This paper describes the Model for Outcome Classification in Health Promotion and Prevention adopted by Health Promotion Switzerland (SMOC, Swiss Model for Outcome Classification) and the process of its development. The context and method of model development, and the aim and objectives of the model are outlined. Preliminary experience with application of the model in evaluation planning and situation analysis is reported. On the basis of an extensive literature search, the model is situated within the wider international context of similar efforts to meet the challenge of developing tools to assess systematically the activities of health promotion and prevention.

The SIB Swiss Institute of Bioinformatics (www.isb-sib.ch) provides world-class bioinformatics databases, software tools, services and training to the international life science community in academia and industry. These solutions allow life scientists to turn the exponentially growing amount of data into knowledge. Here, we provide an overview of SIB's resources and competence areas, with a strong focus on curated databases and SIB's most popular and widely used resources. In particular, SIB's Bioinformatics resource portal ExPASy features over 150 resources, including UniProtKB/Swiss-Prot, ENZYME, PROSITE, neXtProt, STRING, UniCarbKB, SugarBindDB, SwissRegulon, EPD, arrayMap, Bgee, SWISS-MODEL Repository, OMA, OrthoDB and other databases, which are briefly described in this article.

肽聚糖识别(PGRPs)蛋白家族作为一种主要的模式识别受体,在免疫识别中发挥重要的作用。本研究以栉孔扇贝为对象,采用分子生物学手段,原核重组肽聚糖识别蛋白,并对其功能进行深入研究。 用SWISS-MODEL预测蛋白三维结构, 显示CfPGRP-S1与其它PGRPs、Ⅱ型噬菌体酰胺酶有着类似的晶体结构。由于含有酰胺酶活性所必需的保守氨基酸位点H112, H221, C229和T277,CfPGRP-S1很有可能是一个N-乙酰胞壁酸-L-丙氨酸酰胺酶。 在糖结合试验中,CfPGRP-S1蛋白不可逆的结合肽聚糖和几丁质的作用,证实了CfPGRP-S1对肽聚糖和几丁质两种微生物表面保守分子模式的识别功能。在后续的菌结合试验中,CfPGRP-S1蛋白能与一系列的革兰氏阳性菌、革兰氏阴性菌和真菌结合,其中包括溶壁微球菌、枯草芽孢杆菌、鳗弧菌、大肠杆菌和毕赤酵母菌。由此可见,CfPGRP-S1不仅仅识别肽聚糖,还能识别其他一些分子,如几丁质,并对多种类型的微生物起着免疫识别作用。 CfPGRP-S1蛋白对溶壁微球菌、枯草芽孢杆菌、大肠杆菌有直接杀灭作用,最小抑菌浓度分别为2 μg/mL、2 μg/mL、4 μg/mL,对革兰氏阳性菌的抑菌活性大于革兰氏阴性菌,而对真菌只有微弱的抑制作用,这可能是因为菌细胞壁组分和结构上的差异造成的。CfPGRP-S1蛋白对栉孔扇贝病原菌鳗弧菌的生长却没有任何影响,推测该蛋白可能在非致病性菌的免疫防御中起到了重要作用。 CfPGRP-S1蛋白既具有免疫识别功能,又起到免疫效应分子的作用,对多种非致病菌具有直接杀灭作用。本研究结果为深入了解栉孔扇贝固有免疫的机制提供理论基础。

A novel manganese superoxide dismutase (MnSOD) was cloned from bay scallop Argopecten irradians by 3' and 5' rapid amplification of cDNA ends (RACE) PCR. The full-length cDNA of MnSOD was of 1207 bp with a 678 bp open reading frame encoding 226 amino acids. The deduced amino acid sequence contained a putative signal peptide of 26 amino acids. Sequence comparison showed that the MnSOD of A. irradians shared high identity with MnSOD in invertebrates and vertebrates, such as MnSOD from abalone Haliotis discus discus (ABG88843) and frog Xenopus laevis (AAQ63483). Furthermore, the 3D structure of bay scallop MnSOD was predicted by SWISS-MODEL Protein Modelling Server and compared with those of other MnSODs. The overall structure of bay scallop MnSOD was similar to those of zebrafish Danio rerio, fruit fly Drosophila melanogaster, Chinese shrimp Fenneropenaeus chinensis, human Homo sapiens, and had the highest similarity to scallop Mizuhopecten yessoensis and abalone H. discus discus. A quantitative real-time PCR (qRT-PCR) assay was developed to detect the mRNA expression of MnSOD in different tissues and the temporal expression in haemocytes following challenge with the bacterium Vibrio anguillarum. A higher-level of mRNA expression of MnSOD was detected in gill and mantle. The expression of MnSOD reached the highest level at 3 h post-injection with V. anguillarum and then slightly recovered from 6 to 48 h. The results indicated that bay scallop MnSOD was a constitutive and inducible protein and thus could play an important role in the immune responses against V anguillarum infection. An aspartic endopeptidase was purified in our laboratory from Rhipicephalus (Boophilus) microplus eggs [Logullo, C., Vaz, I.S., Sorgine, M.H., Paiva-Silva, G.O., Faria, F.S., Zingali, R.B., De Lima, M.F., Abreu, L., Oliveira, E.F., Alves, E.W, Masuda, H., Gonzales, J.C., Masuda, A., and Oliveira, P.L., 1998. Isolation of an aspartic proteinase precursor from the egg of a hard tick, Rhipicephalus (Boophilus) microplus. Parasitology 116, 525-532]. Boophilus yolk cathepsin (BYC) was tested as component of a protective vaccine against the tick, inducing a significant immune response in cattle [da Silva, VI., Jr., Logullo, C., Sorgine, M., Velloso, F.F., Rosa de Lima, M.F., Gonzales, J.C., Masuda, H., Oliveira, P.L., and Masuda, A., 1998. Immunization of bovines with an aspartic proteinase precursor isolated from Rhipicephalus (Boophilus) microplus eggs. Vet. Immunol. Immunopathol. 66,331-341]. In this work, BYC was cloned and its primary sequence showed high similarity with other aspartic endopeptidases. In spite of this similarity, BYC sequence shows many important differences in relation to other aspartic peptidases, the most important being the lack of the second catalytic Asp residue, considered to be essential for the catalysis of this class of endopeptidases. When we determined BYC cleavage specificity by LC-MS, we found out that it presents a preference for hydrophobic residues in P1 and P1` in accordance to most aspartic endopeptidases. Also, when analyzed by circular dicroism, BYC presented high beta sheet content, also a characteristic of aspartic endopeptidases. On the other hand, although both native and recombinant BYC are catalytically active, they present a very low specific activity, what seems to indicate that this peptidase will digest its natural substrate, vitellin, very slowly. We speculate that such a slow Vn degradative process might constitute an important strategy to preserve egg protein content to the hatching larvae. (c) 2007 Elsevier Inc. All rights reserved.

Insect digestive chymotrypsins are present in a large variety of insect orders but their substrate specificity still remains unclear. Ewer insect chymotrypsins from 3 different insect orders (Dictyoptera, Coleoptera and two Lepidoptera) were isolated using affinity chromatography. Enzymes presented molecular masses in the range of 20 to 31 kDa and pH optima in the range of 7.5 to 10.0. Kinetic characterization. using different, colorimetric and fluorescent substrates indicated that insect chymotrypsins differ from, bovine chymotrypsin in their primary specificity toward small substrates (like N-benzoyl-L-Tyr p-nitroanilide) rather than on their preference for large substrates (exemplified by Succynil-Ala-Ala-Pro-Phe P-nitroanilide). Chloromethyl ketones (TPCK, N-alpha-tosyl-L-Phe chloromethyl ketone and Z-GGF-CK, N-carbobenzoxy-Gly-Gly-phe-CK) inactivated all chymotrypsins legated. Inactivation rates follow apparent first-order kinetics with variable second order rates (TPCK, 42 to 130 M(-1)s(-1); Z-GGF-CK, 150 to 450 M(-1)s(-1) that may be remarkably low for S. frugiperda chymotrypsin (TPCK, 6 M(-1)s(-1); Z-GGF-CK, 6.1 M(-1) s(-1)). Homology modelling and sequence alignment showed that. in lepidopteran chymotrypsins, differences in the amino acid residues in the neighborhood of the catalytic His 57 may affect its pKa, value. This is Proposed as the cause of the decrease in His 57 reactivity toward chloromethyl ketones. Such amino acid replacement in the active site is proposed. to be an adaptation to the presence of dietary ketones. (C) 2009 Wiley Periodicals, Inc.

Trehalase (EC 3.2.1.28) hydrolyzes only alpha, alpha`- trehalose and is present in a variety of organisms, but is most important in insects and fungi. Crystallographic data showed that bacterial trehalase has 0312 and E496 as the catalytical residues and three Arg residues in the active site. Those residues have homologous in all family 37 trehalases including Spodoptera frugiperda trehalase (0322, E520, R169, R227, R287). To test the role of these residues, mutants of trehalase were produced. All mutants were at least four orders of magnitude less active than wild type trehalase and no structural difference between these mutants and wild type enzyme were discernible by circular dichroism. D322A and E520 pH-activity profile lacked the alkaline arm and the acid arm, respectively, suggesting that D322 is the acid and E520 the basic catalyst. Azide increases E520A activity three times, confirming its action as the basic catalyst. Taking into account the decrease in activity after substitution for alanine residue, the three arginine residues are as important as the catalytical ones to trehalase activity. This clarifies the previous misidentification of an Arg residue as the acid catalyst. As far as we know, this is the first report on the functional identification residues important for trehalase activity. (C) 2010 Elsevier Ltd. All rights reserved.

The major beta-1,3-glucanase from Tenebrio molitor (TLam) was purified to homogeneity (yield, 6%; enrichment, 113 fold; specific activity, 4.4 U/mg). TLam has a molecular weight of 50 kDa and a pH optimum of 6. It is an encloglucanase that hydrolyzes beta-1,3-glucans as laminarin and yeast beta-1,3-1,6-glucan, but is inactive toward other polysaccharides (as unbranched beta-1,3-glucans or mixed beta-1,3-1,4-glucan from cereals) or disaccharides. The enzyme is not inhibited by high substrate concentrations and has low processivity (0.6). TLam has two ionizable groups involved in catalysis, and His, Tyr and Arg residues plus a divalent ion at the active site. A Cys residue important for TLam activity is exposed after laminarin binding. The cDNA coding for this enzyme was cloned and sequenced. It belongs to glycoside hydrolase family 16, and is related to other insect glucanases and glucan-binding proteins. Sequence analysis and homology modeling allowed the identification of some residues (E174, E179, H204, Y304, R127 and R181) at the active site of the enzyme, which may be important for TLam activity. TLam efficiently lyses fungal cells, suggesting a role in making available walls and cell contents to digestion and in protecting the midgut from pathogen infections. (C) 2009 Elsevier Ltd. Serines proteinases inhibitors (PIs) are widely distributed in nature and are able to inhibit both in vitro and in vivo enzymatic activites. Seed PIs in than leguminous are classified in seven families, Bowman-Birk and Kunitz type families that most studied representing an important role in the first line of defense toward insects pests. Some Kunitz type inhibitors possess activities serine and cysteine for proteinases named bifunctional inhibitor, as ApTKI the inhibitor isolate from seed of Adenanthera pavonina. The A. pavonina inhibitor presenting the uncommon property and was used for interaction studies between proteinases serine (trypsin) and cysteine (papain). In order to determinate the in vitro interaction of ApTKI against enzymes inhibitor purification was carried cut by using chromatographic techniques and inhibition assays. The 3D model of the bifunctional inhibitor ApTKI was constructed SWISS-MODEL program by homology modeling using soybean trypsin inhibitor (STI, pdb:1ba7), as template which presented 40% of identity to A. pavonina inhibitor. Model quality was evaluated by PROCHECK program. Moreover in silico analyzes of formed complex between the enzymes and ApTKI was evaluated by HEX 4.5 program. In vitro results confirmed the inhibitory assays, where the inhibitor presented the ability to simultaneously inhibit trypsin and papain. The residues encountered in the inhibitor model of folder structural three-dimensional that make contact to enzymes target coud explain the specificity pattern against serine and cysteine proteinases

O vírus Influenza é o responsável pela gripe, uma doença que ocasiona milhões de mortes e hospitalizações todos os anos. Nas infecções severas, especialmente em pessoas com risco para complicações, os antivirais tornam-se os principais meios para o manejo clínico, merecendo especial destaque os inibidores da neuraminidase (INAs). De fato, na pandemia de 2009 a Organização Mundial da Saúde (OMS) recomendou o uso do oseltamivir para o tratamento dos doentes. Porém, devido à evolução genética viral, surgiram cepas com mutações no gene codificador da neuraminidase (NA) responsáveis por substituições aminoacídicas que levam à resistência aos fármacos INAs. Assim, a OMS passou a recomendar a vigilância de resistência genotípica para os vírus Influenza. Este trabalho teve como objetivos verificar a ocorrência de mutações no gene codificador da NA dos vírus Influenza A (H1N1) pandêmico que possam estar relacionadas à resistência aos INAs em cepas circulantes na mesorregião metropolitana de Belém no período de maio de 2009 a maio de 2012 e analisar, através da modelagem de proteínas, as substituições aminoacídicas da NA que possam estar influenciando na conformação protéica. Durante o período de estudo, foram recebidas no Laboratório de Vírus Respiratórios 2619 amostras clínicas de pacientes que apresentavam sinais e sintomas de infecção respiratória aguda com até cinco dias de evolução. Para a detecção do genoma viral foi feita a extração do RNA viral, seguida de RT-PCR em tempo real utilizando marcadores específicos para Influenza A H1N1pdm, resultando em 744 (28,4%) positivas. Parte das amostras positivas foram então inoculadas em células MDCK. Para as amostras isoladas em cultura de células, foi feita uma nova extração do RNA viral seguida de uma RT-PCR e semi-nested (PCR) utilizando iniciadores específicos para o gene NA, e posterior análise em sequenciador automático ABI Prism 3130xl (Applied Biosystems). A modelagem molecular da NA foi realizada através dos softwares SWISS-MODEL, MODELLER 9.10, PROCHECK, VERIFY3D e PYMOL. A análise parcial das sequências da neuraminidase nas amostras sequenciadas mostrou que não houve a circulação de cepas de vírus H1N1pdm com a mutação H275Y, a principal envolvida na resistência ao oseltamivir. Porém, em duas amostras foi identificada a substituição D199N que já foi relatada em vários estudos mostrando uma possível associação com o aumento da resistência ao oseltamivir. As amostras de 2012 apresentaram duas substituições (V241I e N369K) que estão relacionadas com um possível papel na compensação dos efeitos negativos causados pela mutação H275Y. A modelagem molecular mostrou que na mutação D199N houve uma alteração na estrutura da proteína NA próxima ao sítio de ligação ao antiviral. A análise filogenética revelou que as amostras de 2012 formaram um cluster isolado, demonstrando uma variação muito mais temporal do que geográfica. Este representa o primeiro estudo de resistência dos vírus Influenza H1N1pdm na mesorregião metropolitana de Belém, representando um importante instrumento para que os profissionais de saúde adotem estratégias mais eficazes no manejo da doença e no desenvolvimento de novos fármacos anti-influenza.
