This study intended to evaluate the maze test accuracy in cognitive deficit screening in elderly with or without neuropsychological pathology. The sample included 40 healthy young (18-25 years old; mean- 21 ± 1.6), 40 healthy old (60-77 years old; mean- 67 ± 5.1) and 18 patients with probable diagnosis of Alzheimer s disease initial stage (52-90 years old; mean- 78 ± 9.2). Data analysis was made using Anova with Tukey s post hoc, multiple linear regression analysis and ROC curve analysis. According to Tukey s test Alzheimer patients spent more time (46843 ± 37926 ms) to execute the test than healthy young (5482 ± 2873 ms; p= 0.0001) and elderly (17978 ± 13700; p= 0.0001); healthy young executed test n lower time (p= 0.035). According to the regression analysis of age, education level and cognitive performance of the three groups, the cognitive performance was the predictor of the execution time. When analyzing young and elderly only age was the predictor and the cognitive performance was the only factor to influence the test of old aged healthy and patients. The ROC curve analysis indicated 72% accuracy for young and elderly and 36% for healthy and elderly patients. The maze execution time represented a better balance between sensibility (75%) and the specificity (61%) was near 13575 ms, indicating that those subjects that execute the maze in a time higher to this value may show cognitive deficit related to the executive function. According to the results it is suggested that the maze test used in this study shows a good accuracy in the cognitive deficit tracking and may discriminate age changes

The increase of elderly population in the world and in Brazil has indicated the necessity of health systems capable to evaluate, to diagnose and to intervene in the conditions of health and disease of that segment. During that stage of human development, physical and cognitive changes happen and they are capable to influence the functional acting. It s important to distinguish the limit between the normal and the pathological. Besides the common changes during the aging, biological rhithmicity changes happen, as alterations in the cycle vigil-sleep that can influence in certain tasks performance. This study aimed to verify the influence of the age, of the sex and of the hour in a maze test performance. Eighty individuals were evaluated, 40 youths (20 men and 20 women) and 40 senior (20 men and 20 women). They were separated in 2 different groups that were tested at 9:00 o clock and at 15:00 o clock. Initially they were submitted to health evaluation, cognitive evaluation and of sleep quality and chronotype. They were instructed to accomplish the maze test whose time of execution was timed and registered. Significant differences were observed according to age for the masculine group between elderly in the morning and in the afternoon and in the feminine group between youth and elderly in the test accomplished in the morning and in the afternoon. Significant differences were not observed according to sex and hour of the day and also between attempts. In compare between the 30th and the31st, accomplished in a 15minutes of interval, significant difference was observed just for the elderly group in the morning and in the afternoon. We observed significant correlations in the maze test performance with the cronotype, with the age, with the education and with the cognitive acting. The maze test was capable to detect differences between age in the acting profile and in the evaluation of the information maintenance after 15 minutes, however it was not possible to verify difference between sex and hour of the day. Finally the correlations of the maze test with another varied may indicate your importance as coadjutant instrument in those functions evaluation

Metilmercúrio (MeHg), a forma orgânica do mercúrio, é um dos poluentes de maior risco ao ambiente. MeHg é uma potente neurotoxina, principalmente durante o desenvolvimento do sistema nervoso central. A neurotoxicidade induzida pelo MeHg no período pré-natal pode causar desordens mentais, paralisia cerebral e convulsões. Nós investigamos o imunoconteúdo de S100B no fluído cerebroespinal (FCE) e no tecido encefálico, uma proteína ligante de cálcio produzida e secretada pelos astrócitos, na qual tem uma atividade trófica e tóxica, dependendo da sua concentração. Ratas grávidas foram expostas ao MeHg (5 mg/kg/dia) no 12°, 13° e 14° dias de gestação. O fluído cerebroespinal e o tecido encefálico (mais especificamente hipocampo, córtex cerebral e cerebelo) foram obtidos dos neonatos no 1°, 15° e 30° dia pós-natal. O acúmulo de MeHg foi medido do tecido encefálico após o nascimento e aos 30 dias de vida. Um aumento da S100B no FCE foi observado aos 15 dias de vida pós-natal, mas desapareceu aos 30 dias. No tecido hipocampal mostrou um aumento da S100B (e redução da proteína ácida fibrilar glial) imediatamente após nascimento, mas não posteriormente. Nenhuma mudança foi observada no teste cognitivo (labirinto aquático) desses ratos em idade adulta. Nossos resultados reforçam o envolvimento glial na neurotoxicidade induzida pelo MeHg. As mudanças no hipocampo ao nascimento, poderiam estar relacionadas com as desordens cognitivas e epilépticas atribuídas ao MeHg. O aumento da S100B no FCE reforça a hipótese de que o aumento da S100B está relacionado a danos no sistema nervoso central. Embora, o mecanismo celular envolvido no aumento do conteúdo da S100B no FCE seja desconhecido, os resultados sugerem que a S100B possa ser usada como um marcador periférico na injúria induzida pelo MeHg.

Para medir possíveis influências da mastigação e do estilo de vida sedentário em modelo murino senil, impusemos um de três regimes de dieta aos diferentes grupos experimentais do 21º dia pós-natal até 6 ou 18 meses de vida: dieta sólida tipo pellet; dieta em pellet seguida por uma em pó, farelada; ou dieta peletizada seguida de pó e novamente pellet, com intervalos de tempo iguais em cada dieta. Para mimetizar o estilo de vida sedentário ou ativo, os animais foram criados, respectivamente, em gaiolas-padrão (ambiente empobrecido-AP) ou em gaiolas enriquecidas (ambiente enriquecido-AE). Para medir os efeitos da dieta, do ambiente e da idade sobre a atividade exploratória, realizamos o teste do campo aberto, onde camundongos jovens de AP que sofreram alteração da atividade mastigatória demonstraram maior preferência pela zona periférica, mas no envelhecimento e no AE essas diferenças foram minimizadas. Nos velhos de AE, essas diferenças reapareceram. Já sobre as influências na aprendizagem e memória espacial, aplicamos o labirinto aquático de Morris e vimos que a redução da atividade mastigatória, independente do ambiente, diminuiu a taxa média de aprendizado espacial e sua reabilitação recuperou as perdas associadas em animais jovens e, quando combinada ao AE, melhorou a taxa de aprendizado em velhos. Não se encontrou correlação entre taxa de aprendizado e velocidade de nado dos camundongos sugerindo que os déficits são de natureza cognitiva. Concluímos assim, que a alteração da atividade mastigatória influencia o padrão de exploração por zonas no campo aberto e a estimulação ambiental acentua os seus efeitos no envelhecimento, privilegiando a preferência pela zona periférica e a redução da atividade mastigatória prejudica a memória espacial durante o teste do labirinto aquático de Morris e a sua reabilitação é capaz de recuperar as habilidades espaciais, mas em idosos é necessária a combinação com um AE.

O mercúrio inorgânico é facilmente absorvido por ingestão ou via cutânea. Entretanto, uma quantidade relativamente pequena de Hg2+ atravessa a barreira hematoencefálica ou as membranas biológicas, sendo em ratos adultos, o transporte axonal retrógrado a única via para a absorção de Hg2+ por neurônios, apresentando um forte potencial neurotóxico. Desta forma, o presente estudo objetivou investigar os efeitos da exposição crônica ao cloreto de mercúrio em memória social e emocional de ratos adultos. Para isso utilizou-se ratos Wistar, machos (n=40), com 5 meses de idade, distribuídos em dois grupos, um dos quais foi exposto ao Cloreto de Mercúrio (HgCl2) via oral, por gavagem intra-gástrica (0,375mg/Kg), durante 45 dias. O outro grupo, denominado grupo controle (n=20) recebeu água destilada por gavagem. Foram utilizados os seguintes testes comportamentais: teste do campo aberto, teste de reconhecimento social para avaliação de memória social; o Teste do Labirinto em T Elevado (LTE) foi usado para avaliar o aprendizado do estado de esquiva e as memórias de curta e longa-duração. Após a finalização dos testes, os animais foram sacrificados para a dosagem do mercúrio total no hipocampo e através de um Espectrofotômetro de Absorção Atômica. Os resultados revelaram que os animais submetidos à exposição ao cloreto de mercúrio não manifestaram déficits em atividade exploratória. Nos dados do Teste de Reconhecimento Social, observamos que não houve alteração em memória social. No teste do LTE, o grupo exposto ao HgCl2 necessitou de um número maior de exposições para aquisição do critério de esquiva (p<0,05) e apresentaram latência maior no braço aberto do aparato (p<0,05). Após 24 horas, verificou-se que os animais expostos passaram menos tempo no braço fechado em relação ao grupo controle, sugerindo déficits de memória de longa duração. Ao observar apenas o grupo HgCl2, percebeu-se uma melhora no reteste, indicando preservação na memória de curta duração. Os dados de espectrometria de absorção atômica mostraram uma maior deposição de mercúrio no hipocampo de animais intoxicados, em relação aos animais do grupo controle.

Introduction: Aortic insufficiency (AoI), a volume overload, is characterized by the diastolic reflux of blood from the regurgitating aorta to the left ventricle. This effect results from malfunctioning aortic cusps. The main cause of AoI in developing countries is rheumatic fever, including Brazil, and valvar degeneration in developed countries. There is a strong association between cardiovascular diseases and depression. Selective serotonin reuptake inhibitors (SSRI) are one of the most prescribed antidepressants in the world. Previous studies of our laboratory showed that the utilization of a SSRI, paroxetine, improved cardiac function in rats with sub-chronic AoI and reduced the daily ingestion of hypertonic sodium (NaCl 0,3M). Cardiovascular diseases can determine behavior changes like increase of anxiety, and it is yet unknown if AoI would determine anxiety or anhedonia, incapacity of obtaining pleasure through physical or sensorial experiences. A possible target for SSRI action could be a change in the expression of enzyme isoforms that collaborate in the contractile function of the heart muscle, like the heavy chains of myosine, the sarcoplasmatic reticulum Ca2+/ATPase (SERCA) and its regulator protein, phospholamban (PLB). Objectives: Evaluation of behavior parameters for anxiety and anhedonia state and genic expression of a-myosine, b-myosine, SERCA2a and PLB in the heart tissue of rats with subchronic AoI that received treatment with an SSRI (paroxetine) for 4 weeks. Methods: Surgery to induce AoI was performed on male Wistar rats, anxiety was evaluated by the elevated plus-maze (EPM) and state of anhedonia was tested by ingestion of 2% sucrose solution. After euthanasia the heart tissue was collected and total RNA was extracted to be analyzed by the RT-qPCR method. Ethanol withdrawn individuals present a wealth of signs and symptoms, some of them related with anxiety. To better understand brain areas involved in anxiety caused by ethanol abstinence, preclinical studies have been employing models of ethanol consumption followed by withdrawal in rodents submitted to behavioral tests of anxiety, such as the elevated plus-maze. The aim of this study was to investigate if short- or long-term ethanol withdrawal could alter both anxiety-related behaviors in the elevated plus-maze (EPM) and open field tests and the number of serotonin immunorreactive cels in the dorsal raphe nucleus, a midbrain area associated with anxiety. Female Wistar rats (90 days old) were submitted to increasing concentrations of ethanol (2% for 3 days, 4% for 3 days and 6% for 15 days) as the only source of liquid diet and the control group received water ad libitum. Both groups received food ad libitum. In the behavioral experiments, on 21st day of consumption, ethanol was substituted by water (withdrawal) and 72 h or 21 days after withdrawal animals were submitted to the EPM, where it was evaluated the percentage of time and entries in the open arms and the entries in the enclosed arms during 5 minutes. Twenty and four hours after testing in the EPM, animals were submitted to the open field test for 15 minutes, where the distance traveled by the animals was observed along this period. During the first 5 minutes, the distance traveled, entries and time spent in the center of the test were analyzed. In the immunohistochemistry study, animals were submitted to 21 days of consumption of ethanol followed or not by 72 hours and 21 days of withdrawal previously perfusion, brain tissue preparation and quantification of serotonin dyed cells in the dorsal and caudal portions in the dorsal raphe nucleus. Behavioral data showed that both short- and long-term ethanol withdrawals reduced the open arms exploration in the EPM. In the open field test there were no locomotor activity changes during the total 15 minutes; however, longterm ethanol withdrawal reduced the exploration in the center of the open field during the first 5 minutes. In the immunohistochemistry step, there were no differences, when short- and long-term withdrawn groups were compared with control group; nevertheless, the chronic consumption of ethanol decreased the number of serotonergic immunorreactive cells in the dorsal part of dorsal raphe nucleus. Taken together, results here obtained suggest that both short- and long-term ethanol withdrawals promoted an anxiogenic-like effect that was not related with changes in the serotonin immunorreactivity in the dorsal and caudal parts of the dorsal raphe nucleus.

Anxiety is an emotional phenomenon, and normally it is interpreted as an adaptative behavior front to adversities. In its pathological form, anxiety can severely affect aspects related to the personal and professional life. Studies have shown a close relationship between anxiety disorders and aversive memory processing. Considering that the pharmacotherapy of anxiety disorders is still limited, innovative anxiolytic agents are needed. In this regard, neuropeptides systems are interesting therapeutic targets to the treatment of psychopathologies. Neuropeptide S (NPS), a 20-aminoacid peptide, is the endogenous ligand of a G-protein coupled receptor (NPSR), which has been reported to evoke hyperlocomotion, awakefull states, besides anxiolysis and memory improvements in rodents. This study aimed to investigate the effects of biperiden (BPR; an amnesic drug), diazepam (DZP; an anxiolytic drug) and NPS at three distinct times: pre-training, post-training, and pre-test, in order to assess anxiety and memory process in the same animal model. The elevated Tmaze (ETM) is an apparatus derived from the elevated plus-maze test, which consists of one enclosed and two open arms. The procedure is based on the avoidance of open spaces learned during training session, in which mice were exposed to the enclosed arm as many times as needed to stay 300 s. In the test session, memory is assessed by re-exposing the mouse to the enclosed arm and the latency to enter an open arm was recorded. When injected pre-training, BPR (1 mg/kg) impaired learning and memory processing; DZP (1 and 2 mg/kg) evoked anxiolysis, but only at the dose of 2 mg/kg impaired memory; and NPS 0.1 nmol induced anxiolysis without affecting memory. Post-training injection of DZP (2 mg/kg) or BPR (1 and 3 mg/kg) did not affect memory consolidation, while the post-trainning administration of NPS 1 nmol, but not 0.1 nmol, improved memory in mice. Indeed, pre-trainning administration of NPS 1 nmol did not prevent memory impairment elicited by BPR (2 mg/kg, injected before training). In the open field test, BPR 1 mg/kg and NPS 1 nmol induced hyperlocomotion in mice. In conclusion, the proposed ETM task is practical for the detection of the anxiolytic and amnesic effects of drugs. The anxiolytic and memory enhancement effects of NPS were detected in the ETM task, and reinforce the role of NPS system as an interesting therapeutic target to the treatment of anxiety disorders

PURPOSE: To compare the 2% ibopamine provocative test with the water drinking test as a provocative test for glaucoma. METHODS: Primary open-angle glaucoma patients and normal individuals were selected from CEROF-Universidade Federal de Goiânia UFG, and underwent the 2% ibopamine provocative test and the water drinking test in a randomized fashion, at least 1 week apart. Intraocular pressure (IOP) before and after both tests, Bland-Altman graph, sensitivity and specificity (as mesured by ROC curves) were obtained for both methods. RESULTS: Forty-seven eyes from 25 patients were included (27 eyes from 15 glaucoma patients and 20 eyes from 10 normal individuals), with a mean age of 54.2 ± 12.7 years. The mean MD of glaucoma patients was -2.8 ± 2.11 dB. There was no statistically difference in the baseline IOP (p=0.8) comparing glaucoma patients, but positive after the provocative tests (p=0.03), and in the IOP variation (4.4 ± 1.3 mmHg for ibopamine and 3.2 ± 2.2 mmHg for water drinking test, p=0.01). There was no difference in all studied parameters for normal individuals. The Bland-Altman graph showed high dispersion comparing both methods. The areas under the ROC curve were 0.987 for the ibopamine provocative test, and 0.807 for the water-drinking test. CONCLUSION: In this selected subgroup of glaucoma patients with early visual field defect, the ibopamine provocative test has shown better sensitivity/specificity than the water drinking test.

The first idea of this study is trat there may be a disturbance of the visuo-spatial function in cerebral palsy. This idea is based upon the theories of Luria and Piaget. The second idea of this study is that the computer can help the investigation of visuo-spatial function. With the intention of evaluating the spatial function in cerebral palsy, the literature related to these themes was revised. We justify the proposal of a new test to study visuo-spatial function. This test uses straight lines in four different orientations and the subject has to recognize and arrange them individually or sets of them. This was done through the use of pencil and papel, and computer. The results confirm that there is a spatial disturbance and that a better understanding of the subject's difficulties was made possible through the use of the computer and the Logo computer language.
