Alteration in the relative percentage of survival (RPS) rate of rainbow trout (Oncorhynchus mykiss) exposed to 5, 10 and 25µg ml-1 levamisole for 2 h against Yersinia ruckeri was investigated. The average weight of the 120 fish used in this study was 6.3g. Upon challenge with a virulent strain, the relative survival percentage of respectively 83.3%, 86.7% and 76.6% was recorded. The results suggest that the application of levamisole in fish farms could increase resistance to infection of fish and offer economic benefits.

In the aquatic environment, fish are exposed to various stimuli at once and have developed different response mechanisms to deal with these multiple stimuli. The current study assessed the combined impacts of estrogens and bacterial infection on the physiological status of fish. Juvenile rainbow trout were exposed to two different concentrations of 17 beta-estradiol (E2) (2 or 20 mg/kg feed) and then infected with three concentrations of Yersinia ruckeri, a bacterial pathogen causing massive losses in wild and farmed salmonid populations. Organism-level endpoints to assess the impact of the single and combined treatments included hepatic vitellogenin transcript expression to evaluate the E2 exposure efficiency and survival rate of pathogen-challenged fish. The two E2 doses increased vitellogenin levels within the physiological range. Infection with Y. ruckeri caused mortality of trout, and this effect was significantly enhanced by a simultaneous exposure to high E2 dose. The hormone reduced survival at intermediate and high (10(4) and 10(6) colony forming units, cfu) bacterial concentrations, but not for a low one (10(2) cfu). Analysis of hepatic gene expression profiles by a salmonid 2 k cDNA microarray chip revealed complex regulations of pathways involved in immune responses, stress responses, and detoxicification pathways. E2 markedly reduced the expression of several genes implicated in xenobiotic metabolism. The results suggest that the interaction between pathogen and E2 interfered with the fish's capability of clearing toxic compounds. The findings of the current study add to our understanding of multiple exposure responses in fish.

Intensified aquaculture has strong impact on fish health by stress and infectious diseases and has stimulated the interest in the orchestration of cytokines and growth factors, particularly their influence by environmental factors, however, only scarce data are available on the GH/IGF-system, central physiological system for development and tissue shaping. Most recently, the capability of the host to cope with tissue damage has been postulated as critical for survival. Thus, the present study assessed the combined impacts of estrogens and bacterial infection on the insulin-like growth factors (IGF) and tumor-necrosis factor (TNF)-α. Juvenile rainbow trout were exposed to 2 different concentrations of 17β-estradiol (E2) and infected with Yersinia ruckeri. Gene expressions of IGF-I, IGF-II and TNF-α were measured in liver, head kidney and spleen and all 4 estrogen receptors (ERα1, ERα2, ERβ1 and ERβ2) known in rainbow trout were measured in liver. After 5 weeks of E2 treatment, hepatic up-regulation of ERα1 and ERα2, but down-regulation of ERß1 and ERß2 were observed in those groups receiving E2-enriched food. In liver, the results further indicate a suppressive effect of Yersinia-infection regardless of E2-treatment on day 3, but not of E2-treatment on IGF-I whilst TNF-α gene expression was not influenced by Yersinia-infection but was reduced after 5 weeks of E2-treatment. In spleen, the results show a stimulatory effect of Yersinia-infection, but not of E2-treatment on both, IGF-I and TNF-α gene expressions. In head kidney, E2 strongly suppressed both, IGF-I and TNF-α. To summarise, the treatment effects were tissue- and treatment-specific and point to a relevant role of IGF-I in infection.

Data on the occurrence of Yersinia species, other than Y. pestis in Brazil are presented. Over the past 40 years, 767 Yersinia strains have been identified and typed by the National Reference Center on Yersinia spp. other than Y. pestis, using the classical biochemical tests for species characterization. The strains were further classified into biotypes, serotypes and phagetypes when pertinent. These tests led to the identification of Yersinia cultures belonging to the species Y. enterocolitica, Y. pseudotuberculosis, Y. intermedia, Y. frederiksenii and Y. kristensenii. Six isolates could not be classified in any of the known Yersinia species and for this reason were defined as Non-typable (NT). The bio-sero-phagetypes of these strains were diverse. The following species of Yersinia were not identified among the Brazilian strains by the classical phenotypic or biochemical tests: Y. aldovae, Y. rhodei, Y. mollaretti, Y. bercovieri and Y. ruckeri. The Yersinia strains were isolated from clinical material taken from sick and/or healthy humans and animals, from various types of food and from the environment, by investigators of various Institutions localized in different cities and regions of Brazil.

Research on the endocrine role of estrogens has focused on the reproductive system, while other potential target systems have been less studied. Here, we investigated the possible immunomodulating role of 17beta-estradiol (E2) using rainbow trout (Oncorhynchus mykiss) as a model. The aims of the study were to examine a) whether estrogens can modulate immune gene transcription levels, and b) whether this has functional implications for the resistance of trout towards pathogens. Trout were reared from fertilization until 6 months of age under (1) control conditions, (2) short-term E2-treatment (6-month-old juveniles were fed a diet containing 20 mg E2/kg for 2 weeks), or c) long-term E2-treatment (twice a 2-h-bath-exposure of trout embryos to 400 mug 17beta-estradiol (E2)/L, followed by rearing on the E2-spiked diet from start-feeding until 6 months of age). Analysis of plasma estrogen levels indicated that the internal estrogen concentrations of E2-exposed fish were within the physiological range and analysis of hepatic vitellogenin mRNA levels indicated that the E2 administration was effective in activating the endogenous estrogen receptor pathway. However, expression levels of the hepatic complement components C3-1, C3-3, and Factor H were not affected by E2-treatment. In a next step, 6-month-old juveniles were challenged with pathogenic bacteria (Yersinia ruckeri). In control fish, this bacterial infection resulted in significant up-regulation of the mRNA levels of hepatic complement genes (C3-1, C3-3, Factor B, Factor H), while E2-treated fish showed no or significantly lower up-regulation of the complement gene transcription levels. Apparently, the E2-treated trout had a lower capacity to activate their immune system to defend against the bacterial infection. This interpretation is corroborated by the finding that survival of E2-treated fish under bacterial challenge was significantly lower than in the control group. In conclusion, the results from this study suggest that estrogens are able to modulate immune parameters of trout with functional consequences on their ability to cope with pathogens.

Lotes de camundongos suiços convencionais foram inoculados tanto por via intragástrica (IG) quanto por via intravenosa (IV) com Yersinia enterocolitica dos sorotipos 0:3, 0:8 e 0:9 e com amostras de yersinias atípicas. Foi mantido um lote de animais não inoculados como controle. Verificou-se qual o período de permanência dessas bactérias no intestino dos animais inoculados. Yersinia enterocolitica dos sorotipos 0:3, 0:8 e 0:9, considerados adaptados ao homem, permaneceram no intestino dos animais inoculados por um período muito maior do que as amostras de Yersinia não adaptadas, quer inoculadas por via intragástrica, quer por via intravenosa.

Foi feita análise da metodologia empregada e dos resultados alcançados em pesquisa de Yersinia pestis, em material de 24.703 roedores e outros pequenos mamíferos oriundos dos focos pestosos do Nordeste do Brasil, no período de 1966 a 1982. Concluiu-se ser necessário haver maior rapidez na realização dos exames para que os dados obtidos sejam convenientemente aplicados nas atividades de vigilância e controle da peste.

OBJETIVO: Conocer la existencia de Yersinia enterocolitica en suínos visiblemente sanos y sacrificados para el consumo humano. MÉTODOS: Fueron estudiadas 100 muestras de tejido linfático obtenidas en el momento del sacrifício, en un matadero del Estado de México. Fueron realizados muestreos pilotos de 20 casos, de los cuales 20% fueron positivos, permitiendo obtener una muestra estudiada (n=100). Las muestras colectadas de tejido linfático fueron acondicionadas para el aislamiento de Yersinia enterocolitica en caldo de Rappaport y en medio de cultivo de Salmonella-Shigella y MacConkey. Las identificaciones fueron efectuadas por medio de pruebas bioquímicas y serológicas, utilizándose en el caso los antisueros O:3, O:8 y O:9 para la biotipificación correspondiente. RESULTADOS: Fueron obtenidos 22 aislamientos tipificándose 8 serotipos pertenecientes al O:3 y 8 al O:9 correspondientes al biotipo 1; y, en 6 muestras no fue posible la serotipificación. No se encontró en el total de los aislados el serotipo O:8. CONCLUSIONES: En base en la metodología, se registró la presencia de Y. enterocolitica y sus serogrupos en tejido linfático de porcinos por la primera vez en México; esto es importante porque el patógeno y sus serotipos aislados están comprometidos con mayor frecuencia con problemas de salud pública.

No período de 1966 a 1982 foram isoladas 861 cepas de Yersinia pestis sendo 471 originadas de material de roedores e outros pequenos mamíferos, 236 de lotes de pulgas, 2 de lotes de Ornithodorus e 152 de seres humanos dos focos pestosos do Nordeste do Brasil. Entre os roedores, a espécie que concorreu para o maior número de isolamentos foi o Zygodontomys lasiurus pixuna que, também, forneceu o maior número de lotes de pulgas naturalmente infectados, principalmente do gênero Polygenis. O isolamento da Yersinia pestis de material proveniente de 13 Municípios do Estado de Pernambuco, 7 do Ceará, 3 da Paraíba, 1 do Piauí e 1 da Bahia, evidencia que o problema da peste nos focos brasileiros é bastante atual e merecedor de atenção. O maior número de cepas isoladas e de localidades afetadas, registradas no Estado de Pernambuco não significa maior incidência da peste no mesmo, mas é conseqüência da pesquisa mais intensa da Yersinia pestis e da existência de laboratórios melhor preparados para o seu diagnóstico neste Estado.

Pathogenic Yersinia pestis isolates were collected during a plague outbreak at the Paraiba State in 1986. The Y. pestis isolates were investigated for the presence of virulence-associated factors and plasmid content. All strains analysed were proficient in the expression of the VW and fraction 1 antigens, pigment adsorption and pesticin-fibronolysin-coagulase production. A similar plasmid profile composed by four plasmid with molecular weight of 60, 44, 14.9, and 6.4 Megadaltons (MD) was found in all strains. DNA cleavage with EcoRI restriction enzyme further demonstrated the uniform plasmid content of the Y. pestis isolates. Seven additional Y. pestis strains, previously isolated in the same region but in an endemic state, showed the same plasmid fingerprint. The lack of any detectable difference between epidemic and endemic isolates as well as the value of plasmid fingerprints in epidemiology of Y. pestis is discussed.

In the present study three techniques for obtaining outer membrane enriched fractions from Yersinia pestis were evaluated. The techniques analysed were: differential solubilization of the cytoplasmic membrane with Sarkosyl or Triton X-100, and centrifugation in sucrose density gradients. The sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) of outer membrane isolated by the different methods resulted in similar protein patterns. The measurement of NADH-dehydrogenase and succinate dehydrogenase (inner membrane enzymes) indicated that the outer membrane preparations obtained by the three methods were pure enough for analytical studies. In addition, preliminary evidences on the potential use of outer membrane proteins for the identification of geographic variants of Y. pestis wild isolates are presented.
