This study aimed to evaluate the effect of aqueous extract of neem on germination and fungi incidence on seeds of three cultivars (Serrinha, BR 17 and Maranhão) of cowpea. Neem leaves were dryed, crushed and prepared dilutions of 0.5; 1.0; 2.0, 4.0 g dm -3and control. The fungi incidence was evaluated by the test filter paper and germination according to the Rules for Seeds Testing (Regras para Análise de Sementes). In the three cultivars analyzed, reduction in the incidence of Aspergillus sp and Fusarium sp was observed. In relation to the influence of extracts of neem leaves on seed germination, significant effect of extract in Maranhão cultivar was observed, where all concentrations differed from the control, and propovided a considerable increase in the percentage of normal seedlings. It was concluded that the leaf extract of neem was effective in controlling Aspergillus sp, Fusarium sp , Phoma sp and Macrophomina phaseolina at different concentrations in different cultivars and seed germination was stimulated for the Maranhão cultivar.

Molybdenum is one of the essential micronutrients for soybeans, acting directly on nitrogen metabolism as enzyme cofactor of nitrogenase. Usually, this nutrient is supplied to the plants through seed treatment or foliar application. The aim of this study was to evaluate the molybdenum effects by foliar in the physiological potential of soybean seeds and verify its interference in the enzyme activities involved in nitrogen metabolism. Soybean seeds of BMX Turbo cultivar were used, produced in Erechim, RS, harvest 2013, from plants treated with the following Mo concentrations: 0; 25; 50 and 75 g ha-1, supplied through two commercial products (Biomol and Molybdate) and stored during 0 and 6 months in uncontrolled conditions. The first experiment was conducted in Seedtes Seed Analysis Laboratory in Pato Branco, PR. The used design was completely randomized in a factorial analysis 4 x 2 x 2 with four replications each. The physiological potential of the seeds was evaluated by the germination test, seedling growth, accelerated aging and emergence on the soil. The second experiment was conducted in a greenhouse, where the seeds derived from treatments with different concentrations of Mo: 0; 25; 50 and 75 g ha-1 supplied through two commercial products (Biomol and Molybdate) were grown in vases. The used design was completely randomized in a factorial analysis 4 x 2 with four replications. Evaluations were performed when the plants reached the R1 phenological stage concerning the nodulation, dry matter of root and shoot of the plants and the determination of the activity of the enzymes glutamine synthetase and glutamate synthetase and the content of total soluble proteins. The data were submitted to variance analysis and when significant they were assessed by Tukey’s test for comparison of products and seed storage and with regression study to the concentrations at 5% probability. Analyses were performed using SISVAR statistical software. The soybean seed storage under uncontrolled conditions affected the seed vigour produced with Mo, regardless of the commercial product used during production. The application of Mo through foliar positively influences the production of soya beans which presented increasing responses in the germination and vigour with the application of Mo above 25 g ha-1 . The enrichment of Mo through foliar did not affect the nodulation of plants of the next generation, however, the use of Mo above 25 g ha-1 provided an increase in the activity of enzymes involved in nitrogen metabolism as well as on the total protein content.

Seeds of carrot, groundnut, lettuce, oilseed rape and onion were stored hermetically in laminated aluminium foil packets in four environments (dry or ultra-dry moisture contents combined factorially with temperatures of 20 degrees C or -20 degrees C), replicated at several sites. After ten years' hermetic storage, seed moisture content, equilibrium relative humidity, viability (assessed by ability to germinate normally in standard germination tests) and vigour were determined. After a decade, the change in seed moisture content of samples stored at -20 degrees C was small or nil. Except for groundnut and lettuce (where loss in viability was about 8 and 3%, respectively), no loss in viability was detected after 10 years' hermetic storage at -20 degrees C. In all cases, there was no difference in seed survival between moisture contents at this temperature (P > 0.25). Comparison of seed vigour (root length and rate of germination) also confirmed that drying to moisture contents in equilibrium with 10-12% r.h. had no detrimental effect to longevity when stored at -20 degrees C: the only significant (P < 0.05) differences detected were slightly greater root lengths for ultra-dry storage of four of the six seed lots. Seed moisture content had increased after a decade at 20 degrees C (generally to the level in equilibrium with ambient relative humidity). Hence, sub-zero temperature storage helped maintain the long-term integrity of the laminated aluminium foil packets, as well as that of the seeds within.

Early establishment of endophytes can play a role in pathogen suppression and improve seedling development. One route for establishment of endophytes in seedlings is transmission of bacteria from the parent plant to the seedling via the seed. In wheat seeds, it is not clear whether this transmission route exists, and the identities and location of bacteria within wheat seeds are unknown. We identified bacteria in the wheat (Triticum aestivum) cv. Hereward seed environment using embryo excision to determine the location of the bacterial load. Axenic wheat seedlings obtained with this method were subsequently used to screen a putative endophyte bacterial isolate library for endophytic competency. This absence of bacteria recovered from seeds indicated low bacterial abundance and/or the presence of inhibitors. Diversity of readily culturable bacteria in seeds was low with 8 genera identified, dominated by Erwinia and Paenibacillus. We propose that anatomical restrictions in wheat limit embryo associated vertical transmission, and that bacterial load is carried in the seed coat, crease tissue and endosperm. This finding facilitates the creation of axenic wheat plants to test competency of putative endophytes and also provides a platform for endophyte competition, plant growth, and gene expression studies without an indigenous bacterial background.

The accelerated ageing (AA) test is widely used to evaluate seed vigor in various species including vegetable crops. The objective of this work was to study procedures for conducting AA to evaluate seed vigor of pumpkin and zucchini seed lots. Five seed lots of commercially acceptable quality (germination) standards were selected from both cv. `Menina Brasileira' (zucchini) and `Barbara' Hybrid (pumpkin). The following tests were conducted: standard germination, the first count of germination, and AA with and without saline solution (periods of 48, 72 and 96 hours). For each combination of temperature and ageing period, seeds were placed in a single layer on a screen in a germination plastic box with either 40 mL deionized water or 40g NaCl/100mL of water. Both AA methods identified lots 2 and 5 of `Menina Brasileira' and lot 10 of `Barbara' hybrid as having the lowest physiological quality. The temperature 41 C promoted a more drastic reduction in germination than did 38 C. This observation was more marked after 96 hours. Based on the results obtained, it can be concluded that the combination 41 degrees C/96 hours should be used to evaluate the physiological potential of pumpkin and zucchini seeds; however, additional studies are necessary to evaluate other combinations and confirm this indication.

It is fundamentally important that adequate tests are used to evaluate the physiological quality of produced and commercialized seeds. The objective of this work was to study accelerated ageing and controlled deterioration to evaluate seed vigour in beetroot, seeking to associate these results with seedling emergence in the field. Consequently, five seed lots of Top Tall Early Wonder cultivar were submitted to the tests of germination, seedling emergence in the field, accelerated ageing (using periods of 24, 48 and 72 hours at 42 degrees C) and controlled deterioration (seeds with moisture contents of 22, 24 and 26% at 41 degrees C and 45 degrees C for 12, 24 and 36 hours). Combinations of 72h at 42 degrees C for the accelerated ageing test and 45 degrees C with 24% moisture content for 24h for the controlled deterioration test were sensitive enough to evaluate the physiological potential of beetroot seeds, providing information that was compatible with results of the seedling emergence in the field.

The slow and uneven germination of Syagrus oleracea seeds represents a problem for its propagation, especially for commercial production. This work aimed to study the effects of substrate and mechanical scarification on the germination of S. oleracea seeds. Germination (% G), emergence rate (ER), leaf area (cm(2)), root length (cm), leaf dry mass (g) and root dry mass (g) were also determined. Non-scarified seeds sown in vermiculite germinated faster and showed higher germination (65%), as well as greater root length (16.55 cm). leaf dry mass (5.07 g) and root dry mass (12.41 g), when compared to the scarified seeds.
