No presente documento é registrada a ocorrência de infestações preocupantes pela mosca-dos-estábulos, Stomoxys calcitrans L. (Diptera: Muscidae), em propriedades pecuárias próximas de usinas sucroalcooleiras na região Sul de Mato Grosso do Sul, no ano de 2009. A biologia do inseto; sua importância epidemiológica, econômica e veterinária e; possíveis alternativas de controle são apresentadas e discutidas. Entre as alternativas de controle sugere-se, em especial, o revolvimento completo duas vezes por semana do material de compostagem nas usinas e a drenagem do local onde é executada, assim como o enterrio parcial da palha pós-colheita da cana-de-açúcar com uso de cultivador. Nas propriedades pecuárias recomenda-se a limpeza de locais de criação da mosca, representado por acúmulo de dejetos de animais domésticos e ou de resíduos alimentares, especialmente em confinamentos e leiterias, dentre outras recomendações. Somente com ações integradas será possível resolver o problema e contribuir para o desenvolvimento sustentável de duas importantes cadeias produtivas do agronegócio brasileiro.

Bovine besnoitiosis, caused by the cyst-forming apicomplexan Besnoitia besnoiti, is commonly reported in some restricted regions of South-Western Europe, and in larger regions of Africa and Asia. This infection is thought to be transmitted by blood feeding insects and is responsible for major economic losses in cattle production. A recent emergence in Europe, notified in the Centre of France, Spain and Germany, has attracted more attention to this disease. Clinical signs could appear in some animals; however, many infected cattle remain asymptomatic or show scleral-conjunctival cysts (SCC) only. Recent development of serological methods allows carrying out seroepidemiological field studies. In this respect, a long-term investigation was performed in a dairy cattle farm localized in an enzootic area of besnoitiosis of South-western France between March 2008 and May 2009. The objective was to estimate the seasonal pattern of B. besnoiti infections based on the presence of SCC and serology (ELISA and Western blot). In parallel, an entomological survey was conducted to describe population dynamics of Stomoxys calcitrans and Tabanidae species. The seroprevalence determined by Western blot in a cohort of 57 animals continuously present during the whole survey increased from 30% in March 2008 to 89.5% in May 2009 and was always higher than the prevalence based on clinically assessed SCC. New positive B. besnoitia seroconversions occurred throughout the year with the highest number in spring. In addition, many seroconversions were reported in the two months before turn-out and could be associated with a high indoors activity of S. calcitrans during this period.

We have cloned and sequenced two defensins, Smd1 and Smd2, from anterior midgut tissue of the blood-sucking fly Stomoxys calcitrans. The DNA and N-terminal protein sequences suggest both are produced as prepropeptides. Smd1 differs from the classic defensin pattern in having an unusual six-amino acid-long N-terminal sequence. Both Smd1 and Smd2 have lower pI points and charge than insect defensins derived from fat body/hemocytes. Northern analysis shows both of these defensin molecules are tissue specific; both are produced by the anterior midgut tissue and, unlike the other insect defensins reported to date, neither appears to be expressed in fat body or hemocytes. Northern analysis also shows that mRNAs for both defensins are constitutively produced in the anterior midgut tissues and that these transcripts are up-regulated in response to sterile as well as a lipopolysaccharide-containing blood meal. However, anti-Gram-negative biological activity in the midgut is substantially enhanced by lipopolysaccharide. These findings suggest that the insect midgut has its own tissue-specific immune mechanisms and that this invertebrate epithelium is, like several vertebrate epithelia, protected by specific antibacterial peptides.

Con el objetivo de determinar la prevalencia de hemoparásitos en bovino de leche y su efecto sobre la productividad de los animales; se identificó la presencia de hemoparásitos y se analizaron las diferencias agroecológicas que permiten la presencia de estas parasitosis en la Finca Los Robles ubicada en el municipio de San Rafael del Norte, Jinotega y la Finca Vista Hermosa ubicada en el municipio de San Pedro de Lovago, Chontales. Para el estudio se hizo un análisis descriptivo, con distribuciones de frecuencia; el estudio se realizó en vacas de ordeño con un rango de peso de 425 a 352 kg en ambas fincas para un total de 12 animales (6 por finca), sin aplicar ningún tipo de medicamento en un periodo de 63 días. Efectuando 3 muestreos con intervalos de 21 días entre los meses de Marzo y Abril, para un total 36 exámenes; Durante el estudio se realizó el pesaje de la leche semanalmente, el pesaje corporal de las vacas, y la realización de hematocrito para determinar la anemia causada por los hemoparásitos, la toma de muestras se realizó mediante venopunción yugular, se extrajo 2ml de sangre, trasladándose estas al laboratorio de parasitología (FACA-UNA) donde se elaboraron 2 frotis sanguíneos por cada muestra, usando el método de tinción de Giemsa. Obteniendo 2 especies de hemoparásitos Babesia y Anaplasma. Se determinó que en la Finca Los Robles, en el primer muestreo la prevalencia de Anaplasma marginale y centrale fue de un 100 % y en Vista Hermosa fue de 50% a Anaplasma marginale y el 16.6% A.centrale, resultando nula a Babesia; En el segundo y tercer muestreo se obtuvo en los Robles el 100% de las muestras a Anaplasma marginale y centrale y el 50% positivas a Babesia, mientras que en Vista hermosa se obtuvo el 83.33 % A. Marginale y el 33.33 % Anaplasma Centrale, siendo nula la presencia de Babesia; la prevalencia de Anaplasma puede ser mayor a la de Babesia puesto que la garrapata no es el único vector y su presencia está ligada a los insectos picadores como Tábanus y Stomoxys calcitrans. El hematocrito indicó que los individuos de Los Robles presentaron una disminución gradual en el cuadro hemático, siendo un individuo el más afectado con un 19 % debido a que presentaba infección mixta Babesia y Anaplasma, en Vista Hermosa se mantuvieron en el rango fisiológico (46 - 24), identificándose una con el rango más bajo 27 %. Las pérdidas económicas en el periodo de estudio (63 días) en un lote de 6 animales fueron de C$ 3251 en leche y C$ 1092 en peso, para Los Robles y en Vista Hermosa fue de C$ 2268 en leche y C$ 3360 en peso. Para realizar un control eficaz de los hemoparásitos se debe contar con un plan de control de ectoparásitos, efectuar exámenes complementarios frecuentemente, para determinar la especie de hemoparásito y así aplicar un tratamiento específico, además mantener un plan de vigilancia permanente.

Esta é a primeira descrição de um surto de Trypanosomavivax ocorrido no Estado de São Paulo, no município de Lins. Animais acometidos apresentaram febre, icterícia, diminuição da produção de leite, perda de peso, diarreia profusa, abortos, anemia, leucocitose e hiperfibrigenemia. Foram registrados 31 óbitos de vacas e bezerros em 1.080 bovinos no total. Três vacas apresentaram sintomatologia nervosa, como dismetria, ataxia e fraqueza muscular, além de ptialismo, aumento de linfonodos e edema submandibular. Hemoparasitas flagelados foram observados em esfregaços sanguíneos, e a espécie de tripanossomo foi diagnosticada como T.vivax por PCR. A cepa de T. vivax mostrou ser resistente ao tratamento com aceturato de diaminozeno e a infecção disseminou rapidamente no rebanho. Pelo ELISA, observou-se que 98,36% (599) das amostras de soro colhidas apresentaram títulos positivos para IgG anti-T.vivax. O surto ocorreu em condições de baixa precipitação pluviométrica, fato que indica que outros fatores estavam envolvidos na ocorrência desse surto, como a ausência de tabanídeos e a grande presença de Haematobia irritans e Stomoxys calcitrans, cujo aumento populacional pode ser devido ao uso de resíduos de usinas de açúcar e álcool nos canaviais que circundavam a granja leiteira.

De larvas e pupas de Musca domestica, Chrysomya albiceps, Cochliomyia homivorax, Stomoxys calcitrans e Syntesiomyia nudiseta coletadas em diversos ambiente, em São Paulo, Paraná, Mato Grosso, Mato Grosso do Sul e Minas Gerais, foram obtidas dez espécies de microhimenópteros parasitóides da supermamília Chalcidoidea, algumas assinaladas pela primeira vez no Brasil.

This is the first description of a trypanosoma vivax outbreak in the state of Sao Paulo (municipality of Lins). From the ELISA test, 599 serum samples (98.36%) were positive for anti-T. vivax IgG antibodies. This outbreak occurred during a very dry period, which indicates that other factors were involved in the outbreak, such as absence of tabanids and large populations of Haematobia irritans and Stomoxys calcitrans. The increases in these populations may have been due to the use of biosolid waste from sugar and ethanol plants in the sugarcane plantations surrounding the dairy farm.

Fluorescence spectrophotometry can reliably detect levels of the pteridine 6-biopterin in the heads of individual Drosophila serrata Malloch 1927. Pteridine content in both laboratory and field captured flies is typically a level of magnitude higher than the minimally detectable level (mean(lab)=0.54 units, mean(field)=0.44 units, minimum detectable level=0.01 units) and can be used to predict individual age in laboratory populations with high certainty (r(2)=57%). Laboratory studies of individuals of known age ( from 1 to 48 days old) indicate that while pteridine level increases linearly with age, they also increase in a linear manner with rearing temperature and ambient light levels, but are independent of sex. As expected, the longevity of laboratory-reared males ( at least 48 days) is higher than the range of predicted ages of wild-caught males based on individual pteridine levels (40 days). However, the predictive equation based on pteridine level alone suggested that a number of wild-caught males were less than 0 days old, and the 95% confidence for these predictions based on the inverse regression broad. The age of the oldest wild-caught male is to fall within the range of 2 to 50 days. The effects of temperature and light intensity determined in laboratory study (effect sizes omega(2)=14.3 and respectively) suggests that the calibration of the prediction equation for field populations would significantly improved when combined with fine scaled studies of habitat temperature and light conditions. ability to determine relative age in individual wild-caught D. serrata presents great opportunities for a variety evolutionary studies on the dynamics of populations.

Diatom culture and larval feeding experiments were conducted to test the viability and acceptability of preserved algal concentrates. C. calcitrans is characterised by the presence of setae which keep them suspended in cultures and make autoflocculation very difficult. Flocculation was induced by the addition of a floc-forming chemical. Using the optimum conditions, it was possible to harvest the algae within 1-h settling time and with about 84% recovery. The viability of frozen Chaetoceros was determined by actual cell reproduction. Preliminary feeding experiments showed that Chaetoceros can be successfully used as a substitute for fresh diatoms as feed for Penaeus monodon larvae. Simple freezing techniques, with or without the use of protectants has been found convenient for preserving algal concentrates in small volumes for both feeding and culture purposes.

Batch cultures of C. calcitrans were maintained indoors at a temperature range of 21 to 25 C and continuously illuminated by 40-watt daylight fluorescent lights. Cultures were exposed to 5 different intensities from 200 to 25,000 lux.Population counts show that light intensity affects growth and reproduction of the algae cultivated. A comparison of population peak growths showed cultures illuminated by 12,000 lux to have higher cell counts than those exposed to higher or lower light intensities.

In natural systems phytoplankton interact with planktonic (free living) and attached epiphytic bacteria both synergistically and antagonistically. The specificity of the association with micro algae and bacteria differs in terms of adhesion mechanisms and metabolic cooperation. Present research was carried out to study the effect of bacterial isolates namely Bacillus sp. and Pseudomonas sp. from algal culture systems on the growth of micro algae such as Chaetoceros calcitrans and Nannochloropsis oculata. C. calcitrans (F= 15.34; P<0.05) and N. oculata (F=12.52; P<0.05) showed significantly higher growth, in treatments with Bacillus sp. and Pseudomonas sp when compared to control.

A novel technique was developed for the flocculation of marine microalgae commonly used in aquaculture. The process entailed an adjustment of pH of culture to between 10 and 10.6 using NaOH, followed by addition of a non-ionic polymer Magnafloc LT-25 to a final concentration of 0.5 mg L-1. The ensuing flocculate was harvested, and neutralised giving a final concentration factor of between 200- and 800-fold. This process was successfully applied to harvest cells of Chaetoceros calcitrans, C. muelleri, Thalassiosira pseudonana, Attheya septentrionalis, Nitzschia closterium, Skeletonema sp., Tetraselmis suecica and Rhodomonas salina, with efficiencies >=80%. The process was rapid, simple and inexpensive, and relatively cost neutral with increasing volume (cf. concentration by centrifugation). Harvested material was readily disaggregated to single cell suspensions by dilution in seawater and mild agitation. Microscopic examination of the cells showed them to be indistinguishable from corresponding non-flocculated cells. Chlorophyll analysis of concentrates prepared from cultures of Concentrates of T. pseudonana prepared using pH-induced flocculation gave better growth of juvenile Pacific oysters (Crassostrea gigas) than concentrates prepared by ferric flocculation, or centrifuged concentrates using a cream separator or laboratory centrifuge. In follow up experiments, concentrates prepared from 1000 L Chaetoceros muelleri cultures were effective as supplementary diets to improve the growth of juvenile C. gigas and the scallop Pecten fumatus reared under commercial conditions, though not as effective as the corresponding live algae. The experiments demonstrated a proof-of-concept for a commercial application of concentrates prepared by flocculation, especially for use at a remote nursery without on-site mass-algal culture facilities.

Diatoms were collected from Buyuan Bay, and from the hatchery tanks at Tigbauan, to determine the commonly occurring species, the feasibility of culturing these species, and the potential of these selected species as food for larval P. monodon. The commonly occurring diatoms were identified as Chaetoceros calcitrans, Navicula grimmei, Nitzchia seriata, Nitzchia closterium and Amphiprora sp. These diatoms were isolated and unialgal cultures prepared. Protein content analysis using the micro-Kjildahl method gave the following result: C. calcitrans, 11 . 78%; Nitzchia seriata, 25%; Nitzchia closterium, 30 . 5%; Navicula grimmei, 9 . 06% and Amphiprora sp. 8 . 96%. Feeding experiments were conducted to determine acceptability of the different diatom species and percentage survival of larval stages Z SUB-1 -M SUB-2 . Larvae were placed in 4-l capacity plastic containers with a stocking density of 10/l. The results of several feeding trials using the different mass-produced diatoms are summarized. From the data gathered, C. calcitrans appears to be the most promising candidate as feed for zoea and mysis stages of P. monodon. The average percentage survival of C. calcitrans was 63 . 76% for the 3 trials, and as high as 82 . 22% in the third trial. Comparatively high percentage survival of larvae was also recorded when Nitzchia seriata (48 . 17%) and Nitzchia closterium (67 . 6%) were given as feed, while both Amphiprora sp. and Navicula grimmei gave 0% survival. The poor results with Amphiprora sp. and Navicula grimmei may be due to their low protein content (8 . 96% and 9 . 06%, respectively) and the inability of the larvae to ingest them. Navicula and Amphiprora were observed to cling to the appendages of the larvae and to settle down in the medium making them unavailable to the larvae. Low survival was also noted when frozen C. calcitrans was used (14 . 25%). This may be due partly to the effect of the floculating agent (ALSO SUB-4 . 25 g/l) used in concentrating the diatoms. When protein contents of C. calcitrans, N. seriata and N. closterium are compared, the 2 Nitzchia species have relatively higher protein contents than C. calcitrans and, therefore, could be the more desirable feed candidates. However, few feeding trials were made using Nitzchia so that additional investigations will have to be done on this aspect.
