Plants containing condensed tannins (CTs) may hold promise as alternatives to synthetic anthelmintic (AH) drugs for controlling gastrointestinal nematodes (GINs). However, the structural features that contribute to the AH activities of CTs remain elusive. This study probed the relationships between CT structures and their AH activities. Eighteen plant resources were selected based on their diverse CT structures. From each plant resource, two CT fractions were isolated and their in vitro AH activities were measured with the Larval Exsheathment Inhibition Assay, which was applied to Haemonchus contortus and Trichostrongylus colubriformis. Calculation of mean EC50 values indicated that H. contortus was more susceptible than T colubriformis to the different fractions and that the F1 fractions were less efficient than the F2 ones, as indicated by the respective mean values for H.contortus F1 = 136.9 ± 74.1 µg/ml; and for H.contortus F2 = 108.1 ± 53.2 µg/ml and for T colubriformis F1 = 233 ± 54.3 µg/ml and F2=166 ± 39.9 µg/ml. The results showed that the AH activity against H. contortus was associated with the monomeric subunits that give rise to prodelphinidins (P < 0.05) and with CT polymer size (P < 0.10). However, for T. colubriformis AH activity was correlated only with prodelphinidins (P < 0.05). These results suggest that CTs have different modes of action against different parasite species.

The purpose of the experiment was to evaluate infective Trichostrongylus colubriformis larvae vertical migration in two forage grass species. Experimental modules formed by eight plots, established with Brachiaria decumbens cv. Australian and Panicum maximum cv. Aruana, were used in the study, totaling four plots for each grass species. Each plot was divided into six 30 x 30 cm subplots. Larval migration was evaluated in the four seasons of the year, in different plant strata (0-7, 7-14, 14-21, 21-28 and above 28 cm). Four feces deposits were made, one in each season of the year, in the middle of 30-cm tall forage. The feces were collected from the forage ten days after each feces deposit in the experimental subplots. Grass height was measured in each of the strata immediately before the collections. The forage of the different strata was cut from an area measuring 10-cm in radius. The feces were collected manually from the subplots. There was a grass species and grass stratum interaction in the deposit made in autumn (P < 0.05). During that season, most of the larvae were recovered from the Brachiaria grass base; meanwhile, at the forage apex, the biggest average was registered in the aruana grass. Infective larvae (U) recovery was similar among the different strata during spring. In springtime, the biggest L3 recovery occurred at the 21-28 cm stratum from both forage species. No L3 was recovered from any of the No L3 was recovered from any of the grass strata during winter and summer. Study results show that migration of T colubriformis larvae was more influenced by weather conditions Than by forage species.

O experimento teve como objetivo avaliar a recuperação de larvas infectantes (L3) de Trichostrongylus colubriformis em Brachiaria decumbens cv. Australiana, Cynodon dactilon cv. Coast-cross e Panicum maximum cv. Aruana. Foram utilizados módulos experimentais constituídos por seis canteiros de 32,4 m² cada, perfazendo dois canteiros por espécie forrageira. Cada canteiro foi dividido em 36 parcelas, de 30 x 30 cm, de forma a permitir seis repetições por espécie e por altura da forragem em cada semana de colheita de material. A sobrevivência larval foi avaliada do meio do verão e até meados do outono, sob o efeito de duas alturas de poda das forragens: baixa, 5 cm e alta, 30 cm. A poda foi realizada imediatamente antes da deposição das fezes contaminadas com ovos de T. colubriformis, obtidos de ovinos, que ocorreu no dia 05/02/ 2004. A colheita das fezes e da forragem foi realizada uma, duas, quatro, oito, 12 e 16 semanas após a deposição das fezes nos canteiros experimentais. A altura da forragem foi medida em cada uma das subdivisões imediatamente antes da colheita. A forragem foi cortada rente ao solo, de uma área delimitada com o auxílio de um círculo de 10 cm de raio. As fezes foram recolhidas manualmente dos canteiros. O número de larvas infectantes recuperado foi muito pequeno em comparação com a quantidade de larvas produzidas nas culturas controle, mantidas no laboratório (máximo de 6,7% no capim Aruana com 30 cm de altura). Arecuperação de L3 das amostras fecais foi maior quando as fezes foram depositadas em meio ao capim alto (com 30 cm - P<0,05). Porém, a recuperação de L3 das forragens foi similar em ambos os cortes. em relação à concentração de L3 (número de L3/kg de matéria seca), não houve diferença entre o corte baixo e alto (P>0,05) em nenhuma das semanas experimentais. Dentre as espécies forrageiras, o capim aruana foi o que, no geral, apresentou maiores concentrações de L3 de T. colubriformis.

Avaliaram-se a associação entre o número de células inflamatórias no intestino delgado e a resistência à infecção por Trichostrongylus colubriformis em ovinos de três raças (Santa Inês, Suffolk e Ile de France), naturalmente infectados. Mastócitos, eosinófilos e leucócitos globulares foram quantificados na mucosa intestinal. A concentração de histamina foi estimada em amostras teciduais do intestino, bem como foi determinado o comprimento de machos e fêmeas de T. colubriformis. A resposta celular foi similar na mucosa intestinal das três raças ovinas (P>0,05). Houve grande variação entre os ovinos em relação aos resultados parasitológicos e celulares, mesmo nos animais de mesma raça. em geral, os animais que apresentaram número menor de células inflamatórias tiveram cargas parasitárias maiores, contagens de ovos por grama de fezes mais altas e exemplares de T. colubriformis maiores. Os resultados indicaram que mastócitos, eosinófilos e leucócitos globulares prejudicaram o estabelecimento, o desenvolvimento e a sobrevivência dos parasitas.

The objective of this study was to determine the level of resistance of Haemonchus contortus and Trichostrongylus colubriformis in sheep to levamisole albendazole ivermectin moxidectin closantel and trichlorfon The parasites were isolated from sheep naturally infected by gastrointestinal nematodes and were then kept in monospecifically infected lambs for production of infective larvae (L3) of both species Forty-two lambs at three months of age were simultaneously artificially infected with 4000 L3 of H contortus and 4000 L3 of T colubriformis The animals were allocated Into seven groups with six animals each that received one of the following treatments Group 1-control no treatment Group 2-moxidectin (0 2 mg/kg body weight (BW)) Group 3-closantel (10 mg/kg BW) Group 4-trichlorfon (100 mg/kg BW) Group 5-levamisole phosphate (4 7 mg/kg BW) Group 6-albendazole (5 0 mg/kg BW) and Group 7-ivermectin (0 2 mg/kg BW) Nematode fecal egg counts (FEC) were carried out on the day of treatment and again at 3 7 10 and 14 days post-treatment on the same occasions composite fecal cultures were prepared for each group for production of L3 which were identified into genus The animals were sacrificed for worm counts at 14 days after treatment The efficacy of each treatment was calculated from the arithmetic mean of the FEC or worm burden of the treated group compared with the values of the control group Only trichlorfon and moxidectin treatments resulted in a significant reduction of H contortus recorded at necropsy (73% and 45% respectively) Moxidectin reduced T colubriformis worm burdens by 82% and albendazole by 19% All other anthelmintics resulted in no significant reduction in the numbers of worms found at necropsy In conclusion the Isolates of H contortus and T colubriformis showed multiple resistance to all groups of anthelmintics tested This is the first report based on the controlled efficacy test to show resistance of T colubriformis to macrocyclic lactones in Brazil (C) 2010 Elsevier B.V. The survival of infective larvae (L3) of Trichostrongylus colubriformis was evaluated on Brachiaria, Coast-cross and Aruana forage grasses. Feces of sheep parasitized exclusively by T. colubriformis were deposited in winter and spring on experimental plots whose grasses were cut at two heights: 5 cm and 30 cm. One, two, four, eight, 12 and 16 weeks after depositing the feces, fecal and forage samples were collected for the retrieval and quantification of L3. Retrieval of L3 from feces and forage was negligible in winter due to the dry weather, although a few larvae were retrieved in the last larval collections. However, L3 retrieval from fecal samples was greater in spring, especially two weeks after feces were deposited on 30 cm high grasses. At this time, the L3 retrieval rate from the three forage grasses differed significantly (P < 0.05), with Aruana grass showing the highest average L3 retrieval rate, followed by Coast-cross and Brachiaria. In conclusion, the winter drought proved very unfavorable for the presence of L3 in the environment, and the microclimate of Aruana pastureland was generally the most favorable for the retrieval of infective larvae.

Objective To establish the prevalence of anthelmintic resistance in ovine gastrointestinal nematodes in southern Queensland. Design An observational parasitological study using the faecal egg count reduction test. Methods Sheep farms (n = 20) enrolled in this study met the twin criteria of using worm testing for drench decisions and having concerns about anthelmintic efficacy. On each farm, 105 sheep were randomly allocated to one of six treatment groups or an untreated control group. Faecal samples were collected on day 0 and days 10–14 for worm egg counts and larval differentiation. Single- and multi-combination anthelmintics, persistent and non-persistent, oral liquid or capsule, pour-on and injectable formulations were tested. Monepantel was not tested. Farmers also responded to a questionnaire on drenching practices. Results Haemonchus contortus was the predominant species. Efficacy <95% was recorded on 85% of farms for one or more anthelmintics and on 10% of farms for six anthelmintics. No resistance was identified on three farms. The 4-way combination product was efficacious (n = 4 farms). Napthalophos resistance was detected on one farm only. Resistance to levamisole (42% of farms), moxidectin injection (50% of farms) and the closantel/abamectin combination (67% of farms) was identified. Moxidectin oral was efficacious against Trichostrongylus colubriformis, which was predominant on only one farm. Of the farms tested, 55% ran meat breeds, 60% dosed more than the recommended dose rate and 70% always, mostly or when possible practised a ‘drench and move’ strategy. Conclusion This level of anthelmintic resistance in southern Queensland will severely compromise worm control and force increased use of monepantel.
