Cyclotides are mini-proteins of 28-37 amino acid residues that have the unusual feature of a head-to-tail cyclic backbone surrounding a cystine knot. This molecular architecture gives the cyclotides heightened resistance to thermal, chemical and enzymatic degradation and has prompted investigations into their use as scaffolds in peptide therapeutics. There are now more than 80 reported cyclotide sequences from plants in the families Rubiaceae, Violaceae and Cucurbitaceae, with a wide variety of biological activities observed. However, potentially limiting the development of cyclotide-based therapeutics is a lack of understanding of the mechanism by which these peptides are cyclized in vivo. Until now, no linear versions of cyclotides have been reported, limiting our understanding of the cyclization mechanism. This study reports the discovery of a naturally occurring linear cyclotide, violacin A, from the plant Viola odorata and discusses the implications for in vivo cyclization of peptides. The elucidation of the cDNA clone of violacin A revealed a point mutation that introduces a stop codon, which inhibits the translation of a key Asn residue that is thought to be required for cyclization. The three-dimensional solution structure of violacin A was determined and found to adopt the cystine knot fold of native cyclotides. Enzymatic stability assays on violacin A indicate that despite an increase in the flexibility of the structure relative to cyclic counterparts, the cystine knot preserves the overall stability of the molecule. Cyclotides are a fascinating family of plant-derived peptides characterized by their head-to-tail cyclized backbone and knotted arrangement of three disulfide bonds. This conserved structural architecture, termed the CCK (cyclic cystine knot), is responsible for their exceptional resistance to thermal, chemical and enzymatic degradation. Cyclotides have a variety of biological activities, but their insecticidal activities suggest that their primary function is in plant defence. In the present study, we determined the cyclotide content of the sweet violet Viola odorata, a member of the Violaceae family. We identified 30 cyclotides from the aerial parts and roots of this plant, 13 of which are novel sequences. The new sequences provide information about the natural diversity of cyclotides and the role of particular residues in defining structure and function. As many of the biological activities of cyclotides appear to be associated with membrane interactions, we used haemolytic activity as a marker of bioactivity for a selection of the new cyclotides. The new cyclotides were tested for their ability to resist proteolysis by a range of enzymes and, in common with other cyclotides, were completely resistant to trypsin, pepsin and thermolysin. The results show that while biological activity varies with the sequence, the proteolytic stability of the framework does not, and appears to be an inherent feature of the cyclotide framework. The structure of one of the new cyclotides, cycloviolacin O14, was determined and shown to contain the CCK motif. This study confirms that cyclotides may be regarded as a natural combinatorial template that displays a variety of peptide epitopes most likely targeted to a range of plant pests and pathogens.

Cyclotides, a family of approximately 50 mini-proteins isolated from various Violaceae and Rubiaceae plants, are characterized by their circular peptide backbone and six conserved cysteine residues arranged in a cystine knot motif. Cyclotides show a wide range of biological activities, making them interesting targets for both pharmaceutical and agrochemical research, but little is known about their natural function and the events that trigger their expression. An investigation of the geographical and seasonal variations of cyclotide profiles has been performed, using the native Australian violet, Viola hederacea, and the Swedish sweet violet, Viola odorata, as model plants. The results showed that in the Australian violet the relative peptide levels of some cyclotides remained almost constant throughout the year, while other cyclotides were present only at certain times of the year. Therefore, it appears that V. hederacea expresses a basic armory of cyclotides as well as special add-ons whose levels are influenced by external factors. In the Swedish violet, cyclotide levels were increased up to 14 times during the warmest period of the year. The larger variation in expression levels of the Swedish plants may be a reflection of a greater climatic variation.

High-resolution palynological analysis on annually laminated sediments of Sihailongwan Maar Lake (SHL) provides new insights into the Holocene vegetation and climate dynamics of NE China. The robust chronology of the presented record is based on varve counting and AMS radiocarbon dates from terrestrial plant macro-remains. In addition to the qualitative interpretation of the pollen data, we provide quantitative reconstructions of vegetation and climate based on the method of biomization and weighted averaging partial least squares regression (WA-PLS) technique, respectively. Power spectra were computed to investigate the frequency domain distribution of proxy signals and potential natural periodicities. Pollen assemblages, pollen-derived biome scores and climate variables as well as the cyclicity pattern indicate that NE China experienced significant changes in temperature and moisture conditions during the Holocene. Within the earliest phase of the Holocene, a large-scale reorganization of vegetation occurred, reflecting the reconstructed shift towards higher temperatures and precipitation values and the initial Holocene strengthening and northward expansion of the East Asian summer monsoon (EASM). Afterwards, summer temperatures remain at a high level, whereas the reconstructed precipitation shows an increasing trend until approximately 4000 cal. yr BP. Since 3500 cal. yr BP, temperature and precipitation values decline, indicating moderate cooling and weakening of the EASM. A distinct periodicity of 550-600 years and evidence of a Mid-Holocene transition from a temperature-triggered to a predominantly moisture-triggered climate regime are derived from the power spectra analysis. The results obtained from SHL are largely consistent with other palaeoenvironmental records from NE China, substantiating the regional nature of the reconstructed vegetation and climate patterns. However, the reconstructed climate changes contrast with the moisture evolution recorded in S China and the mid-latitude (semi-)arid regions of N China. Whereas a clear insolation-related trend of monsoon intensity over the Holocene is lacking from the SHL record, variations in the coupled atmosphere-Pacific Ocean system can largely explain the reconstructed changes in NE China.

1. Late glacial and postglacial sediments from three former lakes in the Lake Garda area (Southern Alps) were investigated. 2. The pollen diagram from Bondone (1550 m) shows an older phase rich in NAP. A younger one corresponds with the Younger Dryas time according to two radiocarbon determinations. In the Preboreal no climatic deterioration could be found. 3. At first plants, which are nowadays typical for snow-ground, pioneer and dwarf shrub associations, immigrated into the surroundings of Bondone. In Alleröd times larch and pine appeared as the first trees. At the beginning of the Preboreal dense forest existed in that region. During the Alleröd timber line was at about 1500 m. 4. In the pollen diagrams from Saltarino (194 m) and Fiavè (654 m) an oldest period rich in NAP is followed by two stadial and two interstadial phases. Tree birches and larches immigrated during the oldest interstadial phase. 5. In the case of Saltarino and Fiavè only a preliminary dating could be made. A correlation seems to be possible with diagrams published by Zoller as well as with the diagram of Bondone. Discrepances in dating, which arise then, are discussed. According to the two possibilities of dating the youngest stadial is synchronous either with the so-called Piottino stadial or the Younger Dryas time. Consequently the oldest interstadial phase of Saltarino corresponds either with the Bölling or with a pre-Bölling interstadial. The last possibility seems to be more probable. 6. In the southern part of the Lake Garda area reforestation was preceded by a long shrub phase mainly with Juniperus. At about 650 m there was a period with Pinus mugo and only with a small amount of Juniperus before reforestation. A phase with Betula nana well known from areas north of the Alps could nowhere be found. 7. In the area under study larch appeared as the first tree. Lateron it has been the most important constituent of the forests near timber line. Birch, which plays an important role as a pioneer tree in Denmark - for instance at the transition of the pollen zones III/IV - as well as in Southern Germany during Bölling time, was of less importance at the southern border of the Alps. In that area the spreading of Pinus occurred very early causing dense forests. 8. During the last stadial phase (probably Younger Dryas time) dense forests with Pinus and Larix existed at 650 m. In the lower part of the Lake Garda area, however, both thermophilous trees as Quercus and herbs frequently occurred. This leads to the conclusion that during this time tree growth was limited by dryness in lower altitudes of the border of the Southern Alps. Pinus and Juniperus, however, do not show higher values in this period, a fact which cannot yet be explained. 9. A list of plants, which were found in the sediments, is compiled. Helodium lanatum, Dictamnus albus, Mercurialis cf. ovata, Buxus, Cerinthe cf. minor, Onosma, Anthericum and Asphodelus albus are findings, which are of special interest for the history of the flora of that region.

The cyclotides are the largest family of naturally occurring circular proteins. The mechanism by which the termini of these gene-encoded proteins are linked seamlessly with a peptide bond to form a circular backbone is unknown. Here we report cyclotide-encoding cDNA sequences from the plant Viola odorata and compare them with those from an evolutionarily distinct species, Oldenlandia affinis. Individual members of this multigene family encode one to three mature cyclotide domains. These domains are preceded by N-terminal repeat regions (NTRs) that are conserved within a plant species but not between species. We have structurally characterized peptides corresponding to these NTRs and show that, despite them having no sequence homology, they form a structurally conserved alpha-helical motif. This structural conservation suggests a vital role for the NTR in the in vivo folding, processing, or detoxification of cyclotide domains from the precursor protein.

The performance of visual speech recognition (VSR) systems are significantly influenced by the accuracy of the visual front-end. The current state-of-the-art VSR systems use off-the-shelf face detectors such as Viola- Jones (VJ) which has limited reliability for changes in illumination and head poses. For a VSR system to perform well under these conditions, an accurate visual front end is required. This is an important problem to be solved in many practical implementations of audio visual speech recognition systems, for example in automotive environments for an efficient human-vehicle computer interface. In this paper, we re-examine the current state-of-the-art VSR by comparing off-the-shelf face detectors with the recently developed Fourier Lucas-Kanade (FLK) image alignment technique. A variety of image alignment and visual speech recognition experiments are performed on a clean dataset as well as with a challenging automotive audio-visual speech dataset. Our results indicate that the FLK image alignment technique can significantly outperform off-the shelf face detectors, but requires frequent fine-tuning.

Chromolaena, or Siam weed, is a serious problem in several tropical and sub-tropical areas around the world. In our own region, it is a serious weed in New Guinea, East Timor and Indonesia and is also under an eradication regime in North Queensland. The chapter summarises current knowledge about the taxonomy, biology, distribution, ecology, impacts and control of the weed. Biological control has been a major multinational initiative against this weed in recent years and these efforts are described in some detail. Interestingly agents have not been universally effective because of weed biotype differences and climate. Considerable success has been achieved in New Guinea, principally with the tephritid fly Cecidocares connex and by the efforts of Michael Day, Rachel McFadyen and Graham Donnelly from Alan Fletcher Research Station.
