Genetically modified foods are a major concern around the world due to the lack of information concerning their safety and health effects. This work evaluates differences, at the proteomic level, between two types of crop samples: transgenic (MON810 event with the Cry1Ab gene, which confers resistance to insects) and non-transgenic maize flour commercialized in Brazil. The 2-D DIGE technique revealed 99 differentially expressed spots, which were collected in 2-D PAGE gels and identified via mass spectrometry (nESI-QTOF MS/MS). The abundance of protein differences between the transgenic and non-transgenic samples could arise from genetic modification or as a result of an environmental influence pertaining to the commercial sample. The major functional category of proteins identified was related to disease/defense and, although differences were observed between samples, no toxins or allergenic proteins were found.

In the present work, the development of a genosensor for the event-specific detection of MON810 transgenic maize is proposed. Taking advantage of nanostructuration, a cost-effective three dimensional electrode was fabricated and a ternary monolayer containing a dithiol, a monothiol and the thiolated capture probe was optimized to minimize the unspecific signals. A sandwich format assay was selected as a way of precluding inefficient hybridization associated with stable secondary target structures. A comparison between the analytical performance of the Au nanostructured electrodes and commercially available screen-printed electrodes highlighted the superior performance of the nanostructured ones. Finally, the genosensor was effectively applied to detect the transgenic sequence in real samples, showing its potential for future quantitative analysis.

Plants kept under competition tend to modify their morphology to optimize the use of production factors. This study aimed to evaluate the effects of competition between transgenic maize and five weed species on the growth and yield of transgenic maize hybrid. The experiment used a randomized block design with four replicates in a factorial 5 x 2 + 6 scheme consisting of a combination of maize under competition with five weed species (Bidens pilosa, Commelina benghalensis, Brachiaria brizantha, Sorghum arundinaceum and Ipomoea triloba) in two weed densities (15 or 30 plants m-2) plus six treatments corresponding to maize and weed species without competition. All the means for dry matter accumulated by maize plants in the stem and leaf in the density of 15 plants m ² were higher than the means for plants in coexistence with 30 plants m-². Number of kernels, diameter and length of cob were not affected by competition with weeds. The weeds that most interfered with maize biomass production were S.arundinaceum and B.brizantha. Leaf dry mass accumulation was more sensitive than the production of stem. It was observed that maize was usually very competitive with weeds, and there was a strong decrease in dry matter accumulation of all the weeds in the study when in coexistence with the crop.

To determine if Bt maize seedlings are protected against lesser cornstalk borer damage, Bt hybrids at the 3 and 4 leaf stages were tested under temperatures between 20-32oC and artificial infestation. A high level of resistance was reported in all Bt maize as expressed by larval survival, larval weight, damage score, and number of surviving plants. All Bt maize protected plants against lesser cornstalk borer damage. Also, the resistance present in the Bt maize was not affected by daytime temperature.

A área brasileira plantada com milho geneticamente modificado (GM) expressando genes Cry derivados da bactéria do solo Bacillus thuringiensis (Bt) aumentou de 4,9% (5,0 milhões de hectares) da área total plantada em 2009 para 81,4% (15,83 milhões de hectares) em 2014. No entanto, estudos sobre os efeitos da tecnologia Bt-milho sobre microrganismos não alvo em solos tropicais são incipientes. Dessa forma, foi realizado experimento de campo para avaliar a atividade fisiológica das comunidades bacterianas associadas com genótipos de milho Bt plantados em Latossolo Vermelho Escuro do Cerrado e solos hidromórficos da planície com inundações localizadas. Um híbrido não transgênico (30F35) e seus homólogos transgênicos 30F35Y (Cry1Ab) e 30F35H (Cry1F) foram plantados com delineamento de blocos casualizados com quatro repetições. Solos rizosféricos e não rizosféricos coletados de plantas no estádio de florescimento foram submetidos aos ensaios de diversidade metabólica com Biolog e atividades enzimáticas de urease, arginase, fosfatase ácida e fosfatase alcalina. Solos rizosféricos apresentaram maior atividade microbiana e não foram detectadas diferenças significativas entre os genótipos em todos os parâmetros bioquímicos e de solo avaliados. Os resultados sugerem que o milho Bt não afeta negativamente a comunidade microbiana dos solos tropicais.

ABSTRACT The cultivation of genetically modified crops in Brazil has led to the need to assess the impacts of this technology on non-target species. Under field conditions, the potential effect on insect biodiversity was evaluated by comparing a homogeneous corn field with conventional and transgenic maize, expressing different Bt proteins in seven counties of Minas Gerais, Brazil. The richness pattern of non-target insect species, secondary pests and natural enemies were observed. The results do not support the hypothesis that Bt protein affects insect biodiversity. The richness and diversity data of insects studied were dependent on the location and other factors, such as the use of insecticides, which may be a major factor where they are used.

The Brazilian government has approved many transgenic maize lines for commercialization and has established a threshold of 1% for food labeling, which underscores need for monitoring programs. Thirty four samples including flours and different types of nacho chips were analyzed by conventional and real-time PCR in 2011 and 2012. The events MON810, Bt11, and TC1507 were detected in most of the samples, and NK603 was present only in the samples analyzed in 2012. The authorized lines GA21, T25, and the unauthorized Bt176 were not detected. All positive samples in the qualitative tests collected in 2011 showed a transgenic content higher than 1%, and none of them was correctly labeled. Regarding the samples collected in 2012, all positive samples were quantified higher than the threshold, and 47.0% were not correctly labeled. The overall results indicated that the major genetically modified organisms detected were MON810, TC1507, Bt11, and NK603 events. Some industries that had failed to label their products in 2011 started labeling them in 2012, demonstrating compliance with the current legislation observing the consumer rights. Although these results are encouraging, it has been clearly demonstrated the need for continuous monitoring programs to ensure consumers that food products are labeled properly.

As the most commercially valuable cereal grown worldwide and the best-characterized in genetic terms, maize was predictably the first target for transformation among the important crops. Indeed, the first attempt at transformation of any plant was conducted on maize (1). These early efforts, however, were inevitably unsuccessful, since at that time, there were no reliable methods to permit the introduction of DNA into a cell, the expression of that DNA, and the identification of progeny derived from such a “transgenic” cell (2). Almost 20 years later, these technologies were finally combined, and the first transgenic cereals were produced. In the last few years, methods have become increasingly efficient, and transgenic maize has now been produced from protoplasts as well as from Agrobacterium-medieited or “Biolistic” delivery to embryogenic tissue (for a general comparison of methods used for maize, the reader is referred to a recent review—ref. 3). The present chapter will describe probably the simplest of the available procedures, namely the delivery of DNA to the recipient cells by vortexing them in the presence of silicon carbide (SiC) whiskers (this name will be used in preference to the term “fiber,” since it more correctly describes the single crystal nature of the material).

ABSTRACT Production of the polyketide antimicrobial metabolite 2,4-diacetyl-phloroglucinol (DAPG) is a key factor in the biocontrol activity of Pseudomonas fluorescens CHA0. Strain CHA0 carrying a translational phlA'-'lacZ fusion was used to monitor expression of the phl biosynthetic genes in vitro and in the rhizosphere. Expression of the reporter gene accurately reflected actual production of DAPG in vitro and in planta as determined by direct extraction of the antimicrobial compound. In a gnotobiotic system containing a clay and sand-based artificial soil, reporter gene expression was significantly greater in the rhizospheres of two monocots (maize and wheat) compared with gene expression in the rhizospheres of two dicots (bean and cucumber). We observed this host genotype effect on bacterial gene expression also at the level of cultivars. Significant differences were found among six additional maize cultivars tested under gnotobiotic conditions. There was no difference between transgenic maize expressing the Bacillus thuringiensis insecticidal gene cry1Ab and the near-isogenic parent line. Plant age had a significant impact on gene expression. Using maize as a model, expression of the phlA'-'lacZ reporter gene peaked at 24 h after planting of pregerminated seedlings, and dropped to a fourth of that value within 48 h, remaining at that level throughout 22 days of plant growth. Root infection by Pythium ultimum stimulated bacterial gene expression on both cucumber and maize, and this was independent of differences in rhizosphere colonization on these host plants. To our knowledge, this is the first comprehensive evaluation of how biotic factors that commonly confront bacterial inoculants in agricultural systems (host genotype, host age, and pathogen infection) modulate the expression of key biocontrol genes for disease suppression.

La creciente implantación de maíz transgénico en España está ocasionando cambios importantes en las poblaciones de taladros de maíz y en su ecofisiología, cambios que están por estudiar. En el evento de maíz transgénico Bt 176, hoy en día no autorizado, la concentración de proteína de Bacillus thuringiensis decrecía en el maíz senescente de forma que en otoño se podían encontrar en campos con este evento alguna larva de taladros. La implantación de maíz con el evento MON 810 sugirió la posibilidad de estudiar la posible supervivencia de larvas en maíz senescente y algunos aspectos de la diapausa de estas larvas. Para ello se infestaron 6 variedades de maíz, una con el evento Bt 176, dos con el evento MON 810 y las tres isogénicas correspondientes, con larvas neonatas de Sesamia nonagrioides (Lefébvre), confiando en una infestación natural de Ostrinia nubilalis (Hübner). Las larvas de S. nonagrioides recogidas en el evento Bt176 suponían el 14% del total de larvas de esta especie muestreadas, mientras que las de O. nubilalis recogidas en el maíz Bt suponían un 25% del total de esta especie. Sin embargo en las variedades con el evento MON 810 apenas se recogieron un 0,25% y un 1,9% de larvas de S. nonagrioides y O. nubilalis respectivamente. Las larvas recogidas en el evento Bt 176 no mostraron diferencias en su intensidad de diapausa con las recogidas en las isogénicas correspondientes. El efecto del fotoperiodo en la terminación de la diapausa fue diferente en ambas especies: mientras que el fotoperiodo largo aceleraba la terminación de diapausa en S. nonagrioides pero no en O. nubilalis pareciendo tener un efecto más pronunciado la oscuridad completa. El parasitismo por parte de Lydella thompsoni (Herting) fue del 5% para el total de larvas de O. nubilalis mientras que para el total de larvas de S. nonagrioides fue del 0,4% lo que indica la preferencia de esta taquínido por el pirálido.

The present review compiles positive MS fragmentation data of selected carotenoids obtained using various ionization techniques and matrices. In addition, new experimental data from the analysis of carotenoids in transgenic maize and rice callus are provided. Several carotenes and oxygen-functionalized carotenoids containing epoxy, hydroxyl, and ketone groups were ionized by atmospheric pressure chemical ionization (APCI)-tandem mass spectrometry (MS/MS) in positive ion mode. Thus, on the basis of the information obtained from the literature and our own experiments, we identified characteristic carotenoid ions that can be associated to functional groups in the structures of these compounds. In addition, pigments with a very similar structure were differentiated through comparison of the intensities of their fragments. The data provide a basis for the structural elucidation of carotenoids by mass spectrometry (MS).

In der vorliegenden Arbeit ging es um die Erarbeitung, Anwendung und Beurteilung von quantitativen Analysenverfahren / Methoden für ein Monitoring von durch Bt-Mais verursachbaren Umwelteffekten im Boden. Die Ausgangsthese besagte, dass sich transgene Maisstreu beim mikrobiellen Abbau anders verhält als konventionelle. Bezugnehmend auf die These wurden zwei Freilandversuche (Freilandmikrokosmenmethode nach Raubuch 1997 über 2 Jahre, Quantifizierung des Maisstreuabbaus mit Hilfe kleiner Bodensäulen über 1 Jahr) und zwei Inkubationsversuche im Labor (INK bei drei verschiedenen Temperaturen über 49 Tage und INK mit verschiedenen landwirtschaftlich genutzten Böden über 49 Tage mit jeweils kontinuierlicher Respirationsratenermittlung nach Isermeyer 1952) sowie Inhaltsstoffbestimmungen der Maisstreu durchgeführt. Für alle Untersuchungen wurde Streu der vier Maissorten Novelis (transgen, Monsanto 810), Nobilis (Isolinie von Novelis), Valmont (transgen, Bt 176, Fa. Syngenta) und Prelude (Isolinie von Valmont) eingesetzt. Nach Beendigung der Laborversuche sowie des Freilandversuches nach der Freilandmikrokosmenmethode wurden mikrobielle Messgrößen wie Adenylategehalt, Ergosterolgehalt, Cmik- und Nmik-Gehalt am Boden-Streu-Gemisch bestimmt. Der Einsatz der Isotopentechnik (Bestimmung von 13C/12C an gemahlenem Boden-Streu-Gemisch bzw. gefriergetrocknetem K2SO4 als Extrakt aus dem Boden-Streu-Gemisch) ermöglichte eine genaue Quantifizierung der abgebauten Maisstreu und brachte dadurch Aufschluss über das Abbauverhalten verschiedener Maissorten. Bezüglich der Ermittlung der mikrobiellen Messgrößen ergab sich für die transgene Sorte Novelis* stets eine durchschnittlich geringere pilzliche Biomasse. Langfristig ergaben sich bei der Kohlenstoff- und Stickstoffdynamik keine Trends hinsichtlich transgener bzw. konventioneller Maisstreu. Sowohl im Freilandversuch nach der Mikrokosmenmethode als auch in den Inkubationsversuchen trat das Phänomen der kurzzeitigen Respirationsratenerhöhung der Mikroorganismen nach Zugabe der transgenen Maissorten auf, welches nicht bei Zugabe der konventionellen Maisstreu auszumachen war. ______________________________
