O extrato de folhas de estévia (Stevia rebaudiana Bertoni) é o único edulcorante utilizado na substituição da sacarose, que pode ser produzido totalmente no Brasil. O objetivo deste estudo foi determinar os comportamentos de características temporais dos estímulos doce e amargo da estevia em doçuras equivalentes a soluções de sacarose (DESS) a 3 e 10%. As curvas tempo-intensidade (T-I) para cada substância foram coletadas utilizando-se o programa Sistema de Coleta de Dados Tempo-Intensidade - SCDTI para Windows, onde os provadores registravam com auxílio do mouse a percepção de cada estímulo solicitado em função do tempo, para cada amostra. Os parâmetros das curvas T-I coletados foram: tempo de intensidade máxima (TImax), intensidade máxima (Imax), tempo onde a intensidade máxima começa a declinar (Td), tempo de platô (Platô), área sob a curva (Área) e tempo total de duração do estímulo (Ttot). Os parâmetros Td, Ttot, Área e Platô das curvas T-I para o estímulo doce nos dois níveis de doçura foram significativamente superiores para estévia, enquanto TImax e Imax foram significativamente menores (p£0,05), sendo que as diferenças entre os valores para as duas substâncias foram muito maiores a DESS a 10%. A sacarose não apresentou nenhum registro para o estímulo amargo tanto a 3 como a 10%, enquanto a estévia apresentou curvas T-I características, com intensidade e o tempo total de duração dependente da concentração.

The microorganism Sclerotinia was isolated from roots of Stevia rebaudiana (Bert.) Bertoni in plantations in the northwest of Parana and submitted to the cultivation in the presence of extracts and vegetable balsams of Tarragon (Artemisia draconculus), Thyme (Thymus vulgaris), Manjerona (Origanum majorona), Mint citrata (Mintpiperita var. citrata), Purple Basil (Ocimum basilicum L.), Andiroba (Carapa guanensis) and Copaíba (Copaifera reticulata Ducke). The first five oils were extracted by steam drags, after the drying of the vegetable in greenhouse with circulation of air at 45°C. The last two were used in natura. A suspension (100ìl) of fungus previously cultivated, was added to each plate. The results show that after 7 days of incubation the thyme oils 10ìl, purple basil 25ìl, manjerona 25ìl, mint citrata 50ìl, tarragon 50ìl were capable to inhibit the growth of Sclerotinia, while the andiroba oil only reached this result with 200ìl. The copaiba balsam, even in the concentration of 500ìl, was unable to inhibit the growth of the microorganism.

Stevia rebaudiana is a traditional crop of Latin America, now rising to international interest thanks to its specific glycosides that are very sweet and still poor of calories making its potential interesting for dietary recommendations. Its short growing cycle makes this species possible as an annual crop also in non tropical areas, including the Mediterranean Basin and parts of Italy, where rainfall or irrigation is available during late spring and summer. One of the main concerns is the quantity of water necessary for the crop and the ideal number of plants per hectare in order to maximize water use efficiency. The present research was carried out in central Italy to compare yields and water consumption in plots with three plant densities (50, 25, 16 plants per square metre). The results show that the most dense plots can give the best results in the area of the trial.

The demand for an alternative and a high potency sweetener to substitute sugar increases year in year out, more so as a high percentage of the world population becomes increasingly diabetic. The alternative natural sweetener at hand has been Stevia rebaudiana Bertoni, a plant species, native to Paraguay and a member of the family compositae. Stevia is usually propagated by stem cuttings due to low percentage (10 %) seed germination, thus limiting large scales cultivation. To cultivate this crop en mass therefore, there is need to evolve efficient rooting techniques. Influences of irradiation from light, and hormones on rooting have been reported. The rooting efficacy in stem cuttings of this crop under varying light wavelengths, dark and hormone factors was investigated. Evaluated parameters include- (i) day of root emergent, (ii) percentage of rooted cuttings, (iii) average number, (iv) length and (v) width, of roots. Analysis of variance at p<.05 revealed that the number, length and width, of roots differed significantly in each case at p<0.000. Light irradiation was highly effective and a necessary factor for rooting in stems cuttings of this crop. The red light-IBA combined factors served best in stem micro-cutting practice and facilitation of effective mass cultivation in stevia crop.

El presente trabajo de grado hace referencia al uso de fertilizantes nitrogenados de lenta liberaci?n, los cuales buscan soluciones de conservaci?n del medio ambiente y pretenden reducir los costos en la aplicaci?n de los fertilizantes. En Colombia, para el establecimiento de productos como la papa, del costo total de producci?n por hect?rea, se encuentra destinado aproximadamente un 29.6% a la aplicaci?n de fertilizantes, mientras que en cultivos como el cacao es el 8% (SAC, 2006). En el Valle del Cauca, se observan rangos que van desde el 11,5% para la arveja, hasta el 41,9% en el repollo (SEDAMA,2010); por lo tanto, existen sectores en los cuales los efectos ser?n mucho m?s cr?ticos, debido al gran consumo de fertilizantes nitrogenados que demanda el desarrollo adecuado del cultivo. Gran parte de los productores agr?colas aplican los fertilizantes de forma emp?rica de acuerdo con la respuesta del cultivo, y en ocasiones se cae en el error de aplicar m?s cantidad de fertilizante que el necesario por el cultivo, lo cual se traduce en una posterior contaminaci?n del medio ambiente y p?rdidas econ?micas para el agricultor.

The demand for natural sweeteners has been gaining more and more importance due to the great controversy associated with the use of some synthetic sweeteners as cyclamates, aspartame and acesulfame-K. The steviol glycosides (E 960) are a group of natural sweeteners of generalized use; these compounds are obtained from Stevia rebaudiana Bertoni, a sweet plant native from South America (Carocho et al., 2015). However, Stevia rebaudiana Bertoni may have other uses to be exploited, in particular due to its antioxidant capacity. This plant is already produced in Portugal but it is important to evaluate if the plant chemical composition is maintained regardless of culture conditions. Therefore, in this study, stevia samples were cultivated in Braganca (northeastern of Portugal) in a field trial with defined culture conditions. After harvesting, the plants were submitted to two different treatments: kept fresh by freezing (-20°C) and oven-dried (30°C). The antioxidant profile of the samples was studied through evaluation of free radicals scavenging activity, reducing power, phenolic compounds (HPLC-DAD-ESI/MS), tocopherols (HPLC-fluorescence) and free sugars (HPLC-RI). Significant differences were observed: while oven-dried samples showed the highest antioxidant activity and phenolic compounds concentration (mainly 5-O-caffeoylquinic acid and 3,5-O-dicaffeoylquinic acid), the frozen fresh samples had the highest values of total tocopherols and total sugars. These results confirm that the plants grown in Bragança have excellent bioactive secondary metabolites responsible for the observed antioxidant capacity.

Las hojas de Stevia rebaudiana y sus glucósidos recientemente se han comenzado a utilizar de manera importante como edulcorantes. Sin embargo, existen reportes acerca del efecto antihiperglucemiante de extractos y un componente glucósido. El objetivo de este trabajo fue cuantificar los glucósidos de S. rebaudiana, evaluar la citotoxicidad y el efecto de la administración aguda y crónica del extracto sobre la glucemia en modelos animales como en humanos. Los glucósidos de los extracto de las variedades Morita II y Criolla se cuantificaron por HPLC, empleando una columna C18 (250 mm x 4.6 mm y tamaño de partícula de 5μm), con detector UV a 210 nm, fase móvil de acetonitrilo/amortiguador fosfato de sodio 10 mmol/L, pH 2.6 (32:68 v/v). Se realizó un estudio de citotoxicidad en células Vero, una prueba de tolerancia a la glucosa intraperitoneal y ensayo de consumo crónico (4 semanas) en un modelo animal de diabetes y finalmente, se determinó el índice glicémico (I.G) en individuos sanos. El contenido de glucósidos fue mayor en la variedad Morita II aunque la CC50 en ambas es >300 μg/mL. Las áreas bajo la curva de la IPGTT así como la glucosa en ayuno de los animales no fueron significativamente diferentes (p>0.05) y el I.G. del extracto fue 11.11%, lo cual lo clasifica como I.G. bajo. El extracto de S. rebaudiana Morita II es de bajo índice glicémico y, en las dosis evaluadas, no es citotóxico ni posee efecto agudo o crónico sobre la glucemia, lo cual lo hace un edulcorante inocuo.

The food industry is moving towards the use of natural sweeteners such as those produced by Stevia rebaudiana due to the number of health and safety concerns surrounding artificial sweeteners. Despite the fact that these sweeteners are natural; they cannot be assumed safe. Steviol glycosides have a steroidal structure and therefore may have the potential to act as an endocrine disruptor in the body. Reporter gene assays (RGAs), H295R steroidogenesis assay and Ca(2+) fluorimetry based assays using human sperm cells have been used to assess the endocrine disrupting potential of two steviol glycosides: stevioside and rebaudioside A, and their metabolite steviol. A decrease in transcriptional activity of the progestagen receptor was seen following treatment with 25,000 ng/ml steviol in the presence of progesterone (157 ng/ml) resulting in a 31% decrease in progestagen response (p=<0.01). At the level of steroidogenesis, the metabolite steviol (500-25,000 ng/ml) increased progesterone production significantly by 2.3 fold when exposed to 10,000 ng/ml (p=<0.05) and 5 fold when exposed to 25,000 ng/ml (p=<0.001). Additionally, steviol was found to induce an agonistic response on CatSper, a progesterone receptor of sperm, causing a rapid influx of Ca(2+). The response was fully inhibited using a specific CatSper inhibitor. These findings highlight the potential for steviol to act as a potential endocrine disruptor.

Special diets are used to mitigate many human diseases. When these diets require changes in carbohydrate content, then sweetness becomes an important characteristic. The range of low-calorie sweeteners available to the food industry is expanding. It is essential to have an exact knowledge of the relative sweetness of various sweeteners in relation to different sucrose concentrations. The objective of this study was to determine the variation on the relative sweetness of aspartame (APM), stevia [Stevia rebaudiana (Bert.) Bertoni] leaf extract (SrB) and the mixture cyclamate/saccharin - two parts of cyclamate and one part of saccharin - (C/S) with the increase in their concentrations, and in neutral and acid pH in equi-sweet concentration to 10% sucrose, using magnitude estimation. Sweetness equivalence of SrB in relation to sucrose concentrations of 20% or higher and of APM and C/S to sucrose concentrations of 40% or higher could not be determined, because a bitter taste predominated. The potency of all sweeteners decreased as the level of sweetner increased. In equi-sweet concentration of sucrose at 10%, with pH 7.0 and pH 3.0, the potency was practically the same for all sweeteners evaluated.

La demanda creciente de los edulcorantes como sustitutos del azúcar y de los productos acalóricos o con bajas calorías de los consumidores españoles (entre 2008 y 2012 la demanda de edulcorantes creció en España un 36,5%) ha sido la base para la realización de este trabajo. En él se ha estudiado el cultivo de Stevia rebaudiana Bertoni, una planta compuesta originaria de Paraguay de la que se obtiene un edulcorante natural y acalórico del mismo nombre, stevia. Se ha analizado la viabilidad del cultivo en el sur de España, concretamente en Andalucía, mediante el estudio y la clasificación de una colección de más de 50 suelos de esta región según su índice de viabilidad para el cultivo de stevia. De este modo, se ha podido señalar la aptitud de las distintas comarcas de Andalucía para el cultivo de esta especie. De acuerdo con los resultados obtenidos, un 32% de las comarcas son de buena aptitud, un 53% son de aptitud media y un 15% son de baja aptitud para este cultivo.
