The role of Vibrio parahaemolyticus in food borne gastroenteritis outbreaks associated primarily with the consumption of contaminated seafoods has been well documented. Information pertaining to various aspects of its occurrence in seafoods, procedures for isolation and identification, generation time and inactivation profiles is discussed. Emphasis has been given to the response of V. parahaemolyticus to low temperatures, heating and antibacterial agents. The public health hazard posed by the pathogen is outlined and the guidelines for control are reviewed in detail.

Qualitative studies on the microflora of slime and guts of prawns and of sea water off Nagapattinam showed the presence of Vibrio in the slime and sea water. They were further tested for Vibrio parahaemolyticus types and related bio-types. Evidence of its occurrence is given. This points to the need for further studies on the distribution of this organism in terms of public health significance.

The strains of Vibrio parahaemolyticus isolated from water, sediment, plankton, fish and prawn of Cochin backwater were tested for sensitivity to ten antibiotics namely, ampicillin, chloramphenicol, gentamycin, kanamycin, neomycin, oxytetracycline, penicillin, polymyxin-B, streptomycin and sulphadiazine. Of the 120 isolates tested, 96.7 and 93.3% were sensitive to gentamycin and chloramphenicol respectively. No strain was sensitive to penicillin and only 5% were sensitive to kanamycin. Isolates from fish and prawn showed higher resistance to ampicillin and none of them was sensitive to kanamycin. Multiple resistant V. parahaemolyticus strains were more in prawn than in other samples.

Seasonal variation of Vibrio parahaemolyticus in fish (Etroplus sauratensis) and prawn (Metapenaeus dobsoni) was monitored from March 1982 to February 1983. Analyses of total viable count, vibrio-like organisms, V. parahaemolyticus like organisms and V. parahaemolyticus showed that they occur more in prawn than in fish. In a more polluted environment, the counts of V. parahaemolyticus associated with fish were found to be higher than in prawn.

A study was conducted in the Cochin area of India to determine the effect of drinking water on Vibrio parahaemolyticus, a bacterium that contaminates fish harvested from marine and estuarine environments. Times of fresh-water exposure required to inactivate these bacteria are given. Findings indicate that the washing of fish and equipment used to handle the fish in drinking water may decrease in the number of viable Vibrio cells and thus aid in prevention of food poisoning.

Grateloupia turuturu, previously known as Grateloupia doryphora, has been widely reported to be an invasive algal species. There are no studies to relate the impact of its existence on its surrounding environment. In this paper, we present our results to show that about 70% of individuals collected from the field could turn Vibrio parahaemolyticus into non-culturable state on both selective (TCBS) and non-selective (2216E) culture medium in 24 h in the presence of light in live algal culture. Total bacteria counts on TCBS and 2216E plates dropped from the initial 565 (174) and 1192 (60) cfu ml(-1) respectively to zero in 24 h. This effect disappeared when the alga was grown in darkness. The same effect was not found in two other intertidal macroalgae Laminaria japonica and Palmaria palmata. Further tests showed that the settlement ability of bacteria in seawater was impaired significantly in the presence of this alga in comparison with three other algal species. (c) 2006 Elsevier B.V. Microbiological studies on the incidence, behaviour, activity and ecological implications of marine micro~organisms, particularly microbial pathogens in coastal waters and estuaries exhibit the increasing concern and awareness of environmental impacts on health and wealth. Marine microbiologists have been active in investigating on the distribution, kinds of organisms and their activity in the environment. However, informations on the effect of environment on the ecology or on the distribution (spatial/temporal) of microbial comunity and competition among groups inhabiting the ecosystem are sparE§L Estuarine environment are complex with respect to diversity of habitats, variation in physicochemical parameters and contamination by terrestrial bacterial species. Being the organisms of‘public health significance, ecological studies on total coliforms, faecal coliforms, faecal streptococci, §. ggli and X. parahaemolyticus have great relevance as studies of these types would provide a wealth of information to environmentalists and to fishery industry. Outbreaks of mass mortality in postlarval abalone, Haliotis diversicolor supertexta (L.), have swept across south China since 2002 and in turn have resulted in many abalone farms closing. Twenty-five representative bacterial isolates were isolated from a sample of five diseased postlarval abalone, taken 15 d postfertilization during an outbreak of postlarval disease in Sanya, Hainan Province, China in October 2004. A dominant isolate, referred to as Strain 6, was found to be highly virulent to postlarvae in an experimental challenge test, with a 50% lethal dose (LD50) value of 3.2 x 10(4) colony forming units (CFU)/mL, while six of the other isolates were weakly virulent with LD50 values ranging from 1 x 10(6) to 1 x 10(7) CFU/mL, and the remaining 18 isolates were classified as avirulent with LD50 values greater than 1 x 10(8) CFU/mL. Using both an API 20E kit and 16S recombinant DNA sequence analysis, Strain 6 was shown to be Vibrio parahaemolyticus. It was sensitive to 4 and intermediately sensitive to 5 of the 16 antibiotics used when screening the antibiotic sensitivities of the bacterium. Extracellular products (ECPs) prepared from the bacterium were lethal to postlarvae when used in a toxicity test at a concentration of 3.77 mg protein/mL, and complete liquefaction of postlarvae tissues occurred within 24 h postexposure. Results from this study implicate V. parahaemolyticus as the pathogen involved in the disease outbreaks in postlarval abalone in Sanya and show that the ECPs may be involved in the pathogenesis of the disease.

This thesis deals initially with a literature reference survey ,taxonomy, their incidence in selected food fishes and shellfishes, and their incidence and distribution, their survival during different types of processing, their heat survival at temperatures of 50 ,55 and 60 degree centigrade their growth initiation at different low levels of pHs(4.0 to 10) ,and their developmental resistance to various chemical agents. The trials for the study were collected from various landing centre at cochin and the retail outlets. Based on these data collections the researcher was able to obtain more knowledge of the processing technology and the survival of pathogens like salmonella and vibrio parahaemolyticus.

In India much work has been done on different aspects of quality of processed fish and fishery products (Pillai et al., 1965; Iyer and Chaudhuri, 1966; Iyer et al., 1966; Sreenivasan and Joseph, 1966; Pillai and Rao, 1969; Mathen et al., 1975; Valsan et al., 1985) and standards have been formulated for almost all the fishery products. Quality standards for fresh fish cover only a few fish species available in the markets. For the formulation and recommendation of quality standards for fish and fishery products for domestic trade detailed background informations are necessary. Khot et al., (1982), Valsan et al. J (1985) and Iyer et al. (1986) have reported on the bacterial flora of sea foods at retail level in Bombay. Cochin is a major fish landing centre of the south west coast of India, and an average of 25,000. tonnes of fish/shell fish are landed annually at this harbour which is about 10 percent of the total catch of fish in Kerala (Lakshmanan -et -al., 1984). All the varieties of marine, brackish water and fresh water fishes are available in Cochln throughout the year for catering to Its cosmopolitan. population and informations regarding their bacteriological quality are scanty. Hence the present study was undertaken to investigate the bacteriological quality of fish and fishery products available in the markets and cold storages situated in and around Cochin meant for internal consumption.

The incidence of various human pathogenic bacteria in commercially available and home-made shrimp feeds used on some farms in India was analyzed. The Total Heterotrophic Bacteria in the commercial feed samples ranged between 103–105 cfu g-1 and those in the farm-made feeds between 106-107 cfu g-1. No bacteria of significance to human health were found to be associated with any of the commercial feed samples analyzed, while farm-made feeds analyzed during the study showed a high incidence of various human pathogens such as Vibrio parahaemolyticus, V. cholerae, Escherichia coli and Staphylococcus aureus. Possible modes of contamination in feeds and ways to prevent them are discussed.

The interaction between microorganisms and host defense mechanisms is a decisive factor for the survival of marine bivalves. They rely on cell-mediated and humoral reactions to overcome the pathogens that naturally occur in the marine environment. In order to understand host defense reactions in animals inhabiting extreme environments we investigated some of the components from the immune system of the deep sea hydrothermal vent mussel Bathymodiolus azoricus. Cellular constituents in the hemolymph and extrapallial fluid were examined and led to the identification of three types of hemocytes revealing the granulocytes as the most abundant type of cell. To further characterize hemocyte types, the presence of cell surface carbohydrate epitopes was demonstrated with fluorescent WGA lectin, which was mostly ascribed to the granulocytes. Cellular reactions were then investigated by means of phagocytosis and by the activation of putative MAPKs using the microbial compounds zymosan, glucan, peptidoglycan and lipopolysaccharide. Two bacterial agents, Bacillus subtilis and Vibrio parahaemolyticus, were also used to stimulate hemocytes. The results showed that granulocytes were the main phagocytic cells in both hemolymph and extrapallial fluid of B. azoricus. Western blotting analyses using commercially available antibodies against ERK, p38 and JNK, suggested that these putative kinases are involved in signal transduction pathways during experimental stimulation of B. azoricus hemocytes. The fluorescent Ca2+ indicator Fura-2 AM was also insightful in demonstrating hemocyte stimulation in the presence of laminarin or live V. parahaemolyticus. Finally, the expression of the antibacterial gene mytilin was analyzed in gill tissues by means of RT-PCR and whole-mount in situ hybridization. Mytilin transcripts were localized in hemocytes underlying gill epithelium. Moreover, mytilin was induced by exposure of live animals to V. parahaemolyticus. These findings support the premise of a conserved innate immune system in B. azoricus. Such system is comparable to other Bivalves and involves the participation of cellular and humoral components. © 2008 Elsevier Inc. All rights reserved.

Microcosm studies were performed to evaluate the survival of Escherichia coli, Salmonella paratyphi and Vibrio parahaemolyticus in water and sediment collected from the freshwater region of Vembanad Lake (9 35◦N 76 25◦E) along the south west coast of India. All three test microorganisms showed significantly (p < 0.01) higher survival in sediment compared to overlying water. The survival in different sediment types with different particle size and organic carbon content revealed that sediment with small particle size and high organic carbon content could enhance their extended survival (p < 0.05). The results indicate that sediments of the Lake could act as a reservoir of pathogenic bacteria and exhibit a potential health hazard from possible resuspension and subsequent ingestion during recreational activities. Therefore, the assessment of bacterial concentration in freshwater Lake sediments used for contact and non contact recreation has of considerable significance for the proper assessment of microbial pollution of the overlying water, and for the management and protection of related health risk at specific recreational sites. Besides, assessment of the bacterial concentration in sediments can be used as a relatively stable indicator of long term mean bacterial concentration in the water column above

Prevalence of faecal indicator bacteria, Escherichia coli and pathogenic bacteria, Vibrio cholerae, Vibrio parahaemolyticus and Salmonella were analysed in Vembanadu lake (98350N 768250E), along south west coast of India for a period of one year from ten stations on the southern and northern sides of a salt water regulator constructed in Vembanadu Lake in order to prevent incursion of seawater during certain periods of the year. While the northern side of the lake has a connection to the sea, the southern side is enclosed when the salt water regulator is closed. The results revealed the water body is polluted with high faecal coliform bacteria with mean MPN value ranging from 1718-7706/100 ml. E. coli, V. cholerae, V. parahaemolyticus and Salmonella serotypes such as S. paratyphi A, B, C and S. newport were isolated and this is the first report on the isolation of these Salmonella serovars from this lake. E. coli showed highest percentage of incidence (85.6–86.7%) followed by Salmonella (42–57%), V. choleare (40–45%) and V. parahaemolyticus (31.5–32%). The increased prevalence of indicator and pathogenic bacteria in the enclosed southern part of Vembanadu Lake may be resulting from the altered flow patterns due to the salt water regulator.

Prevalence of faecal coliform bacteria and the survival of Escherichia coli, Vibrio parahaemolyticus and Salmonella paratyphi were studied in the water and sediment from Vembanadu Lake in the presence and absence of protozoan predators. The density of faecal coliform bacteria ranged between mean MPN value 5080–9000/100 ml in water and 110,000–988,000/1 g in sediment (p <0.01), which was 110 times greater than in overlying water. The laboratory microcosm studies revealed that E. coli, V. parahaemolyticus and S. paratyphi showed significantly higher survival (p <0.05) potential in sediment than in overlying water both in the presence and absence of protozoan predators. The results indicate that Vembanadu Lake sediment constitutes a reservoir of pathogenic bacteria and exhibits potential health hazard from possible resuspension and subsequent ingestion during recreational activities. Therefore, assessment of bacterial concentration in freshwater lake sediments used for contact and non-contact recreation is of considerable significance for the proper assessment of microbial pollution of the overlying water and the management and protection of related health risk at specific recreational sites. In addition, assessment of the bacterial concentration in sediments can be used as a relatively stable indicator of long-term mean bacterial concentration in the water column above.
