Part I. The cellular slime mold Dictyostelium discoideum is a simple eukaryote which undergoes a multi-cellular developmental process. Single cell myxamoebae divide vegetatively in the presence of a food source. When the food is depleted or removed, the cells aggregate, forming a migrating pseudoplasmodium which differentiates into a fruiting body containing stalk and spore cells. I have shown that during the developmental cycle glycogen phosphorylase, aminopeptidase, and alanine transaminase are developmentally regulated, that is their specific activities increased at a specific time in the developmental cycle. Phosphorylase activity is undetectable in developing cells until mid-aggregation whereupon it increases and reaches a maximum at mid-culmination. Thereafter the enzyme disappears. Actinomycin D and cycloheximide studies as well as studies with morphologically aberrant and temporally deranged mutants indicate that prior RNA and concomitant protein synthesis are necessary for the rise and decrease in activity and support the view that the appearance of the enzyme is regulated at the transcriptional level. Aminopeptidase and alanine transaminase increase 3 fold starting at starvation and reach maximum activity at 18 and 5 hours respectively.

The cellular DNA s of D. discoideum were characterized by CsC1 buoyant density gradient centrifugation and by renaturation kinetics. Whole cell DNA exhibits three bands in CsCl: ρ = 1.676 g/cc (nuclear main band), 1.687 (nuclear satellite), and 1.682 (mitochondrial). Reassociation kinetics at a criterion of Tm -23°C indicates that the nuclear reiterated sequences make up 30% of the genome (Cot1/2 (pure) 0.28) and the single-copy DNA 70% (Cot1/2(pure) 70). The complexity of the nuclear genome is 30 x 109 daltons and that of the mitochondrial DNA is 35-40 x 106 daltons (Cot1/2 0.15). rRNA cistrons constitute 2.2% of nuclear DNA and have a ρ = 1.682.

RNA extracted from 4 stages during developmental cycle of Dictyostelium was hybridized with purified single-copy nuclear DNA. The hybrids had properties indicative of single-copy DNA-RNA hybrids. These studies indicate that there are, during development, qualitative and quantitative changes in the portion of the single-copy of the genome transcribed. Overall, 56% of the genome is represented by transcripts between the amoeba and mid-culmination stages. Some 19% are sequences which are represented at all stages while 37% of the genome consists of stage specific sequences.

Part II. RNA and protein synthesis and polysome formation were studied during early development of the surf clam Spisula solidissima embryos. The oocyte has a small number of polysomes and a low but measurable rate of protein synthesis (leucine-3H incorporation). After fertilization, there is a continual increase in the percentage of ribosomes sedimenting in the polysome region. Newly synthesized RNA (uridine-5-3H incorporation) was found in polysomes as early as the 2-cell stage. During cleavage, the newly formed RNA is associated mainly with the light polysomes.

RNA extracted from polysomes labeled at the 4-cell stage is polydisperse, nonribosomal, and non-4 S. Actinomycin D causes a reduction of about 30% of the polysomes formed between fertilization and the 16-cell stage.

In the early cleavage stages the light polysomes are mostly affected by actinomycin.

The piezoelectric composite material could engender stress concentration resulting from small cracks during layers easily, as the cracks growth will lead to the failure of the whole structure. In this paper, a finite element model for piezoelectric composite materials by ABAQUS including interlayer crack was established, and the J integral and crack tip stress of different types PZT patches were calculated by using the equivalent integral method. Then, the J integral for adhesive layers with different thickness, elastic modulus considering and not considering piezoelectricity was investigated. The results show that the J integral of mode I, II reduces with thicker adhesive layer and lower elastic modules, and the J integral of mode II decreases more sharply than that of mode I.

Thermal tuning of the localized surface plasmon resonance (LSPR) of Ag nanoparticles on a thermochromic thin film of VO2 was studied experimentally. The tuning is strongly temperature dependent and thermally reversible. The LSPR wavelength lambda(SPR) shifts to the blue with increasing temperature from 30 to 80 degrees C, and shifts back to the red as temperature decreases. A smart tuning is achievable on condition that the temperature is controlled in a stepwise manner. The tunable wavelength range depends on the particle size or the mass thickness of the metal nanoparticle film. Further, the tunability was found to be enhanced significantly when a layer of TiO2 was introduced to overcoat the Ag nanoparticles, yielding a marked sensitivity factor Delta lambda(SPR)/Delta n, of as large as 480 nm per refractive index unit (n) at the semiconductor phase of VO2.

A engenharia de tecidos é um domínio tecnológico emergente em rápido desenvolvimento que se destina a produzir substitutos viáveis para a restauração, manutenção ou melhoria da função dos tecidos ou órgãos humanos. Uma das estratégias mais predominantes em engenharia de tecidos envolve crescimento celular sobre matrizes de suporte (scaffolds), biocompatíveis e biodegradáveis. Estas matrizes devem possuir não só elevadas propriedades mecânicas e vasculares, mas também uma elevada porosidade. Devido à incompatibilidade destes dois parâmetros, é necessário desenvolver estratégias de simulação de forma a obter estruturas optimizadas. A previsão real das propriedades mecânicas, vasculares e topológicas das matrizes de suporte, produzidas por técnicas de biofabricação, é muito importante para as diversas aplicações em engenharia de tecidos. A presente dissertação apresenta o estado da arte da engenharia de tecidos, bem como as técnicas de biofabricação envolvidas na produção de matrizes de suporte. Para o design optimizado de matrizes de suporte foi adoptada uma metodologia de design baseada tanto em métodos de elementos finitos para o cálculo do comportamento mecânico, vascular e as optimizações topológicas, como em métodos analíticos para a validação das simulações estruturais utilizando dados experimentais. Considerando que as matrizes de suporte são estruturas elementares do tipo LEGO, dois tipos de famílias foram consideradas, superfícies não periódicas e as superfícies triplas periódicas que descrevem superfícies naturais. Os objectivos principais desta dissertação são: i) avaliar as técnicas existentes de engenharia de tecidos; ii) avaliar as técnicas existentes de biofabricação para a produção de matrizes de suporte; iii) avaliar o desempenho e comportamento das matrizes de suporte; iv) implementar uma metodologia de design de matrizes de suporte em variáveis tais como a porosidade, geometria e comportamento mecânico e vascular por forma a auxiliar o processo de design; e por fim, v) validar experimentalmente a metodologia adoptada.

This work introduces two major changes to the conventional protocol for designing plastic antibodies: (i) the imprinted sites were created with charged monomers while the surrounding environment was tailored using neutral material; and (ii) the protein was removed from its imprinted site by means of a protease, aiming at preserving the polymeric network of the plastic antibody. To our knowledge, these approaches were never presented before and the resulting material was named here as smart plastic antibody material (SPAM). As proof of concept, SPAM was tailored on top of disposable gold-screen printed electrodes (Au-SPE), following a bottom-up approach, for targeting myoglobin (Myo) in a point-of-care context. The existence of imprinted sites was checked by comparing a SPAM modified surface to a negative control, consisting of similar material where the template was omitted from the procedure and called non-imprinted materials (NIMs). All stages of the creation of the SPAM and NIM on the Au layer were followed by both electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). AFM imaging was also performed to characterize the topography of the surface. There are two major reasons supporting the fact that plastic antibodies were effectively designed by the above approach: (i) they were visualized for the first time by AFM, being present only in the SPAM network; and (ii) only the SPAM material was able to rebind to the target protein and produce a linear electrical response against EIS and square wave voltammetry (SWV) assays, with NIMs showing a similar-to-random behavior. The SPAM/Au-SPE devices displayed linear responses to Myo in EIS and SWV assays down to 3.5 μg/mL and 0.58 μg/mL, respectively, with detection limits of 1.5 and 0.28 μg/mL. SPAM materials also showed negligible interference from troponin T (TnT), bovine serum albumin (BSA) and urea under SWV assays, showing promising results for point-of-care applications when applied to spiked biological fluids.

O presente estudo teve como objetivos (i) avaliar a validade do emprego do teste SMART, em Drosophila melanogaster, como indicador da contaminação de amostras de água superficial associada a misturas complexas, (ii) detectar a atividade tóxico-genética de dejetos industriais, lançados no rio Caí, empregando o cruzamento aprimorado. Dentro desta perspectiva, pretendeu também (iii) comparar os dados obtidos para as amostras sob influência de despejos industriais com aqueles previamente observados para amostras sob influência de dejetos de origem urbana, provenientes das cidades de Montenegro e São Sebastião do Caí (Silva., 1999). Na tentativa de avaliar a genotoxicidade, associada ao curso final do rio Caí, foram selecionados os seguintes pontos de coleta de despejos industriais: Km 18,6 - situado na foz do arroio Bom Jardim, próximo à área de disposição do efluente final líquido e da drenagem das áreas de disposição dos resíduos sólidos do complexo industrial – e Km 13,6 - no canal da bacia de acumulação e segurança 7 do pólo industrial Neste ensaio genético, cada amostra industrial foi administrada às larvas de terceiro estágio em duas diluições (25% e 50%), bem como na sua forma crua (100%) - sendo avaliados um total de 40 indivíduos por amostra por concentração, totalizando a análise de 11.712.000 células por amostra. Foram utilizados dois controles negativos, o controle de campo – representado pela nascente de um riacho localizada em uma área conservada com fraca ação antrópica e próxima aos pontos do rio – assim como o diluente água destilada. Uma vez que as freqüências das diferentes categorias de manchas não foram significantemente superiores àquelas observadas nos controles negativos (água destilada), os pontos Km 18,6 e Km 13,6 foram caracterizados como destituídos de ação genotóxica nos três meses de coleta : março, junho e setembro. Estes achados sugerem que, nas condições experimentais empregadas, os dejetos de origem industrial não foram capazes de induzir lesões do tipo mutação gênica, cromossômica, assim como eventos relacionados com recombinação mitótica. Por outro lado, a comparação dos dados obtidos no presente estudo com os observados por Silva (1999) para dejetos urbanos, revelou a validade do emprego do teste SMART como uma ferramenta para detecção de contaminação ambiental. De fato, as amostras urbanas referentes aos meses de março (Km 52, 78 e 80) e setembro (Km 52) – coletadas concomitantemente com as de origem industrial – foram diagnosticadas como indutoras de aneuploidias e/ou de grandes deleções cromossômicas. As potências genotóxicas médias estimadas mostraram que o Km 80 foi o local com o maior grau de genotoxicidade – seguido pelos Km 78 e 52 – que apresentaram potências semelhantes Considerando os resultados obtidos, em cinco pontos situados ao longo do curso final do rio Caí, conclui-se que os prejuízos causados pelos dejetos urbanos podem ser tão ou mais nocivos que os impostos pelos de origem industrial – especialmente em função de seu grande volume de lançamento.

Considerando não apenas a importância das antraciclinas na terapêutica do câncer, mas também os efeitos colaterais associados ao uso destas drogas, o presente estudo procurou avaliar a atividade genotóxica de seis antraciclinas em uso clínico - doxorrubicina (DOX), daunorrubicina (DNR), epirrubicina (EPI), idarrubicina (IDA), além dos análogos de última geração, pirarrubicina (THP) e aclarrubicina (ACLA). Para tanto, foi empregado o Teste de Mutação e Recombinação Somática (SMART) em Drosophila melanogaster, que permite a detecção simultânea de mutação gênica e cromossômica, assim como de eventos relacionados com recombinação mitótica - possibilitando quantificar a contribuição deste último parâmetro genético para a genotoxicidade total induzida pelas drogas em estudo. Os dados obtidos a partir desta análise demonstraram que todas as antraciclinas estudadas induziram acréscimos significativos, relacionados tanto à mutação, quanto à recombinação nas células somáticas deste inseto. Além disso, a recombinação mitótica - entre cromossomos homólogos - foi o evento responsável por, aproximadamente, 62 a 100% da toxicidade genética observada. A comparação do potencial genotóxico dos diferentes análogos, através da padronização do número de danos genéticos por unidade de tratamento (mM), caracterizou a ACLA e o THP como as drogas mais potentes – sendo cerca de 20 vezes mais efetivas, como genotoxinas, do que a DOX, o análogo menos potente. Já que a principal ação genotóxica desta família de compostos está relacionada à inibição da topoisomerase II (topo II) – uma enzima que atua no relaxamento da supertorção da dupla hélice de DNA, através da quebra e posterior religação de suas fitas - as diferenças observadas podem ser atribuídas ao mecanismo envolvido neste bloqueio Enquanto os análogos DOX, DNR, EPI, IDA e THP atuam como venenos de topo II - tornando permanentes as quebras induzidas pela enzima - a ACLA inibe a função catalítica desta enzima, impedindo a sua ligação ao DNA. Cabe ainda ressaltar que a genotoxicidade da ACLA não está restrita à sua atividade catalítica sobre a topo II, mas também à sua ação como veneno de topo I e à sua habilidade de intercalar-se na molécula de DNA. Quando a potência genotóxica destas drogas foi associada a suas estruturas químicas, observou-se que substituições no grupamento amino-açúcar levaram a uma maior atividade tóxico-genética, quando comparadas a modificações no cromóforo. Cabe ainda ressaltar que as modificações estruturais, presentes nos análogos DOX, DNR, EPI, IDA e THP, não alteraram a sua ação recombinogênica. No entanto, no que se refere a ACLA, observaram-se decréscimos significativos na indução de recombinação mitótica - que podem ser atribuídas às múltiplas substituições presentes tanto no grupamento amino-açúcar quanto no cromóforo. O conjunto destas observações evidencia que a genotoxicidade total das drogas em estudo está centrada na indução de recombinação homóloga - um evento predominantemente envolvido tanto na iniciação, quanto na progressão do câncer. A alta incidência de tumores secundários, em pacientes submetidos ao tratamento com as antraciclinas, pode, pois, ser atribuída à ação preferencial destas drogas sobre a recombinação mitótica – embora a atividade mutagênica não possa ser desconsiderada.

The synthesis, characterization and application of aqueous dispersions of superparamagnetic/polymer hybrid nanoparticles and capsules is described. Implementation of the superparamagnetic moiety into the polymer matrix enables a response of the nanomaterials towards an external magnetic field. Application of the external field is used for two main purposes: i) As heat generator, when an alternating magnetic field is applied. ii) As structuring agent to self-assemble superparamagnetic nanoparticles in the external field.rnIn the first part, superparamagnetic nanoparticles were used as heat generators in order to achieve a magnetic field induced release of an active compound from nanocontainers. To achieve such a release in remote-controlled fashion, the encapsulation of superparamagnetic nanoparticles into polymer nanocapsules was combined with the integration of a thermolabile compound into the shell of the nanocontainers. The magnetic nanoparticles acted as generators for heat, which decomposed the thermolabile compound. Pores were created in the degrading shell and an active substance was released.rn Additionally, the self-assembly of polymer nanoparticles, which were labeled with a superparamagnetic moiety as structuring agent, could be demonstrated. A combination of a magnetic field induced self-assembly and a sintering of neighboring particles upon an increase in temperature above the glass transition temperature of the polymer was used to form stable architectures. Various structures with tunable periodicity could be obtained ranging from smooth linear nanofibers to zigzag fibers. Besides solely creating linear architectures, the frugal process additionally allowed the creation of arrangements in analogy to more complex polymer architectures: By the introduction of defined junction points, the generation of branched structures and networks was demonstrated. Additionally, by tailoring the interaction of differently sized particles, the preparation of nanoparticle arrangements in statistical or block copolymer fashion was shown. Moreover, a reversible linear assembly and linkage of the nanoparticles was demonstrated following a lock/unlock mechanism. Therefore, the particles were locked in their linear assembly by a stable iron(III) hydroxamato-complex and unlocked by addition of a reducing agent and formation of a less stable iron(II)-complex.Further, in various projects with collaboration partners, nanoparticles and nanocapsules were labeled with a superparamagnetic moiety for their use as contrast agents in magnetic resonance imaging or as magnetically separable dispersions.

This paper presents the design of a bat-like micro aerial vehicle with actuated morphing wings. NiTi shape memory alloys (SMAs) acting as artificial biceps and triceps muscles are used for mimicking the morphing wing mechanism of the bat flight apparatus. Our objective is twofold. Firstly, we have implemented a control architecture that allows an accurate and fast SMA actuation. This control makes use of the electrical resistance measurements of SMAs to adjust morphing wing motions. Secondly, the feasibility of using SMA actuation technology is evaluated for the application at hand. To this purpose, experiments are conducted to analyze the control performance in terms of nominal and overloaded operation modes of the SMAs. This analysis includes: (i) inertial forces regarding the stretchable wing membrane and aerodynamic loads, and (ii) uncertainties due to impact of airflow conditions over the resistance–motion relationship of SMAs. With the proposed control, morphing actuation speed can be increased up to 2.5 Hz, being sufficient to generate lift forces at a cruising speed of 5ms−1.

Advances in electronics nowadays facilitate the design of smart spaces based on physical mash-ups of sensor and actuator devices. At the same time, software paradigms such as Internet of Things (IoT) and Web of Things (WoT) are motivating the creation of technology to support the development and deployment of web-enabled embedded sensor and actuator devices with two major objectives: (i) to integrate sensing and actuating functionalities into everyday objects, and (ii) to easily allow a diversity of devices to plug into the Internet. Currently, developers who are applying this Internet-oriented approach need to have solid understanding about specific platforms and web technologies. In order to alleviate this development process, this research proposes a Resource-Oriented and Ontology-Driven Development (ROOD) methodology based on the Model Driven Architecture (MDA). This methodology aims at enabling the development of smart spaces through a set of modeling tools and semantic technologies that support the definition of the smart space and the automatic generation of code at hardware level. ROOD feasibility is demonstrated by building an adaptive health monitoring service for a Smart Gym.

This work describes a semantic extension for a user-smart object interaction model based on the ECA paradigm (Event-Condition-Action). In this approach, smart objects publish their sensing (event) and action capabilities in the cloud and mobile devices are prepared to retrieve them and act as mediators to configure personalized behaviours for the objects. In this paper, the information handled by this interaction system has been shaped according several semantic models that, together with the integration of an embedded ontological and rule-based reasoner, are exploited in order to (i) automatically detect incompatible ECA rules configurations and to (ii) support complex ECA rules definitions and execution. This semantic extension may significantly improve the management of smart spaces populated with numerous smart objects from mobile personal devices, as it facilitates the configuration of coherent ECA rules.

The deployment of the Ambient Intelligence (AmI) paradigm requires designing and integrating user-centered smart environments to assist people in their daily life activities. This research paper details an integration and validation of multiple heterogeneous sensors with hybrid reasoners that support decision making in order to monitor personal and environmental data at a smart home in a private way. The results innovate on knowledge-based platforms, distributed sensors, connected objects, accessibility and authentication methods to promote independent living for elderly people. TALISMAN+, the AmI framework deployed, integrates four subsystems in the smart home: (i) a mobile biomedical telemonitoring platform to provide elderly patients with continuous disease management; (ii) an integration middleware that allows context capture from heterogeneous sensors to program environment¿s reaction; (iii) a vision system for intelligent monitoring of daily activities in the home; and (iv) an ontologies-based integrated reasoning platform to trigger local actions and manage private information in the smart home. The framework was integrated in two real running environments, the UPM Accessible Digital Home and MetalTIC house, and successfully validated by five experts in home care, elderly people and personal autonomy.
