Toxoplasma gondii, un protozoaire très répandu dans le monde, peut infecter de nombreuses espèces homéothermes incluant les mammifères et les oiseaux qui développent alors une toxoplasmose. L’impact de la toxoplasmose en termes de santé publique est majeur, particulièrement chez les personnes immunodéprimées et les foetus. Les niveaux d’infection humaine dans certaines régions de l’Arctique Canadien sont parmi les plus élevés au monde et ce, malgré l’absence de félidés qui sont les seuls hôtes capables d’excréter T. gondii. Plusieurs études ont suggéré la consommation de viande crue de mammifères marins et notamment de phoques comme source d’infection des Inuits. Notre travail de recherche visait à comprendre les mécanismes de dispersion de T. gondii dans les écosystèmes aquatiques menant à la contamination du milieu marin de l’Arctique par des oocystes, et à évaluer l’importance de cette voie de dispersion dans l’infection des phoques et conséquemment dans celle des Inuits. Notre hypothèse était que les oocystes de T. gondii, excrétés durant l’hiver par des félidés dans le Subarctique et transportés par les rivières pendant la fonte printanière, contaminaient les estuaires de l’Arctique Canadien. Dans un premier temps, une étude transversale de séroprévalence chez les phoques de l’Arctique Canadien a montré que ces populations étaient infectées par T. gondii et pouvaient ainsi a priori constituer une source d’infection pour les Inuit. Des variations spatio-temporelles de la séroprévalence étaient observées suggérant un lien potentiel avec des variations dans la contamination environnementale par les oocystes. Un schéma conceptuel explicitant les mécanismes de transport et de devenir des oocystes de T. gondii, du phénomène de la fonte de la neige jusqu’à l’exposition des organismes marins, a été proposé dans le chapitre suivant. Des interactions entre les différents mécanismes identifiés, qui agissent sur des échelles spatio-temporelles variées, devraient favoriser l’apparition de concentrations relativement élevées aux estuaires permettant ainsi l’exposition et potentiellement l’infection de phoques. Pour évaluer la contamination environnementale par les oocystes excrétés par la population de lynx du bassin versant de l’Arctique Canadien (les seuls félidés majoritairement distribués dans ce vaste territoire), nous avons mené une étude sérologique de type transversale dans cette population. Cette étude a permis de montrer que des lynx étaient infectés par T. gondii et a également suggéré que la dynamique des cycles de populations lynx-lièvres pouvait être un processus important dans la transmission de T. gondii. Finalement, la modélisation du transport hydrique des oocystes a indiqué que les concentrations hypothétiques d’oocystes dans l’eau de la fonte pourraient être suffisantes pour permettre l’exposition au niveau des estuaires de bivalves filtreurs, qui sont des proies pour les phoques et donc potentiellement des sources infectieuses pour ces derniers. Dans des écosystèmes nordiques en pleine mutation, la compréhension des mécanismes de transmission d’agents pathogènes d’origine hydrique comme T. gondii est plus que nécessaire, notamment dans le but de protéger les populations fragilisées de ces régions.

A toxoplasmose é uma das parasitoses mais comuns em gatos sendo que a sua seroprevalência mundial varia entre 5,5% e 97,4%. Em Portugal, existem poucos estudos sobre a prevalência da toxoplasmose e, por isso, este estudo tem como objetivo principal contribuir para o conhecimento da seroprevalência de base clínica no nosso país. Para a sua realização, foram colhidas amostras de soro de 63 gatos e recolhidos dados epidemiológicos sobre a idade, a raça, o género, o tipo de alimentação, o acesso ao exterior e o tipo de habitat dos progenitores. As amostras de soro foram analisadas através da técnica de imunofluorescência indireta, tendo sido encontrados 25 gatos seropositivos, correspondendo a uma seroprevalência de base clínica de 41%. O único fator de risco identificado foi o acesso ao exterior. A idade do animal não foi estatisticamente significativa como fator de risco, mas observou-se um aumento da prevalência em animais até aos 6 anos de idade. Os gatos de progenitores com acesso ao exterior, animais de género feminino e com alimentação de risco apresentaram maior número de seropositivos, no entanto, estes resultados não foram estatisticamente significativos.

A toxoplasmose congénita é uma doença infecciosa, causada pelo parasita Toxoplasma gondii e, adquirida por transmissão materno-fetal, a qual pode acarretar sequelas neurológicas e oculares muito graves, no recém-nascido. O presente estudo incide sobre as linhas de prevenção da doença, em Portugal. A base da prevenção define-se como primária, através da determinação do estatuto imunológico da mulher, do aconselhamento e adopção de medidas higiénico-dietéticas das mulheres seronegativas, de forma a evitar a infecção materna. A vigilância serológica, na detecção de uma possível infecção materna, e a instituição da terapêutica de profilaxia, constituem a prevenção secundária, de modo a evitar a infecção fetal. A prevenção terciária recai, sobre o estabelecimento de um novo esquema terapêutico, dotado de alguma teratogenicidade, com o intuito de minimizar as sequelas da infecção. Em Portugal, existem muitas mulheres seronegativas, mal informadas acerca da doença, e que não tomam medidas preventivas correctas, para evitar a infecção. Esta problemática é decrescente, de norte para sul do país. A prevenção da doença pode ser bem-sucedida, através da implementação de directrizes específicas, dirigidas aos diferentes grupos de risco e da orientação correcta, pelos profissionais de saúde. A realização de estudos, em várias áreas de intervenção da doença, optimiza a sua prevenção e a sua relação de custo-benefício.

Toxoplasmosis, provoked by the intracellular parasite Toxoplasma gondii, is one of the most prevalent parasitoses in the world. In humans, transmission occurs by three evolutionary forms of the parasite: oocysts, tissue cysts and tachyzoites. Wild and domestic felines are definitive hosts. The ocular form of toxoplasmosis can be of congenital origin with early or late clinical manifestations, or acquired after birth. T. gondii is considered the main culprit for most cases of infectious uveitis. This study aimed at assessing ocular toxoplasmosis, relating it to factors associated to the patient s lifestyle and describing the epidemic-serological and clinical profile of affected individuals. A cross-sectional study was conducted with a population of 159 patients. Univariate analysis (odds ratio) was used to evaluate the data, with a confidence interval of 95% and p-value < 0.05. A prevalence of 4% of ocular toxoplasmosis was observed in the population of patients treated at an ophthalmological clinic. Of patients directly examined by immunoenzymatic assay (MEIA-AxSYM®- Microparticle Enzyme Immune Assay), considering only uveitis, a frequency of anti-T. gondii of 73%, most of whom exhibited titulation between 40-99 UI IgG/mL. With respect to location of ocular lesions, bilaterality was observed in 57% of patients assessed by the ophthalmoscopy technique. When compared with the results of an active search of medical records, a similarity in ocular toxoplasmosis (74%) and bilateral lesion location (55%) was observed. Type I lesion was the most frequent type observed, with intraocular disposition in the macula. An epidemiological survey revealed that direct contact with cats; consuming raw or poorly cooked meat and direct contact with the soil were significantly associated with greater likelihood of acquiring ocular toxoplasmosis. Sample characterization in relation to age range was significant for patients between 31 and 40 years [χ², chi-square test (p = 0.04)], but population traits such as schooling, sanitary district, and monthly income were not significant. Results confirm that ocular toxoplasmosis is widely distributed in the metropolitan area of Natal, Brazil, with significant prevalence of ocular lesions provoked by T.gondii. It is suggested that sanitary authorities exert greater control in order to minimize the risk of toxoplasmic infection, mainly in pregnant women.

Toxoplasmosis is a zoonosis of worldwide distribution caused by the protozoan Toxoplasma gondii, triggering dangerous complications in immunocompromised patients and pregnant women, as well as having great economic impact for the livestock. So far the control of toxoplasmosis is made primarily by chemotherapy. However, most drugs used routinely have some limitations. In order to control this disease, several research groups, including ours, has been working to develop a medical-veterinary vaccine based on parasite antigens, vectors and protocols of immunization. In this study were implemented and standardized methodologies for amplification and cloning of recombinant immunogens in the system for the development of a prototype vaccine, based on the surface antigens of T. gondii and recombinant adenovirus encoding these antigens. Genes encoding BAG1, GRA2 and SAG1 proteins were amplified. We established a strategy for cloning SAG1, SAG2, SAG3 and TgAMA1- genes in recombinant system. The genes encoding SAG1 and SAG2 were cloned and their sequences showed high similarity with sequences from GenBank. The virtual translation of these proteins showed polymorphisms in the amino acid sequence, which can be correlated with levels of antigenicity. Simultaneously, the adenovirus encoding the SAGs (HAdSAGs) were expanded, purificated and characterizated. Immunization of C57bl/6 mice, using viral supernatant was not enought to elicit immune responses at high levels, being required HAdSAGs titration for future immunizations. Therefore, this study allowed the cloning of the two genes important for the development of a prototype vaccine. Besides, implementations methodologies that permit advancements in the development of a vaccine against toxoplasmosis using adenovirus to express proteins of the parasite

Inquiries about toxoplasmosis are essential because its zoonotic potential and pathogenicity in pets and production animals. The dog can be involved in the mechanical transmission of the infection by Toxoplasma gondii and in spite of not being the definitive host, it has an epidemiological role in this disease. The high rate of dogs naturally infected by T gondii and the great possibility of adquiring this protozoan, justify the importance of attempting against for the occurrence of this illness. The aim of this article was to demonstrated through literature revision, some clinical aspects, epidemic and pathological of the toxoplasmosis in the canine species.

Congenital Toxoplasmosis results in severe systemic disease. If mother is infected for the first time during gestation, she can infect the fetus causing substantial damage. However, relatively little is known about the seroprevalence and epidemiological and economic factors of Toxoplasmosis infection in pregnancy in the most state in northeastern Brazil and knowledge about this can be essential in determining effective and acceptable prevention strategies. Our aim was to determine the prevalence of Toxoplasmosis in pregnant woman consulted by reference Maternity Escola Januário Cicco in Natal, a city in Northeastern Brazil, which belongs to the public health system, correlating to the risk factors involved in the infection and to accomplish active Search in the Hospital of Pediatrics Profº Heriberto Bezerra of the damages caused by the Toxoplasmic infection in children up to 12 years of age. The study was conducted from March to December 2007 and sera obtained from 190 pregnant women were tested for IgM and IgG antibodies avidity to Toxoplasma by Microparticle enzyme immunoassay (Abbott AxSYM system - Abbott Laboratories, Chicago, IL, USA). Data were examined with univariate analysis. Chi-squared (x2) and Odds ratio was calculated (IC 95% p 0,05). Of these women, 126 (66,3%) had only IgG antibodies high-avidity against T. gondii; 01 (0,52%) had a IgM and IgG high-avidity antibodies against T. gondii and 63 (33,1%) have neither IgM nor IgG against T. gondii. Our studies shown that the direct contact with cats or dogs was highly associated with the Toxoplasma gondii infection (OR, 2.72, p<0.001, 95% CI 1.46 5.02). The years school (p<0,001), socioeconomic status and knowledge about the disease (both p value 0.05) also were associated with Toxoplasmosis. The pattern of risk factors for infection presents regional variations, however our data corroborate others studies in Brazil. In children up to 12 years, one case of Congenital Toxoplasmosis was just registered in seven years (2000 - 2006). There were several suggestive cases, with signs and characteristic symptoms, but that the infection was not confirmed due to lack in the researches through laboratorial and images exams that addressed that it zoonosis

Com o objetivo de estudar a soroprevalência de vírus linfotrópico de células T humanas I/II (HTLV-I/II), vírus da imunodeficiência humana, sífilis e toxoplasmose, em gestantes atendidas em unidade básicas de saúde do município de Botucatu - São Paulo - Brasil, bem como os fatores de risco para a infecção pelo HTLV -I/II, foram realizados inquérito sorológico e avaliação dos resultados de exames solicitados na rotina do prénatal. em 913 gestantes, a soroprevalência de HTLV- I e de HTLV- II foi de 0,1%. Sífilis, toxoplasmose e infecção pelo HIV foram encontradas. Nenhum dos fatores de risco pesquisados mostrou-se seguro para identificar gestantes com infecção pelo HTLV- I/II. A comparação da proporção de gestantes infectadas e de doadores de sangue da região sudeste do Brasil com testes reagentes para HTLV- I/II não mostrou diferença estatística.

The frequency of anti-T.gondii IgG antibodies was evaluated and compared in 253 sera samples of equines by modified agglutination test (MAT) and indirect immunofluorescent antibody test (IFAT). Fifteen samples (5.9%) were positive by IFAT and 32 (12.6%) by MAT. Significant difference between the two methods was found when a cutoff titer of 16 was established. There was no correlation with breed, gender, and age of horses examined.

Foram avaliados pela reação de imunofluorescência indireta (RIFI) para detecção de anticorpos-IgG anti-Toxoplasma, soros de 191 gatos de 3 diferentes municípios do Estado de São Paulo e um município do Estado do Paraná, enviados ao Serviço de Diagnóstico de Zoonoses, da Faculdade de Medicina Veterinária e Zootecnia, UNESP, Campus de Botucatu. Destes animais, 56 (29,3%) possuíam raça definida e 135 (70,7%) sem raça definida (SRD), sendo 88 (46,1%) fêmeas e 103 (53,9%) machos, com idades variando de dois meses a dezoito anos. Obtiveram-se 37 (19,4%) animais reagentes à RIFI, com títulos de 1/16 (n = 10;27,0%), 1/64 (n = 18;48,6%) e 1/256 (n = 9;24,3%). Não houve associação significativa entre sexo, raça, procedência ou idade (p > 0,05), entretanto, a taxa de infecção foi maior em animais mais velhos, com 13,2% dos animais entre dois meses e três anos de idade, 23,5% entre três e seis anos, 17,6% entre seis e nove anos, 25% entre nove e doze anos, 20% entre doze e quinze anos e 50% entre quinze e dezoito anos de idade.

Com o objetivo de comparar variáveis epidemiológicas na toxoplasmose em ovinos e caprinos, amostras de soro de animais de propriedades localizadas em duas regiões do Estado de Pernambuco, Brasil, foram testadas pela reação de imunofluorescência indireta (RIFI) para detecção de anticorpos anti-Toxoplasma gondii. Dos 173 soros ovinos testados, em 35,3% foram encontrados resultados positivos, enquanto 40,4% dos 213 soros caprinos foram positivos à RIFI. em ovinos, associações significativas foram encontradas para sexo e raça, mas não para região, tipo de manejo ou falha reprodutiva. Os resultados da RIFI para os caprinos foram significativamente associados ao sexo, raça, região, tipo de manejo e de exploração, mas não com a ocorrência de falhas reprodutivas.
