The essential oils isolated by hydrodistillation from trunk bark and leaves of Talauma ovata A. St. Hil. (Magnoliaceae), collected in four seasons, were analyzed by capillary GC and GC/MS. Altogether 52 components were identified, The oils were characterized by predominance of cyclic sesquiterpenes. The main components were linalool, trans-beta-guaiene, germaerene D, germacrene B, spathulenol, caryophyllene oxide, viridiflorol and alpha-endesmol. The content of individual components was variable during the year. All oils were screened against several strains of bacteria and yeasts, using the agar well-diffusion technique. The antimicrobial activity of oils showed strong dependence with the season. Significant activity was found for oils obtained in the spring and summer.

Tire volatile constituents of the fresh leaves of Talauma ovata (Magnoliaceae) were extracted by hydrodistillation and analyzed by GC and GC/MS. Twenty-seven compounds were identified representing 98.8% of the total oil. The oil from this species showed to be constituted by monoterpenes and sesquiterpenes in the same ratio. Among the monoterpenes, limonene (34.8%) and alpha-pinene (11.3%) were the main constituents, and beta-bisabolene (10.7%) was the major substance for the sesquiterpene fraction.

Some plants are widely distributed, ranging in different vegetation types. Since bird distribution can be directly associated to vegetational cover, we expect that seed dispersers can also vary following phytophysiognomy for some ornithochorous plants. The purpose of this paper was to describe potential seed disperser birds of Talauma ovata (Magnoliaceae) in semideciduous Atlantic forest and cerrado fragments in south-east Brazil. In 79 h of focal observations we recorded 1085 visits of 25 different bird species consuming T. ovata diaspores. Although most bird species observed consuming T. ovata diaspores present generalist habits, they were potential seed dispersers, presenting high visits rate and do not staying a long time on the plants, favoring seed dispersal efficiency. Fruit consumption was not different between species in the study sites and the time spending on the plants was not different between species in semideciduous forest, but was different in cerrado. Agonistic encounters were observed mainly in the end of fruiting season, but was not relevant to decrease the number of seeds dispersed by birds. The occurrence of different potential seed dispersers between areas, such as Cyanocorax cristatellus (Corvidae), Schistochlamys ruficapillus (Emberizidae) and Mimus saturninus (Mimidae), which are endemic from the cerrado ecosystem and could rarely be found in semideciduous forest, suggests that seed dispersers can vary according to phytophysiognomy characteristics. Our results also indicate that the evolution of a generalist seed dispersal system such as observed for T. ovata can make possible the colonization of different ambients. Furthermore, the high number of visits recorded for both areas suggests that the fleshy arils of T. ovata are important feeding resources for birds.

Two new lignans, magnovatins A (1) and B (2), along with nine known compounds, were isolated from the leaves of Magnolia ovata. The known compounds were identified as acuminatin (3), licarin A (4), kadsurenin M, 4-O-demethylkadsurenin M, oleiferin A, oleiferin C, spathulenol, parthenolide, and 11,13-dehydrocompressanolide. In addition, compounds I and 2 yielded four new derivatives (1a, 1b, 2a, and 2b). The structures of the new compounds were established on the basis of spectrometric data evaluation. Free-radical scavenging and antimicrobial activities of the major compounds 1, 3, and 4 were investigated.

Aim of the study: Magnolia ovata (A.St.-Hil.) Spreng (formerly Talauma ovata), known as ""pinha-do-brejo"" or ""baguacu"", is a large tree widely distributed in Brazil. Its trunk bark has been used in folk medicine against fever. However, no data have been published to support the antipyretic ethnopharmacological use. This study investigated the antipyretic and anti-inflammatory effects of the ethanolic extract (EEMO). dichloromethane fraction (DCM), and the isolated compound costunolide. Materials and methods: The antipyretic and anti-inflammatory activities were evaluated in experimental models of fever and inflammation in mice. Results: The oral administration of EEMO, DCM and costunolide inhibited carrageenan (Cg)-induced paw oedema (ID(50) 72.35 (38.64-135.46) mg/kg, 5.8 (2.41-14.04) mg/kg and 0.18 (0.12-0.27) mg/kg, respectively) and was effective in abolishing lipopolysaccharide (LPS)-induced fever (30 mg/kg, 4.5 mg/kg and 0.15 mg/kg, respectively). EEMO was also effective in reducing cell migration in the pleurisy model. Intraplantar injection of costunolide also reduced the paw oedema, myeloperoxidase and N-acetyl-glucosaminidase activity induced by Cg in mice. Conclusions: Collectively, these results show, for the first time, that extracts obtained from Magnolia ovata possess antipyretic and anti-inflammatory properties, and costunolide appears to be the compound responsible for these effects. The volatile and non-volatile constituents of the unripe fruits of Magnolia ovata (A. St.-Hil.) Spreng. (Magnoliaceae) were studied. The essential oils were obtained by hydrodistillation of the fruit of two plant populations (A and B) and analyzed by GC/FID and GC/MS. The oil of sample A was rich in sesquiterpenes, mainly spathulenol (19.3%), while the oil of sample B showed a predominance of aliphatic compounds, mainly hexadecanoic acid (52.0%). Extracts of the dried fruit contained fourteen known compounds including nine lignoids (magnovatin A, magnovatin B, acuminatin, licarin A, oleiferin A, oleiferin C, kadsurenin M, 4-O-demethylkadsurenim M and 7-epi-virolin), two sesquiterpene lactones (parthenolide and michelenolide) and three alkaloids (lysicamine, lanuginosine and O-methylmoschatoline). Michelenolide, 7-epi-virolin and lisycamine are reported for the first time in the species, while the remaining compounds have already been reported in the leaves and/or trunk bark of Magnolia ovata. Acetylation of oleiferin A yielded a new compound, acetyl oleiferin A, whose NMR data and that of michelenolide are furnished.

Magnolia ovata seeds have been reported as desiccation sensitive. In order to test if the drying rate would affect the assessment of storage behaviour of these seeds, the effect of different drying rates and storage times on the viability was tested. Seeds were dried over activated silica gel (fast drying) or salt solutions for different periods (slow drying) and stored at -20°C. Partial drying transiently increased the final germination and the germination speed index, but further drying resulted in reduction of these parameters. Drying rate affected the final germination and vigour. Seeds that were slow-dried to 0.10 g H 2O ̇ g -1 dw retained high viability when compared with seeds desiccated to the same water content level by the fast drying method, although their vigour was reduced. Only slow-dried seeds could be stored at -20°C for 90 d without reduction of viability. These data suggested that the storage behaviour of seeds of M. ovata seeds should be classified as intermediate.

Le fioriture algali rappresentano un problema emergente che colpisce le attività ricreative costiere negli ultimi decenni e molta responsabilità è attribuita alla pressione antropica sulle coste, anche se fondamentalmente i fenomeni di fioriture dannose rimangono imprevedibili. Un tempo confinate in aree tropicali e sub-tropicali, nuove alghe tossiche produttrici di composti palitossina-simili, quali Ostreopsis spp., si sono diffuse anche in regioni temperate, come il Mediterraneo. Le condizioni di sviluppo di queste fioriture sono legate a fattori ambientali quali irraggiamento, temperatura, concentrazione di nutrienti e alle caratteristiche biologiche delle singole specie. Per valutare il rischio reale sulla salute dell’uomo, vi è necessità di conoscere i parametri ambientali ottimali alla proliferazione delle specie, in termini di temperatura, intensità luminosa e disponibilità di nutrienti, così come di chiarire la struttura delle tossine e le condizioni che possono favorire la loro produzione. In questo studio un ceppo della dinoflagellata Ostreopsis ovata e un ceppo della dinoflagellata Prorocentrum lima sono stati coltivati in colture batch a tre diverse temperature (20°C, 25°C, 30°C) come monocolture e come colture miste. La raccolta è avvenuta nel periodo della loro presunta fase stazionaria. Le informazioni circa il comportamento delle microalghe nelle diverse colture sono state raccolte tramite calcolo del tasso di crescita, analisi del consumo di nutrienti, valutazione dell’efficienza fotosintetica, produzione di tossine. E’ stato applicato un nuovo metodo di misura per il calcolo dei biovolumi. Tra le due microalghe, O. ovata ha esibito maggiore variabilità morfologica, tassi di crescita più elevati e migliore efficienza fotosintetica, mostrando però di subire stress alla temperatura di 30°C. P. lima ha presentato tassi di crescita inferiori, incompleto utilizzo dei nitrati e minore efficienza fotosintetica, accompagnata da grande dissipazione di energia sotto forma di calore. Nelle colture miste, l’uptake più rapido di nitrati da parte di O. ovata ha fatto sì che il numero di cellule di P. lima fosse notevolmente inferiore a quello sviluppatosi nelle colture pure. Ciononostante, la produzione di acido okadaico da parte di P. lima non si è modificata in presenza di O. ovata. Il ceppo di O. ovata utilizzato è stato in grado di produrre ovatossina-a, la tossina maggiormente presente nelle cellule, e quantità circa quattro volte inferiori di ovatossine d+e, oltre a piccole quantità di palitossina putativa. Non è stato invece in grado di produrre le altre due ovatossine conoscisute, l’ovatossina-b e l’ovatossina-c. La produzione di tossine ha dimostrato di essere correlata positivamente con l’aumento della temperatura quando espressa su unità di biovolume, ed è stata inferiore nelle colture miste rispetto alle colture pure. Grazie alla maggiore capacità di crescita, al miglior utilizzo di nutrienti e alla migliore efficienza fotosintetica, O. ovata si dimostra in grado di dare origine a importanti eventi di fioritura, mentre per P. lima i parametri di crescita studiati non evidenziano questa capacità. Le analisi in corso circa la quantità di tossine emesse nel mezzo di coltura permetterano di completare il quadro delineato dal presente studio. Allo stato attuale, non si può concludere che esista attività allelopatica di una specie nei confronti dell’altra.

The benthic dinoflagellate O. ovata represents a serious threat for human health and for the ecology of its blooming areas: thanks to its toxicity this microalga has been responsible for several cases of human intoxication and mass mortalities of benthic invertebrates. Although the large number of studies on this dinoflagellate, the mechanisms underpinning O. ovata growth and toxin production are still far to be fully understood. In this work we have enriched the dataset on this species by carrying out a new experiment on an Adriatic O. cf. ovata strain. Data from this experiment (named Beta) and from another comparable experiment previously conducted on the same strain (named Alpha), revealed some interesting aspects of this dinoflagellate: it is able to grow also in a condition of strong intracellular nutrient deficiency (C:P molar ratio > 400; C:N > 25), reaching extremely low values of chlorophyll-a to carbon ratio (0.0004). Was also found a significant inverse relationships (r > -0.7) between cellular toxin to carbon and cellular nutrient to carbon ratios of experiment Alpha. In the light of these result, we hypothesized that in O. cf. ovata nutrient-stress conditions (intended as intracellular nutrient deficiency) can cause: i) an increase in toxin production; ii) a strong decrease in chlorophyll-a synthesis; iii) a lowering of metabolism associated with the formation of a sort of resting stage. We then used a modelling approach to test and critically evaluate these hypotheses in a mechanistic way: newly developed formulation describing toxin production and fate, and ad hoc changes in the already existent formulations describing chlorophyll synthesis, rest respiration, and mortality, have been incorporated in a simplified version of the European Regional Seas Ecosystem Model (ERSEM), together with a new ad hoc parameterization. The adapted model was able to accurately reproduce many of the trends observed in the Alpha experiment, allowing us to support our hypotheses. Instead the simulations of the experiment Beta were not fully satisfying in quantitative terms. We explained this gap with the presumed different physiological behaviors between the algae of the two experiments, due to the different pre-experimental periods of acclimation: the model was not able to reproduce acclimation processes in its simulations of the experiment Beta. Thus we attempt to simulate the acclimation of the algae to nutrient-stress conditions by manual intervention on some parameters of nutrient-stress thresholds, but we received conflicting results. Further studies are required to shed light on this interesting aspect. In this work we also improve the range of applicability of a state of the art marine biogeochemical model (ERSEM) by implementing in it an ecological relevant process such as the production of toxic compounds.

Negli ultimi 10 anni i blooms attribuibili alla dinoflagellata bentonica Ostreopsis cf. ovata sono aumentati in termini di frequenza ed intensità lungo le coste del Mediterraneo, avendo ripercussioni negative sulla salute umana e forti impatti sulle comunità marine bentoniche, ciò a seguito della produzione di potenti tossine (composti palitossina-simili) da parte della microalga. Tra i fattori ecologici che innescano o regolano le dinamiche dei bloom tossici le interazioni tra microalghe e batteri sono in misura sempre maggiore oggetto di ricerca. In questo studio è stata analizzata la struttura filogenetica della comunità batterica associata ad O. cf. ovata in colture batch e valutate le dinamiche successionali della stessa in relazione alle differenti fasi di crescita della microalga (oltre che in relazione alle dinamiche di abbondanza virale). Lo studio filogenetico è stato effettuato tramite l’ausilio di metodiche molecolari di sequenziamento di next generation (Ion Torrent). Le abbondanze dei batteri e delle particelle virali sono state determinate tramite microscopia ad epifluorescenza; l’abbondanza cellulare algale è stata stimata tramite metodo Uthermohl. Il contributo della frazione batterica ad elevata attività respiratoria è stato determinato tramite doppia colorazione con coloranti DAPI e CTC. Dai dati emersi si evince che la comunità batterica attraversa due fasi di crescita distinte, una più marcata e concomitante con la fase esponenziale di O. cf. ovata, l'altra quando la microalga è in fase media stazionaria. Per quanto concerne la composizione filogenetica della comunità sono stati rilevati 12 phyla, 17 classi e 150 generi, sebbene i dati ottenuti abbiano rilevato una forte dominanza del phylum Proteobacteria con la classe Alphaproteobacteria, seguita dal phylum Bacteroidetes con la classe Sphingobacteria. Variazioni nella struttura filogenetica della comunità batterica, a livello di generi, tra le diverse fasi di crescita della microalga ha permesso di evidenziare ed ipotizzare particolari interazioni di tipo mutualistico e di tipo competitivo.
