Se describe una nueva subespecie de Teline stenopetala (Webb & Berthel.) Webb & Berthel.: T. stenopetala subsp. santoantaoi Marrero-Rodr. subsp. nov., para la isla de Santo Antão, Islas de Cabo Verde. Se comenta las relaciones taxonómicas con las otras subespecies descritas. Igualmente se hace un breve comentario sobre su hábitat, ecología y estado de conservación de sus poblaciones.

Six isolates of an unknown Gram-positive, catalase-negative, chain-forming, coccus-shaped organism isolated from ovine and caprine mastitis were characterized by phenotypic and molecular taxonomic methods. On the basis of cellular morphology and the results of biochemical tests, the organism was tentatively identified as a streptococcal species. Comparative 16S rRNA gene sequencing studies confirmed that the organism is a member of the genus Streptococcus, with Streptococcus equi as its closest phylogenetic relative (98(.)8% similarity). DNA-DNA pairing studies showed that the unidentified organism displayed more than 70% relatedness to the type strains of S. equi subsp. equi and subsp. zooepidemicus. Despite the relatively high DNA-DNA reassociation values, biotyping and ribotyping allowed clear differentiation of the unknown bacterium from the two recognized subspecies of S. equi. On the basis of phenotypic and molecular genetic evidence, it is proposed that the unknown Streptococcus isolates from ovine and caprine mastitis be classified as a novel subspecies, Streptococcus equi subsp. ruminatorum subsp. nov. The type strain is CECT 5772(T) (=CCUG 47520(T) = Mt 167(T)).

Gram-negative, aerobic, motile, rod-shaped bacteria were isolated from the intestines of freshwater fish on two separate occasions. Colonies of both strains, JF3835(T) and JF4413, produced non-diffusible green pigment following 4-5 days incubation on Luria-Bertani agar. The most abundant fatty acids were summed feature 3 (comprising C(16 : 1)ω7c and/or C(15 : 0) iso 2-OH), C(16 : 0) and C(18 : 1)ω7c. The DNA G+C content was 62.9 mol%. Sequence analysis of the 16S rRNA gene indicated 100 % sequence similarity between the two strains. In comparison with recognized species, the new strains exhibited the greatest degree of sequence similarity with members of the Pseudomonas chlororaphis subspecies: P. chlororaphis subsp. chlororaphis (99.84 %), P. chlororaphis subsp. aurantiaca (99.75 %) and P. chlororaphis subsp. aureofaciens (99.40 %). While DNA-DNA relatedness confirmed the placement of strains JF3835(T) and JF4413 as members of the species P. chlororaphis, multilocus sequencing indicated that the strains formed a distinct cluster within it. On the basis of genotypic and phenotypic evidence, strains JF3835(T) and JF4413 represent a novel subspecies of the species P. chlororaphis, for which the name Pseudomonas chlororaphis subsp. piscium subsp. nov. is proposed. The type strain is JF3835(T) (=NCIMB 14478(T)=DSM 21509(T)).

Aeromonas are widely distributed in the aquatic environment, and are considered to be emerging organisms that can produce a series of virulence factors. The present study was carried out in a sanitary sewage stabilization pond treatment system, located in Lins, State of Sao Paulo, Brazil. Most probable number was applied for estimation of the genus Aeromonas. Colony isolation was carried out on blood agar ampicillin and confirmed by biochemical characterization. Aeromonas species were isolated in 72.4% of influent samples, and in 55.2 and 48.3% of effluent from anaerobic and facultative lagoons, respectively. Thirteen Aeromonas species were isolated, representing most of the recognized species of these organisms. Even though it was possible to observe a tendency of decrease, total elimination of these organisms from the studied system was not achieved. Understanding of the pathogenic organism`s dynamics in wastewater treatment systems with a reuse potential is especially important because of the risk it represents.

É comentada a variabilidade no colorido elitral de Eburodacrys havanensis Chevrolat, 1862.

Early Cretaceous planktonic foraminiferal assemblages include rare planispiral and pseudoplanispiral morphotypes with elongate chambers that BouDagher-Fadel et al. assigned to Schackoina or accommodated in the new genus Claviblowiella. New findings of well-preserved planktonic foraminiferal faunas from the Lesches en Diois (SE France) section, the Cismon core (NE Italy), the Calabianca (NW Sicily) section and the Upper Aptian of Deep Sea Drilling Project (DSDP) Site 545 drilled off Morocco, have allowed a better understanding of the morphological features of these rare, unevenly distributed taxa. Our data demonstrate that each small planispiral species with globular chambers has a corresponding "clavate" morphotype which (as the "normal" forms) exhibits a smooth, finely perforate wall. Consequently, the latter have been assigned here to the genus Globigerinelloides and treated as subspecies of the "non-clavate" taxa. The (clavate) subspecies belonging to the genus Globigerinelloides here retained are G. duboisi sigali Longoria, G. maridalensis elongatus subsp. nov., G. blowi lobatus subsp. nov. and G. paragottisi clavatus subsp. nov., while Globigerinelloides minai Obregòn de la Parra is not retained. In addition, a new genus, Pseudoschackoina, type species Planomalina saundersi Bolli (senior synonym of Hastigerinoides cepedai Obregòn de la Parra, has been formalised for individuals possessing elongate, pointed, laterally compressed chambers, bearing tubulospines arranged on a pseudoplanispiral (dysaxial) coiling mode. Stratigraphically, in the sections studied the first taxon to appear is Pseudoschackoina saundersi, in the uppermost part of the Selli Level (=OAE1a), immediately followed, just above the OAE1a, by all the "clavate" globigerinelloidids. Regarding the last occurrences, Pseudoschackoina saundersi and G. maridalensis elongatus disappear in the lower part of the Globigerinelloides algerianus Zone, Globigerinelloides paragottisi clavatus at the top of the same zone, while Globigerinelloides blowi lobatus and G. duboisi sigali range up to the lower part of the Ticinella bejaouaensis Zone.

Introducción: La rápida detección e identificación bacteriana es fundamental para el manejo de los pacientes críticos que presentan una patología infecciosa, esto requiere de métodos rápidos para el inicio de un correcto tratamiento. En Colombia se usan pruebas microbiología convencional. No hay estudios de espectrofotometría de masas en análisis de muestras de pacientes críticos en Colombia. Objetivo general: Describir la experiencia del análisis microbiológico mediante la tecnología MALDI-TOF MS en muestras tomadas en la Fundación Santa Fe de Bogotá. Materiales y Métodos: Entre junio y julio de 2013, se analizaron 147 aislamientos bacterianos de muestras clínicas, las cuales fueron procesadas previamente por medio del sistema VITEK II. Los aislamientos correspondieron a 88 hemocultivos (60%), 28 urocultivos (19%), y otros cultivos 31 (21%). Resultados: Se obtuvieron 147 aislamientos con identificación adecuada a nivel de género y/o especie así: en el 88.4% (130 muestras) a nivel de género y especie, con una concordancia del 100% comparado con el sistema VITEK II. El porcentaje de identificación fue de 66% en el grupo de bacilos gram negativos no fermentadores, 96% en enterobacterias, 100% en gérmenes fastidiosos, 92% en cocos gram positivos, 100% bacilos gram negativos móviles y 100% en levaduras. No se encontró ninguna concordancia en bacilos gram positivos y gérmenes del genero Aggregatibacter. Conclusiones: El MALDI-TOF es una prueba rápida para la identificación microbiológica de género y especie que concuerda con los resultados obtenidos de manera convencional. Faltan estudios para hacer del MALDI-TOF MS la prueba oro en identificación de gérmenes.

As a consequence of the transfer of the type species Conglomeromonas largomobilis subsp. largomobilis to the genus Azospirillum, the name of the genus Conglomeromonas must be changed in accordance with Rule 37a(1) of the International Code of Nomenclature of Bacteria. Consequently, it is proposed that the subspecies Conglomeromonas largomobilis subsp, parooensis be transferred to the genus Skermanella gen, nov. as the type species Skermanella parooensis gen, nov., sp, nov. This taxon belongs to an isolated subline of descent in the Azospirillum branch of the alpha-Proteobacteria. The spelling of the specific epithet of Azospirillum largomobile is corrected to Azospirillum largimobile.

Analysis of the 16S rDNA sequence of Conglomeromonas largomobilis subsp. largomobilis supports a phylogenetic relationship with the species of the genus Azospirillum. This confirms results of previous nucleic acid hybridization studies (FALK, E. C., J. L. JOHNSON, V. D. L. BALDANI, J. DOBEREINER, and N. R. KRIEG. 1986. Int. J. Syst. Bacteriol. 36: 80-85). Conglomeromonas largomobilis subsp. largomobilis was most closely related to the species Azospirillim lipoferum and Azospirillum brasilense but sufficiently distant to warrant separate species status. Conglomeromonas largomobilis subsp. parooensis was more distantly related to the existing species of Azospirillum and represents an isolated subline of descent. On the basis of the phylogenetic evidence a prosposal is made to transfer the subspecies Conglom-eromonas largomobilis subsp. largomobilis to the genus Azospirillum as Azospirillum largomobile comb. nov. and to retain the genus Conglomeromonas by elevating the subspecies C. largomobilis subsp. parooensis to the type species of Conglomeromonas as Conglomeromonas parooensis sp. nov.

The Mycoplasma mycoides cluster consists of six pathogenic mycoplasmas causing disease in ruminants, which share many genotypic and phenotypic traits. The M. mycoides cluster comprises five recognized taxa: Mycoplasma mycoides subsp. mycoides Small Colony (MmmSC), M. mycoides subsp. mycoides Large Colony (MmmLC), M. mycoides subsp. capri (Mmc), Mycoplasma capricolum subsp. capricolum (Mcc) and M. capricolum subsp. capripneumoniae (Mccp). The group of strains known as Mycoplasma sp. bovine group 7 of Leach (MBG7) has remained unassigned, due to conflicting data obtained by different classification methods. In the present paper, all available data, including recent phylogenetic analyses, have been reviewed, resulting in a proposal for an emended taxonomy of this cluster: (i) the MBG7 strains, although related phylogenetically to M. capricolum, hold sufficient characteristic traits to be assigned as a separate species, i.e. Mycoplasma leachii sp. nov. (type strain, PG50(T) = N29(T) = NCTC 10133(T) = DSM 21131(T)); (ii) MmmLC and Mmc, which can only be distinguished by serological methods and are related more distantly to MmmSC, should be combined into a single subspecies, i.e. Mycoplasma mycoides subsp. capri, leaving M. mycoides subsp. mycoides (MmmSC) as the exclusive designation for the agent of contagious bovine pleuropneumonia. A taxonomic description of M. leachii sp. nov. and emended descriptions of M. mycoides subsp. mycoides and M. mycoides subsp. capri are presented. As a result of these emendments, the M. mycoides cluster will hereafter be composed of five taxa comprising three subclusters, which correspond to the M. mycoides subspecies, the M. capricolum subspecies and the novel species M. leachii.

Conditions have been developed for genetic transformation and insertional mutagenesis in Leifsonia xyli subsp. xyli (Lxx), the causal organism of ratoon stunting disease (RSD), one of the most damaging and intractable diseases of sugarcane internationally. Transformation frequencies ranged from 1 to 10 colony forming units (CFU)/mug of plasmid DNA using Clavibacter/Escherichia coli shuttle vectors pCG188, pDM302, and pDM306 and ranged from 50 to 500 CFU/mug using cosmid cloning vectors pLAFR3 and pLAFR5-km. The transformation/transposition frequency was 0 to 70 CFU/mug of DNA, using suicide vectors pUCD623 and pSLTP2021 containing transposable elements Tn4431 and Tn5, respectively. It was necessary to grow Lxx in media containing 0.1% glycine for electroporation and to amplify large plasmids in a dam(-)/dcm(-) E. coli strain and purify the DNA by anion exchange. To keep selection pressure at an optimum, the transformants were grown on nitrocellulose filters (0.2-mum pore size) on media containing the appropriate antibiotics. Transposon Tn4431 containing a promoterless lux operon from Vibrio fischeri and a tetracycline-resistance gene was introduced on the suicide vector pUCD623. All but 1% of the putative transposon mutants produce light, indicating transposition into functional Lxx genes. Southern blot analysis of these transformants indicates predominantly single transposon insertions at unique sites. The cosmid cloning vector pLAFR5-km was stably maintained in Lxx. The development of a transformation and transposon mutagenesis system opens the way for molecular analysis of pathogenicity determinants in Lxx.

Después de estudiar comparativamente la morfología, corología, ecología, número cromosomático y aceite esencial de los táxones Thymus loscosii Willk. y Thymus loscosii Willk. subsp. fontqueri Jalas, se concluye, de forma razonada, que el status más adecuado para este último es el de especie, combinándose de nuevo cpmo Thymus fontqueri (Jalas) Molero & Rovira; también se propone su inclusión en la sección Serpyllum (Miller) Bentham. En el estudio del aceite esencial de ambos tomillos por CGL e IR se han identificado 14 de sus componentes, destacando el p-citral, mirceno, cineol y alcanfor como mayoritarios. El cromatograma que presentan es muy parecido, pudiéndose atribuir a un quimótipo común P-citral (y otro compuesto no identificado). Entre los componentes la oscilación es máxima para el mirceno, claramente predominante en el T. fontqueri.

O objetivo deste trabalho foi acompanhar a evolução clínica, o desempenho produtivo e reprodutivo e descrever as lesões de caprinos intoxicados por Ipomoea carnea subsp. fistulosa após a retirada dos locais onde ocorre a planta. Para isso foram utilizados 37 caprinos, divididos em 4 grupos. O Grupo 1 era composto por 14 caprinos adquiridos em uma propriedade onde ocorria a planta e que apresentavam condição corporal ruim e sinais clínicos nervosos da intoxicação, que variavam de discretos a acentuados. O Grupo 2 era composto por 10 cabras adquiridas em uma propriedade onde não ocorria a planta e também apresentavam condição corporal ruim. O Grupo 3 era composto por dois caprinos com sinais clínicos da intoxicação, que foram abatidos na fazendo onde tinham se intoxicado. O Grupo 4 era composto por 11 caprinos que serviram como controle para o estudo das lesões macroscópicas e histológicas. Os animais dos Grupos 1 e 2 foram avaliados por um período de 12 meses em uma propriedade localizada no município de Castanhal, onde não ocorre a planta. Durante esse período os animais recebiam o mesmo manejo. Seis meses após, os animais do Grupo 1 continuavam com condição corporal ruim, pelo áspero, maior susceptibilidade à infestações por parasitas gastrintestinais e permaneciam com sinais nervosos. Nos animais que apresentavam sinais nervosos discretos houve diminuição desses sinais, principalmente do tremor de intenção, que passou a ser menos perceptível. Nesse mesmo período os caprinos do Grupo 2 ganharam, em média, 13 kg. Das 8 cabras do Grupo 1 que permaneceram na propriedade experimental somente 4 emprenharam e pariram, sendo que 3 cabritos morreram logo após o nascimento, enquanto que todas as cabras do Grupo 2 emprenharam e pariram cabritos sadios. Nos encéfalos dos caprinos do Grupo 1, 3 e 4 foram realizados estudos histológico, morfológico e morfométrico. Macroscopicamente dois animais apresentaram atrofia cerebelar. No estudo morfométrico, as principais alterações histológicas observadas nos animais dos Grupos 1 e 3 foram diminuição dos neurônios de Purkinje do cerebelo. Conclui-se que caprinos cronicamente intoxicados por I. carnea que deixam de ingerir a planta apresentam sinais permanentes, mesmo que diminuídos de intensidade, fraco desempenho produtivo e reprodutivo e alta susceptibilidade aos parasitas gastrintestinais. Sugere-se que os produtores ao iniciar um plano de controle da intoxicação eliminem todos os animais que em um prazo de até 15 dias não apresentam regressão total dos sinais. O sinal permanente mais frequente é o tremor de intenção, associado à perda de neurônios de Purkinje, que poderia ser o principal responsável pela desnutrição dos animais e as conseqüentes falhas reprodutivas e maior susceptibilidade às parasitoses gastrintestinais.

Abstract Background Xanthomonads are plant-associated bacteria responsible for diseases on economically important crops. Xanthomonas fuscans subsp. fuscans (Xff) is one of the causal agents of common bacterial blight of bean. In this study, the complete genome sequence of strain Xff 4834-R was determined and compared to other Xanthomonas genome sequences. Results Comparative genomics analyses revealed core characteristics shared between Xff 4834-R and other xanthomonads including chemotaxis elements, two-component systems, TonB-dependent transporters, secretion systems (from T1SS to T6SS) and multiple effectors. For instance a repertoire of 29 Type 3 Effectors (T3Es) with two Transcription Activator-Like Effectors was predicted. Mobile elements were associated with major modifications in the genome structure and gene content in comparison to other Xanthomonas genomes. Notably, a deletion of 33 kbp affects flagellum biosynthesis in Xff 4834-R. The presence of a complete flagellar cluster was assessed in a collection of more than 300 strains representing different species and pathovars of Xanthomonas. Five percent of the tested strains presented a deletion in the flagellar cluster and were non-motile. Moreover, half of the Xff strains isolated from the same epidemic than 4834-R was non-motile and this ratio was conserved in the strains colonizing the next bean seed generations. Conclusions This work describes the first genome of a Xanthomonas strain pathogenic on bean and reports the existence of non-motile xanthomonads belonging to different species and pathovars. Isolation of such Xff variants from a natural epidemic may suggest that flagellar motility is not a key function for in planta fitness.

An international collection of the sugarcane ratoon stunting disease pathogen, Leifsonia xyli subsp. xyli, was analysed to assess genetic diversity. DNA fingerprinting using BOX primers was performed on 105 isolates, comprising 65 Australian isolates and an additional 40 isolates from Indonesia (n = 8), Japan (n = 1), USA (n = 3), Brazil (n = 2), Mali (n = 2), Zimbabwe (n = 13), South Africa (n = 9) and Reunion (n = 2). Sixty-two of these isolates were also screened using ERIC primers. No variation was found among any of the isolates. The intergenic spacer (IGS) region of the ribosomal RNA genes from 54 isolates was screened for sequence variation using single-stranded conformational polymorphism (SSCP), but none was observed. Direct sequencing of the IGS from a subset of nine isolates, representing all of the countries sampled in this study, confirmed the results of the SSCP analysis. Likewise, no sequence variation was found in the 16S ribosomal RNA genes of the same subset. Four Colombian isolates from sugarcane, morphologically similar to L. xyli subsp. xyli, were putatively shown to be an undescribed Agrococcus species of unknown pathogenicity. The lack of genetic variation among L. xyli subsp. xyli isolates, independent of time of sampling, cultivar of isolation, or country of origin, suggests the worldwide spread of a single pathogenic clone, and further suggests that sugarcane cultivars resistant to ratoon stunting disease in one area should retain this property in other regions.
