Informe que mide la implantación de los servicios de subtitulado y audiodescripción en las televisiones de la TDT, así como el nivel de satisfacción de los usuarios y que también analiza las tecnologías que hacen posible la accesibilidad audiovisual. En él se desvela que la evolución de la prestación de los servicios de subtitulado ha sido muy significativa y en cuatro años prácticamente se ha duplicado la cantidad de programación subtitulada en la TDT. En torno al 90% en las televisiones de ámbito público y al 75% en los canales comerciales han implantado el servicio de subtitulado. Los servicios de audiodescripción también han aumentado desde las 27 horas en 2011 a las 120 del 2014. Sin embargo, aún no se cumple con la obligación legal de audiodescribir 10 horas semanales en los canales públicos y 2 horas semanales en los canales comerciales. Con respecto a la opinión de los usuarios se observa un claro avance en la satisfacción global de éstos con el subtitulado, aunque también reclaman la mejora en su calidad, que se incorpore en las diferentes plataformas audiovisuales, que su distribución en franjas horarias sea acorde al consumo que se hace de la televisión, y que la señalización sea correcta. Respecto a la la audidescripción, la satisfacción de los usuarios también mejora respecto a la encuesta de 2010, pero todavía se encuentra en niveles muy bajos.

Este artículo detalla el diseño, implementación y validación de un sistema abierto de Televisión Digital accesible para personas con deficiencia visual. La solución facilita que este colectivo pueda acceder a los contenidos de la guía electrónica de programación recibidos a través de la TDT, pudiendo configurar a demanda la interfaz de usuario gráfica (IGU) y la síntesis de voz (TTS). Se presenta el análisis de sistemas existentes, requisitos demandados por este colectivo y tecnologías disponibles, tanto de TDT como de TTS, con el fin de elegir las más apropiadas con criterios de accesibilidad, interoperabilidad y bajo coste. Los resultados obtenidos en la investigación demuestran que el sistema desarrollado es congruente con los criterios del Diseño para Todos según constata la validación realizada. El sistema ofrece combinaciones de colores y fuentes (contraste, tamaño) para diferentes necesidades de la deficiencia visual, y utiliza TTS local y adaptable para las personas con ceguera.

Este proyecto consiste en el diseño completo, de una red de distribución de TDT, a nivel local, mediante difusión SFN, Single Frequency Network. Este tipo de difusión, tiene la capacidad de difundir los servicios de televisión en una única frecuencia, cubriendo un área, ya sea local o estatal, aprovechando en las zonas de interferencia los rebotes de la señal y así evitar el uso de una frecuencia distinta por cada centro de emisión, todos los que componen un área de cobertura. Para el diseño de la red, se ha optado por diseñar una red IP, mediante distribución multicast, ya que esta es la tecnología imperante a día de hoy, quedando obsoleta ya, la distribución analógica, ya que consume muchos más recursos y por consiguiente mucho más costosa de implementar. El documento se divide en cuatro capítulos. En el primer capítulo se realizará una introducción teórica a las redes de distribución SFN, centrándose en el cálculo de los retardos, punto fundamental en el diseño de este tipo de redes. Se continuará unas nociones básicas de redes IP y el protocolo multicast, en el que se basa el trasporte de la señal. El capítulo dos, se centra en el diseño de la red, desde los centros de producción, donde se generan los programas a emitir, hasta los diferentes centros de difusión que cubrirán todo el área de cobertura requerida, pasando por el centro de multiplexación, donde se sitúa la cabecera que compondrá el múltiplex a difundir. Se describirán los equipos y el diseño de los distintos centros que conforman la red, centros de producción, multiplexación y difusión. A demás se realizará el cálculo de retardo de la señal, necesario en este tipo de redes. Se continuará con el capítulo tres, donde se describirá la configuración de la red, tanto a nivel de equipamiento, como el diseño y asignación IP de toda la red, separando la red de servicio de la red de gestión para una mayor confiabilidad y eficiencia de la red. Se finalizará con la descripción de la gestión de la red, que mediante diferentes herramientas, proporcionan un monitoreado en tiempo real de todo el sistema, dando la posibilidad de adelantarsey previniendo posibles incidencias que, puedan causar alguna deficiencia en el servicio que se entrega al usuario final. ABSTRACT. This project involves the complete design of a network´s TDT distribution, locally, by broadcast SFN (Single Frequency Network). This type of broadcast, has the ability to broadcast television´s services on a single frequency, covering an area, whether local or state, drawing on the interference zones, signal´s rebounds, to avoid the use of a different frequency each broadcast center, all those who make a coverage area. For the design of the network, has been chosen to design an IP network using multicast distribution, since this is the prevailing technology today, as the analogue distribution, consumes more resources and therefore, much more costly to implement. The document is divided into four chapters. In the first chapter you can find a theoretical introduction to SFN distribution networks, focusing on the calculation of delays, fundamental point, in the design of these networks. A basic understanding of IP networks and the multicast protocol, in which the transport of the signal is based, will continue. Chapter two focuses on the design of the network, from production centers, where the programs are created to broadcast, to different distribution centers covering the entire area of coverage required, through the multiplexing center, where the head is located, which comprise the multiplex. Also, the equipment and design of the various centers in the network, production centers, multiplexing center and distribution centers, are described. Furthermore, the calculation of signal delay, necessary in such networks, is performed. We will continue with the chapter three, where the network configuration will be described, both in termsofequipment, such as design IP mapping of the entire network, separating the service network and management network, for increased the reliability and efficiency of the network. It will be completed with the description of the management of the network, using different tools provide real-time monitoring of the entire system, making it possible, to anticipate and prevent any incidents that might cause a deficiency in the service being delivered to final user.

La presente tesis doctoral, consistente en una investigación basada en la técnica de encuesta, tiene el propósito de estudiar la relación entre la práctica del deporte y el desarrollo de determinadas competencias genéricas en jóvenes universitarios. Su utilidad e interés se justifica en último extremo, en argumentos de calidad, en la dimensión de funcionalidad de la universidad (Orden, Garduño, Asensio, Mafokozi, González & Muñoz, 2002, Orden 2009), que para realizar su misión de manera excelente, además de conseguir la eficacia y eficiencia, debe dar respuesta a las necesidades de la sociedad en la que se halla inserta. Las universidades se ven inmensas en un proceso de rendición de cuentas que las obliga a evaluar la calidad de sus procesos y de sus productos. En este sentido, se han establecido sistemas de garantía interna de calidad y se llevan a cabo tareas de acreditación y certificación, con el fin de garantizar que se cumplen unos determinados criterios de gestión de calidad. Estas evaluaciones, si bien están diseñadas para que repercutan en la mejora de la enseñanza y, por ende, de los resultados educativos de los estudiantes, se centran más minuciosamente en la evaluación docente, la evaluación de titulación o la evaluación de los niveles de satisfacción de los usuarios del servicio educativo, especialmente los alumnos. En la evaluación de calidad, uno de los aspectos que se contemplan es la empleabilidad de los estudiantes, que resulta clave en un proceso evaluativo de unas instituciones cuyos egresados se ven obligados a responder ante la apremiante presión de eficiencia que demanda el mercado laboral, en donde se busca que el aspirante a cualquier puesto de trabajo haya alcanzado las competencias específicas y genéricas que le habiliten como profesional cualificado...

Bananas are susceptible to a diverse range of biotic and abiotic stresses, many of which cause serious production constraints worldwide. One of the most destructive banana diseases is Fusarium wilt caused by the soil-borne fungus, Fusarium oxysporum f. sp. cubense (Foc). No effective control strategy currently exists for this disease which threatens global banana production. Although disease resistance exists in some wild bananas, attempts to introduce resistance into commercially acceptable bananas by conventional breeding have been hampered by low fertility, long generation times and association of poor agronomical traits with resistance genes. With the advent of reliable banana transformation protocols, molecular breeding is now regarded as a viable alternative strategy to generate disease-resistant banana plants. Recently, a novel strategy involving the expression of anti-apoptosis genes in plants was shown to result in resistance against several necrotrophic fungi. Further, the transgenic plants showed increased resistance to a range of abiotic stresses. In this thesis, the use of anti-apoptosis genes to generate transgenic banana plants with resistance to Fusarium wilt was investigated. Since water stress is an important abiotic constraint to banana production, the resistance of the transgenic plants to water stress was also examined. Embryogenic cell suspensions (ECS) of two commercially important banana cultivars, Grand Naine (GN) and Lady Finger (LF), were transformed using Agrobacterium with the anti-apoptosis genes, Bcl-xL, Bcl-xL G138A, Ced-9 and Bcl- 2 3’ UTR. An interesting, and potentially important, outcome was that the use of anti-apoptosis genes resulted in up to a 50-fold increase in Agrobacterium-mediated transformation efficiency of both LF and GN cells over vector controls. Regenerated plants were subjected to a complete molecular characterisation in order to detect the presence of the transgene (PCR), transcript (RT-PCR) and gene product (Western blot) and to determine the gene copy number (Southern blot). A total of 36 independently-transformed GN lines (8 x Bcl-xL, 5 x Bcl-xL G138A, 15 x Ced-9 and 8 x Bcl-2 3’ UTR) and 41 independently-transformed LF lines (8 x Bcl-xL, 7 x BclxL G138A, 13 x Ced-9 and 13 x Bcl-2 3’ UTR) were identified. The 41 transgenic LF lines were multiplied and clones from each line were acclimatised and grown under glasshouse conditions for 8 weeks to allow monitoring for phenotypic abnormalities. Plants derived from 3 x Bcl-xL, 2 x Ced-9 and 5 x Bcl-2 3’ UTR lines displayed a variety of aberrant phenotypes. However, all but one of these abnormalities were off-types commonly observed in tissue-cultured, non-transgenic banana plants and were therefore unlikely to be transgene-related. Prior to determining the resistance of the transgenic plants to Foc race 1, the apoptotic effects of the fungus on both wild-type and Bcl-2 3’ UTR-transgenic LF banana cells were investigated using rapid in vitro root assays. The results from these assays showed that apoptotic-like cell death was elicited in wild-type banana root cells as early as 6 hours post-exposure to fungal spores. In contrast, these effects were attenuated in the root cells of Bcl-2 3’ UTR-transgenic lines that were exposed to fungal spores. Thirty eight of the 41 transgenic LF lines were subsequently assessed for resistance to Foc race 1 in small-plant glasshouse bioassays. To overcome inconsistencies in rating the internal (vascular discolouration) disease symptoms, a MatLab-based computer program was developed to accurately and reliably assess the level of vascular discolouration in banana corms. Of the transgenic LF banana lines challenged with Foc race 1, 2 x Bcl-xL, 3 x Ced-9, 2 x Bcl-2 3’ UTR and 1 x Bcl-xL G138A-transgenic line were found to show significantly less external and internal symptoms than wild-type LF banana plants used as susceptible controls at 12 weeks post-inoculation. Of these lines, Bcl-2 3’ UTR-transgenic line #6 appeared most resistant, displaying very mild symptoms similar to the wild-type Cavendish banana plants that were included as resistant controls. This line remained resistant for up to 23 weeks post-inoculation. Since anti-apoptosis genes have been shown to confer resistance to various abiotic stresses in other crops, the ability of these genes to confer resistance against water stress in banana was also investigated. Clonal plants derived from each of the 38 transgenic LF banana plants were subjected to water stress for a total of 32 days. Several different lines of transgenic plants transformed with either Bcl-xL, Bcl-xL G138A, Ced-9 or Bcl-2 3’ UTR showed a delay in visual water stress symptoms compared with the wild-type control plants. These plants all began producing new growth from the pseudostem following daily rewatering for one month. In an attempt to determine whether the protective effect of anti-apoptosis genes in transgenic banana plants was linked with reactive oxygen species (ROS)-associated programmed cell death (PCD), the effect of the chloroplast-targeting, ROS-inducing herbicide, Paraquat, on wild-type and transgenic LF was investigated. When leaf discs from wild-type LF banana plants were exposed to 10 ìM Paraquat, complete decolourisation occurred after 48 hours which was confirmed to be associated with cell death and ROS production by trypan blue and 3,3-diaminobenzidine (DAB) staining, respectively. When leaf discs from the transgenic lines were exposed to Paraquat, those derived from some lines showed a delay in decolourisation, suggesting only a weak protective effect from the transgenes. Finally, the protective effect of anti-apoptosis genes against juglone, a ROS-inducing phytotoxin produced by the causal agent of black Sigatoka, Mycosphaerella fijiensis, was investigated. When leaf discs from wild-type LF banana plants were exposed to 25 ppm juglone, complete decolourisation occurred after 48 hours which was again confirmed to be associated with cell death and ROS production by trypan blue and DAB staining, respectively. Further, TdT-mediated dUTP nick-end labelling (TUNEL) assays on these discs suggested that the cell death was apoptotic. When leaf discs from the transgenic lines were exposed to juglone, discs from some lines showed a clear delay in decolourisation, suggesting a protective effect. Whether these plants are resistant to black Sigatoka is unknown and will require future glasshouse and field trials. The work presented in this thesis provides the first report of the use of anti-apoptosis genes as a strategy to confer resistance to Fusarium wilt and water stress in a nongraminaceous monocot, banana. Such a strategy may be exploited to generate resistance to necrotrophic pathogens and abiotic stresses in other economically important crop plants.

The progesterone receptor (PR) is a candidate gene for the development of endometriosis, a complex disease with strong hormonal features, common in women of reproductive age. We typed the 306 base pair Alu insertion (AluIns) polymorphism in intron G of PR in 101 individuals, estimated linkage disequilibrium (LD) between five single-nucleotide polymorphisms (SNPs) across the PR locus in 980 Australian triads (endometriosis case and two parents) and used transmission disequilibrium testing (TDT) for association with endometriosis. The five SNPs showed strong pairwise LD, and the AluIns was highly correlated with proximal SNPs rs1042839 (Δ2 = 0.877, D9 = 1.00, P < 0.0001) and rs500760 (Δ2 = 0.438, D9 = 0.942, P < 0.0001). TDT showed weak evidence of allelic association between endometriosis and rs500760 (P = 0.027) but not in the expected direction. We identified a common susceptibility haplotype GGGCA across the five SNPs (P = 0.0167) in the whole sample, but likelihood ratio testing of haplotype transmission and non-transmission of the AluIns and flanking SNPs showed no significant pattern. Further, analysis of our results pooled with those from two previous studies suggested that neither the T2 allele of the AluIns nor the T1/T2 genotype was associated with endometriosis.

A whole-genome scan was conducted to map quantitative trait loci (QTL) for BSE resistance or susceptibility. Cows from four half-sib families were included and 173 microsatellite markers were used to construct a 2835-cM (Kosambi) linkage map covering 29 autosomes and the pseudoautosomal region of the sex chromosome. Interval mapping by linear regression was applied and extended to a multiple-QTL analysis approach that used identified QTL on other chromosomes as cofactors to increase mapping power. In the multiple-QTL analysis, two genome-wide significant QTL (BTA17 and X/Y ps) and four genome-wide suggestive QTL (BTA1, 6, 13, and 19) were revealed. The QTL identified here using linkage analysis do not overlap with regions previously identified using TDT analysis. One factor that may explain the disparity between the results is that a more extensive data set was used in the present study. Furthermore, methodological differences between TDT and linkage analyses may affect the power of these approaches.

The power of testing for a population-wide association between a biallelic quantitative trait locus and a linked biallelic marker locus is predicted both empirically and deterministically for several tests. The tests were based on the analysis of variance (ANOVA) and on a number of transmission disequilibrium tests (TDT). Deterministic power predictions made use of family information, and were functions of population parameters including linkage disequilibrium, allele frequencies, and recombination rate. Deterministic power predictions were very close to the empirical power from simulations in all scenarios considered in this study. The different TDTs had very similar power, intermediate between one-way and nested ANOVAs. One-way ANOVA was the only test that was not robust against spurious disequilibrium. Our general framework for predicting power deterministically can be used to predict power in other association tests. Deterministic power calculations are a powerful tool for researchers to plan and evaluate experiments and obviate the need for elaborate simulation studies.

Population-wide associations between loci due to linkage disequilibrium can be used to map quantitative trait loci (QTL) with high resolution. However, spurious associations between markers and QTL can also arise as a consequence of population stratification. Statistical methods that cannot differentiate between loci associations due to linkage disequilibria from those caused in other ways can render false-positive results. The transmission-disequilibrium test (TDT) is a robust test for detecting QTL. The TDT exploits within-family associations that are not affected by population stratification. However, some TDTs are formulated in a rigid-form, with reduced potential applications. In this study we generalize TDT using mixed linear models to allow greater statistical flexibility. Allelic effects are estimated with two independent parameters: one exploiting the robust within-family information and the other the potentially biased between-family information. A significant difference between these two parameters can be used as evidence for spurious association. This methodology was then used to test the effects of the fourth melanocortin receptor (MC4R) on production traits in the pig. The new analyses supported the previously reported results; i.e., the studied polymorphism is either causal of in very strong linkage disequilibrium with the causal mutation, and provided no evidence for spurious association.

Utilizing DNA samples from 91 Afrikaner nuclear families with one or more affected children, five genomic regions on chromosomes 2p, 8q, 11q, 20q, and 21q that gave evidence for association with GTS in previous case-control association studies were investigated for linkage and association with GTS. Highly polymorphic markers with mean heterozygosity of 0.77 were typed and resulting genotypes evaluated using single marker transmission disequilibrium (TDT), single marker haplotype relative risk (HRR), and multi-marker "extended" TDT and HRR methods. Single marker TDT analysis showed evidence for linkage or association, with p-values near 0.05, for markers D2S139, GATA28F12, and D11S1377 on chromosomes 2p11, 8q22 and 11q23-24, respectively. Extended, two-locus TDT and HRR analysis provided further evidence for linkage or association on chromosome 2 with p-values of 0.007 and 0.025, and chromosome 8 with p-values of 0.059 and 0.013, respectively. These results provide important additional evidence for the location of GTS susceptibility loci.

Topic detection and tracking (TDT) is an area of information retrieval research the focus of which revolves around news events. The problems TDT deals with relate to segmenting news text into cohesive stories, detecting something new, previously unreported, tracking the development of a previously reported event, and grouping together news that discuss the same event. The performance of the traditional information retrieval techniques based on full-text similarity has remained inadequate for online production systems. It has been difficult to make the distinction between same and similar events. In this work, we explore ways of representing and comparing news documents in order to detect new events and track their development. First, however, we put forward a conceptual analysis of the notions of topic and event. The purpose is to clarify the terminology and align it with the process of news-making and the tradition of story-telling. Second, we present a framework for document similarity that is based on semantic classes, i.e., groups of words with similar meaning. We adopt people, organizations, and locations as semantic classes in addition to general terms. As each semantic class can be assigned its own similarity measure, document similarity can make use of ontologies, e.g., geographical taxonomies. The documents are compared class-wise, and the outcome is a weighted combination of class-wise similarities. Third, we incorporate temporal information into document similarity. We formalize the natural language temporal expressions occurring in the text, and use them to anchor the rest of the terms onto the time-line. Upon comparing documents for event-based similarity, we look not only at matching terms, but also how near their anchors are on the time-line. Fourth, we experiment with an adaptive variant of the semantic class similarity system. The news reflect changes in the real world, and in order to keep up, the system has to change its behavior based on the contents of the news stream. We put forward two strategies for rebuilding the topic representations and report experiment results. We run experiments with three annotated TDT corpora. The use of semantic classes increased the effectiveness of topic tracking by 10-30\% depending on the experimental setup. The gain in spotting new events remained lower, around 3-4\%. The anchoring the text to a time-line based on the temporal expressions gave a further 10\% increase the effectiveness of topic tracking. The gains in detecting new events, again, remained smaller. The adaptive systems did not improve the tracking results.

一. 小麦相关基因ver203F cDNA全序列克隆与功能分析 根据本实验室通过差异筛选技术克隆到的与春化相关的基因cDNA verc203的序列,设计PCR 5’端PCR引物,利用RACE(rapid amplification of cDNA ends)克隆策略,得到春化相关基因ver203基因的同源基因ver203F cDNA的3’端序列,长度为1,197bp。Northern分析表明ver203F全长约为1.5 kb,且其表达具有春化处理的特异性。根据3’RACE克隆的ver203F 3 ’端核苷酸序列设计了3’端PCR引物,利用5’RACE克隆到该基因的5’端片段,经DNASIS核酸分析软件分析将5’45RACE和3’RACE DNA序列拼接合并,得到ver203F全长cDNA,从TdT加尾5’末端到poly A全长为1,561 bp,5’端起始密码子ATG上游非编码区-1~-192共了192bp,终止密码子TGA到poly A的非编码区有253bp,cDNA编码区全长1,119 bp,推测编码373氨基酸残基。国际基因序列数据库检索表明该基因序列(GenBank/EMBL/DDBJ:AB012013)与大麦茉莉酸诱导基因有部分同源性。因上推测该基因在调控开花过程中可能参与茉莉酸介导的信号传导途径,ver203F作用的发挥可能需要其它蛋白的参与,或ver203F本身就是一个受体蛋白。 为了研究ver203F基因的功能,将通过3’RACE克隆到的ver203F 3’端序到分别构建正义和反义植物表达载体,通过花粉管通道法、农杆菌介导的叶圆片法以及农杆菌介导的真空转化法分别转化小麦、烟草和拟南芥菜。获得转基因植株后,PCR、DNA Dob Blot、Southern Blot分析以及GUS活性检测证明外源基因已整合到转基因植株中,并得到表达。在获得的小麦、烟草和拟南芥菜转基因植株中,它们开花时间都相应地推迟,表明正常植物体内该基因在控制营养生长向生殖生长的转变中起作用。ver203F可以影响小麦和拟南茶菜花序的发育,首先无论正义还是反义都使得花序的发育受到抑制,在小麦中表现为顶部小穗退化,在拟南芥菜中表现为顶花。其次在转化正义基因的转基因拟南芥菜中,观察到产生的顶花为对称的两朵或以对称的两朵顶花基部为生长点长出丛生花,这种对称花的麦型小麦小穗中小花的表型相类似,说明ver203F基因可能在小麦小花的发育过程中也起着重要作用。 二. 春化相关基因ver17在开花过程中功能的分析 以春化处理冬小麦(京冬1号)幼苗cDNA为材料,通过减法杂交与差异筛选得到春化相关cDNA克隆verc17。为了研究该基因的功能,以包含CaMV 35S启动子的pBI221为载体,将ver17cDNA片段分别从两个方向插入pBI221的BamH I-Sma I, Xba I-BamH I间,构建正义和反义表达质粒:p17S和p17X,通过花粉管通道法转化小麦。对T0和T1两代转基因小麦的观察发现,在转化反义基因p17X的转基因小麦首先表现为抽穗推迟,其次穗的顶部和基部小穗严重退化,另外还发现转化反义基因的小麦败育现象严重(主要是花粉败育),因此推测ver17基困能可具有以下几方面的特点:a.春化诱导型表达;b.促进植物开花;c.促进穗顶端和基部花的发育,减少其退化;d.影响雄蕊的发育。

C1q family proteins with C1q domain have been reported in vertebrates, but their biological roles are currently unknown. In this study, a C1q-like factor, designated Carassius auratus gibelio ovary-specific C1q-like factor (CagOC1q-like), was identified as a cortical granules component. Immunofluorescence localization revealed that the C1q family member was specifically expressed in follicular epithelial cells, and associated with cortical granules in fully grown oocytes. Moreover, it was discharged to the perivitelline space and egg envelope upon fertilization. As it is the first identified C1q family member that is expressed in follicular cells that surround oocyte, CagOC1q-like was applied to detection of follicular cell apoptosis and deletion. The entire cytological process of follicular cell apoptosis and deletion was clearly seen from double visualizations of follicular cells with CagOC1q-like immunofluorescence and apoptotic follicular cells labeled by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) during oocyte maturation and ovulation.
