In aquaculture, application of fish hybrids has increased. This technique permits improvement of the fish production by providing specimens showing better growth rate when compared to the parental species. Indeed, sterile individuals are highly demanded because quite frequently parental fish mature before they reach the market size, which impairs their growth and decrease their economic value. Throughout the last years, the commercial and scientific interest in salmonids has increased rapidly, among them, the brook trout (Salvelinus fontinalis), Arctic charr (Salvelinus alpinus) are species that can be crossed to produce hybrids that might by cultured in the fish farms. In the present thesis, we have assessed chromosome numbers and evaluate gonadal sex in the brook trout X Arctic charr hybrid progenies. In our populations, the karyotype of the brook trout comprises 84 chromosomes: 16 bi-armed chromosomes (meta-submetacentric) and 68 one-armed chromosomes (telo-acrocentrics) and the chromosome arm number, NF= 100. Arctic charr karyotype shows variation related to the chromosome number (2n= 81-82) and stable chromosome arm number (NF= 100). 2n= 81 chromosomes consisted of 19 bi-armed and 62 one-armed chromosomes, while 2n= 82 karyotype was organized into 18 meta-submetacentric and 64 acrocentrics. The cytogenetic and histological analysis of the brook trout X Arctic charr hybrids (sparctics) was carried out to asses chromosome and chromosome arm number and gonadal sex of the studied specimens. Diploid chromosome number in the hybrids varied from 81 to 84 and individuals with 83 and 84 chromosomes were predominant. Most of the fish had chromosome arm number equal to 100. Robertsonian fusion in the Arctic charr and chromosome behaviour in the hybrid fish cells might lead to the observed variation in chromosome numbers in the hybrids. Among studied fish, 12 were males, 3 were females and 9 had intersex gonads. No correlation between chromosome number and disturbances in the gonadal development was found. This might suggest that intersex gonads might have been developed as a consequence of disturbances in the genetic sex determination process. Genetic sex determination acts properly in the parental species but in the hybrids this may not be as efficient.

Eleven minisatellite DNA locus specific probes, isolated from Atlantic salmon (Salmo salar) and brown trout (Salmo trutta) partial genomic DNA libraries, were tested for cross-hybridization to eleven other salmonid species, i.e. sockeye salmon (Oncorhynchus nerka); coho salmon (O. kisutch), chum salmon (O. keta); pink salmon (O. gorbuscha); chinook salmon (O. tshawytscha); rainbow trout (O. mykiss); brook trout (Salvelinus fontinalis); Arctic charr (S. alpinus); grayling (Thymallus thymallus); huchen (Hucho hucho); pollan (Coregonus autumnalis). Simple single locus profiles for each of these species were revealed by, from two to ten SLPs. These markers are likely to be of great value in addressing several problems in aquaculture of these species.

We hypothesized that the spatial distribution of groundwater inflows through river bottom sediments is a critical factor associated with the selection of coaster brook trout (a life history variant of Salvelinus fontinalis,) spawning sites. An 80-m reach of the Salmon Trout River, in the Huron Mountains of the upper peninsula of Michigan, was selected to test the hypothesis based on long-term documentation of coaster brook trout spawning at this site. Throughout this site, the river is relatively similar along its length with regard to stream channel and substrate features. A monitoring well system consisting of an array of 27 wells was installed to measure subsurface temperatures underneath the riverbed over a 13-month period. The monitoring well locations were separated into areas where spawning has and has not been observed. Over 200,000 total temperature measurements were collected from 5 depths within each of the 27 monitoring wells. Temperatures within the substrate at the spawning area were generally cooler and less variable than river temperatures. Substrate temperatures in the non-spawning area were generally warmer, more variable, and closely tracked temporal variations in river temperatures. Temperature data were inverted to obtain subsurface groundwater velocities using a numerical approximation of the heat transfer equation. Approximately 45,000 estimates of groundwater velocities were obtained. Estimated velocities in the spawning and non-spawning areas confirmed that groundwater velocities in the spawning area were primarily in the upward direction, and were generally greater in magnitude than velocities in the non-spawning area. In the non-spawning area there was a greater occurrence of velocities in the downward direction, and velocity estimates were generally lesser in magnitude than in the spawning area. Both the temperature and velocity results confirm the hypothesis that spawning sites correspond to areas of significant groundwater influx to the river bed.

Monogeneans (flatworms) are among the most host-specific of parasites in general and may be the most host-specific of all fish parasites. Specificity, in terms of a restricted spatial distribution within an environment, is not unique to parasites and is displayed by some fungi, insects, birds, symbionts and pelagic larvae of free-living marine invertebrates. The nature of cues, how habitats are recognised and how interactions between partners are mediated and maintained is of interest across these diverse associations. We review some experiments that demonstrate important factors that contribute to host-specificity at the level of infective stages (larvae of oviparous monogeneans; juveniles of viviparous gyrodactylids) and adult parasites. Recent research on immune responses by fish to monogenean infections is considered. We emphasise the critical importance of host epidermis to the Monogenea. Monogeneans live on host epidermis, they live in its products (e.g. mucus), monopisthocotyleans feed on it, some of its products are attractants and it may be an inhospitable surface because of its immunological activity. We focus attention on fish but reference is made to amphibian hosts. We develop the concept for a potential role in host-speciality by the anterior adhesive areas, either the specialised tegument and/or anterior secretions produced by monogeneans for temporary but firm attachment during locomotion on host epithelial surfaces. Initial contact between the anterior adhesive areas of infective stages and host epidermis may serve two important purposes. (1) Appropriate sense organs or receptors on the parasite interact with a specific chemical or chemicals or with surface structures on host epidermis. (2) A specific but instant recognition or reaction occurs between component(s) of host mucus and the adhesive(s) secreted by monogeneans. The chemical composition of fish skin is known to be species-specific and our preliminary analysis of the chemistry of some monogenean adhesives indicates they are novel proteins that display some differences between parasite families and species. The objective of this article is to review the populations of Arctic charr in the south of Scotland which have become locally extinct, and the reasons for their demise. In the British Isles, the Arctic charr in individual lakes have been isolated from each other for thousands of years and have developed a variety of phenotypic characteristics which are probably genetically based. About 200 populations of Arctic charr have been recorded from different parts of the British Isles: approximately 12 in England, 50 in Ireland, 175 in Scotland and four in Wales. The threats to charr from acidification, afforestation, engineering schemes, angling and fish farming are assessed, and the establishment of new populations is proposed as a method of replacing extinct stocks or providing an additional safeguard for valuable stocks in threatened waters.

The Arctic charr of the British Isles are all non-migratory and are near their most southernmost range. Windermere is one of the few lowland lakes at southerly latitudes to contain a substantial proportion of Arctic charr. The first recorded mention of charr in Windermere was made around 1540 but it was not till the 17th century that different "sons" of charr were recognized, based on differences in their breeding behaviour. In the 1960's, the presence of two distinct populations, autumn spawners and spring spawners were discovered. In the 1980's it was shown that there were at least four races of charr in Windermere, based on genetic characteristics. Recently, the lake has changed due to inputs of phosphorus from treated sewage released into the lake resulting in eutrophication particularly in the south basin. Since the mid-1980's the numbers of charr caught in the south basin have declined.

Arctic char Salvelinus alpinus farmed in different places in Austria and free of the viral diseases viral haemorrhagic septcaemia (VHS), infectious haematopoietic necrosis (IHN) and infectious pancreatic necrosis (IPN) experienced disease and mortality. Diseased fish showed skin ulceration and pathological signs of sepsis. Aeromonas sp. was isolated as pure culture from the kidney of freshly euthanized diseased fish. Three independent isolates from outbreaks that occurred on 2 of the affected farms were analyzed phylogenetically by DNA sequence analysis of the rrs and gyrB genes and phenotypically with biochemical reactions. All 3 isolates were identified as Aeromonas salmonicida subsp. smithia. Analysis of virulence genes in these isolates revealed the presence of a Type III secretion system as well as several related virulence effector genes including aexT, encoding the Aeromonas exotoxin AexT, aopP and aopH. These genes are characteristic for virulent strains of typical and atypical subspecies of A. salmonicida.

Tagging, displacemenat nd recapture, and ultrasonict racking of displaced mature Sunapee trout (Salvelinusa Ipinus) in Floods Pond, Maine, demonstrated that rapid within-season homing occurs in this relict form of Arctic char. Of the trout displaced about 1.8 km from their spawning ground from 1972 to 1975, 9% to 32% were recaptured one to four times within the same spawning season in trap nets set on the spawning ground. Eight of 14 trout tracked ultrasonically in 1975 homed in 2.5 to 10.0 h. Movements of the homing fish were variable; some trout homed paralleling the shoreline, others homed in open water or used a combination of near-shore and open-water movements. Behavior was similar between the sexes and during day and night, although two fish did begin to move just at sundown. Swimming speeds ranged from 15 to 35 cm s- 1 and averaged about 0 .6 body lengths s -1•. Swimming directions were not influenced by wind and wave direction, nor were swimming speeds within individual tracks influenced by cloud cover, wave height, or water depth. Heavy overcast at night m&y have inhibited movement. Sunapee trout are apparently familiar with the entire lake and travel widely within it. Visual features are postulated as orientational cues, though use of such cues is not clearly demonstrated by our experiments.
