-thaw embryo morphology. Exposure to single-agent cryoprotectants (10% concentration for 15 min) was not toxic to embryos, whereas for higher concentrations (20 and 30%) and a longer duration of exposure (30 min), DMSO and PG were better tolerated than the other cryoprotectants. Among nine cryoprotectant mixtures, the combination of 20% DMSO + 10% PG + 10% MeOH had the lowest toxicity after exposure for 10 min or 15 min. High percentages of morphologically intact embryos, 50.6 +/- 16.7% (mean +/- S.D.) and 77.8 +/- 15.5%, were achieved by the straw vitrifying method (20.5% DMSO + 15.5% acetamide + 10% PG, thawing at 43 degrees C and washing in 0.5 M sucrose solution for 5 min) and by the solid surface vitrification method (40% GLY, thawing at 22 degrees C and washing in 0.5 M sucrose solution for 5 min). After thawing, morphological changes in the degenerated embryos included shrunken yolks and ruptured chorions. Les infections à Salmonella Enteritidis chez les humains sont associées à la consommation d’œufs ou d’ovoproduits contaminés. La vaccination est un outil utilisé pour diminuer les risques d’infection à SE chez la volaille, mais avec des résultats variables. Au Canada deux bactérines, MBL SE4C et Layermune, sont couramment utilisées pour lutter contre SE. Cependant, leur efficacité n’a pas été complètement déterminée chez les poules pondeuses plus âgées. Par ailleurs, la capacité de ces vaccins à prévenir la transmission verticale et horizontale n’a pas encore été étudiée. L’objectif principal de cette étude était d’évaluer l’effet des deux bactérines sur la réponse immunitaire chez les poules pondeuses, de vérifier la protection conférée par ces vaccins contre l’infection expérimentale à SE, et d’identifier des protéines immunogènes afin de développer un vaccin sous-unitaire. Les oiseaux ont été vaccinés avec deux protocoles d’immunisation en cours d’élevage (soit à 12 et 18, ou à 16 semaines d’âge). Le groupe contrôle a été injecté avec la solution saline. Les oiseaux ont été inoculés per os avec 2 x 109 CFU de la souche SE lysotype 4 à 55 ou à 65 semaines d’âge. Les anticorps (IgG et IgA) ont été mesurés à différents temps avec un ELISA maison en utilisant l’antigène entier de SE. La phagocytose, flambée oxydative, les populations des splénocytes B et T ont été analysées en utilisant la cytométrie en flux. Les signes cliniques, l’excrétion fécale, la contamination des jaunes d’œufs et l’invasion des salmonelles dans les organes ont été étudiés pour évaluer l’efficacité de protection. La transmission horizontale a aussi été étudiée en évaluant l’infection à SE chez les oiseaux mis en contact avec les oiseaux inoculés. Les protéines immunogènes ont été identifiées par SDS-PAGE et Western blot à l’aide d’antisérums prélevés suite à la vaccination et/ou à l’infection expérimentale/naturelle, puis caractérisées par la spectrométrie de masse. Le protocole de vaccination avec deux immunisations a généré un niveau élevé de séroconversion à partir de 3 jusqu’à 32-34 semaines post-vaccination par rapport à celui avec une seule immunisation (p < 0.02), mais il n’y avait plus de différence entre les groupes à 54 et 64 semaines d’âge. Il n’y a pas eu de corrélation entre les niveaux d’IgG et les taux d’isolement des salmonelles dans les organes et des jaunes d’œuf. La production des IgA n’a été observée que chez les oiseaux vaccinés avec 2 injections de MBL SE4C (p ≤ 0.04). Après l’infection expérimentale, la production des IgA a été significativement plus élevée aux jours 1 et 7 p.i dans l’oviducte des oiseaux vaccinés (sauf pour le groupe vacciné avec 2 injections de Layermune) par comparaison avec le groupe contrôle (p ≤ 0.03). Seule la bactérine MBL SE4C a eu un effet protecteur contre la contamination des jaunes d’œuf chez les oiseaux infectés. Ce vaccin réduit partiellement en utilisant deux immunisations, le taux d’excrétion fécale des salmonelles chez les oiseaux inoculés et les oiseaux horizontalement infectés (p ≤ 0.02). Cinq des protéines identifiées par la spectrométrie de masse sont considérées comme des protéines potentiellement candidates pour une étude plus approfondie de leur immonogénicité: Lipoamide dehydrogenase, Enolase (2-phosphoglycerate dehydratase) (2-phospho-D-glycerate hydro-lyase), Elongation factor Tu (EF-Tu), Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) et DNA protection during starvation protein. En général, les bactérines ont induit une immunité humorale (IgG et IgA) chez les poules pondeuses. Cette réponse immunitaire a protégé partiellement les oiseaux quant à l’élimination des salmonelles, la contamination des jaunes d’œuf, ainsi que la transmission horizontale. Dans cette étude, la bactérine MBL SE4C (avec deux immunisations) s’est montrée plus efficace pour protéger les oiseaux que la bactérine Layermune. Nos résultats apportent des informations objectives et complémentaires sur le potentiel de deux bactérines pour lutter contre SE chez les poules pondeuses. Étant donné la protection partielle obtenue en utilisant ces vaccins, l’identification des antigènes immunogènes a permis de sélectionner des protéines spécifiques pour l’élaboration éventuelle d’un vaccin plus efficace contre SE chez les volailles.

La campylobactériose est une zoonose causée par Campylobacter jejuni, une bactérie commensale du poulet, considérée comme la principale source de contamination humaine. C. jejuni est rarement retrouvé dans le tube digestif des poulets avant deux ou trois semaines d'âge. Ce qui pourrait s'expliquer par la transmission d'une immunité maternelle (anticorps IgY) transmise aux poussins via le jaune d'œuf. À la Chaire de recherche en Salubrité des Viandes (CRSV), la caractérisation d'anticorps IgY extraits de jaunes d'œufs frais a montré des niveaux de production d’anticorps différents selon le mode d’immunisation et suggère, in vitro, des effets sur ce pathogène. Ce qui laisse penser qu'en tant qu'additif alimentaire, une poudre de jaunes d'œuf potentialisée permettrait de lutter contre C. jejuni chez le poulet à griller. Dans ce travail, le processus de fabrication de l'additif (déshydratation par « Spray dry » puis encapsulation) a été évalué et les différents modes d'immunisation des poules pondeuses ont également été comparés. Les anticorps ont été extraits des différentes poudres de jaunes d'œuf ou du produit final encapsulé, et caractérisés in vitro (dosage / ELISA, test de mobilité, bactéricidie, western blot). Puis, une évaluation in vivo de la capacité de ces poudres encapsulées, incorporée à 5 % dans la moulée, afin de réduire ou de bloquer la colonisation intestinale des oiseaux par C. jejuni a été testée. In vitro, les résultats ont montré des concentrations d'anticorps et d'efficacité variables selon le type de vaccination. Dans cette étude, on a observé que le « Spray dry » a concentré les anticorps dans les poudres et que ces anticorps sont restés fonctionnels contre C. jejuni. On a également observé que l'encapsulation n’entraîne pas une perte quantitative des anticorps contenus dans les poudres. Malgré les résultats in vitro encourageants, les résultats in vivo ne révèlent aucune inhibition ou réduction de la colonisation des oiseaux par C. jejuni. L’absence d’efficacité la poudre de jaunes d’œuf encapsulée dans notre étude n’est pas due à une perte quantitative et/ou qualitative des anticorps comme soutenu dans les expériences in vitro. Ce qui démontre que les recherches doivent être poursuivies afin de déterminer les conditions optimales de l'utilisation de la poudre de jaune d'œuf in vivo, en tant qu'additif alimentaire chez les poulets

In this work, cholesterol oxide formation and alteration of fatty acid composition were analyzed in n-3 enriched eggs under different storage periods and two temperatures. The eggs enriched with n-3 fatty acids were stored at 5 or 25 degrees C for 45 days and subsequently boiled or fried. For each treatment, 12 yolks were analyzed every 15 days including time zero. The concentrations of the cholesterol oxides 7-ketocholesterol, 7 beta-hydroxycholesterol, and 7 alpha-hydroxycholesterol increased during the storage period and were higher in fried eggs. Only the 7-ketocholesterol was affected by the storage temperature, and its concentration was highest in eggs stored at 25 degrees C. There was no significant difference in the contents of cholesterol and vitamin E at the different storage periods; however, the concentration of vitamin E decreased with thermal treatment. In addition, the n-3 polyunsaturated fatty acids, especially 18:3, 20:5, and 22:6, were reduced throughout the storage at 5 and 25 degrees C.

Vitamin A deficiency is a serious public health problem in developing countries, and it causes death and blindness among children in the developing countries. The fortification of food could be an important source of vitamins to control deficiency. 60 Coturnix coturnix japonica quails were used in a randomized design with duration of seven weeks. The birds were assigned into five treatments with four repetitions. The objective was to evaluate the influence of the supplementation with different levels of retinyl palmitate (2,000 IU, 4,000 IU, 8,000IU and 16,000 IU) in quails under the levels of retinyl in egg yolks. The method used to dose retinyl in yolks of quail eggs was High Performance Liquid Chromatography and the enzymatic method to quantify the cholesterol concentration. The weight and production of eggs was significantly modified by the supplementation with retinyl in the birds. The results showed a gradual increase in the incorporation of retinyl in the egg yolk as a response to the supplementation, reaching values 384% higher than the control values. By the end of the supplementations a significant reduction in the concentrations of retinyl in the eggs yolk was observed. The most lasting supplementations were with 8,000 IU and 16,000 IU which lasted for three weeks. The cholesterol content in eggs was not significantly modified. The consumption of one egg enriched with 16000UI of retinol palmitate in the present study, by day, would probably reach 10 and 7,3% of the daily recommendations of this micronutrient for children of 1 to 3 years of age, and for 4 to 8 years, respectively.
