Today all kinds of innovations and research work is done by partnerships of competent entities each having some specialized skills. Like the development of the global economy, global innovation partnerships have grown considerably and form the basis of most of the sophisticated innovations today. To further streamline and simplify such cooperation, several innovation networks have been formed, both at local and global levels. This paper discusses the different types of innovations and how cooperation can benefit innovation in terms of pooling of resources and sharing of risks. One example of an open global co-innovation network promoted by Tata Consultancy Services, the TCS COIN is taken as a case. It enables venture capitalists, consultants, research agencies, companies and universities form nodes of the network so that each entity can play a meaningful role in the innovation network. Further, two innovation projects implemented using the COIN are discussed. Innovation Networks like these could form the basis of a unique global innovation network, which is not owned by any company and is used by innovation partners globally to collaborate and conduct research and development.

L'enseignement, l'apprentissage et l'évaluation du raisonnement clinique en sciences infirmières est un défi pour les éducateurs de cette profession et leurs étudiants. Depuis plusieurs décennies, les chercheurs et les éducateurs dans le domaine des sciences de la santé ont travaillé pour élaborer des instruments d'évaluation dans le but de pouvoir mesurer le raisonnement clinique (Charlin, Bordage & Van der Vleuten, 2003). Plusieurs études semblent appuyer le test de concordance de script (TCS) en termes de validité, fiabilité, faisabilité et applicabilité pour plusieurs disciplines et différents contextes (Carrière & al, 2009). Deschênes et ses collaborateurs (2006; Deschênes, Charlin, Gagnon & Goudreau, 2011) ont mis au point et validé un TCS spécifiquement pour le raisonnement clinique en sciences infirmières (RCI). Comme l'évaluation a un impact important sur les stratégies d'apprentissage des étudiants (Sibert et al, 2001; Durak, Caliskan & Bor, 2007), les outils d'évaluation valides et fiables qui permettraient l'identification des problèmes spécifiques dans le développement du raisonnement clinique en sciences infirmières seraient très utiles pour guider les décisions concernant l'éducation (Gierl, 2007). Nous avons donc mené une étude pour explorer le potentiel diagnostique des questions d'un TCS. La question de recherche est la suivante: «Dans quelle mesure chaque question d’un TCS visant à évaluer le RCI peut-elle être reliée à des catégories et des stratégies de pensée spécifiques?" Avec une sous-question: «Comment peut-on décrire le potentiel diagnostique d’un TCS pour évaluer le RCI?". Nous avons fait une deuxième analyse de contenu des données qui ont été obtenues dans une étude précédente dans laquelle cinq vignettes (15 questions) du TCS de Deschênes (2006) ont été utilisées. Les résultats ont montré les catégories et stratégies de pensées utilisées pour répondre à certaines questions du TCS selon les groupes de participants. Aussi, nos résultats ont permis de découvrir des disparités importantes entre les groupes, notamment que le RCI des expertes est si différent des étudiantes, qu’il ne peut servir de référence. Enfin, cette étude démontre que le TCS a un potentiel diagnostique niveau par niveau (1ère, 2e, 3e année et expertes) et non d’un niveau à un autre.

Mit dem zunehmenden Einsatz von E-Learning-Plattformen rücken verstärkt Wirtschaftlichkeitsaspekte in den Betrachtungsmittelpunkt, die Methoden zur Ermittlung systembedingter Kosten voraussetzen. Durch die zunehmende Serviceorientierung und Integration von LMS mit bestehenden Komponenten der Anwendungsarchitektur sind hierfür jedoch neue Methoden notwendig, welche die Defizite traditioneller Total Cost of Ownership-Modelle abbauen. Einen Ansatzpunkt hierfür bietet das ITIL-Referenzmodell, das einen Rahmen für taktische und operative IT-Services vorgibt und somit die Grundlage für eine serviceorientierte Gesamtkostenermittlung in Form der Total Cost of Services (TCS) liefert.

Streptococcus sanguinis is a commensal pioneer colonizer of teeth and an opportunistic pathogen of infectious endocarditis. The establishment of S. sanguinis in host sites likely requires dynamic fitting of the cell wall in response to local stimuli. In this study, we investigated the two-component system (TCS) VicRK in S. sanguinis (VicRKSs), which regulates genes of cell wall biogenesis, biofilm formation, and virulence in opportunistic pathogens. A vicK knockout mutant obtained from strain SK36 (SKvic) showed slight reductions in aerobic growth and resistance to oxidative stress but an impaired ability to form biofilms, a phenotype restored in the complemented mutant. The biofilm-defective phenotype was associated with reduced amounts of extracellular DNA during aerobic growth, with reduced production of H2O2, a metabolic product associated with DNA release, and with inhibitory capacity of S. sanguinis competitor species. No changes in autolysis or cell surface hydrophobicity were detected in SKvic. Reverse transcription-quantitative PCR (RT-qPCR), electrophoretic mobility shift assays (EMSA), and promoter sequence analyses revealed that VicR directly regulates genes encoding murein hydrolases (SSA_0094, cwdP, and gbpB) and spxB, which encodes pyruvate oxidase for H2O2 production. Genes previously associated with spxB expression (spxR, ccpA, ackA, and tpK) were not transcriptionally affected in SKvic. RT-qPCR analyses of S. sanguinis biofilm cells further showed upregulation of VicRK targets (spxB, gbpB, and SSA_0094) and other genes for biofilm formation (gtfP and comE) compared to expression in planktonic cells. This study provides evidence that VicRKSs regulates functions crucial for S. sanguinis establishment in biofilms and identifies novel VicRK targets potentially involved in hydrolytic activities of the cell wall required for these functions.

Background: Treacher Collins syndrome (TCS) is an autosomal dominant craniofacial disorder caused by frameshift deletions or duplications in the TCOF1 gene. These mutations cause premature termination codons, which are predicted to lead to mRNA degradation by nonsense mediated mRNA decay (NMD). Haploinsufficiency of the gene product (treacle) during embryonic development is the proposed molecular mechanism underlying TCS. However, it is still unknown if TCOF1 expression levels are decreased in postembryonic human cells. Methods: We have estimated TCOF1 transcript levels through real time PCR in mRNA obtained from leucocytes and mesenchymal cells of TCS patients (n = 23) and controls (n = 18). Mutational screening and analysis of NMD were performed by direct sequencing of gDNA and cDNA, respectively. Results: All the 23 patients had typical clinical features of the syndrome and pathogenic mutations were detected in 19 of them. We demonstrated that the expression level of TCOF1 is 18-31% lower in patients than in controls (p < 0.05), even if we exclude the patients in whom we did not detect the pathogenic mutation. We also observed that the mutant allele is usually less abundant than the wild type one in mesenchymal cells. Conclusions: This is the first study to report decreased expression levels of TCOF1 in TCS adult human cells, but it is still unknown if this finding is associated to any phenotype in adulthood. In addition, as we demonstrated that alleles harboring the pathogenic mutations have lower expression, we herein corroborate the current hypothesis of NMD of the mutant transcript as the explanation for diminished levels of TCOF1 expression. Further, considering that TCOF1 deficiency in adult cells could be associated to pathologic clinical findings, it will be important to verify if TCS patients have an impairment in adult stem cell properties, as this can reduce the efficiency of plastic surgery results during rehabilitation of these patients.

Objective: To describe a new syndrome of X-linked myoclonic epilepsy with generalized spasticity and intellectual disability (XMESID) and identify the gene defect underlying this disorder. Methods: The authors studied a family in which six boys over two generations had intractable seizures using a validated seizure questionnaire, clinical examination, and EEG studies. Previous records and investigations were obtained. Information on seizure disorders was obtained on 271 members of the extended family. Molecular genetic analysis included linkage studies and mutational analysis using a positional candidate gene approach. Results: All six affected boys had myoclonic seizures and TCS; two had infantile spasms, but only one had hypsarrhythmia. EEG studies show diffuse background slowing with slow generalized spike wave activity. All affected boys had moderate to profound intellectual disability. Hyperreflexia was observed in obligate carrier women. A late-onset progressive spastic ataxia in the matriarch raises the possibility of late clinical manifestations in obligate carriers. The disorder was mapped to Xp11.2-22.2 with a maximum lod score of 1.8. As recently reported, a missense mutation (1058C>T/P353L) was identified within the homeodomain of the novel human Aristaless related homeobox gene (ARX). Conclusions: XMESID is a rare X-linked recessive myoclonic epilepsy with spasticity and intellectual disability in boys. Hyperreflexia is found in carrier women. XMESID is associated with a missense mutation in ARX. This disorder is allelic with X-linked infantile spasms (ISSX; MIM 308350) where polyalanine tract expansions are the commonly observed molecular defect. Mutations of ARX are associated with a wide range of phenotypes; functional studies in the future may lend insights to the neurobiology of myoclonic seizures and infantile spasms.

Realizada no bojo do processo de diagn??stico e reforma dos TCs, o Programa de Moderniza????o do Sistema de Controle Externo dos Estados e do Distrito Federal (Promoex), esta pesquisa revela quais s??o os problemas que afetam o desempenho administrativo e a legitimidade institucional desses ??rg??os fiscalizadores, bem como as suas qualidades, as quais podem ser utilizadas como motor de sua reformula????o. A partir da interpreta????o das opini??es dos atores entrevistados, a an??lise final procura revelar quais caminhos podem ser trilhados para se modernizarem os Tribunais de Contas subnacionais.

De acordo com o contrato estabelecido entre a Administração do Porto de Sines, S.A. e a PSA Sines, concessionária do Terminal de Contentores de Sines (TCS), cabe à Autoridade Portuária a proteção necessária à operação portuária. Neste sentido, foi estabelecido um planeamento faseado do desenvolvimento do Molhe Leste do Porto de Sines, de acordo com o desenvolvimento do Terminal XXI. Tendo já o concessionário procedido à ampliação do cais de 380 metros para 730 metros, cabe à Autoridade Portuária proceder às operações necessárias que proporcionem as adequadas condições de abrigo à agitação marítima. Com um investimento de €32 500 000,00, por parte da APS, S.A., surge a empreitada “2.ª Fase de Ampliação do Molhe Leste do Porto de Sines”. Trata-se portanto, de uma oportunidade única para um aluno de Engenharia Civil no ramo de Hidráulica, proceder ao acompanhamento e fiscalização da empreitada numa empresa de referência a nível Mundial. A finalidade deste relatório será enunciar o procedimento adotado na tarefa de fiscalização de controlo de custos, durante a fase de execução da “2.ª Fase de Ampliação do Molhe Leste do Porto de Sines”, do ponto de vista do Dono de Obra (APS, S.A.), descrevendo todas as etapas decorrentes do processo de fiscalização de controlo de custos de uma obra pública de acordo da legislação em vigor, nomeadamente o Código dos Contratos Público (C.C.P.).

We show that a self-generated set of combinatorial games, S. may not be hereditarily closed but, strong self-generation and hereditary closure are equivalent in the universe of short games. In [13], the question "Is there a set which will give a non-distributive but modular lattice?" appears. A useful necessary condition for the existence of a finite non-distributive modular L(S) is proved. We show the existence of S such that L(S) is modular and not distributive, exhibiting the first known example. More, we prove a Representation Theorem with Games that allows the generation of all finite lattices in game context. Finally, a computational tool for drawing lattices of games is presented. Hind-limb ischemia has been used in type 1 diabetic mice to evaluate treatments for peripheral arterial disease or mechanisms of vascular impairment in diabetes [1]. Vascular deficiency is not only a pathophysiological condition, but also an obvious circumstance in tissue regeneration and in tissue engineering and regenerative medicine (TERM) strategies. We performed a pilot experiment of hind-limb ischemia in streptozotocin(STZ)-induced type 1 diabetic mice to hypothesise whether diabetes influences neovascularization induced by biomaterials. The dependent variables included blood flow and markers of arteriogenesis and angiogenesis. Type 1 diabetes was induced in 8-week-old C57BL/6 mice by an i.p. injection of STZ (50 mg/kg daily for 5 days). Hind-limb ischemia was created under deep anaesthesia and the left femoral artery and vein were isolated, ligated, and excised. The contralateral hind limb served as an internal control within each mouse. Non-diabetic ischaemic mice were used as experiment controls. At the hind-limb ischemia surgical procedure, different types of biomaterials were placed in the blood vessels gap. Blood flow was estimated by Laser Doppler perfusion imager, right after surgery and then weekly. After 28 days of implantation, surrounding muscle was excised and evaluated by histological analysis for arteriogenesis and angiogenesis. The results showed that implanted biomaterials were promote faster restoration of blood flow in the ischemic limbs and improved neovascularization in the diabetic mice. Therefore, we herein demonstrate that the combined model of hind-limb ischemia in type 1 diabetes mice is suitable to evaluate the neovascularization potential of biomaterials and eventually tissue engineering constructs.

Higher-dimensional automata constitute a very expressive model for concurrent systems. In this paper, we discuss ``topological abstraction" of higher-dimensional automata, i.e., the replacement of HDAs by smaller ones that can be considered equivalent from the point of view of both computer science and topology. By definition, topological abstraction preserves the homotopy type, the trace category, and the homology graph of an HDA. We establish conditions under which cube collapses yield topological abstractions of HDAs.

Se indagará principalmente acerca del rol de los procesos neutrales, como la deriva génica, de procesos selectivos, como la selección natural mediada por polinizadores y de procesos históricos (geológicos y climáticos del pasado) en la diversificación floral tanto a escala microevolutiva como macroevolutiva. La heterogeneidad ambiental que se presenta en amplios rangos geográficos puede promover la diferenciación entre poblaciones debido a las diferencias en condiciones físicas y biológicas. De esta manera, especies ampliamente distribuidas ofrecen la oportunidad de explorar la dinámica de los procesos evolutivos que tienen lugar a nivel interpoblacional (Dobzhansky 1970, Thompson 1999). El estudio comparativo entre especies hermanas permite comprender cómo la selección natural (adaptación) y la inercia filogenética (herencia ancestral) han modelado los rasgos de las especies que observamos en la actualidad (Díaz 2002, Schluter 2000, Futuyma 2005). Uno de los usos más importantes de la información filogenética es el de reconstruir la historia del cambio evolutivo en caracteres adaptativos mediante su mapeo en la filogenia y la reconstrucción del estado de estos caracteres en el ancestro. Así, la asociación entre transición de caracteres y transiciones en grupos funcionales es una evidencia directa de la hipótesis adaptativa de que los rasgos son seleccionados por grupos funcionales de polinizadores. Una aproximación filogenética puede permitir identificar la dirección y el tiempo de evolución. Todos estos aspectos señalan la necesidad de adoptar una perspectiva conceptualmente integrada (morfológica, genética, filogenética, filogeográfica y ecológica) en el estudio de la biología evolutiva de las flores. Estudiar como actúan los procesos micro- y macroevolutivos en las interacciones planta-polinizador, en una dimensión espacial y temporal, arrojará resultados importantes tanto en el campo teórico como en el de la conservación. Por una parte, permitirá poner a prueba hipótesis relevantes sobre la adaptación de caracteres, mientras que explorará los procesos evolutivos que subyacen a las tramas de las interacciones planta-polinizador; por otro lado, comprender el rol de los cambios climáticos pasados en la diversificación biológica es interesante tanto desde una aproximación evolutiva como desde la biología de la conservación (Avise 2000; Moritz et al. 2000; Petit et al. 2003; Hewitt 2004). Géneros a ser estudiados en este proyecto: 1- Anarthrophyllum (Fabaceae,15 spp), 2- Monttea (Plantaginaceae, 3 spp), 3- Caleolaria (Calceolariaceae 3 spp), 4- Centris (Apidae, 1 spp), 5- Jaborosa (Solanaceae, 23 spp). Metodología: Mapeado de las poblaciones. Elenco de polinizadores, frecuencia. Obtención y medición de caracteres fenotípicos florales. Néctar: concentración y vol. Aceites (peso); Morfometría geométrica (Zelditch et al. 2005). Éxito reproductivo (Dafni & Kevan 2003). Caracteres genéticos: extracción, amplificación y secuenciación: en Calceolaria se utilizarán 2 genes de cloroplasto trnH-psbA y trnS-trnG y genes anónimos nucleares de copia única (scnADN), para Jaborosa se utilizarán 3 genes de cloroplasto (trnH-psbA, TrnD-trnT y ndhF-rp32) y el gen nuclear GBSSI waxy. Finalmente para Centris cineraria se usaría el tRNA ILE y NADH Deshidrogenada subunidad 2. Análisis filogenéticos de parsimonia (Goloboff et al. 2000, Kitching et al. 1998, Nixon 2002, Farris et al. 1996, Sorenson 1999); Filogeografía: reconstrucción de redes por parsimonia (Clement et al. 2000; Posada et al. 2000), análisis de clados anidados (NCPA). Se usarán las claves de inferencia (Templeton 2004). Para todos estos análisis se utilizarán los siguientes programas: DnaSP, Network, Arlequin, MrBayes, Paup, ModelTest, Beast, TNT, WinClada TCS y GeoDis. Estadística multivariada: Los diferentes rasgos florales mencionados se analizarán utilizando distancias de Gower (datos cualitativos) y euclídeas (datos cuantitativos) mediante la técnica multivariada ACoP.

Report for the scientific sojourn carried out at the l’ Institute for Computational Molecular Science of the Temple University, United States, from 2010 to 2012. Two-component systems (TCS) are used by pathogenic bacteria to sense the environment within a host and activate mechanisms related to virulence and antimicrobial resistance. A prototypical example is the PhoQ/PhoP system, which is the major regulator of virulence in Salmonella. Hence, PhoQ is an attractive target for the design of new antibiotics against foodborne diseases. Inhibition of the PhoQ-mediated bacterial virulence does not result in growth inhibition, presenting less selective pressure for the generation of antibiotic resistance. Moreover, PhoQ is a histidine kinase (HK) and it is absent in animals. Nevertheless, the design of satisfactory HK inhibitors has been proven to be a challenge. To compete with the intracellular ATP concentrations, the affinity of a HK inhibidor must be in the micromolar-nanomolar range, whereas the current lead compounds have at best millimolar affinities. Moreover, the drug selectivity depends on the conformation of a highly variable loop, referred to as the “ATP-lid, which is difficult to study by X-Ray crystallography due to its flexibility. I have investigated the binding of different HK inhibitors to PhoQ. In particular, all-atom molecular dynamics simulations have been combined with enhanced sampling techniques in order to provide structural and dynamic information of the conformation of the ATP-lid. Transient interactions between these drugs and the ATP-lid have been identified and the free energy of the different binding modes has been estimated. The results obtained pinpoint the importance of protein flexibility in the HK-inhibitor binding, and constitute a first step in developing more potent and selective drugs. The computational resources of the hosting institution as well as the experience of the members of the group in drug binding and free energy methods have been crucial to carry out this work.
