Chitosan-DNA nanoparticles employed in gene therapy protocols consist of a neutralised, stoichiometric core and a shell of the excess of chitosan which stabilises the particles against further coagulation. At low ionic strength, these nanoparticles possess a high stability; however, as the ionic strength increases, it weakens the electrostatic repulsion which can play a decisive part in the formation of highly aggregated particles. In this study, new results about the effect of ionic strength on the colloidal stability of chitosan-DNA nanoparticles were obtained by studying the interaction between chitosans of increasing molecular weights (5, 10, 16, 29, 57 and 150 kDa) and calf thymus DNA. The physicochemical properties of polyplexes were investigated by means of dynamic light scattering, static fluorescence spectroscopy, optic microscopy, transmission electronic microscopy and gel electrophoresis. After subsequent addition of salt to the nanoparticles solution, secondary aggregation increased the size of the polyplexes. The nanoparticles stability decreased drastically at the ionic strengths 150 and 500 mM, which caused the corresponding decrease in the thickness of the stabilising shell. The morphologies of chitosan/DNA nanoparticles at those ionic strengths were a mixture of large spherical aggregates, toroids and rods. The results indicated that to obtain stable chitosan-DNA nanoparticles, besides molecular weight and N/P ratio, it is quite important to control the ionic strength of the solution. © 2013 Copyright Taylor and Francis Group, LLC.

In this work, mesoporous titania is prepared by templating latex sphere arrays with four different sphere diameters at the micrometric scale (phi > 1 mu m). The mesoporous titania homogeneously covers the latex spheres and substrate, forming a thin coating characterized by N-2 adsorption isotherm, small angle X-rays scattering, atomic force, field emission and transmission electronic microscopies. Mesoporous titania has been templated into different shapes such as hollow particles and monoliths according to the amount of sol used to fill the voids of the close packed latex spheres. Titania topography strongly depends on the adsorption of polymeric segments over latex spheres surface, which could be decreased by changing the dimensions of latex spheres (phi = 9.5 mu m) generating a lamellar architecture. Thus, micrometric latex sphere arrays can be used to achieve new surface patterns for mesoporous materials via a fast and inexpensive chemical route for construction of functional devices in different technological fields such as energy conversion, inclusion chemistry and biomaterials. Black flies are opportunistic sugar-feeders. They take sugar meals from Homopteran honeydew secretions or plant nectars, depending on availability. Homopteran honeydew secretions contain both simple and complex carbohydrates while plant nectars contain primarily simple carbohydrates. In order to determine whether honeydew secretions offer more energy than plant nectars to their insect visitors a study of wild-caught black flies was undertaken in Algonquin Provincial Park, Canada during the spring of 1 998 and 1 999. It was hypothesized that female black flies maintained on honeydew sugars will survive longer, produce more eggs and have a greater parasite vectoring potential than those maintained on artificial nectar or distilled water. Results demonstrated that: (1) host-seeking female Prosimulimfuscum/mixtum and Simulium venustum maintained on artificial honeydew did not survive longer than those maintained on artificial nectar when fed ad libitum; (2) fiiUy engorged S. venustum and Simulium rugglesi maintained on artificial honeydew did not produce more eggs than those maintained on artificial nectar when fed ad libitum; and (3) S. rugglesi did not have a greater vectoring potential of Leucocytozoon simondi when maintained on artificial honeydew as opposed to artificial nectar when fed ad libitum. However, all flies maintained on the two sugars (artificial honeydew and artificial nectar) survived longer, produce more eggs and had greater vectoring potential than those maintained on distilled water alone.

Beaucoup d'efforts dans le domaine des matériaux polymères sont déployés pour développer de nouveaux matériaux fonctionnels pour des applications spécifiques, souvent très sophistiquées, en employant des méthodes simplifiées de synthèse et de préparation. Cette thèse porte sur les polymères photosensibles – i.e. des matériaux fonctionnels qui répondent de diverses manières à la lumière – qui sont préparés à l'aide de la chimie supramoléculaire – i.e. une méthode de préparation qui repose sur l'auto-assemblage spontané de motifs moléculaires plus simples via des interactions non covalentes pour former le matériau final désiré. Deux types de matériaux photosensibles ont été ciblés, à savoir les élastomères thermoplastiques à base de copolymères à blocs (TPE) et les complexes d'homopolymères photosensibles. Les TPEs sont des matériaux bien connus, et même commercialisés, qui sont généralement composés d’un copolymère tribloc, avec un bloc central très flexible et des blocs terminaux rigides qui présentent une séparation de phase menant à des domaines durs isolés, composés des blocs terminaux rigides, dans une matrice molle formée du bloc central flexible, et ils ont l'avantage d'être recyclable. Pour la première fois, au meilleur de notre connaissance, nous avons préparé ces matériaux avec des propriétés photosensibles, basé sur la complexation supramoléculaire entre un copolymère tribloc simple parent et une petite molécule possédant une fonctionnalité photosensible via un groupe azobenzène. Plus précisément, il s’agit de la complexation ionique entre la forme quaternisée d'un copolymère à blocs, le poly(méthacrylate de diméthylaminoéthyle)-poly(acrylate de n-butyle)-poly(méthacrylate de diméthylaminoéthyle) (PDM-PnBA-PDM), synthétisé par polymérisation radicalaire par transfert d’atomes (ATRP), et l'orange de méthyle (MO), un composé azo disponible commercialement comportant un groupement SO3 -. Le PnBA possède une température de transition vitreuse en dessous de la température ambiante (-46 °C) et les blocs terminaux de PDM complexés avec le MO ont une température de transition vitreuse élevée (140-180 °C, en fonction de la masse molaire). Des tests simples d'élasticité montrent que les copolymères à blocs complexés avec des fractions massiques allant de 20 à 30% présentent un caractère élastomère. Des mesures d’AFM et de TEM (microscopie à force atomique et électronique à ii transmission) de films préparés à l’aide de la méthode de la tournette, montrent une corrélation entre le caractère élastomère et les morphologies où les blocs rigides forment une phase minoritaire dispersée (domaines sphériques ou cylindriques courts). Une phase dure continue (morphologie inversée) est observée pour une fraction massique en blocs rigides d'environ 37%, ce qui est beaucoup plus faible que celle observée pour les copolymères à blocs neutres, dû aux interactions ioniques. La réversibilité de la photoisomérisation a été démontrée pour ces matériaux, à la fois en solution et sous forme de film. La synthèse du copolymère à blocs PDM-PnBA-PDM a ensuite été optimisée en utilisant la technique d'échange d'halogène en ATRP, ainsi qu’en apportant d'autres modifications à la recette de polymérisation. Des produits monodisperses ont été obtenus à la fois pour la macroamorceur et le copolymère à blocs. À partir d'un seul copolymère à blocs parent, une série de copolymères à blocs partiellement/complètement quaternisés et complexés ont été préparés. Des tests préliminaires de traction sur les copolymères à blocs complexés avec le MO ont montré que leur élasticité est corrélée avec la fraction massique du bloc dur, qui peut être ajustée par le degré de quaternisation et de complexation. Finalement, une série de complexes d'homopolymères auto-assemblés à partir du PDM et de trois dérivés azobenzènes portant des groupes (OH, COOH et SO3) capables d'interactions directionnelles avec le groupement amino du PDM ont été préparés, où les dérivés azo sont associés avec le PDM, respectivement, via des interactions hydrogène, des liaisons ioniques combinées à une liaison hydrogène à travers un transfert de proton (acidebase), et des interactions purement ioniques. L'influence de la teneur en azo et du type de liaison sur la facilité d’inscription des réseaux de diffraction (SRG) a été étudiée. L’efficacité de diffraction des SRGs et la profondeur des réseaux inscrits à partir de films préparés à la méthode de la tournette montrent que la liaison ionique et une teneur élevée en azo conduit à une formation plus efficace des SRGs.

A joint use of experimental and theoretical techniques allows us to understand the key role of intermediate- and short-range defects in the structural and electronic properties of ZnO single crystals obtained by means of both conventional hydrothermal and microwave-hydrothermal synthesis methods. X-ray diffraction, Raman spectra, photoluminescence, scanning electronic and transmission electron microscopies were used to characterize the thermal properties, crystalline and optical features of the obtained nano and microwires ZnO structures. In addition, these properties were further investigated by means of two periodic models, crystalline and disordered ZnO wurtzite structure, and first principles calculations based on density functional theory at the B3LYP level. The theoretical results indicate that the key factor controlling the electronic behavior can be associated with a symmetry breaking process, creating localized electronic levels above the valence band.

High resolution thermogravimetry has been used to evaluate the carbonaceous content in a commercial sample of single-walled carbon nanotube (SWNT). The content of SWNTs in the sample was found to be at least 77mass% which was supported by images obtained with scanning and transmission electron microscopies (SEM and TEM). Furthermore, the influence of SWNT addition on the thermal stability of graphite in mixtures of SWNT/graphite at different proportions was investigated. The graphite stability decreased with the increased of SWNT content in the overall range of composition. This behavior could be due to the close contact between these carbonaceous species as determined by SEM analysis.

Results of an interlaboratory comparison on size characterization of SiO2 airborne nanoparticles using on-line and off-line measurement techniques are discussed. This study was performed in the framework of Technical Working Area (TWA) 34—“Properties of Nanoparticle Populations” of the Versailles Project on Advanced Materials and Standards (VAMAS) in the project no. 3 “Techniques for characterizing size distribution of airborne nanoparticles”. Two types of nano-aerosols, consisting of (1) one population of nanoparticles with a mean diameter between 30.3 and 39.0 nm and (2) two populations of non-agglomerated nanoparticles with mean diameters between, respectively, 36.2–46.6 nm and 80.2–89.8 nm, were generated for characterization measurements. Scanning mobility particle size spectrometers (SMPS) were used for on-line measurements of size distributions of the produced nano-aerosols. Transmission electron microscopy, scanning electron microscopy, and atomic force microscopy were used as off-line measurement techniques for nanoparticles characterization. Samples were deposited on appropriate supports such as grids, filters, and mica plates by electrostatic precipitation and a filtration technique using SMPS controlled generation upstream. The results of the main size distribution parameters (mean and mode diameters), obtained from several laboratories, were compared based on metrological approaches including metrological traceability, calibration, and evaluation of the measurement uncertainty. Internationally harmonized measurement procedures for airborne SiO2 nanoparticles characterization are proposed.

In this work, nanocrystalline Mg-Al-Nd alloys were fabricated using mechanical alloying method. Phase structure of the extrided rods was examined using X-ray diffraction (XRD) and the microstructures were observed using transmission electronic microscopy (TEM). High yield strength was obtained in the alloys with a high Nd content due to grain refinement and Nd rich precipitate phase.

Nesta dissertação, foram preparados materiais poliméricos com atividade bactericida a partir de copolímeros de estireno (Sty) e divinilbenzeno (DVB) e de celulose bacteriana. Três copolímeros à base de Sty e DVB foram sintetizados através da técnica de polimerização em suspensão aquosa. Os copolímeros foram preparados com diferentes estruturas porosas, por meio da variação da composição do sistema diluente. Um copolímero comercial macrorreticulado, Amberlite XAD4, de elevada área superficial, também foi usado neste estudo com o objetivo de comparar sua estrutura polimérica e seu desempenho com o dos copolímeros sintetizados. Os copolímeros de Sty-DVB foram caracterizados por meio da densidade aparente, área específica, diâmetro médio de poros, volume de poros, microscopias ótica e eletrônica de varredura, grau de inchamento, espectrometria de infravermelho com transformada de Fourier (FTIR), análise termogravimétrica e análise elementar. Os copolímeros de Sty-DVB foram modificados quimicamente através da reação de nitração, seguida da reação de redução do grupo nitro a amino e posterior reação de diazotação e hidrólise do grupo sal de diazônio. Os copolímeros modificados contendo os grupos NH2 e OH foram utilizados como suporte para a ancoragem das nanopartículas de prata. As partículas de prata foram obtidas pela redução dos íons Ag+ empregando-se cloridrato de hidroxilamina como redutor e PVP como protetor de colóide. O teor de prata impregnada foi determinado pelo método volumétrico. A celulose bacteriana (BC) foi empregada como uma matriz para impregnação de nanopartículas de prata. Membranas de BC foram impregnadas com solução aquosa de AgNO3, a seguir foi realizada a redução dos íons Ag+ com três diferentes agentes redutores (hidrazina, hidroxilamina e ácido ascórbico) empregando ou não protetor de colóide (PVP ou gelatina). As membranas de BC/Ag0 obtidas foram caracterizadas por difração de raios-X, análise termogravimétrica e microscopia eletrônica de varredura. A capacidade bactericida dos copolímeros de Sty-DVB, dos copolímeros funcionalizados e impregnados com prata e das membranas de BC/Ag0 foi avaliada através do método da contagem em placa contra suspensões de Escherichia coli, em diferentes concentrações (103 a 107 células/mL). Foi observado que os copolímeros de origem e os copolímeros modificados contendo os grupos NO2 e NH2 não apresentaram atividade bactericida contra suspensões de E. coli em nenhuma concentração. Por outro lado, os copolímeros modificados contendo o grupo OH apresentaram alguma atividade bactericida, embora abaixo do esperado devido à baixa incorporação de hidroxilas fenólicas. Os copolímeros modificados impregnados com Ag apresentaram maior ação bactericida do que os copolímeros modificados, o que comprova que a ação bactericida é devida às partículas de prata ancoradas nos copolímeros. Os copolímeros modificados contendo o grupo OH impregnados com Ag mesmo possuindo menores teores de prata apresentaram maior ação bactericida do que os copolímeros modificados contendo o grupo NH2 impregnados com Ag. A maior ação bactericida pode ser atribuída à combinação da atividade bactericida das partículas de prata com a atividade dos grupos hidroxilas fenólicas mesmo que estes grupos estejam pouco incorporados nos copolímeros. Os compósitos de BC/Ag0 exibiram atividade bactericida contra E. coli que foi atribuída à ação das nanoparticulas de prata obtidas quando foi empregada a hidroxilamina como agente redutor e a gelatina como protetor de colóide

Ikerkuntza arlo guztietara zabaldu da ezinezkoak ziruditen erronkak argitu eta ezezagunak ditugun ezagutzak garatzeko asmoz. Lan honetan TiO2 nanohagatxoen sintesia burutu da metodo hidrotermala erabiliz. Sintesi baldintzek lortutako produktuaren egituran eta forman duten eragina kontuan harturik, sintesia gertatzeko ezinbestekoak diren pHa, tenperatura eta denbora aztertu dira. Ondoren, tratamendu kimiko eta termiko ezberdinen bidez konposizio eta egitura ezberdinak lor daitezkeela frogatu da. Bereziki, surfaktante eta pHaren arabera egitura ezberdinak lortzen direla ikusirik. Sintetizatutako laginen karakterizazioa burutzeko X izpien difrakzioa, transmisio bidezko mikroskopia elektronikoa, infragorri espektroskopia, eta termograbimetria teknikak erabili dira. Hauen bidez NaTi3O6·(OH)x·(H2O)y , (TiO2)x(H2O)y , anatasa eta rutilo faseak identifikatu dira, eta nanohagatxoen lodiera eta morfologia ezberdinak ikusi dira.
