Depth data from archival tags on northern rock sole (Lepidopsetta polyxystra) were examined to assess whether fish used tidal currents to aid horizontal migration. Two northern rock sole, out of 115 released with archival tags in the eastern Bering Sea, were recovered 314 and 667 days after release. Both fish made periodic excursions away from the bottom during mostly night-time hours, but also during particular phases of the tide cycle. One fish that was captured and released in an area of rotary currents made vertical excursions that were correlated with tidal current direction. To test the hypothesis that the fish made vertical excursions to use tidal currents to aid migration, a hypothetical migratory path was calculated using a tide model to predict the current direction and speed during periods when the fish was off the bottom. This migration included limited movements from July through December, followed by a 200-km southern migration from January through February, then a return northward in March and April. The successful application of tidal current information to predict a horizontal migratory path not only provides evidence of selective tidal stream transport but indicates that vertical excursions were conducted primarily to assist horizontal migration.

For meroplanktonic larvae that must settle in coastal areas, their successful return to the shore is determined largely by physical transport processes; however, many organisms perform vertical movements to aid successful recruitment. In this study, daytime tidal vertical migration of megalopae of the velvet swimming crab Necora puber was investigated across two different exposures in the shallow waters of Plymouth Sound. Crabs were collected using a plankton net at the surface and near the bottom during flood and ebb tides. Distribution of the pelagic postlarvae was patchy and the abundance varied spatially in tens and thousands of metres. In temporal scales, the annual pattern was dominated by low occurrence of megalopae, punctuated by episodic peaks of high abundance. Most megalopae were collected at the surface irrespective of the tidal phase. The effect of wave exposure on the vertical migration of megalopae was not clear, although there was a general higher abundance of megalopae on exposed shores. Daytime abundance in the water column appears to be regulated by the tidal cycle, as megalopae were more abundant during flood than ebb tides. Although the megalopae do not appear to make large vertical migrations, this behaviour should produce a net shoreward transport. © 2005 Elsevier Ltd. All rights reserved.

O sucesso do recrutamento em espécies com ciclos de vida complexos, i.e. com duas ou mais fases de desenvolvimento, depende do fornecimento de larvas aos locais apropriados do assentamento (mecanismos de pré-assentamento) e na consequente sobrevivência destas larvas até aos estágios bentónicos iniciais (mecanismos de pós-assentamento). O estudo dos principais processos físicos que controlam a variabilidade intra-anual do recrutamento do caranguejo Carcinus maenas foi possível com à obtenção em séries diárias da abundância de megalopas no plâncton e do seu assentamento em substratos artificiais de assentamento, e de parâmetros físicos relacionados com a amplitude de maré e o upwelling, durante os meses da época reprodutiva deste caranguejo. Os nossos resultados demonstraram um padrão cíclico de fornecimento de megalopas de C. maenas aos estuários na costa ocidental de Portugal, relacionado com a amplitude de maré e favorecido pelos ventos de sul. O assentamento de megalopas nos substratos artificiais de assentamento mostrou-se desacoplado do fornecimento nos dois estuários. Os dados obtidos sugerem que as megalopas são transportadas para a costa após a acção de ventos de downwelling, e o fornecimento para os estuários ocorre por transporte selectivo por corrente de maré. A utilização de substratos artificiais para medir densidades de juvenis de Carcinus maenas deve ser ponderada, uma vez que se verificou que em habitats estruturalmente menos complexos os juvenis utilizam os colectores como refúgio; em habitats mais complexos, os juvenis preferiram refugiar-se nas ervas marinhas existentes. No entanto, a estimativa de abundâncias diárias medidas nos colectores permitiu a descrição das abundâncias dos vários estádios de desenvolvimento juvenis, bem como as respectivas taxas de crescimento. As capacidades locomotoras de diferentes classes de tamanho de juvenis de C. maenas foram estimadas mediante o cálculo de um índice de locomoção. Os nossos resultados sugerem que os juvenis mais pequenos estão relativamente protegidos em habitats com vegetação densa, provavelmente porque este tipo de habitats inibe os movimentos dos juvenis de maiores dimensões. Foram também encontradas evidências da existência de segregação temporal na actividade locomotora das diferentes classes de juvenis, que funcionará como um mecanismo que permite reduzir o canibalismo e, consequentemente, aumentar a capacidade de suporte dos habitats juvenis. Foi realizado um estudo sobre o canibalismo nos juvenis de C. maenas na Ria de Aveiro, in situ e utilizando densidades reais. Verificou-se que as presas mais pequenas eram as mais vulneráveis, sendo os juvenis de maiores dimensões os predadores mais eficazes. Por outro lado, as taxas de canibalismo foram menores em habitats com elevadas densidades de refúgio (Zostera noltii). A estimativa das taxas diárias de mortalidade devidas ao canibalismo são aproximadamente metade das taxas diárias de mortalidade devidas aos processos de pré-assentamento, indicando que esta população de C. maenas será regulada pelo fornecimento larvar, i.e., será uma população regulada essencialmente por mecanismos préassentamento.

In the “selective” cholesteryl ester (CE) uptake process, surface-associated lipoproteins [high density lipoprotein (HDL) and low density lipoprotein] are trapped in the space formed between closely apposed surface microvilli (microvillar channels) in hormone-stimulated steroidogenic cells. This is the same location where an HDL receptor (SR-BI) is found. In the current study, we sought to understand the relationship between SR-BI and selective CE uptake in a heterologous insect cell system. Sf9 (Spodoptera frugiperda) cells overexpressing recombinant SR-BI were examined for (i) SR-BI protein by Western blot analysis and light or electron immunomicroscopy, and (ii) selective lipoprotein CE uptake by the use of radiolabeled or fluorescent (BODIPY-CE)-labeled HDL. Noninfected or infected control Sf9 cells do not express SR-BI, show microvillar channels, or internalize CEs. An unexpected finding was the induction of a complex channel system in Sf9 cells expressing SR-BI. SR-BI-expressing cells showed many cell surface double-membraned channels, immunogold SR-BI, apolipoprotein (HDL) labeling of the channels, and high levels of selective HDL-CE uptake. Thus, double-membraned channels can be induced by expression of recombinant SR-BI in a heterologous system, and these specialized structures facilitate both the binding of HDL and selective HDL-CE uptake.

Tidal stream turbines could have several direct impacts upon pursuit-diving seabirds foraging within tidal stream environments (mean horizontal current speeds > 2 ms−1), including collisions and displacement. Understanding how foraging seabirds respond to temporally variable but predictable hydrodynamic conditions immediately around devices could identify when interactions between seabirds and devices are most likely to occur; information which would quantify the magnitude of potential impacts, and also facilitate the development of suitable mitigation measures. This study uses shore-based observational surveys and Finite Volume Community Ocean Model outputs to test whether temporally predictable hydrodynamic conditions (horizontal current speeds, water elevation, turbulence) influenced the density of foraging black guillemots Cepphus grylle and European shags Phalacrocorax aristotelis in a tidal stream environment in Orkney, United Kingdom, during the breeding season. These species are particularly vulnerable to interactions with devices due to their tendency to exploit benthic and epi-benthic prey on or near the seabed. The density of both species decreased as a function of horizontal current speeds, whereas the density of black guillemots also decreased as a function of water elevation. These relationships could be linked to higher energetic costs of dives in particularly fast horizontal current speeds (>3 ms−1) and deeper water. Therefore, interactions between these species and moving components seem unlikely at particularly high horizontal current speeds. Combining this information, with that on the rotation rates of moving components at lower horizontal current speeds, could be used to assess collision risk in this site during breeding seasons. It is also likely that moderating any device operation during both lowest water elevation and lowest horizontal current speeds could reduce the risk of collisions for these species in this site during this season. The approaches used in this study could have useful applications within Environmental Impact Assessments, and should be considered when assessing and mitigating negative impacts from specific devices within development sites.

In this thesis, producing ability of electricity by horizontal tidal current turbines and installing possibility of these turbines on bridge's piers in the marine environments has been studied to reduce primary implementation costs and make the plan, economical. To do this and to study its feasibility, the exerted forces from installing horizontal tidal current turbines were compared with the forces applied to the bridge structure during designing process (given in the Standards). Then, the allowable ranges of the overloading which is tolerable by the piers of the bridge were obtained. Accordingly, it is resulted that for installing these turbines, the piers of the existing bridges are required to be strengthened. Because of increasing usage of renewable powers and as a suggestion, the exerted forces from installing turbine for loading coefficients of different Standards are given. Finally as an example, preliminary designing of a horizontal tidal current turbine was carried out for Gesham Channel and the forces exerted from turbine to the bridge's pier were calculated for the future usage in order to create a test site of real dimensions.

The rapid increase in the number of tidal stream turbine arrays will create novel and unprecedented levels of anthropogenic activity within habitats characterized by horizontal current speeds exceeding 2 ms−1. However, the potential impacts on pursuit‐diving seabirds exploiting these tidal stream environments remain largely unknown. Identifying similarities between the fine‐scale physical features (100s of metres) suitable for array installations, and those associated with foraging pursuit‐diving seabirds, could identify which species are most vulnerable to either collisions with moving components, or displacement from these installations. A combination of vessel‐based observational surveys, Finite Volume Community Ocean Model outputs and hydroacoustic seabed surveys provided concurrent measures of foraging distributions and physical characteristics at a fine temporal (15 min) and spatial (500 m) resolution across a tidal stream environment suitable for array installations, during both breeding and non‐breeding seasons. These data sets were then used to test for associations between foraging pursuit‐diving seabirds (Atlantic puffins Fratercula arctica, black guillemots Cepphus grylle, common guillemots Uria aalge, European shags Phalacrocorax aristotelis) and physical features. These species were associated with areas of fast horizontal currents, slow horizontal currents, high turbulence, downward vertical currents and also hard–rough seabeds. The identity and strength of associations differed among species, and also within species between seasons, indicative of interspecific and intraspecific variations in habitat use. However, Atlantic puffins were associated particularly strongly with areas of fast horizontal currents during breeding seasons, and European shags with areas of rough–hard seabeds and downward vertical currents during non‐breeding seasons. Synthesis and applications. Atlantic puffins’ strong association with fast horizontal current speeds indicates that they are particularly likely to interact with installations during breeding seasons. Any post‐installation monitoring and mitigation measures should therefore focus on this species and season. The multi‐species associations with high turbulence and downward vertical currents, which often coincide with fast horizontal current speeds, also highlight useful pre‐installation mitigation measures via the omission of devices from these areas, reducing the overall likelihood of interactions. Environmental impact assessments (EIA) generally involve once‐a‐month surveys across 2‐year periods. However, the approaches used in this study show that more focussed surveys can greatly benefit management strategies aiming to reduce the likelihood of negative impacts by facilitating the development of targeted mitigation measures. It is therefore recommended that these approaches contribute towards EIA within development sites.

The study outlined in Testing Tidal Turbines Part 1 explains the variation in performance between turbines operating in steady and turbulent flow conditions. However, the impact of turbulence on devices is generally not well understood. Furthermore, the turbulence characteristics of high velocity marine currents have not been extensively studied. Therefore, knowledge of their characteristics must be expanded and methodologies to predict the impact of the characteristics on devices developed and improved. This study examines the measurement of tidal currents at a site used for testing of medium scale tidal turbines. The data being discussed was collected with a point velocimeter (ADV). The processing procedures implemented are discussed and the resulting estimated turbulence spectra and turbulence intensities are presented. The results contribute to the improvement of knowledge regarding tidal current characteristics. This will be fundamental to the optimisation of the design and operation of tidal stream devices.

Hippocampal neurons in culture develop morphological polarity in a sequential pattern; axons form before dendrites. Molecular differences, particularly those of membrane proteins, underlie the functional polarity of these domains, yet little is known about the temporal relationship between membrane protein polarization and morphological polarization. We took advantage of viral expression systems to determine when during development the polarization of membrane proteins arises. All markers were unpolarized in neurons before axonogenesis. In neurons with a morphologically distinguishable axon, even on the first day in culture, both axonal and dendritic proteins were polarized. The degree of polarization at these early stages was somewhat less than in mature cells and varied from cell to cell. The cellular mechanism responsible for the polarization of the dendritic marker protein transferrin receptor (TfR) in mature cells centers on directed transport to the dendritic domain. To examine the relationship between cell surface polarization and transport, we assessed the selectivity of transport by live cell imaging. TfR-green fluorescent protein-containing vesicles were already preferentially transported into dendrites at 2 days, the earliest time point we could measure. The selectivity of transport also varied somewhat among cells, and the amount of TfR-green fluorescent protein fluorescence on intracellular structures within the axon correlated with the amount of cell surface expression. This observation implies that selective microtubule-based transport is the primary mechanism that underlies the polarization of TfR on the cell surface. By 5 days in culture, the extent of polarization on the cell surface and the selectivity of transport reached mature levels.

HIGHLIGHTS FOR FY 2008 1. Completed the first of a two-year Gulf sturgeon population study on the Choctawhatchee River, Florida. The sub adult and adult Gulf sturgeon population was estimated at 2,800 fish. 2. Gulf sturgeon eggs were collected at three hard bottom sites in the Apalachicola River, Florida; two sites were previously confirmed spawning areas and one was a newly confirmed spawning area. 3. Documented 55 potential environmental threats to Gulf sturgeon spawning habitat in the Pea River, Florida and Alabama. 4. Assigned the Eglin AFB Road-Stream Crossing Working Group to guide the closure, repair and maintenance of roads and road stream crossings that impact threatened and endangered species. 5. Conducted 81 assessments of fish and stream invertebrates on and in watersheds surrounding Eglin AFB. 6. Provided technical support for the 5-year status review and reclassification proposed rule for the Okaloosa darter. 7. Initiated an intensive population genetic analysis of the Okaloosa darter throughout its range. Tissues from over 200 Okaloosa darters were collected and analyzed. 8. Established a GIS database to serve as a host for data from any sites sampled for mussels in Northeast Gulf of Mexico drainages. 9. Conducted habitat surveys at 115 locations in the Apalachicola River to assess the effects of drought-related mussel mortality and strandings, evaluate habitat conditions, and assess population demography. 10. A land use/aerial imagery threats assessment data analysis was completed for the Chipola River. A total of 266 impoundments/borrow pits and 471 unpaved road crossings were identified among the threats. 11. Okaloosa darters marked with elastomeric dyes were monitored in Mill Creek, Eglin AFB, to determine movement and habitat use following completion of a fish passage project. 3 12. Partners for Fish and Wildlife funded a streambank and riparian restoration project on Econfina Creek consisting of 3,900 feet of streambank fencing to exclude cattle access. One acre of riparian floodplain was planted with native trees. 13. We provided design and on-the-ground assistance for restoring surface hydrology at St. Vincent NWR. The project restored approximately 1.5 miles of tidal stream and 100 acres of wetlands. 14. A study was completed on 11 coastal streams to document large wood debris relationships with fluvial geomorphic characteristics. 15. We developed a Population Viability Analysis model for the fat threeridge mussel to determine current and future risk of extinction. 17. A Gulf Sturgeon Friends Group, “Gulf Sturgeon Preservation Society” was organized in FY 08. 18. Multiple outreach projects were completed to detail aquatic resource conservation needs and opportunities, including National Fishing Week, Earth Day, several festivals and school outreach.

Este estudo teve como objetivo contribuir com as informações ecológicas e paleoecológicas geradas para a Baía de Guanabara com base na distribuição das assembléias de foraminíferos bentônicos. Para tal foram coletadas 30 amostras de sedimento superficial, ao longo de três perfis distribuídos pela baía e um testemunho (BG28) de 6 m de comprimento retirado próximo a Ilha do Governador. Nas amostras superficiais foram identificados 30 gêneros e 52 espécies das quais as espécies mais constantes foram Amonia tepida e Bolivina translucens que apresentaram a maior constância. Espécies de habitat de plataforma foram identificadas em diversas estações indicando uma boa eficiência no transporte das correntes de fundo para dentro da baía. Das estações superficiais analisadas, 10 localizadas ao redor da Ilha do Governador não continham testas de foraminíferos, possivelmente como resultado da acidificação do sedimento causado pelo derrame de óleo ocorrido em 2000. O índice de confinamento associado às análises de agrupamento e ao DCA indicaram a presença de três setores ambientais influenciadas por COT e granulometria. O primeiro setor entre Copacabana-Itatipú e Aeroporto Santos Dumont Ilha de Boa Viagem foi o ambiente marinho, o segundo setor entre o Aeroporto Santos Dumont - Ilha de Boa Viagem e Ilha do Governador Ilha de Paquetá Litoral de São Gonçalo pode se classificado como um ambiente de estuário inferior ou baía com grande influência marinha e o terceiro setor entre a Ilha do Governador Ilha de Paquetá Litoral de São Gonçalo e fundo da baía como o ambiente mais confinado. No testemunho foram feitas 7 datações indicando uma idade de aproximadamente 5180 40 anos BP. As datações também mostraram que nos últimos anos a taxa de sedimentação aumentou muito podendo estar relacionada com o período de colonização européia. Foram encontradas 18 gêneros e 30 espécies de foraminíferos das quais a espécie mais constante foi a Ammonia tepida seguida pela Buliminella elegantissima. O padrão de distribuição dessas espécies ocorreu com a maior abundância de B. elegantissima nas porções mais inferiores do testemunho e uma abundância maior de A. tepida nas porções mais superiores. Os índices de confinamento junto com as análises de agrupamento e com as curvas de isótopos mostraram que houve poucas oscilações no aporte de água marinha naquela região. As análises dos isótopos de C13 e C14 e O16 e O18 não seguiram um padrão inverso comum em outros estudos, possivelmente influenciado pela proximidade da costa. As análises de agrupamentos indicaram que nos últimos 5180 anos BP a baía não sofreu grandes variações ambientais, ou seja, a região oeste da baía mesmo apresentando alterações ao longo dos anos não foi suficiente para modificar as características de confinamento. As análises nos padrões de distribuição das assembléias de foraminíferos demonstraram ser eficientes ferramentas na caracterização ambiental e paleoambiental da Baía de Guanabara.
