Olfaction, the sense of smell, has many important functions in humans. Human responses to odors show substantial individual variation. Olfactory receptor genes have been identified and other genes may also influence olfaction. However, the proportion of phenotypic variation in odor response due to genetic variation remains largely unknown. Little is also known about which genes modify specific responses to odors. This study aimed to elucidate genetic and environmental influences on human responses to odors. Individuals from Finnish families (n=146) and Australian (n=413), British (n=163), Danish (n=336), and Finnish (n=399) twins rated intensity and pleasantness of a set of 12 (families) or 6 (twins) odors and tried to identify the odors. In addition, the participants rated their own sense of smell and annoyance experienced with different environmental odors. The odor stimuli of a commercial smell test (The Brief Smell Identification Test; banana, chocolate, cinnamon, gasoline, lemon, onion, paint thinner, pineapple, rose, smoke, soap, and turpentine) were presented in the family study. Based on the results of the family study and a literature survey, a new set of odor stimuli (androstenone, chocolate, cinnamon, isovaleric acid, lemon, and turpentine) was designed for the twin studies. In the family sample, heritabilities of the traits were estimated and underlying genomic regions were searched using a genome-wide linkage scan. In the pooled twin sample, variation in the measured traits was decomposed into genetic and environmental components using quantitative genetic modeling. In addition, associations between nongenetic factors (e.g., sex, age, and smoking) and olfactory-related traits were explored. Suggestive evidence for a genetic linkage for pleasantness of cinnamon at a locus on chromosome 4q32.3 emerged from the family sample. High heritability for the pleasantness of cinnamon was found in the family but not the twin study. Heritability of perceived intensity of androstenone odor was determined to be ~30% in the twin sample. A strong genetic correlation between perceived intensity and pleasantness of androstenone, in the absence of any environmental correlation, indicated that only the genetic correlation explained the phenotypic correlation between the traits (r=-0.27) and that the traits were influenced by an overlapping set of genes. Self-rated olfactory function appeared to reflect the odor annoyance experienced rather than actual olfactory acuity or genetic involvement. Results from nongenetic analyses supported the speculated superiority of females' olfactory abilities, the age-related diminishing of olfactory acuity, and the influences of experience-dependent factors on odor responses. This was the first study to estimate heritabilities and perform linkage screens for individual odors. A genetic effect was detected for only a few responses to specific odors, suggesting the predominance of environmental effects in odor perceptions.

Los pinos son afectados por plagas y e nfermedades debido a diferentes factores como: incendios forestales, periodos de sequías prolongados, el manejo inadecuado del bosque y la falta de aplicación de prácticas silviculturales. Uno de los insectos que más se reporta atacando los pinos es el gor gojo descortezador del pino Dendroctonus frontalis. Ataques por este insecto fueron reportados en el departamento de Nueva Segovia en el periodo comprendido entre 1999 al 2001. D. frontalis afectó aproximadamente 32,359.41 ha. de bosques equivalentes al 50 % del área boscosa de Nueva Segovia y unos 3 millones m 3 de madera en rollo. Los daños estimados por este ataque se aproximan a los 39 millones de dólares por pérdida de madera en pie y 56 millones de dólares por costo de oportunidad y un cuantioso daño a mbiental. Ante tal situación y debido a la importancia que tiene el recurso forestal en Nicaragua, se realizó una investigación con el objetivo de describir la fluctuación poblacional de todos los insectos asociados a los pinos y sus enemigos naturales. El estudio se realizó en el Departamento de Nueva Segovia, específicamente en los Municipios de Dipilto y El Jicaro en el periodo comprendido entre Febrero 2004 a Enero 2005. El monitoreo se realizó cada 15 días y se utilizaron trampas del tipo “Lindgren non sticky funnel trap®” de doce embudos con vaso colector, llevando adherido un paquete de feromona sintetica (frontalin ®) y un depósito de aguarrás (pine turpentine). Los resultados obtenidos fueron los siguientes. Se describió la fluctuación poblacional d e insectos de la familia Scolytidae. Las poblaciones de insectos de esta familia fueron estadísticamente similares en ambos municipios. También, se describió la fluctuación poblacional de insectos del Orden Coleóptera y se les identificó hasta el nivel de familia. El número de insectos de la familia Curculionidae, Cerambycidae, Scarabaeidae, Coccinellidae fueron mayores estadísticamente en Dipilto que en El Jicaro. Por el contrario, los insectos de la familia Buprestidae y Elateridae fueron mayores en El Ji caro en comparación con el municipio de Dipilto. El número de insectos de las familias Chrysomelidae, Carabidae, Nitidullidae, Lampiridae, Tenebrionidae fueron estadísticamente similares en ambos municipios. En este mismo estudio, se describió la fluctuaci ón poblacional de insectos depredadores encontrados en el bosque de pino. Los insectos de la familia Staphylinidae fueron estadísticamente mayores en Dipilto que en El Jicaro, por el contrario las poblaciones de insectos Trogostidae, Histeridae y Cucujidae fueron estadísticamente similares en Dipilto y El Jicaro. Es importante señalar que en comparación con los otros depredadores, la familia Trogostidae se encontró en mayor númer o a lo largo de todo el estudio

This report is the second in a series from a project to assess land-based sources of pollution (LBSP) and effects in the St. Thomas East End Reserves (STEER) in St. Thomas, USVI, and is the result of a collaborative effort between NOAA’s National Centers for Coastal Ocean Science, the USVI Department of Planning and Natural Resources, the University of the Virgin Islands, and The Nature Conservancy. Passive water samplers (POCIS) were deployed in the STEER in February 2012. Developed by the US Geological Survey (USGS) as a tool to detect the presence of water soluble contaminants in the environment, POCIS samplers were deployed in the STEER at five locations. In addition to the February 2012 deployment, the results from an earlier POCIS deployment in May 2010 in Turpentine Gut, a perennial freshwater stream which drains to the STEER, are also reported. A total of 26 stormwater contaminants were detected at least once during the February 2012 deployment in the STEER. Detections were high enough to estimate ambient water concentrations for nine contaminants using USGS sampling rate values. From the May 2010 deployment in Turpentine Gut, 31 stormwater contaminants were detected, and ambient water concentrations could be estimated for 17 compounds. Ambient water concentrations were estimated for a number of contaminants including the detergent/surfactant metabolite 4-tert-octylphenol, phthalate ester plasticizers DEHP and DEP, bromoform, personal care products including menthol, indole, n,n-diethyltoluamide (DEET), along with the animal/plant sterol cholesterol, and the plant sterol beta-sitosterol. Only DEHP appeared to have exceeded a water quality guideline for the protection of aquatic organisms.

Le CS fait partie de la famille des SYSADOA (SYmptomatic Slow Acting Drugs for OsteoArthritis) et est utilisé par les patients avec de l’ostéoarthrose de façon chronique pour ses propriétés anti-inflammatoires. Étant donné que ces patients reçoivent d’autres médicaments, il était intéressant de documenter les effets du CS sur le cytochrome P450 et la NADPH-réductase (NADPH). Pour cette étude, deux modèles ont été utilisés: des lapins témoins (LT) et des lapins avec une réaction inflammatoire (LRI) afin de diminuer l’activité et l’expression du CYP. Six groupes contenant chacun cinq lapins ont été utilisés: un groupe sans CS et deux groupes qui ont pris oralement dans l’eau approximativement 20.5 mg/kg/jour de CS pendant 20 et 30 jours; les lapins des trois groupes restants ont pris du CS comme décrit plus haut, mais ont reçu 5 ml sous-cutanées de térébenthine afin de produire une réaction inflammatoire aseptique (RIA) deux jours avant leur sacrifice, c’est-à-dire aux jours -2, 18 et 28. Les hépatocytes ont été isolés pour évaluer l’activité et l’expression du CYP3A6, CYP1A2 et NADPH et aussi le ARNm de ces protéines. In vitro, nous avons étudié l’effet de différentes concentrations de CS-disaccharides sulfatés, 4S, 6S, et 4,6S de CS, sur l’activité et l’expression du CYP1A2 et du CYP3A6. Pour documenter la présence de la réaction inflammatoire, nous avons mesure les mucoprotéines, dans le sérum des lapins avec une réaction inflammatoire. Aussi nous avons mesuré la présence de l’oxide nitrique (NO) chez les hépatocytes de lapins contrôles et chez les hépatocytes des lapins avec une réaction inflammatoire. La translocation nucléaire du NF-κB a été etudiée par fluorescence chez les hépatocytes. Par comparaison aux lapins témoins, l’administration du CS pendant 20 et 30 jours n’affecte pas l’activité du CYP3A6 et du CYP1A2. La RIA a augmenté les mucoprotéines à 95,1±5,7 vs 8,4±1,6 mg/dl dans les lapins témoins (p<0,05). La RIA a diminué l’activité du CYP3A6 de 62% et l’activité du CYP 1A2 de 54%. Le CS n’empêché pas la diminution du CYP1A2 produite par la RIA. Par ailleurs, le CS n’affecte pas l’activité ni l’expression de la NADPH. La translocation nucléaire de NF-κB a été empêche par l’administration chronique de CS aux lapins avec RIA; en plus, la concentration de l’oxide nitrique n’a pas démontré une augmentation en présence de CS; par contre, CS n’empêche pas l’augmentation des séromucoïdes. Au contraire, CS affecte la diminution du CYP3A6 en fonction de temps et secondaire à la RIA. Dans ce group, CS a rétabli le niveau des protéines du CYP3A6 observé dans le group de lapins témoins. Pourtant cette croissance été independante de mRNA qui garde un niveau trés bas. Le plus remarcable a été la manière dont CS a augmenté la protéine du CYP3A6, sans avoir rétabli l’activité de cet isoforme. Finalement, in vitro, CS et ses trois disaccharides sulfatés (4S, 6S et 4,6S) n’affectent ni l’activité ni l’expression de CYP1A2, CYP3A6 et de la NADPH. En conclusion, l’administration chronique de CS n’affecte pas l’activité ni l’expression du CYP1A2, ou la diminution du CYP1A2 produite par la réaction inflammatoire. Le CS n’affecte pas l’activité ni l’expression du NADPH. Cependant, CS empêche la diminution du CYP3A6 en fonction de temps et secondaire à la RIA.

L’activité catalytique du cytochrome P450 dépend de la disponibilité d’électrons produits par la NADPH P450 réductase (NPR). Notre étude a pour but de déterminer comment l’expression de la NPR est modulée chez le lapin. Afin de comprendre comment l’expression de la NPR est modulée, des hépatocytes de lapins témoins ont été incubés pendant 2, 4, 24 et 48 heures en présence de plusieurs activateurs de facteurs de transcription connus du cytochrome P450. De plus, des lapins ayant reçu une injection sous-cutanée de térébenthine afin de produire une réaction inflammatoire aseptique sont sacrifiés 48 heures plus tard dans le but d’étudier les effets de l’inflammation sur l’expression de la NPR. La rosiglitazone, le fénofibrate, l’acétate de plomb et le chlorure de cobalt (des inducteurs des PPAR, PPAR, AP-1 et HIF-1), après 48 heures d’incubation, n’ont provoqué aucun changement d’expression ou d’activité de la NPR. Après 48 heures d’incubation, la dexaméthasone (Dexa) a augmenté la quantité d’ARNm (QT-PCR), l’expression et l’activité de la NPR (p<0,05), en plus d’augmenter l’ARNm des récepteurs nucléaires CAR (récepteur constitutif à l’androstane) et PXR (récepteur X prégnane) (p<0.05). Le phénobarbital (PB) a augmenté seulement l’activité de la NPR (p<0.05). Par contre, après 48 heures d’incubation, la combinaison PB et Dexa a augmenté la quantité d’ARNm, ainsi que l’expression et l’activité de la NPR (p<0.05). La combinaison de PB et Dexa a induit une augmentation d’ARNm des récepteurs nucléaires CAR, PXR et RXR (récepteur X du rétinoïde) plus précocement, soit après 2 heures d’incubation (p<0.05). Le PD098059 (PD), un bloqueur de l’activation de MAPK1 (mitogen-activated protein kinase), et l’acide okadaïque (OA), un inhibiteur de la protéine phosphatase 2A (PP2A), ont bloqué l'augmentation d'expression et d'activité de la NPR induite par le PB après 48 heures d’incubation. La réaction inflammatoire aseptique a diminué l’expression et l’activité de la NPR après 48 heures d’incubation (p<0.05). On conclue que la dexaméthasone et le phénobarbital sont des inducteurs potentiels de la NPR et que les voies de signalisation de CAR, PXR et RXR semblent être impliquées dans le contrôle de cette induction. Des études supplémentaires devront être complétées afin de confirmer ces résultats préliminaires.

The present work had as objective to apply an experimental planning aiming at to improve the efficiency of separation of a new type of mixer-settler applied to treat waste water contaminated with oil. An unity in scale of laboratory, was installed in the Post-graduation Program of Chemical Engineering of UFRN. It was constructed in partnership with Petrobras S.A. This called device Misturador-Decantador a Inversão de Fases (MDIF) , possess features of conventional mixer-settler and spray column type. The equipment is composed of three main parts: mixing chamber; chamber of decantation and chamber of separation. The efficiency of separation is evaluated analyzing the oil concentrations in water in the feed and the output of the device. For the analysis one used the gravimetric method of oil and greases analysis (TOG). The system in study is a water of formation emulsified with oil. The used extractant is a mixture of Turpentine spirit hydro-carbons, supplied for Petrobras. It was applied, for otimization of the efficiency of separation of the equipment, an experimental planning of the composite central type, having as factorial portion fractionary factorial planning 2 5-2, with the magnifying of the type star and five replications in the central point. In this work, the following independents variables were studied: contents of oil in the feed of the device; volumetric ratio (O/A); total flowrate ; agitation in the mixing chamber and height of the organic bed. Minimum and maximum limits for the studied variables had been fixed according previous works. The analysis of variance for the equation of the empirical model, revealed statistically significant and useful results for predictions ends. The variance analysis also presented the distribution of the error as a normal distribution and was observed that as the dispersions do not depend on the levels of the factors, the independence assumption can be verified. The variation around the average is explained by 98.98%, or either, equal to the maximum value, being the smoothing of the model in relation to the experimental points of 0,98981. The results present a strong interaction between the variable oil contents in the feed and agitation in the mixing chamber, having great and positive influence in the separation efficiency. Another variable that presented a great positive influence was the height of the organic bed. The best results of separation efficiency had been obtained for high flowrates when associates the high oil concentrations and high agitation. The results of the present work had shown excellent agreement with the results carried out through previous works with the mixer-settler of phase inversion

During natural gas processing, water removal is considered as a fundamental step in that combination of hydrocarbons and water favors the formation of hydrates. The gas produced in the Potiguar Basin (Brazil) presents high water content (approximately 15000 ppm) and its dehydration is achieved via absorption and adsorption operations. This process is carried out at the Gas Treatment Unit (GTU) in Guamaré (GMR), in the State of Rio Grande do Norte. However, it is a costly process, which does not provide satisfactory results when water contents as low as 0.5 ppm are required as the exit of the GTU. In view of this, microemulsions research is regarded as an alternative to natural gas dehydration activities. Microemulsions can be used as desiccant fluids because of their unique proprieties, namely solubilization enhancement, reduction in interfacial tensions and large interfacial area between continuous and dispersed phases. These are actually important parameters to ensure the efficiency of an absorption column. In this work, the formulation of the desiccant fluid was determined via phases diagram construction, employing there nonionic surfactants (RDG 60, UNTL L60 and AMD 60) and a nonpolar fluid provided by Petrobras GMR (Brazil) typically comprising low-molecular weight liquid hydrocarbons ( a solvent commonly know as aguarrás ). From the array of phases diagrams built, four representative formulations have been selected for providing better results: 30% RDG 60-70% aguarrás; 15% RDG 60-15% AMD 60-70% aguarrás, 30% UNTL L60-70% aguarrás, 15% UNTL L60-15% AMD 60-70% aguarrás. Since commercial natural gas is already processed, and therefore dehydrated, it was necessary to moister some sample prior to all assays. It was then allowed to cool down to 13ºC and interacted with wet 8-12 mesh 4A molecular sieve, thus enabling the generation of gas samples with water content (approximately 15000 ppm). The determination of the equilibrium curves was performed based on the dynamic method, which stagnated liquid phase and gas phase at a flow rate of 200 mL min-1. The hydrodynamic study was done with the aim of established the pressure drop and dynamic liquid hold-up. This investigation allowed are to set the working flow rates at 840 mL min-1 for the gas phase and 600 mLmin-1 for the liquid phase. Grand fir (Abies grandis Lindl.) has been developed as a model system for the study of wound-induced oleoresinosis in conifers as a response to insect attack. Oleoresin is a roughly equal mixture of turpentine (85% monoterpenes [C10] and 15% sesquiterpenes [C15]) and rosin (diterpene [C20] resin acids) that acts to seal wounds and is toxic to both invading insects and their pathogenic fungal symbionts. The dynamic regulation of wound-induced oleoresin formation was studied over 29 d at the enzyme level by in vitro assay of the three classes of synthases directly responsible for the formation of monoterpenes, sesquiterpenes, and diterpenes from the corresponding C10, C15, and C20 prenyl diphosphate precursors, and at the gene level by RNA-blot hybridization using terpene synthase class-directed DNA probes. In overall appearance, the shapes of the time-course curves for all classes of synthase activities are similar, suggesting coordinate formation of all of the terpenoid types. However, closer inspection indicates that the monoterpene synthases arise earlier, as shown by an abbreviated time course over 6 to 48 h. RNA-blot analyses indicated that the genes for all three classes of enzymes are transcriptionally activated in response to wounding, with the monoterpene synthases up-regulated first (transcripts detectable 2 h after wounding), in agreement with the results of cell-free assays of monoterpene synthase activity, followed by the coordinately regulated sesquiterpene synthases and diterpene synthases (transcription beginning on d 3–4). The differential timing in the production of oleoresin components of this defense response is consistent with the immediate formation of monoterpenes to act as insect toxins and their later generation at solvent levels for the mobilization of resin acids responsible for wound sealing.

We report that promoters for two murine acute-phase protein (APP) genes, complement factor 3 (C3) and serum amyloid A3 (SAA3), can increase recombinant protein expression in response to inflammatory stimuli in vivo. To deliver APP promoter-luciferase reporter gene constructs to the liver, where most endogenous APP synthesis occurs, we introduced them into a nonreplicating adenovirus vector and injected the purified viruses intravenously into mice. When compared with the low levels of basal luciferase expression observed prior to inflammatory challenge, markedly increased expression from the C3 promoter was detected in liver in response to both lipopolysaccharide (LPS) and turpentine, and lower-level inducible expression was also found in lung. In contrast, expression from the SAA3 promoter was found only in liver and was much more responsive to LPS than to turpentine. After LPS challenge, hepatic luciferase expression increased rapidly and in proportion to the LPS dose. Use of cytokine-inducible promoters in gene transfer vectors may make it possible to produce antiinflammatory proteins in vivo in direct relationship to the intensity and duration of an individual's inflammatory response. By providing endogenously controlled production of recombinant antiinflammatory proteins, this approach might limit the severity of the inflammatory response without interfering with the beneficial components of host defense and immunity.

Grand fir (Abies grandis) saplings and derived cell cultures are useful systems for studying the regulation of defensive oleoresinosis in conifers, a process involving both the constitutive accumulation of resin (pitch) in specialized secretory structures and the induced production of monoterpene olefins (turpentine) and diterpene resin acids (rosin) by nonspecialized cells at the site of injury. The pathways and enzymes involved in monoterpene and diterpene resin acid biosynthesis are described, as are the coinduction kinetics following stem injury as determined by resin analysis, enzyme activity measurements, and immunoblotting. The effects of seasonal development, light deprivation, and water stress on constitutive and wound-induced oleoresinosis are reported. Future efforts, including a PCR-based cloning strategy, to define signal transduction in the wound response and the resulting gene activation processes are delineated.

Contains instructions for preparing and administering medicine for adults and children, and generalized uses for certain ingredients, written by Dr. Francis Kittredge. Preparations include ointment for scurvy, bone ointment, nerve ointments, procedures to soothe a sore mouth and to stop excessive bleeding, and treatment to kill worms. The materials used to prepare bone ointment include fresh butter, hog fat, chamomile, garlic, and night shade, among other ingredients. The recipe for “simple nerve ointment” instructs the preparer to simmer half a pint of neet foot oil, a pint of rum, and one jell of oil of turpentine over a “gentle fire.” Kittredge writes that oil of St. John’s Wort is effective in treating swelling of the legs, for cold and aches, and for burning and scalds, while oil of Elderflower is indicated for belly aches. The manuscript is housed in a binding created by the Harvard Medical School library. Tipped into the binding is one letter from Frederick O. West, M.D., Harvard Medical School, Boston, Massachusetts, that accompanied his donation of the Kittredge receipt book to the library in 1919. There is also one letter of unknown provenance enclosed with the receipt book, which contains an inventory of the estate of Antipas Brigham, of Grafton, Massachusetts, signed by Worcester County Judge Joseph Wilder on 7 November 1749. It is unclear if this letter has any connection to Frederick O. West or Francis Kittredge.
