Lung cancer is the leading cause of cancer death. However, poor survival using conventional therapies fuel the search for more rational interventions. The objective of this study was to design and implement a 4HPR-radiation interaction model in NSCLC, employing a traditional clinical modality (radiation), a relatively new, therapeutically unexplored agent (4HPR) and rationally combining them based on molecular mechanistic findings pertaining to their interactions. To test the hypothesis that 4HPR sensitizes cells to radiation-induced cell death via G2+M accumulation, we designed a working model consisting of H522 adenocarcinoma cells (p53, K-ras mutated) derived from an NSCLC patient; 4HPR at concentrations up to 10 μM; and X radiation up to 6 Gy generated by a patient-dedicated Phillips RT-250 X ray unit at 250 KV, 15 mA, 1.85 Gy/min. We found that 4HPR produced time- and dose-dependent morphological changes, growth inhibition, and DNA damage-inducing enhancement of reactive oxygen species. A transient G2+M accumulation of cells maximal at 24 h of continuous 4HPR exposure was used for irradiation time scheduling. Our data demonstrated enhanced cell death (both apoptotic and necrotic) in irradiated cells pre-treated with 4HPR versus those with either stressor alone. 4HPR's effect of increased NSCLC cells' radioresponse was confirmed by clonogenic assay. To explore these practical findings from a molecular mechanistic perspective, we further investigated and showed that levels of cyclin B1 and p34cdc2 kinase—both components of the mitosis promoting factor (MPF) regulating the G2/M transition—did not change following 4HPR treatment. Likewise, cdc25C phosphatase was not altered. However, enhanced p34cdc2 phosphorylation on its Thr14Tyr15 residues—indicative of its inactivation and increased expression of MPF negative regulators chk1 and wee1 kinases—were supportive of explaining 4HPR-treated cells' accumulation. Hence, p34cdc2 phosphorylation, chk1, and wee1 warrant further evaluation as potential molecular targets for 4HPR-X radiation combination. In summary, we (1) demonstrated that 4HPR not only induces cell death by itself, but also increases NSCLC cells' subsequent radioresponse, indicative of potential clinical applicability, and (2) for the first time, shed light on deciphering 4HPR-X radiation molecular mechanisms of interaction, including the finding of 4HPR's role as a p34cdc2 inactivator via Thr14Tyr15 phosphorylation. ^

Self-assembled monolayers (SAMs) of alkanethiols on gold surfaces show great promise in controlling the nucleation and growth of inorganic minerals from solution. In doing so, they mimic the role of some biogenic macromolecules in natural biomineralisation processes. Crystallization on SAM surfaces is usually monitored ex-situ; by allowing the process to commence and to evolve for some time, removing the substrate from the mother solution, and then examining it using microscopy, diffraction etc. We present here for the first time, the use of high energy monochromatic synchrotron X-radiation in conjunction with a two dimensional detector to monitor in situ, in a time resolved fashion, the growth of SrCO3 (strontianite) crystals on a SAM substrate.

The effects of irradiation on some members of the family of aromatic polymers with a cardo group, such as polyetherketone with a cardo group (PEK-C) and polyethersulfone with a cardo group (PES-C), were studied. It was found that PEK-C and PES-C can be crosslinked by irradiation under vacuum. Moreover, it was also found that the intensity of the shake-up peak of x-ray photoelectron spectroscopy (XPS) for PEK-C and PES-C varies with irradiation dose. Gelation doses (Rg) of PEK-C and PES-C were estimated from the XPS shake-up peak.

RESUMO - Na radiologia de intervenção, e concretamente em ortopedia, os raios X são intensamente utilizados permitindo a visualização de diversas técnicas de intervenção cirúrgica. Do ponto de vista médico as vantagens dessa prática são enormes, contudo, os profissionais de saúde envolvidos são susceptíveis de estarem expostos a valores de dose de radiação que significam a sua classificação como trabalhadores expostos. O presente estudo realizou-se num hospital e teve como objectivo obter uma estimativa das doses envolvidas em ortopedia de intervenção, utilizando várias metodologias experimentais para caracterização do campo de radiação primário e secundário. Observaram-se distintos níveis de dose de acordo com a zona anatómica exposta à radiação X: (i) gónadas — 0,02 a 3 mGy/h; (ii) cristalino — 0,06 a 1 mGy/h e (iii) mãos — 0,6 mGy/h. Tais resultados evidenciam uma clara necessidade de utilização de equipamentos de protecção e de vigilância dosimétrica pelos profissionais de saúde envolvidos no acto cirúrgico.--------------------------ABSTRACT - In intervention radiology, and more specifically in orthopaedics, X-rays are intensely used allowing the visualization of many acts of clinical intervention. From a clinical perspective, the advantages of that practice are significant; however, involved health care professionals are susceptible of being exposed to radiation dose values that mean their classification as exposed workers. The present study, performed in a hospital, aimed to obtain an estimation of the doses involved in intervention orthopaedics through several experimental methodologies in order to characterise the primary and the secondary radiation fields. Different levels of dose were observed according to the anatomic area exposed to X radiation: (i) gonads — 0.02 a 3 mGy/h; (ii) crystalline lens — 0.06 a 1 mGy/h e (iii) hands – 0.6 mGy/h. In an emergency department (ED), computed tomography (CT) is particularly beneficial in the investigation of high-speed trauma patients. With the advent of multidetector CT (MDCT) scanners, it is becoming faster and easier to conduct scans. In recent years, this has become evident with an increasing number of CT requests. Patients who have multiple CT scans during their hospital stay can receive radiation doses that have an increased theoretical risk of induction of cancer. It is essential that the clinical justification for each CT scan be considered on an individual basis and that due consideration is given to the radiation risk and possible diagnostic benefit. The current lack of a central State or Commonwealth data repository for medical images is a contributing factor to excessive radiation dosage to the population. The principles of justification and radiation risks are discussed in this study.

The authors review the use of photoacoustic and pyroelectric radiation dosimeters. They compare the characteristics and results obtained with a photoacoustic radiation dosimeter (PARD) and a pyroelectric radiation dosimeter (PERD). The PARD and the PERD can be used to measure the energy fluence rate of continuous X-ray beams. In the same way, the single-pulse photoacoustic radiation dosimeter and the single-pulse pyroelectric radiation dosimeter (PPERD) were compared. They can measure the energy fluence of a single pulse of X-radiation. A theoretical model to explain the results obtained with the PPERD is presented and compared with experimental results.

A radiação-x foi descoberta há mais de uma centena de anos e com o avanço tecnológico diversas formas de aplicação têm vindo a ser descobertas. A utilização constante, devido à ajuda preciosa que estes dispositivos fornecem aos clínicos de Medicina Dentária, faz com que o paciente venha, de forma crescente, a ser exposto a este tipo de radiação. Este trabalho de revisão descritiva tem como principal propósito analisar os riscos, as formas de avaliação e de proteção da radiação-x, no âmbito do seu uso em Medicina Dentária. Para o efeito, realizou-se uma revisão da literatura, tendo-se recorrido ao motor de busca EBSCO host©, utilizando as frases boleanas: (X-ray risk OR Radiography risk OR Dental x-ray risk) AND (Dental Medicine OR Oral), foi substituída a palavra risk por “evaluation” e por “protection”. Desta pesquisa resultaram 82 documentos como referências bibliográficas. Esta revisão de literatura permitiu compreender a diferença entre radiação ionizante e não ionizante, quais os dispositivos utlizados na Medicina Dentária e o seu funcionamento geral, quais as unidades de medida da radiação, quais os riscos iminentes à exposição cumulativa e quais as suas consequências biológicas, perceber de que forma a legislação Portuguesa tem evoluído em matéria de radiação ionizante, quais as diversas formas de redução da dose absorvida pelo paciente e quais as medidas de proteção para os profissionais de saúde expostos à radiação ionizante.

The mature dentinoenamel junction (DEJ) is viewed by some investigators and the current authors, not as a fossilized, sharp transition between enamel and dentin, but as a relatively broad structural transition zone including the mantle dentin and the inner aprismatic enamel. In this study, the DEJ structure in bovine incisors was studied with synchrotron microComputed Tomography (microCT) using small cubes cut parallel to the tooth surface. The reconstructions revealed a zone of highly variable punctate contrast between bulk dentin and enamel; the mean linear attenuation coefficients and their standard deviations demonstrated that this zone averaged less mineral than dentin or enamel but had more highly variable structure than either. The region with the punctuate contrast is, therefore, the mantle dentin. The thickness of the mantle dentin seen in a typical data set was about 30 mu m, and the mantle dentin-enamel interface deviated +/- 15 mu m from the average plane over a distance of 520 mu m. In the highest resolution data (similar to 1.5 mu m isotropic voxels, volume elements), tubules in the dentin could be discerned in the vicinity of the DEJ. Contrast sensitivity was high enough to detect differences in mineral content between near-surface and near-DEJ volumes of the enamel. Reconstructions before and after two cubes were compressed to failure revealed cracks formed only in the enamel and did not propagate across the mantle dentin, regardless of whether loading was parallel to or perpendicular to the DEJ. In this work a software developed in the Instituto de Fisica Gleb Wataghin, IFGW, UNICAMP, Campinas, SP, Brazil for obtaining thermal histories using apatite fission track analysis is presented. This software works in Microsoft-Windows environment. It will be freely disposable in the web site of the Departamento de Raios Cosmicos, IFGW, UNICAMP. Thermal histories obtained through this software are compared with those deduced using Monte Trax the software compatible with Apple Macintosh developed by Gallagher. (C) 2001 Elsevier B.V. Ltd.
