The effect of lysine amino acid supplementation on the growth characteristics and morphological pattern of skeletal muscle tissue in Nile tilapia Oreochromis niloticus larvae was evaluated. There were four treatments (T) with increasing levels of lysine supplement (T1 = 0.0%: T2 =1.1%; T3 = 1.7%; T4 = 4.0%) and one treatment with a commercial diet (T5). In all treatments, morphological and histochemical muscle tissue analyses were similar. Two distinct layers were identified: a superficial red layer, more developed in the lateral line region, formed by fibres with intense to moderate NADH-TR reaction and strong acid-stable mATPase activity, and a deep white one, most of the Muscle mass, formed by fibres with weak NADH-TR reaction and strong alkali-stable mATPase activity. There was an intermediate layer between these two layers with fibres exhibiting either weak acid-stable or acid-labile mATPase activity. Body mass increase was significantly higher in T5 than in the lysine treatments (T1-T4). There was no difference in number and diameters of muscle fibres between lysine treatments. In T5, muscle fibre diameter and number were higher. The frequency of red fibres with diameters <= 8 mu m was higher in the lysine treatments, and with diameters between 16 and 24 mu m, was higher in T5. Most white fibre diameters in T5 were significantly larger than 24 mu m and in T1-T4 were between 8 and 16 mu m. Background: It is known that amino acid oxidation is increased in tumor-bearing rat muscles and that leucine is an important ketogenic amino acid that provides energy to the skeletal muscle.Methods: To evaluate the effects of a leucine supplemented diet on the intestinal absorption alterations produced by Walker 256, growing pregnant rats were distributed into six groups. Three pregnant groups received a normal protein diet (18% protein):pregnant (N), tumor-bearing (WN), pair-fed rats (Np). Three other pregnant groups were fed a diet supplemented with 3% leucine (15% protein plus 3% leucine):leucine (L), tumor-bearing (WL) and pair-fed with leucine (Lp). Non pregnant rats (C), which received a normal protein diet, were used as a control group. After 20 days, the animals were submitted to intestinal perfusion to measure leucine, methionine and glucose absorption.Results: Tumor-bearing pregnant rats showed impairment in food intake, body weight gain and muscle protein content, which were less accentuated in WL than in WN rats. These metabolic changes led to reduction in both fetal and tumor development. Leucine absorption slightly increased in WN group. In spite of having a significant decrease in leucine and methionine absorption compared to L, the WL group has shown a higher absorption rate of methionine than WN group, probably due to the ingestion of the leucine supplemented diet inducing this amino acid uptake. Glucose absorption was reduced in both tumor-bearing groups.Conclusions: Leucine supplementation during pregnancy in tumor-bearing rats promoted high leucine absorption, increasing the availability of the amino acid for neoplasic cells and, mainly, for fetus and host utilization. This may have contributed to the better preservation of body weight gain, food intake and muscle protein observed in the supplemented rats in relation to the non-supplemented ones.

Effects of zinc (Zn) and manganese (Mn) supplementation to a tricalcium phosphate (TCP) rich diet for tiger puffer have been investigated. A TCP supplement to the diet decreased the growth of fish compared to the control diet with a Ca supplementation from Ca-lactate. However, addition of either Zn or Mn to the TCP supplemented diet could not improve the growth of tiger puffer. Addition of both zinc and manganese to the TCP supplemented diet improved the growth of tiger puffer.

The cornicle secretion of Myzus persicae reared on artificial diet only elicits an alarm response in plant-reared conspecifics after the young aphids have been transferred to plants for 7days. Acetate in the form of 0.32% sodium acetate has been added to the diet as an early step in synthesis of the alarm pheromone, (E)-β-farnesene (EBF). The cornicle secretion of diet-reared aphids then elicits an alarm response. However, there is no difference in internal EBF concentration between plant- and diet-reared aphids. Puncturing aphids, either plant- or diet-reared, with a pin shows that both can elicit an alarm response, whereas it is reduced by half with diet-reared individuals. Although there is no significant difference in the concentration of EBF produced, the total amount in diet-reared aphids is increased by acetate in the diet to a level similar to that in plant-reared individuals: the size of aphids reared on an acetate-supplemented diet is increased and comparable with the size of those that are plant-reared. Bioassays with a range of EBF concentrations show a high threshold for the alarm response. It is concluded that the different size of aphids reared on plants and standard diet results in them secreting, respectively, above and below the response threshold.

This study evaluated the effects of dietary supplementation with 0.3% Saccharomyces cerevisiae yeast cell wall and of vaccination against Streptococcus agalactiae on the cellular component of acute inflammation induced in the coelomic cavity of Nile tilapia Oreochromis niloticus and on survival of the fish after challenge. A total of 84 tilapia of mean (+/- SD) weight 125.0 +/- 1.5 g were distributed among twelve 310 l fiberglass tanks according to a 2 x 2 x 3 factorial design in the following manner: with and without supplementation; 2 stimulations (oily solution without S. agalactiae vaccine and vaccination); 15 d later all fish were intracoelomically challenged with 10(8) CFU ml(-1) of a homologous strain of S. agalactiae, and evaluated after 6, 24 and 48 h, with 7 replicates. The fish received the non-supplemented or supplemented diet for a total of 77 d. The vaccination was performed on the 60th day, intracoelomically, as a single injection of 0.5 ml of the vaccine containing 10(8) CFU ml(-1). Fifteen days later, all the fish were challenged with S. agalactiae by means of an intracoelomic inoculation of 10(8) CFU ml(-1). No mortality was observed among the supplemented fish. The fish that were fed the non-supplemented diet and immunized with the bacterium presented a mortality rate of 28.5%. Among the non-supplemented and non-immunized fish, the mortality rate was 38.09%. Supplementation, in both vaccinated and non-vaccinated fish, induced larger accumulations of thrombocytes, lymphocytes and macrophages at the inflammatory focus. The results suggest that supplementation with 0.3% yeast cell wall, in both vaccinated and non-vaccinated fish, improved the inflammatory response of the fish and protected against the challenge. Vaccination increased the defense response, but the effect was stronger when associated with supplementation with S. cerevisiae.

Pyridoxine requirements of tilapia (Sarotherodon mossambicus Peters) were studied in two separate experiments using casein-based diets. In Experiment 1, fish on pyridoxine supplemented diet (14.0mg/100g diet) showed no adverse symptoms and remained healthy while fish on a pyridoxine-free diet showed abnormal behaviour with high mortality. Graded dietary pyridoxine (0.13 to 3.52mg/100g diet) was used in Experiment 2. Lower dietary supplementations of pyridoxine resulted in reduced weight increase, high mortality, high ratio of serum glutamate-oxal-acetate transaminase glutamate-pyruvate transaminase, and reduced blood sugar. The results suggest the dietary requirement of pyridoxine may be between 0.5g and 1.17mg/100g diet; higher supplementations did not appear to confer any further benefits

A feeding trial was conducted to study the role of vitamin E in growth of Catla catla fry. Newly hatched larvae of Catla were fed with synthetic diet, supplemented with graded levels of vitamin E α0, 50, 100, 150, 200, 250 mg/Kg of diet. The spawn were fed with five times of their body weight for 30 days. Observation was made on the basis of survival, growth, daily weight gain and food conversion ratio. The significant weight gain and highest survival could be achieved by the diet supplemented with 150 mg of vitamin E per kg of the diet. The weight gain per day in 0, 50, 100, 150, 200 and 250 mg vitamin F/kg supplemented diet were 4.0, 5.2, 6.5, 7.8, 6.8 and 6.3 mg, while survival was 50, 51.8, 52.4, 52.8, 52.2 and 52% respectively.

An experiment was conducted to evaluate the possibility of using inorganic fertilizer triple super phosphate (TSP), inorganic fertilizer 16:20 (a 16:20 grade fertilizer contains 16 percent N and 20 percent P20 5), rice-bran and duck-manure as phosphorus sources in formulated fish feed for Nile tilapia ( Oreochromis niloticus). Experiment was conducted for a period of 2 months in net-cages suspended in fertilized earthen ponds and all male sex-reversed Nile tilapia (9.39- 10.37 g) were used in the experiment. Seven treatments including one non-feed treatment were used in this experiment. Treatment 1 (non-feed), treatment 2 (-P) where fish fed with phosphorus non-supplemented diet acted as control 1 and treatment 3, 4, 5, 6 and 7 where fish fed with 3% di-calcium phosphate (DCP), 3% triple supper phosphate (TSP), 7% 16:20 inorganic fertilizer, 30% rice-bran and 30% duck-manure supplemented diet, respectively. Results showed that the TSP and 16:20 grade inorganic fertilizer supplementation in diets as phosphorus sources were equivalent to DCP (Di-calcium phosphate) supplementation in terms of growth performance, feed utilization efficiency and final body composition of Nile tilapia. Ricebran and duck-manure were not found as good phosphorus sources.

Des études précédentes ont montré qu’une carence en vitamine E prédispose à la myopathie du poulet de chair. L’effet d’un ajout de vitamine E dans la diète commerciale sur la dégénérescence des fibres musculaires de la poitrine et de la cuisse a été étudié chez les poulets de chair. Des poulets mâles ROSS 308 (n = 1100) ont été assignés de façon aléatoire à deux traitements alimentaires (aliment commercial + 25 à 50 mg de vitamine E surajouté par kg vs aliment commercial + 0 mg de vitamine E supplémentaire). Les poulets ont été répartis sur 10 parquets (cinq répétitions par traitement). Le poids corporel et la consommation d’aliment ont été mesurés hebdomadairement. Aux jours j28, j35, j42 et j49, du sang a été prélevé pour mesurer le niveau de vitamine E et l’activité de la créatine kinase (CK). Les muscles Pectoralis superficialis et Adductor magnus ont été prélevés pour des analyses histologiques aux jours j28, j42 et j49; les fibres dégénérées ont été dénombrées sur chaque muscle prélevé. La concentration plasmatique de vitamine E était plus élevée dans le groupe supplémenté (P = 0.001). L’activité de la CK n’était pas différente dans les deux groupes (P = 0.20) mais très élevée, et n’était pas toujours en relation avec les dommages musculaires, à cause de grandes fluctuations de la CK entre les individus du même groupe. Le nombre de fibres endommagées était plus élevé dans le muscle Pectoralis superficialis (poitrine) que dans le muscle Adductor magnus (cuisse) dans les deux groupes; il y avait aussi moins de fibres dégénérées à j28 dans la poitrine des poulets qui ont reçus la diète supplémentée. Ces résultats suggèrent que l’ajout de vitamine E à la diète conventionnelle augmente le niveau de vitamine E dans le plasma et dans les tissus, diminue le nombre de fibres dégénérées dans la poitrine des jeunes poulets sans pour autant modifier la conversion alimentaire. La mesure de l’activité plasmatique de la CK ne saurait suffire à elle seule pour détecter précocement la myopathie nutritionnelle dans les élevages de poulets de chair.

The constitutive production of AMPs in shrimps ensures that animals are able to protect themselves from low-level assaults by pathogens present in the environment. As these molecules play important roles in the shrimp immune defense system, the expression level of these AMPs are possible indicators of the immune state of shrimps. The present study also indicates the antiviral property of AMPs, especially ALF, stressing the importance of their up-regulation through the application of immunostimulants/probiotics as a prophylactic strategy in aquaculture. The present study shows that shrimp defense system is equipped enough to evade WSSV infection to a certain extent, when the animals were maintained on marine yeast and probiotic diet, whereas the control diet fed group succumbed to WSSV infection. This study reveals that marine yeast and probiotic supplemented diet can delay the process of WSSV infection and confer greater protection to the animals. Particularly, the protection conferred by marine yeast, C. haemulonii S27 and Bacillus MCCB101 were highly promising imparting greater hope to the aquaculture community to overcome the prevailing disease problems in aquaculture. It may be inferred from the present study that up-regulation of AMP genes could be effected by the application of immunostimulants and probiotics. Also, AMP expression profile could be used as an effective tool for screening immunostimulants and probiotics for application in shrimp culture. Ultimately, it is likely that no single compound or strategy will provide a solution to the problem of disease within aquaculture and that, in reality, a suite of techniques will be required including the manipulation of the rearing environment, addition of probionts as a matter of routine during culture, and the use of immunostimulants and other supplements during vulnerable growth phases. Finally, the development of good management practices, the control of environmental variables, genetic improvement in the penaeid species, understanding of host-virus interaction, modulation of the shrimp immune system, supported by functional genomics and proteomics of this crustacean, as a whole suggests that the control of WSSV is not far.

EGb 761 is a standardized extract from the Ginkgo biloba leaf and is purported to improve age-related memory impairment. The acute and chronic effect of EGb 761 on synaptic transmission and plasticity in hippocampal slices from young adult (8-12 weeks) and aged (18-24 months) C57B1/6 mice was tested because hippocampal plasticity is believed to be a key component of memory. Acutely applied EGb 761 significantly increased neuronal excitability in slices from aged mice by reducing the population spike threshold and increased the early phase of long-term potentiation, though there was no effect in slices from young adults. In chronically treated mice fed for 30 days with an EGb 761-supplemented diet, EGb 761 significantly increased the population spike threshold and long-term potentiation in slices from aged animals, but had no effect on slices from young adults. The rapid effects of EGb 761 on plasticity indicate a direct interaction with the glutamatergic system and raise interesting implications with respect to a mechanism explaining its effect on cognitive enhancement in human subjects experiencing dementia. Vegetable consumption is associated with a reduced risk of colorectal cancer, which is the second most common cancer after lung/breast cancer within Europe. Some putative protective phytochemicals are found in higher amounts in young sprouts than in mature plants. The effect of an extract of mixed cruciferous and legume sprouts on DNA damage induced by H(2)O(2) was measured in HT29 cells using single cell microgelelectrophoresis (comet). Significant antigenotoxic effect (P < or = 0.05) was observed when HT29 cells were pre-incubated with the extract (100 and 200 microL/mL) for 24 hours and then challenged with H(2)O(2). A parallel design intervention study was carried out on 10 male and 10 female healthy adult volunteers (mean age = 25.5 years) fed 113 g of cruciferous and legume sprouts daily for 14 days. The effect of the supplementation was measured on a range of parameters, including DNA damage in lymphocytes (comet), the activity of various detoxifying enzymes (glutathione S-transferase, glutathione peroxidase, and superoxide dismutase), antioxidant status using the ferric reducing ability of plasma assay, plasma antioxidants (uric acid, ascorbic acid, and alpha-tocopherol), blood lipids, plasma levels of lutein, and lycopene. A significant antigenotoxic effect against H(2)O(2)-induced DNA damage was shown in peripheral blood lymphocytes of volunteers who consumed the supplemented diet when compared with the control diet (P = 0.04). No significant induction of detoxifying enzymes was observed during the study, neither were plasma antioxidant levels or activity altered. The results support the theory that consumption of cruciferous vegetables is linked to a reduced risk of cancer via decreased damage to DNA.

Ginkgo biloba (EGb) has been proposed as a promising candidate for cancer chemoprevention and has shown protective effects on the liver against chemically induced oxidative injury and fibrosis. The potential beneficial effects of EGb were investigated in two rat liver carcinogenesis bioassays induced by diethylnitrosamine (DEN). In a short-term study for anti-initiating screening, male Wistar rats were fed a basal diet or supplemented diet with 500 or 1000 ppm EGb and initiated 14 days later with a single dose of DEN (100 mg/kg i.p.). The respective groups were killed 24 h or 2 weeks after DEN-initiation. Liver samples were collected for the analysis of proliferating cell nuclear antigen (PCNA), transforming growth factor alpha (TGF-alpha), p53, apoptosis and induction of single hepatocytes and minifoci positive for the enzyme glutathione S-transferase P-form (GST-P). In a medium-term study for anti-promoting screening, the animals received a single dose of DEN (200 mg/kg i.p.) and, 2 weeks later, were fed a basal diet or supplemented diet with 500 or 1000 ppm EGb for 6 weeks. All animals underwent 70% partial hepatectomy (PH) at week 3 and killed at week 8. Liver samples were colleted to analyze development of preneoplastic foci of altered hepatocytes (FAH) expressing GST-P. In the short-term study, pretreatment of rats with 1000 ppm EGb significantly reduced the rates of cell proliferation, apoptosis and p53, TGF-a immunoreactivity and the number of GST-P-positive hepatocytes. In the medium-term study, EGb treatment during the post-initiation stage failed to reduce the development of DEN-induced GST-P-positive foci. Thus, EGb presented inhibitory actions during initiation but not promotion of rat liver carcinogenesis induced by DEN. (C) 2008 Elsevier B.V. All rights reserved.
