As marcas globais mantiveram muita atenção no campo do marketing (Kotler, 1997; Holt, Quelch, e Taylor, 2004; Özsomer e Altaras, 2008), enquanto as marcas locais foram subestimadas (Ger, 1999; Schuiling e Kapferer, 2004). No entanto, o debate adaptação contra padronização foi amplamente discutido. Centra-se na definição de como uma empresa internacional deve construir a sua estratégia: ao padronizar sua estratégia de marketing ou, adaptando para melhor atender a cultura e às necessidades locais (Levitt, 1983; Subhash, 1989; Herbig, 1998; Holt, 2004; Melewar e Vemmervik , 2004; Heerden e Barter, 2008). No entanto, este assunto não foi discutido no contexto específico do consumo alternativo oferecido por concorrentes locais específicos. Hoje em dia, um aumento na oferta de produtos alternativos é observado. O consumo socialmente responsável está crescendo (Sen e Bhattacharya, 2001; Holt, 2002; Loureiro, 2002; François-Lecompte e Valette-Florence, 2006). O mercado dos refrigerantes de cola é de interesse particular. Colas alternativas são refrigerantes de cola que surgiram durante a última década em algumas regiões ou zonas específicas do mundo. Estas colas claramente posicionam-se como uma alternativa ao refrigerante global Coca-Cola. A alternativa não é baseada no preço mas nas características especiais dos produtos que constituem uma proposição de valor específica, diferente da Coca-Cola. Na França, desde uma década, o número de colas regionais aumentou, sendo mais de quinze hoje. O refregirante Breizh Cola foi lançado em 2002 e atinge quase uma quota de mercado de 10% na região Bretanha hoje. Em 2009, a Coca-Cola Entreprise iniciou uma campanha de marketing específica, na Bretanha, baseada em recursos visuais e parcerias regionais. Este caso de adaptação em um contexto de concorrência local específico é explorado nesta dissertação que incide sobre as razões da preferência para Breizh Cola, de um lado, e sobre as acções empreendidas pela Coca-Cola na Bretanha, do outro lado. Este estudo mostra que a Coca-Cola anda nos passos de Breizh Cola, a fim de melhor atender às expectativas local.

Normorphine was synthesised from morphine by thermal decomposition of an N-alpha-chloroethylchloroformate adduct, and purified (> 98% purity) using semipreparative HPLC with ultraviolet detection. Normorphine-3-glucuronide (NM3G) was biochemically synthesised using the substrate normorphine, uridine diphosphoglucuronic acid and Sprague-Dawley rat liver microsomes in a 75% yield (relative to normorphine base). The synthesised NM3G was purified by precipitation and washing with acetonitrile. Determinations of purity using HPLC with electrochemical and ultraviolet detection confirmed that the NM3G produced was of high (> 99%) purity. Mass spectrometry, fourier transform infrared spectrophotometry and nuclear magnetic resonance spectrometry confirmed the structure, especially placement of the glucuronide moiety at the 3-phenolic position and not at the 17-nitrogen. Administration of NM3G by the intracerebroventricular (icy) route to rats in doses of 2.5 and 7.5 mu g resulted in the development of central nervous system (CNS) excitatory behavioural effects including myoclonus, chewing, wet-dog shakes, ataxia and explosive motor behaviour. At an icy dose of 7.5 mu g, NM3G also induced short periods of tonic-clonic convulsive activity. Thus, NM3G elicits CNS excitation following supraspinal administration in a manner analogous to morphine-3-glucuronide (M3G), the major metabolite of morphine (1). Further studies are required to determine whether NM3G attenuates morphine-induced antinociception in se similar manner to M3G.

In humans, hydromorphone (HMOR) is metabolised principally by conjugation with glucuronic acid to form hydromorphone-3-glucuronide (H3G), a close structural analogue of morphine-3-glucuronide (M3G), the major metabolite of morphine. In a previous study we described the biochemical synthesis of H3G together with a preliminary evaluation of its pharmacology which revealed that it is a neuro-excitant in rats in a manner analogous to M3G. Thus the aims of the current study were to quantify the neuro-excitatory behaviours evoked by intracerebroventricular (icv) H3G in the rat and to define its potency relative to M3G. Groups of adult male Sprague-Dawley rats received icy injections (1 muL) of H3G (1 - 3 mug), M3G (2 - 7 mug) or vehicle via a stainless steel guide cannula that had been implanted stereotaxically seven days prior to drug administration. Behavioural excitation was monitored by scoring fifteen different behaviours (myoclonic jerks, chewing, wet-dog-shakes, rearing, tonic-clonic-convulsions, explosive motor behaviour, grooming, exploring, general activity, eating, staring, ataxia, righting reflex, body posture, touch evoked agitation) immediately prior to icy injection and at the following post-dosing times: 5, 15, 25, 35, 50, 65 and 80 min. H3G produced dose-dependent behavioural excitation in a manner analogous to that reported previously for M3G by our laboratory and reproduced herein. H3G was found to be approximately 2.5-fold more potent than M3G, such that the mean (+/- S.D.) ED50 values were 2.3 (+/- 0.1) mug and 6.1 (+/- 0.6) mug respectively. Thus, our data clearly imply that if H3G crosses the BBB with equivalent efficiency to M3G, then the myoclonus, allodynia and seizures observed in some patients dosed chronically with large systemic doses of HMOR, are almost certainly due to the accumulation of sufficient H3G in the central nervous system, to evoke behavioural excitation. In this work, a repair technique with adhesively bonded carbon-epoxy patches is proposed for wood members damaged by horizontal shear and under bending loads. This damage is characterized by horizontal crack growth near the neutral plane of the wood beam, normally originating from checks and shakes. The repair consists of adhesively bonded carbon-epoxy patches on the vertical side faces of the beam at the cracked region to block sliding between the beam arms. An experimental and numerical parametric analysis was performed on the patch length. The numerical analysis used the finite element method (FEM) and cohesive zone models (CZMs), with an inverse modelling technique for the characterization of the adhesive layer. Trapezoidal cohesive laws in each pure mode were used to account for the ductility of the adhesive used. To fully reproduce the tests, horizontal damage propagation within the wood beam was also simulated. A good correlation with the experiments was found. Regarding the effectiveness of the repair, for the conditions selected for this work, a full strength recovery was achieved for the bigger value of patch length tested.

The possible interactions between Delta9-tetrahydrocannabinol (THC) and nicotine remain unclear in spite of the current association of cannabis and tobacco in humans. The aim of the present study was to explore the interactions between these two drugs of abuse by evaluating the consequences of THC administration on the somatic manifestations and the aversive motivational state associated to nicotine withdrawal in mice. Acute THC administration significantly decreased the incidence of several nicotine withdrawal signs precipitated by mecamylamine or naloxone, such as wet-dog-shakes, paw tremor and scratches. In both experimental conditions, the global withdrawal score was also significantly attenuated by acute THC administration. THC also reversed conditioned place aversion associated to naloxone precipitated nicotine withdrawal. We have then evaluated whether this effect of THC was due to possible adaptive changes induced by chronic nicotine on CB1 cannabinoid receptors. The stimulation of GTPS-binding proteins by the cannabinoid agonist WIN 55,212-2 and the density of CB1 cannabinoid receptor binding labelled with [3H] CP-55,940 were not modified by chronic nicotine treatment in the different brain structures investigated. Finally, we evaluated the consequences of THC administration on c-Fos expression in several brain structures after chronic nicotine administration and withdrawal. c-Fos was decreased in the caudate putamen and the dentate gyrus after mecamylamine precipitated nicotine withdrawal. However, acute THC administration did not modify c-Fos expression under these experimental conditions. Taken together, these results indicate that THC administration attenuated somatic signs of nicotine withdrawal and this effect was not associated to compensatory changes on CB1 cannabinoid receptors during chronic nicotine administration. In addition, THC also ameliorated the aversive motivational consequences of nicotine withdrawal.

In this study, the behavioral and electroencephalographic (EEG) analysis of seizures induced by the intrahippocampal injection in rats of granulitoxin, a neurotoxic peptide from the sea anemone Bunodosoma granulifera, was determined. The first alterations occurred during microinjection of granulitoxin (8 µg) into the dorsal hippocampus and consisted of seizure activity that began in the hippocampus and spread rapidly to the occipital cortex. This activity lasted 20-30 s, and during this period the rats presented immobility. During the first 40-50 min after its administration, three to four other similar short EEG seizure periods occurred and the rats presented the following behavioral alterations: akinesia, facial automatisms, head tremor, salivation, rearing, jumping, barrel-rolling, wet dog shakes and forelimb clonic movements. Within 40-50 min, the status epilepticus was established and lasted 8-12 h. These results are similar to those observed in the acute phase of the pilocarpine model of temporal lobe epilepsy and suggest that granulitoxin may be a useful tool not only to study the sodium channels, but also to develop a new experimental model of status epilepticus.

Très peu d’études ont été consacrées à l’œuvre de la romancière française Lydie Salvayre, dont le premier roman a paru en 1990. Il me semble pertinent de me pencher plus sérieusement sur son travail, qui manifeste une véritable exigence d’écriture. Un trait de l’œuvre de Salvayre m’intéresse plus spécifiquement : son rapport à l’héritage. J’envisage ce rapport à la fois dans la thématique, dans l’écriture et dans la forme de deux de ses romans, soient La Puissance des mouches (1995) et La Méthode Mila (2005). Dans chacun de ces deux romans, le narrateur a un rapport trouble à la fois avec son héritage familial et avec son héritage culturel, plus spécialement celui de la philosophie et de la satire classiques. Tiraillé entre le monde du quotidien (trivial) et le monde des idées (sublime), le personnage cherche une issue : il cherche comment vivre. Dans La Puissance des mouches, le narrateur voue une haine à son père et avoue un amour pour Pascal. Dans La Méthode Mila, il est déchiré entre sa mère sénile dont il a la charge et Descartes dont il est obsédé. J’étudierai comment se vit cette déchirure et comment le texte la travaille. La façon dont l’auteure met l’héritage en éclats — elle pastiche, bouscule, malmène, détourne les classiques — n’a rien du respect servile. Cette manière d’hériter, de rompre avec l’évidence d’une continuité avec le passé, pose une question essentielle : comment hériter aujourd’hui, et pourquoi ?

Problème: Ma thèse porte sur l’identité individuelle comme interrogation sur les enjeux personnels et sur ce qui constitue l’identification hybride à l’intérieur des notions concurrentielles en ce qui a trait à l’authenticité. Plus précisément, j’aborde le concept des identifications hybrides en tant que zones intermédiaires pour ce qui est de l’alternance de codes linguistiques et comme négociation des espaces continuels dans leur mouvement entre les cultures et les langues. Une telle négociation engendre des tensions et/ou apporte le lien créatif. Les tensions sont inhérentes à n’importe quelle construction d’identité où les lignes qui définissent des personnes ne sont pas spécifiques à une culture ou à une langue, où des notions de l’identité pure sont contestées et des codes communs de l’appartenance sont compromis. Le lien créatif se produit dans les exemples où l’alternance de code linguistique ou la négociation des espaces produit le mouvement ouvert et fluide entre les codes de concurrence des références et les différences à travers les discriminations raciales, la sexualité, la culture et la langue. Les travaux que j’ai sélectionnés représentent une section transversale de quelques auteurs migrants provenant de la minorité en Amérique du Nord qui alternent les codes linguistiques de cette manière. Les travaux détaillent le temps et l’espace dans leur traitement de l’identité et dans la façon dont ils cernent l’hybridité dans les textes suivants : The Woman Warrior de Maxine Hong Kingston (1975-76), Hunger of Memory de Richard Rodriguez (1982), Comment faire l’amour avec un nègre sans se fatiguer de Dany Laferrière (1985), Borderlands/La Frontera de Gloria Anzalduá (1987), Lost in Translation de Eva Hoffman (1989), Avril ou l’anti-passion de Antonio D’Alfonso (1990) et Chorus of Mushrooms de Hiromi Goto (1994). Enjeux/Questions La notion de l’identification hybride est provocante comme sujet. Elle met en question l’identité pure. C’est un sujet qui a suscité beaucoup de discussions tant en ce qui a trait à la littérature, à la politique, à la société, à la linguistique, aux communications, qu’au sein même des cercles philosophiques. Ce sujet est compliqué parce qu’il secoue la base des espaces fixes et structurés de l’identité dans sa signification culturelle et linguistique. Par exemple, la notion de patrie n’a pas les représentations exclusives du pays d’origine ou du pays d’accueil. De même, les notions de race, d’appartenance ethnique, et d’espaces sexuels sont parfois négativement acceptées si elles proviennent des codes socialement admis et normalisés de l’extérieur. De tels codes de la signification sont souvent définis par l’étiquette d’identification hétérosexuelle et blanche. Dans l’environnement généralisé d’aujourd’hui, plus que jamais, une personne doit négocier qui elle est, au sens de son appartenance à soi, en tant qu’individu et ce, face aux modèles locaux, régionaux, nationaux, voire même globaux de la subjectivité. Nous pouvons interpréter ce mouvement comme une série de couches superposées de la signification. Quand nous rencontrons une personne pour la première fois, nous ne voyons que la couche supérieure. D’ailleurs, son soi intérieur est caché par de nombreuses couches superposées (voir Joseph D. Straubhaar). Toutefois, sous cette couche supérieure, on retrouve beaucoup d’autres couches et tout comme pour un oignon, on doit les enlever une par une pour que l’individualité complète d’une personne soit révélée et comprise. Le noyau d’une personne représente un point de départ crucial pour opposer qui elle était à la façon dont elle se transforme sans cesse. Sa base, ou son noyau, dépend du moment, et comprend, mais ne s’y limite pas, ses origines, son environnement et ses expériences d’enfance, son éducation, sa notion de famille, et ses amitiés. De plus, les notions d’amour-propre et d’amour pour les autres, d’altruisme, sont aussi des points importants. Il y a une relation réciproque entre le soi et l’autre qui établit notre degré d’estime de soi. En raison de la mondialisation, notre façon de comprendre la culture, en fait, comment on consomme et définit la culture, devient rapidement un phénomène de déplacement. À l’intérieur de cette arène de culture généralisée, la façon dont les personnes sont à l’origine chinoises, mexicaines, italiennes, ou autres, et poursuivent leur évolution culturelle, se définit plus aussi facilement qu’avant. Approche Ainsi, ma thèse explore la subjectivité hybride comme position des tensions et/ou des relations créatrices entre les cultures et les langues. Quoique je ne souhaite aucunement simplifier ni le processus, ni les questions de l’auto-identification, il m’apparaît que la subjectivité hybride est aujourd’hui une réalité croissante dans l’arène généralisée de la culture. Ce processus d’échange est particulièrement complexe chez les populations migrantes en conflit avec leur désir de s’intégrer dans les nouveaux espaces adoptés, c’est-à-dire leur pays d’accueil. Ce réel désir d’appartenance peut entrer en conflit avec celui de garder les espaces originels de la culture définie par son pays d’origine. Ainsi, les références antérieures de l’identification d’une personne, les fondements de son individualité, son noyau, peuvent toujours ne pas correspondre à, ou bien fonctionner harmonieusement avec, les références extérieures et les couches d’identification changeantes, celles qu’elle s’approprie du pays d’accueil. Puisque nos politiques, nos religions et nos établissements d’enseignement proviennent des représentations nationales de la culture et de la communauté, le processus d’identification et la création de son individualité extérieure sont formées par le contact avec ces établissements. La façon dont une personne va chercher l’identification entre les espaces personnels et les espaces publics détermine ainsi le degré de conflit et/ou de lien créatif éprouvé entre les modes et les codes des espaces culturels et linguistiques. Par conséquent, l’identification des populations migrantes suggère que la « community and culture will represent both a hybridization of home and host cultures » (Straubhaar 27). Il y a beaucoup d’écrits au sujet de l’hybridité et des questions de l’identité et de la patrie, toutefois cette thèse aborde la valeur créative de l’alternance de codes culturels et linguistiques. Ce que la littérature indiquera Par conséquent, la plate-forme à partir de laquelle j’explore mon sujet de l’hybridité flotte entre l’interprétation postcoloniale de Homi Bhabha concernant le troisième espace hybride; le modèle d’hétéroglossie de Mikhail Bakhtine qui englobent plusieurs de mes exemples; la représentation de Roland Barthes sur l’identité comme espace transgressif qui est un modèle de référence et la contribution de Chantal Zabus sur le palimpseste et l’alternance de codes africains. J’utilise aussi le modèle de Sherry Simon portant sur l’espace urbain hybride de Montréal qui établit un lien important avec la valeur des échanges culturels et linguistiques, et les analyses de Janet Paterson. En effet, la façon dont elle traite la figure de l’Autre dans les modèles littéraires au Québec fournisse un aperçu régional et national de l’identification hybride. Enfin, l’exploration du bilinguisme de Doris Sommer comme espace esthétique et même humoristique d’identification situe l’hybridité dans une espace de rencontre créative. Conséquence Mon approche dans cette thèse ne prétend pas résoudre les problèmes qui peuvent résulter des plates-formes de la subjectivité hybride. Pour cette raison, j’évite d’aborder toute approche politique ou nationaliste de l’identité qui réfute l’identification hybride. De la même façon, je n’amène pas de discussion approfondie sur les questions postcoloniales. Le but de cette thèse est de démontrer à quel point la subjectivité hybride peut être une zone de relation créatrice lorsque l’alternance de codes permet des échanges de communication plus intimes entre les cultures et les langues. C’est un espace qui devient créateur parce qu’il favorise une attitude plus ouverte vis-à-vis les différents champs qui passent par la culture, aussi bien la langue, que la sexualité, la politique ou la religion. Les zones hybrides de l’identification nous permettent de contester les traditions dépassées, les coutumes, les modes de communication et la non-acceptation, toutes choses dépassées qui emprisonnent le désir et empêchent d’explorer et d’adopter des codes en dehors des normes et des modèles de la culture contenus dans le discours blanc, dominant, de l’appartenance culturelle et linguistique mondialisée. Ainsi, il appert que ces zones des relations multi-ethniques exigent plus d’attention des cercles scolaires puisque la population des centres urbains à travers l’Amérique du Nord devient de plus en plus nourrie par d’autres types de populations. Donc, il existe un besoin réel d’établir une communication sincère qui permettrait à la population de bien comprendre les populations adoptées. C’est une invitation à stimuler une relation plus intime de l’un avec l’autre. Toutefois, il est évident qu’une communication efficace à travers les frontières des codes linguistiques, culturels, sexuels, religieux et politiques exige une négociation continuelle. Mais une telle négociation peut stimuler la compréhension plus juste des différences (culturelle ou linguistique) si des institutions académiques offrent des programmes d’études intégrant davantage les littératures migrantes. Ma thèse vise à illustrer (par son choix littéraire) l’identification hybride comme une réalité importante dans les cultures généralisées qui croissent toujours aujourd’hui. Les espaces géographiques nous gardent éloignés les uns des autres, mais notre consommation de produits exotiques, qu’ils soient culturels ou non, et même notre consommation de l’autre, s’est rétrécie sensiblement depuis les deux dernières décennies et les indicateurs suggèrent que ce processus n’est pas une tendance, mais plutôt une nouvelle manière d’éprouver la vie et de connaître les autres. Ainsi les marqueurs qui forment nos frontières externes, aussi bien que ces marqueurs qui nous définissent de l’intérieur, exigent un examen minutieux de ces enjeux inter(trans)culturels, surtout si nous souhaitons nous en tenir avec succès à des langues et des codes culturels présents, tout en favorisant la diversité culturelle et linguistique. The aim of the present thesis was to identify management factors that affect the extent of exploratory behaviour (ground pecking, scratching) as well as quantitative and qualitative as-pects of dust-bathing behaviour in laying hens kept in commercial furnished cages (‘small group housing’) and aviaries. Based on the results, it should be considered which management measures can be recommended for farmers to enhance hen welfare. The feasibility of direct observations of dust-bathing behaviour as well as video observations of exploratory and dust-bathing behaviour was tested in two aviaries. The direct observations were judged to be unfeasible under the conditions encountered. For the analysis of the video recordings, different sampling intervals for instantaneous scan sampling, different extents of observation time, and intra- and inter-observer reliabilities were compared and the most ap-propriate observation scheme selected. Applying the selected scheme (observing the first 16 minutes of every hour distributed over two consecutive light days with a sample interval of two minutes), within the range of environmental conditions found in 22 aviaries, pecking, scratching and dust-bathing behaviour was performed on average 25, 2 and 7 % of the obser-vation time. Hen numbers in the litter were positively associated with stocking density and group size. More scratching was performed with increasing litter height as well as in humid litter. If no litter had been provided, thus substrate consisted of dust and faeces, a reduced proportion of dust-bathing was found. The same method was then used in 16 furnished cage systems. On average 12 % of the total hen number were found on the scratching mats. The hens spent 8 % of the observed time pecking at the mat, 4 % dust-bathing and 0.4 % scratch-ing. Higher proportions were found on the mats and more dust-bathing behaviour occurred, if substrate was provided. Also with increasing light intensity and stocking density more hens were observed on the mats. More pecking and scratching occurred in conditions of higher stocking density, probably due to social facilitation, and of increased mat area per hen. With increasing mat numbers per cage less pecking was observed. Wider mats led to increased dust-bathing behaviour. Finally, 129 dust-baths recorded in 17 aviaries were analysed in detail. On average they lasted 17 minutes, with the tossing phase taking 69 % of this time, including on average 2 vertical wing shakes and 3 scratches with one leg per minute tossing phase. Dust-bath duration de-creased with increasing litter height. Litter type influenced all recorded parameters: dust-bath duration was highest on straw and lowest on fine material and fine material mixed with straw, where on both also the proportion of the tossing phase was lowest. The number of vertical wing shakes during the dust-bath was highest on straw and lowest on fine material mixed with straw as against the frequency of vertical wing shaking that was lowest on straw and highest on fine material. If dust-bathing hens were disturbed twice or more, dust-bathing duration decreased. With increasing light intensity a decreased proportion of the tossing phase as well as a reduced number of vertical wing shakes were recorded. Possibly the light stimulated the hens to dust-bath more often with less tossing behaviour per performance. The observed variation of the dust-bathing parameters could reflect successful adaptation or frustration of the hens. The litter and light conditions on the investigated farms were predominantly restrictive in terms of stimulation of exploration and dust-bathing behaviour. Thus, it was only possible to analyse possible associations between these factors and exploration and dust-bathing behav-iour within the range found. Based on the results the following management recommendations can be given: To allow hens in furnished cages more normal activity, substrate availability should be improved and mat space and light intensity increased. With regard to aviaries as well litter availability should be improved. Litter height should not be too low. Disturbances during dust-bathing should be prevented, but no influencing factors could be identified.

En el presente documento se mostraran todos los análisis y estudios realizados para determinar la factibilidad de un negocio de expendio minorista de bebidas y alimentos denominada “Bar frutal” ó ”Juice Bar , específicamente para ofrecer bebidas frescas y nutritivas de origen frutal, a las cuales se le da la opción de adicionarles suplementos vitamínicos benéficos para la salud y ser acompañadas de alimentos saludables y nutritivos. El establecimiento pretende promover un estilo de vida a través de una alimentación sana y balanceada. El proyecto incluiría líneas de alimentos que funcionaran como acompañamiento a los batidos, que por su preparación y sus características cumplirán estrictamente con el concepto de saludable y nutritivo. El bar frutal ofrece la opción de poder refrescarse y al mismo tiempo nutrirse, con la opción de adquirir suplementos nutricionales como vitaminas, minerales y proteínas. Los principales productos que se van a ofrecer serían granizados, batidos y bebidas no alcohólicas preparadas a base de mezclas de frutas naturales, con posibilidad de adicionar ingredientes naturales como soya, yogurt, leche, miel y complementos nutricionales como hierro, vitaminas y proteínas entre otros complementos vitamínicos. La compañía va a seguir un modelo de negocio de procesamiento de bebidas y alimentos de forma inmediata, cuya distribución será selectiva y directa. Kynurenine (KYN) is the first stable metabolite of the kynurenine pathway, the major route of tryptophan. (TRP) metabolIsm. In the liver, cortisol-inducible tIyptophan-2,3-dioxygenase (TDO) is the first enzyme and rate limiting step. In extrahepatic tissues, it is superceded by indoleamine-2,3-dioxygenase (IDO), an enzyme with a wider substrate specificity. Earlier work in this research group has found substantial elevations in plasma KYN in fasting Tourette's Syndrome (TS) patients with normal TRP and neopterin. The aim of our initial pilot study was to confirm this increase in KYN in fasting human TS patients compared with normal controls, and to see how changes in diet :ay influence certain kynurenine pathway variables. However, we failed to detect a change in plasma KYN, TRP, kynurenic acid (KYNA), neopterin or cortisol between the fasting TS and control groups. Moreover, none of the variables was affected by dietary status, and thus candidates selected for the larger cross-sectional study were permitted to eat and drink freely on the day that blood samples were submitted, but were requested to avoid products containing caffeine, aspirin or nicotine. In the cross-sectional study, TS patients exhibited significantly higher plasma KYN concentrations than controls, although the magnitude of the change was much smaller than originally found. This may be due to differences in detection procedure and the seasonal fluctuation of some biochemical variables, notably cortisol. The generalised increase in neopterin in the TS subject group, suggests a difference in the activity of cytokine-inducible IDO as a likely source for this elevated KYN. Other kynurenine pathway metabolites, specifIcally TRP, 3-hydroxykynurenine (HKY), 3-hydroxyanthranilic acid (HAA) and KYNA were unchanged. In view of recent speculation of the potential therapeutic effects of nicotine in TS, the lower KYN concentrations observed in TS smokers, compared with non-smoking TS patients, was another interesting finding. Tic-like movements, such as head-shakes (HS), which occur in rodents both spontaneously and following diverse drug treatments, closely resemble tic behaviours in humans. The animal tic model was used to examine what effects nicotine may have on shaking behaviours and on selected TRP metabolites. Acute systemic administration of nicotine to mice, produced a dose-dependent reduction in HS frequency (induced by the 5-HT2A/2C agonist DOl), which appeared to be mediated via central nicotinic cholinergic receptors, since mecamylamine pretreatment abolished this effect. Conversely, twice daily subcutaneous injections of nicotine for 7 days, led to an increase in spontaneous and DOI-induced HS. Chronic nicotine also caused a significant elevation m plasma and whole brain KYN concentrations, but plasma TRP, HKY, HAA and KYNA were unaltered. In addition, no change in brain 5-HT or 5-HIAA concentrations or 5-HT turnover, was found. Despite contrasting results from human and animal studIes, a role for nicotine in the mediation of tic-like movements is indicated. The relevance of the kynurenine pathway to TS and the potential role played by nicotine in modifying tic-like behaviours is discussed.

Kynurenine (KYN) is the first stable metabolite of the kynurenine pathway, which accounts for over 95% of tryptophan metabolism. Two previous studies by this research group reported elevated plasma KYN in Tourette syndrome (TS) patients when compared with age and sex matched controls and another study showed that KYN potentiated 5-HT2A-mediated head-shakes (HS) in rodents. These movements have been suggested to model tics in TS. This raised the questions how KYN acts in eliciting this response and whether it is an action of its own or of a further metabolite along the kynurenine pathway. In the liver, where most of the kynurenine pathway metabolism takes place under physiological conditions, the first and the rate limiting enzyme is tryptophan-dioxygenase (TDO) which can be induced by cortisol. In extrahepatic tissues the same step of the pathway is catalyzed by indoleamine-dioxygenase (IDO), which is induced by cytokines, predominantly interferon-y (INF-y). Plasma neopterin, which shows parallel increase with KYN following immune stimulation, was also found elevated in one of these studies positively correlating with KYN. In the present work animal studies suggested that KYN potentiates and quinolinic acid (QUINA) dose dependently inhibits the 5-HT2A-mediated HS response in mice. The potentiating effect seen with KYN was suggested to be an effect of KYN itself. Radioligand binding and phosphoinositide (PI) hydrolysis studies were done to explore the mechanisms by which kynurenine pathway metabolites could alter a 5-HT2A-receptor mediated response. None of the kynurenine pathway metabolites tested showed direct binding to 5-HT2A-receptors. PI hydrolysis studies with KYN and QUINA showed that KYN did not have any effect while QUINA inhibited 5-HT2A-mediated PI hydrolysis. Plasma cortisol determination in TS patients with elevated plasma KYN did not show elevated plasma cortisol levels, suggesting that the increase of plasma KYN in these TS patients is unlikely to be due to an increased TDO activity induced by increased cortisol. Attention deficit hyperactivity disorder (ADHD) is commonly associated with TS. Salivary cortisol detected in a group of children primarily affected with ADHD showed significantly lower salivary cortisol levels when compared with age and sex matched controls. Plasma tryptophan, KYN, neopterin, INF-y and KYN/tryptophan ratio and night-time urinary 6-sulphatoxymelatonin (aMT6s) excretion measured in a group of TS patients did not show any difference in their levels when compared with age and sex matched controls, but TS patients failed to show the expected positive correlation seen between plasma INF-y, neopterin and KYN and the negative correlation seen between plasma KYN and night-time urinary aMT6s excretion seen in healthy controls. The relevance of the kynurenine pathway, melatonin secretion and cortisol to Tourette Syndrome and associated conditions and the mechanism by which KYN and QUINA alter the 5-HT2A-receptor mediated HS response are discussed.

Tic-like movements in rodents bear close similarities to those observed in humans both pharmacologically and morphologically. Pharmacologically, tics are modulated by serotonergic and dopaminergic systems and abnormalities of these systems have been reported in Tourette's Syndrome (TS). Therefore, serotonergic and dopaminergic modulation of tics induced by a thyrotrophin-releasing hormone (TRH) analogue were studied as possible models for TS. The TRH analogue MK771 induced a variety of tic like movements in mice; blinking fore-paw-licking and fore-paw-tremor were quantified and serotonergic and dopaminergic modulation was investigated. The selective dopamine D1 receptor antagonists SCH23390 and SCH39166 and dopamine D2 antagonists raclopride and sulpiride had no effect on MK771 induced blinking. The D1 antagonists attenuated fore-paw-tremor and -licking while the D2 antagonists were generally without effect on these behaviours. Ketanserin (5-HT2A/ alpha-1 antagonist) and ritanserin (5-HT2A/2C antagonist) were able to attenuate MK771-induced blinking and ketanserin, mianserin (5-HT2A/2C antagonist) and prazosin (alpha-1 adrenoceptor antagonist) were able to attenuate MK771-induced fore-paw-tremor and -licking. The 5-HT2C/2B antagonist SB200646A was without effect on blinking and fore-paw-licking but dose-dependently potentiated fore-paw-tremor. The 5-HT1A agonists 8-OH DPAT and buspirone attenuated blinking at the lower doses tested but were ineffective at the higher doses; the converse was found for fore-paw-licking and -tremor behaviours.The effects of these ligands appeared to be at a postsynaptic 5-HTlA site since para-chlorophenylalanine was without effect on the manipulation of these behaviours. (S)-W A Y100135 was without effect on MK771-induced behaviours, spontaneous and DOl-induced head shakes. Because kynurenine potentiates head shakes and plasma concentrations are raised in TS patients the effects of kynurenine on the 5-HT2A/2C agonist DOl mediated head shake were established. Kynurenine potentiated the DOl head shake. Attempts were made to correlate serotonergic unit activity with tic like behaviour in cats but this proved unsuccessful. However, the pharmacological understanding of 5-HTlA receptor function has been hampered because of the lack of selective antagonists for this site. For this reason the effects of the novel 5-HTlA antagonists (S)-WA Y- 100135 and WAY -100635 were tested on 5-HT single-unit activity recorded from the dorsal-raphe-nucleus in the behaving cat. Both drugs antagonised the suppression of unit activity caused by 8-0H DPAT. (S)-WA Y-100135 reduced unit activity whereas WAY-100635 increased it. This suggests that WAY-100635 is acting as an antagonist at the 5-HTlA somatodendritic autoreceptor and that (S)W A Y -100135 acts as a partial agonist at this site. Aspects of tic like behaviour and serotonergic control are discussed.
