Background WSUD implementation in the Gold Coast City Council area commenced more than a decade ago. As a result, Council is expected to be in possession of WSUD assets valued at over tens of million dollars. The Gold Coast City Council is responsible for the maintenance and long-term management of these WSUD assets. Any shortcoming in implementation of best WSUD practices can potentially result in substantial liabilities and ineffective expenditure for the Council in addition to reduced efficiencies and outcomes. This highlights the importance of periodic auditing of WSUD implementation. Project scope The overall study entailed the following tasks: * A state-of-the-art literature review of the conceptual hydraulic and water quality treatment principles, current state of knowledge in relation to industry standards, best practice and identification of knowledge gaps in relation to maintenance and management practices and potential barriers to the implementation of WSUD. * Council stakeholder interviews to understand current practical issues in relation to the implementation of WSUD and the process of WSUD application from development application approval to asset management. * Field auditing of selected WSUD systems for condition assessment and identification of possible strengths and weaknesses in implementation. * Review of the Land Development Guidelines in order to identify any gaps and to propose recommendations for improvement. Conclusions Given below is a consolidated summary of the findings of the study undertaken. State-of-the-art literature review Though the conceptual framework for WSUD implementation is well established, the underlying theoretical knowledge underpinning the treatment processes and maintenance regimes and life cycle costing are still not well understood. Essentially, these are the recurring themes in the literature, namely, the inadequate understanding of treatment processes and lack of guidance to ensure specificity of maintenance regimes and life cycle costing of WSUDs. The fundamental barriers to successful WSUD implementation are: * Lack of knowledge transfer – This essentially relates to the lack of appropriate dissemination of research outcomes and the common absence of protocols for knowledge transfer within the same organisation. * Cultural barriers – These relate to social and institutional factors, including institutional inertia and the lack of clear understanding of the benefits. * Fragmented responsibilities – This results from poor administrative integration within local councils in relation to WSUDs. * Technical barriers – These relate to lack of knowledge on operational and maintenance practices which is compounded by model limitations and the lack of long-term quantitative performance evaluation data. * Lack of engineering standards – Despite the availability of numerous guidelines which are non-enforceable and can sometimes be confusing, there is a need for stringent engineering standards. The knowledge gaps in relation to WSUDs are only closing very slowly. Some of the common knowledge gaps identified in recent publications have been recognised almost a decade ago. The key knowledge gaps identified in the published literature are: * lack of knowledge on operational and maintenance practices; * lack of reliable methodology for identifying life cycle issues including costs; * lack of technical knowledge on system performance; * lack of guidance on retrofitting in existing developments. Based on the review of barriers to WSUD implementation and current knowledge gaps, the following were identified as core areas for further investigation: * performance evaluation of WSUD devices to enhance model development and to assess their viability in the context of environmental, economic and social drivers; establishing realistic life cycle costs to strengthen maintenance and asset management practices; * development of guidelines specific to retrofitting in view of the unique challenges posed by existing urban precincts together with guidance to ensure site specificity; establishment of a process for knowledge translation for enhancing currently available best practice guidelines; * identification of drivers and overcoming of barriers in the areas of institutional fragmentation, knowledge gaps and awareness of WSUD practices. GCCC stakeholder interviews Fourteen staff members involved in WSUD systems management in the Gold Coast City Council, representing four Directorates were interviewed using a standard questionnaire. The primary issues identified by the stakeholders were: * standardisation of WSUD terminology; * clear protocols for safeguarding devices during the construction phase; * engagement of all council stakeholders in the WSUD process from the initial phase; * limitations in the Land Development Guidelines; * ensuring public safety through design; * system siting to avoid conflicts with environmental and public use of open space; * provision of adequate access for maintenance; * integration of social and ecosystem issues to ensure long-term viability of systems in relation to both, vandalism and visual recreation; * lack of performance monitoring and inadequacy of the maintenance budget; * lack of technical training for staff involved in WSUD design approvals and maintenance; incentives for developers for acting responsibly in stormwater management. Field auditing of WSUD systems A representative cross section of WSUD systems in the Gold Coast were audited in the field. The following strengths and weaknesses in WSUD implementation were noted: * The implementation of WSUD systems in the field is not consistent. * The concerns raised by the stakeholders during the interviews in relation to WSUD implementation was validated from the observations from the field auditing, particularly in relation to the following: * safeguarding of devices during the construction phase * public safety * accessibility for maintenance * lack of performance monitoring by Council to assess system performance * inadequate maintenance of existing systems to suit site specific requirements. * A treatment train approach is not being consistently adopted. * Most of the systems audited have satisfactorily catered for public safety. Accessibility for maintenance has been satisfactorily catered for in most of the systems that were audited. * Systems are being commissioned prior to construction activities being substantially completed. * The hydraulic design of most systems appears to be satisfactory. * The design intent of the systems is not always clear. Review of Land Development Guidelines The Land Development Guidelines (TDG) was extensively reviewed and the following primary issues were noted in relation to WSUD implementation: * the LDG appears to have been prepared primarily to provide guidance to developers. It is not clear to what extent the guidelines are applicable to Council staff involved in WSUD maintenance and management; * Section 13 is very voluminous and appears to be a compilation of a series of individual documents resulting in difficulties in locating specific information, a lack of integration and duplication of information; * the LDG has been developed with a primary focus on new urban precinct development and the retrofitting of systems in existing developments has not been specifically discussed; * WSUDs are discussed in two different sections in the LDG and it is not clear which section takes precedence as there are inconsistencies between the two sections; there is inconsistent terminology being used; * there is a need for consolidation of information provided in different sections in the LDG; * there are inconsistencies in the design criteria provided; * there is a need for regular updating of the LDG to ensure that the information provided encompasses the state-of-the-art; * there is limited guidance provided for the preparation of maintenance plans and life cycle costing to assist developers in asset handover and to assist Council staff in assessment. * Based on these observations, eleven recommendations have been provided which are discussed below. Additionally, the stakeholder provided the following specific comments during the interviews in relation to the LDG: * lack of flexibility to cover the different stages of the life cycle of the systems; * no differentiation in projects undertaken by developers and Council; * inadequate information with regards to safety issues such as maximum standing water depth, fencing and safety barriers and public access; * lack of detailed design criteria in relation to Crime Prevention through Environmental Design, safety, amenity, environment, surrounding uses and impacts on surroundings; * inadequate information regarding maintenance requirements specific to the assessment and compliance phases; * recommendations for plantings are based primarily on landscape requirements rather than pollutant uptake capability. Recommendations With regards to the Land Development Guidelines, the following specific recommendations are provided: 1. the relevant sections and their extent of applicability to Council should be clearly identified; 2. integration of the different subsections within Section 13 and re-formatting the document for easy reference; 3. the maintenance guidelines provided in Section 13 should be translated to a maintenance manual for guidance of Council staff; 4. should consider extending the Guidelines to specifically encompass retrofitting of WSUD systems to existing urban precincts; 5. Section 3 needs to be revised to be made consistent with Section 13, to ensure priority for WSUD practices in urban precincts and to move away from conventional stormwater drainage design such as kerb and channelling; 6. it would also be good to specify as to which Section takes predominance in relation to stormwater drainage. It is expected that Section 13 would take predominance over the other sections in the LDG; 7. terminology needs to be made consistent to avoid confusion among developers and Council staff. Water Sensitive Urban Design is the term commonly used in Australia for stormwater quality treatment, rather than Stormwater Quality Improvement Devices. This once again underlines the need for ensuring consistency between Section 3 and Section 13; 8. it would also be good if there is a glossary of commonly used terms in relation to WSUD for use by all stakeholders and which should also be reflected in the LDG; 9. consolidation of all WSUD information into one section should be considered together with appropriate indicators in other LDG Sections regarding the availability of WSUD information. Ensuring consistency in the information provided is implied; 10. Section 13 should be updated at regular intervals to ensure the incorporation of the latest in research outcomes and incorporating criteria and guidance based on the state-of-the-art knowledge. The updating could be undertaken, say, in five year cycles. This would help to overcome the current lack of knowledge transfer; 11. the Council should consider commissioning specialised studies to extend the current knowledge base in relation to WSUD maintenance and life cycle costing. Additionally, Recommendation 10 is also applicable in this instance. The following additional recommendations are made based on the state-of-the-art literature review, stakeholder interviews and field auditing of WSUD systems: 1. Performance monitoring of existing systems to assess improvements to water quality, identify modifications and enhancements to improve performance; 2. Appropriate and monitored maintenance during different phases of development of built assets over time is needed to investigate the most appropriate time/phase of development to commission the final WSUD asset. 3. Undertake focussed investigations in the areas of WSUD maintenance and asset management in order to establish more realistic life cycle costs of systems and maintenance schedules; 4. the engagement of all relevant Council stakeholders from the initial stage of concept planning through to asset handover, and ongoing monitoring. This close engagement of internal stakeholders will assist in building a greater understanding of responsibilities and contribute to overcoming constraints imposed by fragmented responsibilities; 5. the undertaking of a public education program to inform the community of the benefits and ecosystem functions of WSUD systems; 6. technical training to impart state-of-the-art knowledge to staff involved in the approval of designs and maintenance and management of WSUD projects; 7. during the construction phase, it is important to ensure that appropriate measures to safeguard WSUD devices are implemented; 8. risks associated with potential public access to open water zones should be minimised with the application of appropriate safety measures; 9. system siting should ensure that potential conflicts are avoided with respect to public and ecosystem needs; 10. integration of social and ecosystem issues to ensure long-term viability of systems; provide incentives to developers who are proactive and responsible in the area of stormwater management.

Thymine DNA glycosylase (TDG) functions in base excision repair, a DNA repair pathway that acts in a lesion-specific manner to correct individual damaged or altered bases. TDG preferentially catalyzes the removal of thymine and uracil paired with guanine, and is also active on 5-fluorouracil (5-FU) paired with adenine or guanine. The rs4135113 single nucleotide polymorphism (SNP) of TDG is found in 10% of the global population. This coding SNP results in the alteration of Gly199 to Ser. Gly199 is part of a loop responsible for stabilizing the flipped abasic nucleotide in the active site pocket. Biochemical analyses indicate that G199S exhibits tighter binding to both its substrate and abasic product. The persistent accumulation of abasic sites in cells expressing G199S leads to the induction of double-strand breaks (DSBs). Cells expressing the G199S variant also activate a DNA damage response. When expressed in cells, G199S induces genomic instability and cellular transformation. Together, these results suggest that individuals harboring the G199S variant may have increased risk for developing cancer.

O câncer de esôfago é uma malignidade altamente freqüente e letal. Uma característica específica das áreas de alta incidência de câncer de esôfago é a grande proporção de duplas mutações no gene TP53, sendo, ao menos uma delas, uma transição G para A em sítios CpG. Essas transições resultam de malpareamentos GT causados pela desaminação espontânea da 5-metilcitosina em ilhotas CpG. A enzima de reparo de DNA Timina-DNA Glicosilase (TDG) é responsável pelo primeiro passo na remoção da timina de malpareamentos GT em CpG. A alta proporção de mutações em sítios CpG em câncer de esôfago das áreas de alta incidência sugere que a via de reparo de DNA iniciada pela TDG pode estar prejudicada. A presença de duplas mutações, sendo ao menos uma delas em CpG, levantou a hipótese de que a primeira mutação no TP53 reduz a atividade da via de reparo iniciada pela TDG, que acarretaria a segunda mutação em sítios CpG. Dessa forma, o objetivo desse trabalho foi analisar o efeito da p53 sobre a expressão e atividade da TDG. Os resultados obtidos mostram que a expressão de TDG é regulada transcricionalmente pela p53 numa gama de linhagens celulares e é induzida pelo dano ao DNA, de forma p53-dependente. Além disto, os resultados apontam um possível papel da proteína p53 ativa na migração nuclear e atividade da TDG. Estes resultados ainda nos levam à conclusão de que o silenciamento de TDG aumenta a sensibilidade à morte celular induzida por MMS quando a p53 é encontrada na forma selvagem, mas não quando esta proteína é mutada, e de que o status mutacional de TP53 parece afetar a expressão de TDG em CEE primários. Juntos esses resultados sugerem que a p53 regula o reparo de DNA mediado pela TDG e que a inativação de p53 em células tumorais pode contribuir para a aquisição de um mutator phenotype.

Diabetes mellitus e doenças periodontais são altamente prevalentes na população mundial. Doenças periodontais (DPs) compreendem um grupo de condições crônicas inflamatórias induzidas por microorganismos que levam à inflamação gengival, à destruição tecidual periodontal e à perda óssea alveolar. Diabetes mellitus (DM) é o termo utilizado para descrever um grupo de desordens metabólicas associadas à intolerância à glicose e ao metabolismo inadequado de carboidratos. Uma vez que DPs poderiam agir de forma similar a outros estados infecciosos sistêmicos, aumentando a severidade do diabetes, uma possível relação entre ambas tem sido considerada em todo o mundo. Polimorfismos genéticos de um único nucleotídeo (SNPs) têm sido estudados em diversas doenças. Nas periodontites, acredita-se que possam estar envolvidos na exacerbação da resposta inflamatória frente ao desafio bacteriano, modificando a susceptibilidade do hospedeiro. Neste estudo, a prevalência de periodontite foi avaliada em portadores de diabetes mellitus tipo I. Posteriormente, o SNP localizado na região promotora do gene TNFA (-1031T>C) foi analisado e sua importância para a doença periodontal destrutiva foi avaliada. O grupo teste foi constituído por diabéticos tipo I (DGT, n=113) enquanto o grupo controle por indivíduos não diabéticos (ND, n=73). Para as análises dos polimorfismos genéticos, um subgrupo foi retirado do grupo teste (DG, n=58) e comparado ao grupo ND. Os seguintes parâmetros clínicos e demográficos foram avaliados: percentual de sítios com profundidade de bolsa  6,0 mm (%PBS6,0 mm); índice gengival (IG); perda óssea radiográfica (POR); fumo; duração do diabetes ; idade; índice de massa corpórea (IMC), n de internações e n de dentes presentes. Amostras de sangue e/ou esfregaço bucal foram colhidas de 58 pacientes do grupo teste e de 73 controles. Após a extração do DNA genômico e amplificação da região genômica de interesse por PCR (Polymerase Chain Reaction), o polimorfismo TNFA 1031T>C foi analisado por BbsI RFLP (Restriction Fragment Length Polymorphism). A análise dos produtos de digestão foi feita por eletroforese em gel de poliacrilamida 8%. A análise estatística das freqüências alélica e genotípica juntamente com os dados clínicos e epidemiológicos entre os 2 grupos foi feita através do teste do Mann-Whitney e do Qui-quadrado. Os grupos de estudo obedecem ao princípio de Hardy-Weinberg. No grupo ND, as seguintes freqüências genotípicas foram encontradas: 78,1% (T/T); 20,5% (T/C) e 1,4% (C/C) enquanto no grupo D foram: 42,4%(T/T); 37,3% (T/C) e 20,3% (C/C). A frequência do alelo T no grupo diabético (D) foi de 0,610 ao passo que no grupo ND foi de 0,883. Não foi possível encontrar uma relação entre o polimorfismo -1031 T>C do gene TNFA e a presença de periodontite em diabéticos tipo I. Entretanto, o polimorfismo estudado se mostrou significativamente relacionado (p<0,0001 e OR= 4.85 95%IC 2,271-10,338) à presença do diabetes tipo I.

We present new Gemini spectra of 14 new objects found within the H?i tails of Hickson Compact Groups (HCGs) 92 and 100. Nine of them are Galaxy Evolution Explorer (GALEX) far-ultraviolet (FUV) and near-ultraviolet (NUV) sources. The spectra confirm that these objects are members of the compact groups and have metallicities close to solar, with an average value of 12+log(O/H) similar to 8.5. They have average FUV luminosities 7 x 10(40)?erg?s-1 and very young ages (<100?Myr), and two of them resemble tidal dwarf galaxy (TDG) candidates. We suggest that they were created within gas clouds that were ejected during galaxygalaxy interactions into the intergalactic medium, which would explain the high metallicities of the objects, inherited from the parent galaxies from which the gas originated. We conduct a search for similar objects in six interacting systems with extended H?i tails: NGC 2623, NGC 3079, NGC 3359, NGC 3627, NGC 3718 and NGC 4656. We found 35 ultraviolet (UV) sources with ages < 100?Myr; however, most of them are on average less luminous/massive than the UV sources found around HCG 92 and HCG 100. We speculate that this might be an environmental effect and that compact groups of galaxies are more favourable to TDG formation than other interacting systems.

Die vorliegende Arbeit beschäftigt sich mit rechtlichen Fragestellungen rund um Bewertungs-portale im Internet. Zentrale Themen der Arbeit sind dabei die Zulässigkeit der Veröffentlichung der von den Nutzern abgegebenen Bewertungen vor dem Hintergrund möglicherweise entgegenstehender datenschutzrechtlicher Bestimmungen und der Persönlichkeitsrechte der Betroffenen. Des weiteren wird der Rechtsschutz der Betroffenen erörtert und in diesem Zusammenhang die haftungsrechtlichen Risiken der Forenbetreiber untersucht. Gegenstand der Arbeit sind dabei sowohl Online-Marktplätze wie eBay, auf denen sowohl der Bewertende als auch der Bewertete registriert und mit dem Bewertungsverfahren grundsätz-lich einverstanden sind (geschlossene Portale), als auch Portale, auf denen – oftmals unter einem Pseudonym und ohne vorherige Anmeldung – eine freie Bewertungsabgabe, zu Pro-dukteigenschaften, Dienstleistungen bis hinzu Persönlichkeitsmerkmalen des Bewerteten möglich ist (offene Portale). Einleitung und Erster Teil Nach einer Einleitung und Einführung in die Problematik werden im ersten Teil die verschie-denen Arten der Bewertungsportale kurz vorgestellt. Die Arbeit unterscheidet dabei zwischen so genannten geschlossenen Portalen (transaktionsbegleitende Portale wie eBay oder Ama-zon) auf der einen Seite und offenen Portalen (Produktbewertungsportale, Hotelbewertungs-portale und Dienstleistungsbewertungsportale) auf der anderen Seite. Zweiter Teil Im zweiten Teil geht die Arbeit der Frage nach, ob die Veröffentlichung der durch die Nutzer abgegebenen Bewertungen auf den offenen Portalen überhaupt erlaubt ist oder ob hier mögli-cherweise das Persönlichkeitsrecht der Betroffenen und hier insbesondere das Recht auf in-formationelle Selbstbestimmung in Form der datenschutzrechtlichen Bestimmungen die freie Bewertungsabgabe unzulässig werden lässt. Untersucht werden in diesem Zusammenhang im einzelnen Löschungs- bzw. Beseitigungsan-sprüche der Betroffenen aus § 35 Abs. 2 Satz 2 Nr. 1 BDSG bzw. §§ 1004 i. V. m. 823 Abs. 1 BGB (allgemeines Persönlichkeitsrecht). Die Arbeit kommt in datenschutzrechtlicher Hinsicht zu dem Schluss, dass die Bewertungen personenbezogene Daten darstellen, die den datenschutzrechtlichen Bestimmungen unterlie-gen und eine Veröffentlichung der Bewertungen nach dem im deutschen Recht geltenden da-tenschutzrechtlichen Erlaubnisvorbehalt grundsätzlich nicht in Betracht kommt. Vor dem Hintergrund dieser den tatsächlichen Gegebenheiten und Interessenlagen im Internet nicht mehr gerecht werdenden Gesetzeslage diskutiert der Autor sodann die Frage, ob die datenschutzrechtlichen Bestimmungen in diesen Fällen eine Einschränkung durch die grund-gesetzlich garantierten Informationsfreiheiten erfahren müssen. Nach einer ausführlichen Diskussion der Rechtslage, in der auf die Besonderheiten der ein-zelnen Portale eingegangen wird, kommt die Arbeit zu dem Schluss, dass die Frage der Zuläs-sigkeit der Veröffentlichung der Bewertungen von einer Interessenabwägung im Einzelfall abhängt. Als Grundsatz kann jedoch gelten: Ist die bewertete Tätigkeit oder Person in Bezug auf die bewertete Eigenschaft ohnehin einer breiten Öffentlichkeit zugänglich, erscheint eine Veröffentlichung der Daten nicht bedenklich. Dagegen wird man einen Löschungs- bzw. Be-seitigungsanspruch bejahen müssen für die Bewertungen, die Tätigkeiten oder Eigenschaften des Bewerteten, die in keinem Zusammenhang mit ihm als öffentlicher Person stehen, betref-fen. Anschließend geht die Arbeit auf die Persönlichkeitsrechte der Betroffenen und der sich hier-aus ergebenden Beseitigungs- und Unterlassungsansprüchen gemäß der §§ 1004 Abs. 1, 823 Abs. 1 BGB ein, verneint jedoch wegen dem Vorrang der spezialgesetzlichen Bestimmungen aus dem Bundesdatenschutzgesetz letztlich eine Anwendbarkeit der Anspruchsgrundlagen. Schließlich wird in diesem Teil noch kurz auf die Zulässigkeit der Bewertung juristischer Per-sonen eingegangen, die im Grundsatz bejaht wird. Dritter Teil Sofern der zweite Teil der Arbeit zu dem Schluss kommt, dass die Veröffentlichung der Be-wertungen zulässig ist, stellt sich im dritten Teil die Frage, welche Möglichkeiten das Recht dem Bewerteten bietet, gegen negative Bewertungen vorzugehen. Untersucht werden, dabei datenschutzrechtliche, deliktsrechtliche, vertragliche und wettbe-werbsrechtliche Ansprüche. Ein Schwerpunkt dieses Teils liegt in der Darstellung der aktuellen Rechtsprechung zu der Frage wann eine Bewertung eine Tatsachenbehauptung bzw. ein Werturteil darstellt und den sich hieraus ergebenden unterschiedlichen Konsequenzen für den Unterlassungsanspruch des Betroffenen. Diejenigen Bewertungen, die eine Meinungsäußerung darstellen, unterstehen dem starken Schutz der Meinungsäußerungsfreiheit. Grenze der Zulässigkeit sind hier im wesentlichen nur die Schmähkritik und Beleidigung. An Tatsachenbehautpungen dagegen sind schärfere Maßstäbe anzulegen. In diesem Zusammenhang wird der Frage nachgegangen, ob vertragliche Beziehungen zwischen den Beteiligten (Bewertenden, Bewertete und Portalbetreiber) die Meinungsäußerungsfreiheit einschränkt, was jedenfalls für die geschlossenen Portale bejaht wird. Vierter Teil Der vierte Teil der Arbeit beschäftigt sich mit den „Zu-gut-Bewertungen“. Es geht dabei um wettbewerbsrechtliche Ansprüche im Falle verdeckter Eigenbewertungen. Solche Eigenbewertungen, die unter dem Deckmantel der Pseudonymität als Werbemittel zur Imageverbesserung in entsprechenden Bewertungsportale verbreitet werden ohne den wahren Autor erkennen zu lassen, sind in wettbewerbsrechtlicher Hinsicht grundsätzlich unzulässig. Fünfter Teil Im letzten Teil der Arbeit wird schließlich der Frage nach der Verantwortlichkeit der Portal-betreiber für rechtswidrige Bewertungen nachgegangen. Zunächst wird die Feststellung getroffen, dass es sich bei den von den Nutzern abgegebenen Bewertungen um fremde Inhalte handelt und somit die Haftungsprivilegierungen der § 11 Abs. 1 TDG, § 9 Abs. 1 MDStV eingreifen, wonach die Forenbetreiber für die rechtswidrigen Bewertungen jedenfalls so lange nicht verantwortlich sind, solange sie hiervon keine Kenntnis haben. Da von dieser Haftungsprivilegierung nach der Rechtsprechung des Bundesgerichtshofs die Störerhaftung nicht umfasst ist, wird die Reichweite die Forenbetreiber aus der Störerhaftung treffenden Überwachungspflichten diskutiert. Die Arbeit kommt hier zu dem Ergebnis, dass in den Fällen, in denen dem Adressaten der Bewertung die Identität des Verfassers bekannt ist, sich die Verpflichtungen der Forenbetrei-ber auf die Beseitigung bzw. Uracil ist eine der am häufigsten vorkommenden DNA-Basenmodifikationen, die über den Mechanismus der Basen-Exzisions-Reparatur (BER) aus dem Genom entfernt wird. Im Verlauf der Reparatur dieser Läsion durch monofunktionelle Uracil-DNA-Glykosylasen (UNG1/2, SMUG1, TDG und MBD4) entstehen AP-Läsionen und Einzelstrangbrüche. Da von beiden bekannt ist, eine Blockade der Transkription verursachen zu können, wurde in dieser Arbeit der Einfluss von Uracil und dessen Exzision auf die Expression eines Gens untersucht. Dafür wurde eine effiziente Methode entwickelt, die DNA-Basenmodifikation spezifisch in den transkribierten oder nicht-transkribierten DNA-Strang eines Reporter-Vektors einzufügen. rnIn Host cell reactivation Assays konnte gezeigt werden, dass Uracil unabhängig davon, ob es mit Adenin gepaart (U:A) oder mit Guanin (U:G) eine Fehlpaarung bildet, keine direkte Blockade der Transkriptions-Maschinerie in menschlichen Zellen auszulösen vermag. Dies kann daraus geschlossen werden, dass die Expression des Reportergens der Uracil-enthaltenen Vektoren im Vergleich zu unmodifizierten Referenz-Vektoren kurze Zeit nach der Transfektion unverändert ist. Die erst mit zunehmender Inkubationszeit in den Wirtszellen progressiv abnehmende Transkription ließ vermuten, dass die intrazelluläre Prozessierung der Läsion über die BER für die verringerte Genexpression verantwortlich ist. In der Tat bewirkte der Knockdown der BER-initiierenden UNG1/2, die Uracil aus der DNA herausschneidet und damit eine AP-Läsion generiert, eine Verringerung des negativen Effektes eines U:A-Basenpaares auf die Genexpression. Dass der Knockdown der SMUG1- oder TDG-Glykosylase hingegen keine Auswirkungen zeigte, beweist, dass UNG1/2 die Hauptglykosylase für die Exzision dieser Läsion und der Auslöser der inhibierten Transkription in HeLa-Zellen darstellt. Der Zusammenhang zwischen dem Maß des Ausschnitts einer DNA-Basenmodifikation im Verlauf der BER und einer verringerten Expression des Reportergens konnte zudem am Beispiel von 5-Hydroxymethyluracil und der für diese Läsion spezifischen SMUG1-Glykosylase nachgewiesen werden. Im Falle einer U:G-Fehlpaarung besaß weder UNG1/2 noch SMUG1 oder TDG einen Einfluss auf die Rate oder das Ausmaß der mit der Zeit abnehmenden Genexpression, was die Beteiligung einer anderen Glykosylase oder eines anderen Reparatur-Mechanismus vermuten lässt. rnDie Tatsache, dass die Stärke der Gen-Suppression unabhängig davon war, ob Uracil im transkribierten oder nicht-transkribierten DNA-Strang positioniert wurde, lässt die Mutmaßung zu, dass keine Blockade der elongierenden RNA-Polymerase, sondern vielmehr ein indirekter Mechanismus der Auslöser für die verringerte Transkription ist. Dieser Mechanismus muss unabhängig von der gut untersuchten transkriptionsgekoppelten Nukleotid-Exzisions-Reparatur erfolgen, da der Knockdown des hierfür benötigten CSB-Gens keine Auswirkungen auf die Inhibition der Genexpression der Uracil-enthaltenen Vektoren hatte. Insgesamt liefert diese Arbeit neue Erkenntnisse über den Beitrag der einzelnen Uracil-DNA-Glykosylasen zur Reparatur der DNA-Basenmodifikation Uracil in humanen Zellen und zeigt, dass die BER über einen indirekten Mechanismus die Hemmung der Genexpression verursacht.

Deamination of 5-methylcytosine residues in DNA gives rise to the G/T mismatched base pair. In humans this lesion is repaired by a mismatch-specific thymine DNA glycosylase (TDG or G/T glycosylase), which catalyzes specific excision of the thymine base through N-glycosidic bond hydrolysis. Unlike other DNA glycosylases, TDG recognizes an aberrant pairing of two normal bases rather than a damaged base per se. An important structural issue is thus to understand how the enzyme specifically targets the T (or U) residue of the mismatched base pair. Our approach toward the study of substrate recognition and processing by catalytic DNA binding proteins has been to modify the substrate so as to preserve recognition of the base but to prevent its excision. Here we report that replacement of 2′-hydrogen atoms with fluorine in the substrate 2′-deoxyguridine (dU) residue abrogates glycosidic bond cleavage, thereby leading to the formation of a tight, specific glycosylase–DNA complex. Biochemical characterization of these complexes reveals that the enzyme protects an ≈20-bp stretch of the substrate from DNase I cleavage, and directly contacts a G residue on the 3′ side of the mismatched U derivative. These studies provide a mechanistic rationale for the preferential repair of deaminated CpG sites and pave the way for future high-resolution studies of TDG bound to DNA.

Este trabajo de investigaci?n pretende mostrar los resultados de un estudio llevado a cabo como subproyecto del macroproyecto "Evaluaci?n de los procesos acad?micos de la Licenciatura en Lenguas Extranjeras de la Universidad del Valle". Se espera que los resultados de la investigaci?n sirvan como insumo al proceso de autoevaluaci?n para la acreditaci?n de alta calidad del Programa. Este subproyecto buscaba analizar el proceso de elaboraci?n de los trabajos de grado (TDG), de la Licenciatura en Lenguas Extranjeras, aprobados durante el periodo 2004 a 2010. El prop?sito fue analizar y describir la manera como los estudiantes vivieron la experiencia de elaboraci?n del TDG, la pertinencia de la formaci?n en investigaci?n y las diferentes tendencias que marcan los trabajos aprobados durante el periodo mencionado. El estudio se enmarca en el enfoque de investigaci?n evaluativa. Se analiz? un corpus de 97 monograf?as. Se administr? una encuesta y una entrevista a un grupo de egresados que participaron en el grupo focal del cual se recogi? informaci?n sobre su experiencia en la elaboraci?n del trabajo de grado. Los resultados mostraron que los estudiantes consideran que la formaci?n en investigaci?n recibida es pertinente m?s no suficiente para algunas de las ?reas del lenguaje. Se encontr? adem?s que la mayor?a de los trabajos de grado corresponden a problem?ticas relacionadas con la ense?anza y el aprendizaje de las lenguas extranjeras, por encima de trabajos en otras ?reas del lenguaje como la literatura, la traducci?n, la socioling??stica, entre otras. El rol de tutor y de lector fue valorado positivamente por los estudiantes, en t?rminos generales, con algunas excepciones espec?ficas que impidieron el normal desarrollo del proceso de elaboraci?n de estos trabajos. Igualmente, se identificaron dificultades relacionadas con aspectos tales como la escritura del informe final y la relaci?n tutor-estudiante-lector. Tambi?n se encontr? que la formaci?n recibida y la experiencia en investigaci?n obtenida durante la elaboraci?n del TDG, tuvo un impacto positivo en los diferentes ?mbitos de la vida de los estudiantes (personal, acad?mico, laboral, profesional). Finalmente, los estudiantes hicieron sugerencias para el mejoramiento de este proceso.

The aim of study was to examine the effects of the world's most challenging mountain ultramarathon (Tor des Geants [TdG]) on running mechanics. Mechanical measurements were undertaken in male runners (n = 16) and a control group (n = 8) before (PRE), during (MID), and after (POST) the TdG. Contact (tc) and aerial (ta) times, step frequency (f), and running velocity (v) were sampled. Spring-mass parameters of peak vertical ground-reaction force (Fmax), vertical downward displacement of the center of mass (Deltaz), leg-length change (DeltaL), and vertical (kvert) and leg (kleg) stiffness were computed. Significant decreases were observed in runners between PRE and MID for ta (P < .001), Fmax (P < .001), Deltaz (P < .05), and kleg (P < .01). In contrast, f significantly increased (P < .05) between PRE and MID-TdG. No further changes were observed at POST for any of those variables, with the exception of kleg, which went back to PRE. During the TdG, experienced runners modified their running pattern and spring-mass behavior mainly during the first half. The current results suggest that these mechanical changes aim at minimizing the pain occurring in lower limbs mainly during the eccentric phases. One cannot rule out that this switch to a "safer" technique may also aim to anticipate further damages.

Los cambios epigenéticos son responsables de la aparición de muchas patologías humanas y sus causas son debido a factores ambientales como genéticos. Se ha descrito en enfermedades crónicas como la Diabetes Mellitus tipo 2 (T2DM) que se caracteriza por los estados de hiperglucemia y el incremento en el estrés oxidativo que conlleva a complicaciones micro y macro vasculares, asociado a una desmetilación global del genoma. Nuestra hipótesis corresponde a que los órganos diana son afectados por las alteraciones como la metilación e hidroximetilación como consecuencia del estrés oxidativo que luego repercuten en la persistencia de la enfermedad. Métodos: A partir de sangre periférica se analizaron los cambios globales en la metilación del DNA que son afectados por el estado metabólico de 60 individuos (40 pacientes, 20 controles sanos). Por técnicas de cuantificación se compararon los resultados obtenidos con los de la expresión de las enzimas involucradas. Por último, se realizó un estudio de microarreglos de metilación del DNA y de expresión obtenidos de la base de datos GEO para así comparar los resultados con nuestros datos experimentales. Resultados: Los pacientes diabéticos con pobre control metabólico presentaron mayores niveles de metilación que el grupo control y no se encontró alteración en las enzimas involucradas en este proceso. Los resultados fueron concordantes con el estudio de microarreglos. Conclusión: Los estudios experimentales y de microarreglos demostraron que la metilación es tejido específico y que existe una mayor oxidación en pacientes. Por ello proponemos una vía alterna de desmetilación no enzimática, basada en la oxidación directa de los grupos metilos generados por los estados oxidativos característicos de esta enfermedad.
