O objetivo deste trabalho foi verificar o efeito da glutationa (GSH – 0,3mg/mL), no processo de separação, lavagem e capacitação espermática sobre a taxa de produção in vitro de embriões bovinos. O sêmen de um touro foi separado através de gradientes de Percoll e centrifugados duas vezes em meio de fecundação in vitro (lavagem). Espermatozóides foram capacitados in meio de FIV durante 1 h em estufa úmida a 38,5°C e 5% de CO2. Os grupos experimentais foram: Controle (não possui a etapa de lavagem; Grupo 1: sem GSH nas três etapas; Grupo2: GSH somente na capacitação; Grupo3: GSH somente na separação e na lavagem (primeira lavagem); Grupo 4: GSH em todas as etapas. Oócitos provenientes de abatedouro foram maturados in vitro e fertilizados após as preparações feitas no sêmen. Os embriões foram cultivados durante 7 dias com análise da clivagem no 2º dia, taxa de blastocisto e número total de células embrionárias no 7º dia. As taxas de desenvolvimento embrionário foram analisadas pelo qui-quadrado (χ2) e a qualidade embrionária pela ANOVA através do programa Bio Estat 3.0 (AYRES, et al., 2003) com nível de significância de 5%. Não houve diferença estatística efeito do uso da GSH nas diferentes etapas do processo de preparação espermática sobre as taxas de clivagem, blastocisto e eclosão (Grupo Controle: 52, 35 e 50%; Grupo 1: 60, 37 e 44%; Grupo 2: 57, 40 e 50; Grupo 3: 56, 37 e 51%; Grupo 4: 47, 30 e 48%, respectivamente, p>0,05). Também não foi observado diferença estatística quanto ao número de células embrionárias (Grupo Controle: 30,9±21,87, Grupo 1: 57,8±18,26; Grupo 2: 66,2±20,14; Grupo 3: 62,1±22,48; Grupo 4: 76,7±29,28; p>0,05).

The Brazilian livestock stands out for having the world largest commercial herd of cattle and leads meat exportation and production of bovine embryos. The in vitro production (IVP) of embryos is considered an effective option to overcome problems such as infertility in cows with high economic value and also for genetic improvement of cattle. The in vitro oocyte maturation is an essential step to the success of IVP, but is still considered poor when compared to in vivo maturation. Recent studies have suggeested an important role of Fibroblast Growth Factor 10 (FGF10) on the in vitro maturation of oocytes, which favored the expression of genes related to oocyte maturation and cumulus cell expansion. Aware that maturity stage influences the final production of blastocysts, we aimed study to verify if the addition of FGF10 into the maturation medium is able to affect positively the IVP of bovine embryos. Hence, FGF10 was added to maturation in five different concentrations: 0.5 ng/mL (group 0.5), 2.5 ng/mL (group 2.5), 5 ng/mL (group 5), 10 ng/mL (group 10) and 50 ng/mL (group 50). Additionally, two other maturation groups were used, group BSA (Bovine Serum Albumin, 4 mg/mL) and group FCS (Fetal Calf Serum, 10%). The rates of cleavage, morula and blastocyst were analyzed by Analysis of Variance (ANOVA), differences of P<0.05 were considered significant. Cleavage rates did not differ between the seven groups. On the other hand, morula rate on FCS group was higher than groups BSA, 0.5, 10 and 50 (P<0.05), but did not differ among groups treated with intermediate doses of FGF10 (2.5 and 5). FCS group presented higher blastocyst rate compared to all other groups that were well below the FCS group (P<0.0001). Therefore, the use of FGF10 during oocyte maturation did not affect positively embryo development on the IVP of bovine embryos

We consider some problems of the calculus of variations on time scales. On the beginning our attention is paid on two inverse extremal problems on arbitrary time scales. Firstly, using the Euler-Lagrange equation and the strengthened Legendre condition, we derive a general form for a variation functional that attains a local minimum at a given point of the vector space. Furthermore, we prove a necessary condition for a dynamic integro-differential equation to be an Euler-Lagrange equation. New and interesting results for the discrete and quantum calculus are obtained as particular cases. Afterwards, we prove Euler-Lagrange type equations and transversality conditions for generalized infinite horizon problems. Next we investigate the composition of a certain scalar function with delta and nabla integrals of a vector valued field. Euler-Lagrange equations in integral form, transversality conditions, and necessary optimality conditions for isoperimetric problems, on an arbitrary time scale, are proved. In the end, two main issues of application of time scales in economic, with interesting results, are presented. In the former case we consider a firm that wants to program its production and investment policies to reach a given production rate and to maximize its future market competitiveness. The model which describes firm activities is studied in two different ways: using classical discretizations; and applying discrete versions of our result on time scales. In the end we compare the cost functional values obtained from those two approaches. The latter problem is more complex and relates to rate of inflation, p, and rate of unemployment, u, which inflict a social loss. Using known relations between p, u, and the expected rate of inflation π, we rewrite the social loss function as a function of π. We present this model in the time scale framework and find an optimal path π that minimizes the total social loss over a given time interval.
