Die vorliegende Arbeit wurde im Rahmen des BMWi-Verbundprojektes Wechselwirkung und Transport von Aktiniden im natürlichen Tongestein unter Berücksichtigung von Huminstoffen und Tonorganika – Wechselwirkung von Neptunium und Plutonium mit natürlichem Tongestein“ durchgeführt. Um die langfristige Sicherheit der nuklearen Endlager beurteilen zu können, muss eine mögliche Migration der radiotoxischen Abfälle in die Umwelt betrachtet werden. Wegen seiner langen Halbwertszeit (24000 a) leistet Pu-239 einen wesentlichen Beitrag zur Radiotoxizität abgebrannter Kernbrennstoffe in einem Endlager. Das redox-sensitive Pu tritt in Lösung unter umweltrelevanten Bedingungen in den Oxidationsstufen +III bis +VI auf und kann nebeneinander in bis zu vier Oxidationsstufen vorliegen. Tonsteinformationen werden als mögliches Wirtsgestein für Endlager hoch-radioaktiver Abfälle betrachtet. Deshalb sind ausführliche Informationen zur Mobilisierung und Immobilisierung des Pu durch/in das Grundwasser aus einem Endlager von besonderer Bedeutung. In dieser Arbeit wurden neue Erkenntnisse über die Wechselwirkung zwischen Pu und dem natürlichen Tongestein Opalinuston (OPA, Mont Terri, Schweiz) mit Hinblick auf die Endlagerung wärmeentwickelnder radioaktiver Abfälle in einem geologischen Tiefenlager gewonnen.rnDer Fokus der Arbeit lag dabei auf der Bestimmung der Speziation von Pu an der Mineraloberfläche nach Sorptions- und Diffusionsprozessen mittels verschiedener synchrotronbasierter Methoden (µ-XRF, µ-XANES/EXAFS, µ-XRD, XANES/EXAFS). rnDie Wechselwirkung zwischen Pu und OPA wurde zunächst in Batch- und Diffusionsexperimenten in Abhängigkeit verschiedener experimenteller Parameter (u.a. pH, Pu-Oxidationsstufe) untersucht. In Sorptionsexperimenten konnte gezeigt werden, dass einige Parameter (z.B. Temperatur, Huminsäure) einen deutlichen Einfluss auf die Sorption von Pu haben.rnDie Speziationsuntersuchungen wurden zum einen an Pulverproben aus Batchexperimenten und zum anderen an OPA-Dünnschliffen bzw. Diffusionsproben in Abhängigkeit verschiedener experimenteller Parameter durchgeführt. Die EXAFS-Messungen an der Pu LIII-Kante der Pulverproben ergaben, dass eine innersphäriche Sorption von Pu(IV) an Tongestein unabhängig von dem Ausgangsoxidationszustand des Plutoniums in Lösung stattgefunden hat. Durch die Kombination der ortsaufgelösten Methoden wurde erstmalig mittels μ-XRF die Verteilung von Pu und anderen in OPA enthaltenen Elementen bestimmt. µ-XANES-Spektren an Pu-Anreicherungen auf OPA-Dünnschliffen und in Diffusionsproben bestätigen, dass das weniger mobile Pu(IV) die dominierende Spezies nach den Sorptions- und Diffusionsprozessen ist. Darüber hinaus wurde zum ersten Mal ein Diffusionsprofil von Pu in OPA mittels µ-XRF gemessen. Die Speziationsuntersuchungen mittels μ-XANES zeigten, dass das eingesetzte Pu(V) entlang seines Diffusionspfades zunehmend zu Pu(IV) reduziert wird. Mit µ-XRD wurde Illit als dominierende Umgebung, in der Pu angereichert wurde, identifiziert und Siderit als eine redoxaktive Phase auftreten kann. Die Ergebnisse dieser Arbeit zeigen, dass die Sicherheit von OPA als Wirtsgestein eines Endlagers hoch-radioaktiver Abfälle positiv zu bewerten ist. rn

Nectogaline shrews are a major component of the small mammalian fauna of Europe and Asia, and are notable for their diverse ecology, including utilization of aquatic habitats. So far, molecular phylogenetic analyses including nectogaline species have been unable to infer a well-resolved, well-supported phylogeny, thus limiting the power of comparative evolutionary and ecological analyses of the group. Here, we employ Bayesian phylogenetic analyses of eight mitochondrial and three nuclear genes to infer the phylogenetic relationships of nectogaline shrews. We subsequently use this phylogeny to assess the genetic diversity within the genus Episoriculus, and determine whether adaptation to aquatic habitats evolved independently multiple times. Moreover, we both analyze the fossil record and employ Bayesian relaxed clock divergence dating analyses of DNA to assess the impact of historical global climate change on the biogeography of Nectogalini. We infer strong support for the polyphyly of the genus Episoriculus. We also find strong evidence that the ability to heavily utilize aquatic habitats evolved independently in both Neomys and Chimarrogale + Nectogale lineages. Our Bayesian molecular divergence analysis suggests that the early history of Nectogalini is characterized by a rapid radiation at the Miocene/Pliocene boundary, thus potentially explaining the lack of resolution at the base of the tree. Finally, we find evidence that nectogalines once inhabited northern latitudes, but the global cooling and desiccating events at the Miocene/Pliocene and Pliocene/Pleistocene boundaries and Pleistocene glaciation resulted in the migration of most Nectogalini lineages to their present day southern distribution. This paper describes the results of a regional study involving the sampling of 60 pumped tubular wells drilled at the Parana sedimentary basin, Brazil, which was carried out with the purpose of evaluating the U speciation in the Botucatu-Piramboia aquifer. Uranium proved to be intensively dissolved even under the enhanced reducing conditions occurring at the most confined zones of the aquifer, and Eh-pH diagrams were utilized to evaluate the influence of temperature and pressure on the migration of the U-species within the aquifer.

The XAS/WAXS time-resolved method was applied for unraveling the complex mechanisms arising from the evolution of several metastable intermediates during the degradation of chlorine layered double hydroxide (LDH) upon heating to 450 °C, i.e., Zn2Al(OH)6·nH2O, ZnCuAl(OH)6·nH2O, Zn2Al 0.75Fe0.25(OH)6·nH2O, and ZnCuAl0.5Fe0.5(OH)6·nH2O. After a contraction of the interlamellar distance, attributed to the loss of intracrystalline water molecules, this distance experiences an expansion (T > 175-225 °C) before the breakdown of the lamellar framework around 275-295 °C. Amorphous prenucleus clusters with crystallo-chemical local order of zinc-based oxide and zinc-based spinel phases, and if any of copper-based oxide, are formed at T > 175-225 °C well before the loss of stacking of LDH layers. This distance expansion has been ascribed to the migration of Zn II from octahedral layers to tetrahedral sites in the interlayer space, nucleating the nano-ZnO or nano-ZnM2O4 (M = Al or Fe) amorphous prenuclei. The transformation of these nano-ZnO clusters toward ZnO crystallites proceeds through an agglomeration process occurring before the complete loss of layer stacking for Zn2Al(OH)6· nH2O and Zn2Al0.75Fe0.25(OH) 6·nH2O. For ZnCuAl(OH)6·nH 2O and ZnCuAl0.5Fe0.5(OH)6· nH2O, a cooperative effect between the formation of nano-CuO and nano-ZnAl2O4 amorphous clusters facilitates the topochemical transformation of LDH to spinel due to the contribution of octahedral CuII vacancy to ZnII diffusion. © 2013 American Chemical Society.

For the safety assessments of nuclear waste repositories, the possible migration of the radiotoxic waste into environment must be considered. Since plutonium is the major contribution at the radiotoxicity of spent nuclear waste, it requires special care with respect to its mobilization into the groundwater. Plutonium has one of the most complicated chemistry of all elements. It can coexist in 4 oxidation states parallel in one solution. In this work is shown that in the presence of humic substances it is reduced to the Pu(III) and Pu(IV). This work has the focus on the interaction of Pu(III) with natural occurring compounds (humic substances and clay minerals bzw. Kaolinite), while Pu(IV) was studied in a parallel doctoral work by Banik (in preparation). As plutonium is expected under extreme low concentrations in the environment, very sensitive methods are needed to monitor its presence and for its speciation. Resonance ionization mass spectrometry (RIMS), was used for determining the concentration of Pu in environmental samples, with a detection limit of 106- 107 atoms. For the speciation of plutonium CE-ICP-MS was routinely used to monitor the behaviour of Pu in the presence of humic substances. In order to reduce the detection limits of the speciation methods, the coupling of CE to RIMS was proposed. The first steps have shown that this can be a powerful tool for studies of pu under environmental conditions. Further, the first steps in the coupling of two parallel working detectors (DAD and ICP_MS ) to CE was performed, for the enabling a precise study of the complexation constants of plutonium with humic substances. The redox stabilization of Pu(III) was studied and it was determined that NH2OHHCl can maintain Pu(III) in the reduced form up to pH 5.5 – 6. The complexation constants of Pu(III) with Aldrich humic acid (AHA) were determined at pH 3 and 4. the logß = 6.2 – 6.8 found for these experiments was comparable with the literature. The sorption of Pu(III) onto kaolinite was studied in batch experiments and it was determine dthat the pH edge was at pH ~ 5.5. The speciation of plutonium on the surface of kaolinite was studied by EXAFS/XANES. It was determined that the sorbed species was Pu(IV). The influence of AHA on the sorption of Pu(III) onto kaolinite was also investigated. It was determined that at pH < 5 the adsorption is enhanced by the presence of AHA (25 mg/L), while at pH > 6 the adsorption is strongly impaired (depending also on the adding sequence of the components), leading to a mobilization of plutonium in solution.

For the safety assessment of radioactive waste, the possibility of radionuclide migration has to be considered. Since Np (and also Th due to the long-lived 232-Th) will be responsible for the greatest amount of radioactivity one million years after discharge from the reactor, its (im)-mobilization in the geosphere is of great importance. Furthermore, the chemistry of Np(V) is quite similar (but not identical) to the chemistry of Pu(V). Three species of neptunium may be found in the near field of the waste disposal, but pentavalent neptunium is the most abundant species under a wide range of natural conditions. Within this work, the interaction of Np(V) with the clay mineral montmorillonite and melanodins (as model substances for humic acids) was studied. The sorption of neptunium onto gibbsite, a model clay for montmorillonite, was also investigated. The sorption of neptunium onto γ-alumina and montmorillonite was studied in a parallel doctoral work by S. Dierking. Neptunium is only found in ultra trace amounts in the environment. Therefore, sensitive and specific methods are needed for its determination. The sorption was determined by γ spectroscopy and LSC for the whole concentration range studied. In addition the combination of these techniques with ultrafiltration allowed the study of Np(V) complexation with melanoidins. Regrettably, the available speciation methods (e.g. CE-ICP-MS and EXAFS) are not capable to detect the environmentally relevant neptunium concentrations. Therefore, a combination of batch experiments and speciation analyses was performed. Further, the preparation of hybrid clay-based materials (HCM) montmorillonitemelanoidins for sorption studies was achieved. The formation of hybrid materials begins in the interlayers of the montmorillonite, and then the organic material spreads over the surface of the mineral. The sorption of Np onto HCM was studied at the environmentally relevant concentrations and the results obtained were compared with those predicted by the linear additive model by Samadfam. The sorption of neptunium onto gibbsite was studied in batch experiments and the sorption maximum determined at pH~8.5. The sorption isotherm pointed to the presence of strong and weak sorption sites in gibbsite. The Np speciation was studied by using EXAFS, which showed that the sorbed species was Np(V). The influence of M42 type melanodins on the sorption of Np(V) onto montmorillonite was also investigated at pH 7. The sorption of the melanoidins was affected by the order in which the components were added and by ionic strength. The sorption of Np was affected by ionic strength, pointing to outer sphere sorption, whereas the presence of increasing amounts of melanoidins had little influence on Np sorption.

This study comprised batch experiments, direct speciation studies via EXAFS, and modelling with the 2SPNE SC/CE model to elucidate the mechanisms of Np(V) sorption on montmorillonite and, for reference, on γ-Al2O3. The sorption of pM 239Np(V) and µM 237Np(V) on montmorillonite (STx-1, 4 g/L) and γ-Al2O3 (0.5 g/L) was studied at room temperature in the presence and absence of ambient CO2 covering a pH-range from 2.5 (STx-1) or 5 (γ-Al2O3) to 10.5 with 0.01 or 0.1M NaClO4 as background electrolyte. The Np(V) uptake was determined by γ spectroscopy of the supernatants and calculated as percentage as well as distribution coefficient Kd. Sorption starts from pH ~6 and, under exclusion of CO2, increases continuously, while, in the presence of ambient air, it reaches a maximum at pH ~8.5 (γ-Al2O3: log Kd max ≈ 4 mL/g; STx-1: log Kd max ≈ 2.7 mL/g). Beyond that it decreases again due to the formation of queous neptunium carbonate complexes. Furthermore, neptunium sorption on montmorillonite is influenced by ionic strength at pH <6 through ion exchange processes pointing towards the formation of outer-sphere surface complexes there. Isotherms measured at the sorption maximum showed the precipitation of resumably neptunium carbonate complexes above 3∙10^-5 M under ambient air conditions. Additionally, they indicated progressive saturation of the sorption sites of γ-Al2O3. At selected pH (STx-1: 5.0, 7.0, 8.0, 8.5, 9.0, 9.5; γ-Al2O3: 8.5, 9.5) EXAFS samples were prepared as wet pastes with µM 237Np and measured at room temperature in fluorescence mode at ANKA and ESRF. Several spectra were averaged and analysed with EXAFSPAK and FEFF 8.20 employing models of NaNpO2(CO3) or soddyite, (UO2)2SiO4∙2(H2O). The shorter atomic distances of the neptunyl ion at pH 5 compared to the others hinted at the retention of the hydration shell and, thus, at outer-sphere sorption. On average the bond lengths for Np(V) sorbed on STx-1 at high pH were Oax ≈ 1.84 Å and Oeq ≈ 2.53 Å. At high pH, ternary neptunyl carbonate surface complexes could be identified for montmorillonite (C ≈ 3.00 Å), but not for γ-Al2O3, where an interaction of neptunium with the aluminium surface atoms according to the soddyite model gave better agreement with the experimental data. However, neither structure as suggested by the two models could be excluded for both systems rendering a combination most likely. Modelling of the sorption data provided further evidence for the existence of ternary neptunium carbonate surface complexes in both cases. The results of this study can aid environmental risk assessment for clay-based nuclear waste repositories by providing valuable input data for simulations of radionuclide migration from a final disposal site.

Die vorliegende Arbeit wurde im Rahmen eines Stipendiums des interdisziplinärenrnGraduiertenkollegs 826 „Spurenanalytik von Elementspezies: Methodenentwicklung und Anwendungen“, gefördert durch die Deutsche Forschungsgemeinschaft (DFG) und das Land Rheinland-Pfalz, angefertigt. Dabei sollten neue Erkenntnisse über die Wechselwirkung zwischen Neptunium und natürlichem Tongestein mit Hinblick auf die Endlagerung wärmeentwickelnder radioaktiver Abfälle in einem geologischen Tiefenlager gewonnen werden. Auf Grund seiner langen Halbwertszeit von mehr als zwei Millionen Jahren wird Np-237 einen signifikanten Anteil an der Radiotoxizität dieser Abfälle nach Lagerzeiten von mehr als 1000 Jahren haben. Np tritt in Lösung unter umweltrelevanten Bedingungen in den Oxidationsstufen +IV und +V auf. Auf Grund der guten Löslichkeit und daher höheren Mobilität ist Np(V) als die gefährlichere Spezies einzustufen. In den Migrationsstudien wurdernOpalinuston (OPA) aus Mont Terri, Schweiz, als natürliches Referenzmaterial verwendet. Der Fokus dieser Arbeit lag dabei auf der Speziation von Np an der Mineraloberfläche mittels synchrotronbasierter Röntgenabsorptionsspektroskopie (EXAFS/XANES).rnDie Wechselwirkung zwischen Np(V) und OPA wurde zunächst in Batch- und Diffusionsexperimenten in Abhängigkeit verschiedener experimenteller Parameter (u.a. pH, Temperatur, Hintergrundelektrolyt, Einfluss von Huminsäure, Konkurrenz mit U(VI), aerobe/anaerobe Bedingungen) untersucht. Die Untersuchung der Np-Speziation erfolgt zum einen an Pulverproben aus Batch-Experimenten unter aeroben und anaeroben Bedingungen, welche mittels EXAFS-Spektroskopie untersucht wurden. Zum anderen wurden ortsaufgelöste μ-XANES-Messungen an Np-Anreicherungen auf OPA-Dünnschliffen und in Diffusionsproben durchgeführt. Durch Kombination der Spektroskopie mit μ-Rötngenfluoreszenzmapping (XRF) und μ-Röntgenbeugung (XRD) konnten zudem Erkenntnisse über die Elementverteilung von Np und anderen im Opalinuston enthaltenen Elementen sowie über kristalline Mineralphasen im Umfeld von Bereichen erhöhter Np-Konzentration erhalten werden.rnSowohl Sorptionsexperimente als auch die spektroskopischen Untersuchungen zeigten eine teilweise Reduktion von Np(V) zu Np(IV) bei der Wechselwirkung mit OPA. Dabei konnte Pyrit als eine der redoxaktiven Phasen identifiziert werden. In diesem Zusammenhang ist die Bildung schwerlöslicher Np(IV)-Spezies mit Hinblick auf die Endlagerung positiv zu bewerten.

Die vorliegende Arbeit wurde im Rahmen des Verbundprojektes „Rückhaltung endlagerrelevanter Radionuklide im natürlichen Tongestein und salinaren Systemen“ (Förderkennzeichen: 02E10981), welches durch das Bundesministerium für Wirtschaft und Energie (BMWi) gefördert wurde, angefertigt. Ziel war es, erstmals Erkenntnisse zur Wechselwirkung zwischen dem Spaltprodukt Technetium und einem natürlichen Tongestein im Hinblick auf ein Endlager für wärmeentwickelnde radioaktive Abfälle zu erlangen. Hierfür wurde der in der Nordschweiz vorkommende Opalinuston aus Mont Terri als Referenzmaterial verwendet. Das Nuklid Technetium-99 liefert auf Grund seiner langen Halbwertszeit einen signifikanten Beitrag zur Radiotoxizität abgebrannter Brennelemente für mehr als tausend Jahre. Im Falle einer Freisetzung aus den Lagerbehältern wird die Geochemie des Technetiums von seiner Oxidationsstufe bestimmt, wobei lediglich die Oxidationsstufen +IV und +VII von Relevanz sind. Auf Grund seiner hohen Löslichkeit und geringen Affinität zur Sorption an Oberflächen von Mineralien ist Tc(VII) die mobilste und somit auch gefährlichste Spezies. Entsprechend lag der Fokus dieser Arbeit auf Diffusionsexperimenten dieser mobilen Spezies sowie auf dem Einfluss von Eisen(II) auf die Mobilität und die Speziation des Technetiums.rnDie Wechselwirkung zwischen Technetium und Opalinuston wurde in Sorptions- und Diffusionsexperimenten unter Variation verschiedener Parameter (pH-Wert, Zusatz verschiedener Reduktionsmittel, Einfluss von Sauerstoff, Diffusionsweg) untersucht. Im Zuge dieser Arbeit wurden erstmals ortsaufgelöste Untersuchungen zur Speziation des Technetiums an Dünnschliffen und Bohrkernen durchgeführt. Dabei konnten ergänzend zur Speziation auch Informationen über die Elementverteilung und die kristallinen Mineralphasen nahe lokaler Anreicherungen des Radionuklides gewonnen werden. Zusätzlich erlaubten Untersuchungen an Pulverproben die Bestimmung der molekularen Struktur des Technetiums an der Tonoberfläche.rnSowohl die Kombination der oben aufgeführten Sorptionsexperimente mit spektroskopischen Untersuchungen als auch die Diffusionsexperimente zeigten unter Sauerstoffausschluss eine Reduktion von Tc(VII) zu immobilen Tc(IV)-Spezies. Weiterhin konnte die Bildung eines Tc(IV)-Sorptionskomplexes an der Tonoberfläche gezeigt werden. Im Hinblick auf ein Endlager in Tongestein sind diese Ergebnisse positiv zu bewerten.

As foundational species, oaks (Quercus : Fagaceae) support the activities of both humans and wildlife. However, many oaks in North America are declining, a crisis exacerbated by the previous disappearance of other hard mast-producing trees. In addition, the economic demands placed on this drought-tolerant group may intensify if climate change extirpates other, relatively mesophytic species. Genetic tools can help address these management challenges. To this end, we developed a suite of 27 microsatellite markers, of which 22 are derived from expressed sequence tags (ESTs). Many of these markers bear significant homology to known genes and may be able to directly assay functional genetic variation. Markers obtained from enriched microsatellite libraries, on the other hand, are typically located in heterochromatic regions and should reflect demographic processes. Considered jointly, genic and genomic microsatellites can elucidate patterns of gene-flow and natural selection, which are fundamental to both an organism's evolutionary ecology and conservation biology. To this end, we employed the developed markers in an FST-based genome scan to detect the signature of divergent selection among the red oaks (Quercus section Lobatae). Three candidate genes with putative roles in stress responses demonstrated patterns of diversity consistent with adaptation to heterogeneous selective pressures. These genes may be important in both local genetic adaptation within species and divergence among them. Next, we used an isolation-with-migration model to quantify levels of gene-flow among four red oaks species during speciation. Both speciation in allopatry and speciation with gene-flow were found to be major drivers of red oak biodiversity. Loci playing a key role in speciation are also likely to be ecologically important within species

Ice ages are known to be the most dominant palaeoclimatic feature occurring on Earth, producing severe climatic oscillations and consequently shaping the distribution and the population structure of several species. Lampreys constitute excellent models to study the colonization of freshwater systems, as they commonly appear in pairs of closely related species of anadromous versus freshwater resident adults, thus having the ability to colonize new habitats, through the anadromous species, and establish freshwater resident derivates. We used 10 microsatellite loci to investigate the spatial structure, patterns of gene flow and migration routes of Lampetra populations in Europe. We sampled 11 populations including the migratory L. fluviatilis and four resident species, L. planeri, L. alavariensis, L. auremensis and L. lusitanica, the last three endemic to the Iberian Peninsula. In this southern glacial refugium almost all sampled populations represent a distinct genetic cluster, showing high levels of allopatric differentiation, reflecting long periods of isolation. As result of their more recent common ancestor, populations from northern Europe are less divergent among them, they are represented by fewer genetic clusters, and there is evidence of strong recent gene flow among populations. These previously glaciated areas from northern Europe may have been colonized from lampreys expanding out of the Iberian refugia. The pair L. fluviatilis/L. planeri is apparently at different stages of speciation in different locations, showing evidences of high reproductive isolation in the southern refugium, and low differentiation in the north.

Leukocytes are critical effectors of inflammation and tumor biology. Chemokine-like factors produced by such inflammatory sites are key mediators of tumor growth that activate leukocytic recruitment and tumor infiltration and suppress immune surveillance. Here we report that the endocrine peptide hormone, relaxin, is a regulator of leukocyte biology with properties important in recruitment to sites of inflammation. This study uses the human monocytic cell line THP-1 and normal human peripheral blood mononuclear cells to define a novel role for relaxin in regulation of leukocyte adhesion and migration. Our studies indicate that relaxin promotes adenylate cyclase activation, substrate adhesion, and migratory capacity of mononuclear leukocytes through a relaxin receptor LGR7-dependent mechanism. Relaxin-stimulated cAMP accumulation was observed to occur primarily in non-adherent cells. Relaxin stimulation results in increased substrate adhesion and increased migratory activity of leukocytes. In addition, relaxin-stimulated substrate adhesion resulted in enhanced chemotaxis to monocyte chemoattractant protein-1. These responses in THP-1 and peripheral blood mononuclear cells are relaxin dose-dependent and proportional to cAMP accumulation.
