Cyclic peptide architectures can be easily synthesized from cysteine-containing peptides with appending maleimides, free or protected, through an intramolecular Michael-type reaction. After peptide assembly, the peptide can cyclize either during the trifluoroacetic acid treatment, if the maleimide is not protected, or upon deprotection of the maleimide. The combination of free and protected maleimide moieties and two orthogonally protected cysteines gives access to structurally different bicyclic peptides with isolated or fused cycles.

A short and efficient synthesis of heptadeuterated 2,2,4,4,5,7,7-d7-cholestane (1) from cholesterol (3) is described. The deuterated material will be useful for the analysis of different sources of petroleum in analytical geochemistry laboratories as internal standard for quantification of steranes via gas chromatography-mass spectrometry (GC-MS).

In this paper we report a simple and environmentally friendly synthesis of silver nanoparticles (AgNps) and their activities towards the oxygen reduction reaction (ORR). Ultraviolet spectroscopy (UV-vis) and transmission electron microscopy confirmed the formation of poly(vinyl pyrrolidone)-protected colloidal AgNps through direct reduction of Ag+ by glycerol in alkaline medium at room temperature. For the ORR tests, the AgNps were directly produced onto carbon to yield the Ag/C catalyst. Levich plots revealed the process to occur via 2.7 electrons, suggesting that the carbon support contributes to the ORR. We discuss here possibilities of improving the catalytic properties of the Ag/C for ORR by optimizing the parameters of the synthesis.

The synthesis of selenium- and tellurium-containing nucleosides, derived from uridine is described herein. This article describes an efficient and easy one-pot route for the synthesis of a wide range of selenol esters from acyl chloride with diselenides in the presence of indium metal. Objectifs : Cette thèse porte sur l’association entre les caractéristiques socioenvironnementales des voisinages (milieux locaux) et la prévalence des limitations d’activités (ou handicap) dans la population québécoise. Elle a trois objectifs principaux : (1) clarifier les enjeux conceptuels et méthodologiques relatifs à l’étude des déterminants socioenvironnementaux des limitations d’activités; (2) décrire les contributions respectives de la composition socioéconomique des voisinages et de facteurs contextuels à la variabilité locale de la prévalence des limitations d’activités; (3) évaluer la présence d’interactions entre la santé fonctionnelle des personnes (incapacité) et des caractéristiques des voisinages en lien avec la prévalence des limitations d’activités. Méthodes : Une analyse de la littérature scientifique a été effectuée en lien avec le premier objectif de la thèse. En lien avec le deuxième objectif, des données pour le Québec du recensement canadien de 2001 (échantillon de 20% de la population) ont été utilisées pour estimer l’association entre la prévalence des limitations d’activités et des caractéristiques des voisinages : classification urbain-rural, composition socioéconomique (défavorisation matérielle et sociale) et facteurs contextuels (qualité des habitations, stabilité résidentielle et utilisation des transports actifs et collectifs). En lien avec le troisième objectif, des données pour la population urbaine du Québec issues de l’Enquête sur la santé dans les collectivités canadiennes (2003, 2005 et 2007/2008) ont permis de tester la présence d’interaction entre la santé fonctionnelle des personnes et des caractéristiques des voisinages (défavorisation matérielle et sociale, qualité des habitations, stabilité résidentielle et densité des services). Pour les analyses associées aux deux derniers objectifs, l’analyse des corrélats de la prévalence des limitations d’activités a été effectuée à l’aide de régressions logistiques multiniveaux. Résultats : Différents éléments conceptuels et opérationnels limitent la possibilité de faire une synthèse des analyses épidémiologiques portant sur les influences socioenvironnementales sur les limitations d’activités. Les résultats des analyses empiriques suggèrent que : (1) la variation géographique de la prévalence des limitations d’activités s’explique en grande partie par la composition socioéconomique des voisinages; (2) des facteurs contextuels sont associés à cette variation géographique; (3) les mesures relatives d’inégalités sous-estiment les disparités contextuelles dans la distribution des nombres absolus de personnes ayant une limitation d’activités; et (4) l’association entre la prévalence des limitations d’activités et la défavorisation sociale pourrait varier selon la santé fonctionnelle des personnes. Conclusions : Différentes caractéristiques socioenvironnementales sont potentiellement associées aux variations géographiques des limitations d’activités au Québec. Le développement d’indicateurs socioenvironnementaux favoriserait une connaissance plus précise de l’influence de ces caractéristiques socioenvironnementales sur les limitations d’activités et des mécanismes par lesquels s’exerce cette influence. L’établissement d’un système national de surveillance des aménagements territoriaux est proposé afin de soutenir la recherche et la prise de décision. Des indicateurs locaux d’accessibilité aux transports, aux espaces publics ainsi qu’aux services de proximité devraient être priorisés. Ces aspects de l’aménagement du territoire sont susceptibles de rejoindre plusieurs enjeux de santé publique et ils ont comme autre avantage d’être inclus dans différentes orientations québécoises ciblant le vieillissement en santé et la réduction des limitations d’activités.

In der vorliegenden Arbeit wird ein prochirales, aus natürlichen Resourcen gewonnenes Azulen, das Guajazulen genutzt, um neuartige chirale Cyclopentadienyl-Systeme aufzubauen. Mit Alkalimetallhypersilaniden als starke und sperrige Nukleophile gelingt es hypersilylsubstituierte Gujazulenide zu synthetisieren. Diese wurden mittels Elementaranalyse, NMR-Spektroskopie und Röntgendiffraktometrie charakterisiert. Durch nachfolgende Metathesen mit Übergangsmetallhalogeniden konnten in vielen Fällen die entsprechenden Metallocene erhalten werden. Die Experimente enthüllen eine ausgeprägte Regioselektivität der Addition des sperrigen Hypersilyanions an das Guajazulen, die durch das eingesetzte Lösungsmittel graduell verändert werden kann. In nicht-koordinierenden Lösungsmitteln findet man ausschließlich eine Addition an der 6-Position, die 6-Hypersilyl-2,6-dihydroguajazulenide (6-Hyp-Hgual) (M=Li 1, K 2, Cs 4) in ausgezeichneten Ausbeuten liefert. In polaren Solventien erhält man hingegen Mischungen der 6- und 8-Regioisomeren: 2 bzw. (8-Hyp-Hgual) (3). 2 bleibt aber hierbei das Hauptprodukt. Röntgenbeugungsexperimente zeigen, dass 1 im Kristall als dimerer Sandwich-Komplex, meso-[Li2(6-Hyp-Hgual)2], und die THF-Solvate (thf)4K(6-Hyp-Hgual) (2a) sowie (thf)4K(8-Hyp-Hgual) (3a) jeweils als Halb-Sandwich-Komplexe in einer racemischen Mischung vorliegen. Die Verbindungen 1, 2, 3 and 4 eignen sich sehr gut dazu, in Metathesereaktionen als Precursor für neuartige chirale Metallozen-Komplexe eingesetzt zu werden. Insbesondere das Kaliumderivat 2 besticht durch die einfache und relativ preiswerte Synthese, die erzielten hohen Ausbeuten (>80%) und seine leichte Handhabbarkeit. In THF als Solvent wurden die Metallocene 5:5-M’(6-Hyp-Hgual)2 (M’ = Mn 5, Fe 6, Ni 8) und 5:5-Fe(8-Hyp-Hgual)2 (7) erhalten. Bei Verwendung einiger redox-aktiver Metallhalogenide beobachtet man jedoch die Zersetzung der Metallocene unter Bildung des oxidativen Kopplungsproduktes (3-Hyp-6-Hgual)2 (9) sowie der Ausscheidung von Metall. Die Umsetzung von Halogeniden der Gruppe 4 (TiCl3 and M’’Cl4 (M’’ = Ti, Zr, Hf)) mit 2 liefert in THF ausschließlich die Metallozendichloride M’’(6-Hyp-Hgual)2Cl2 (M’’ = Ti (10), Zr (11), Hf (12)). Die erhaltenen Metallozenderivate fallen als Diastereomeren-Gemische an, die sich durch fraktionierende Kristallisation teilweise oder vollständig in ihre Bestandteile, das jeweilige R,R-Racemat und das R,S-meso-Diastereomer auftrennen lassen. Die Strukturen der rac-Diastereomere konnten durch Beugungsexperimente aufgeklärt werden. Durch eine Metathese von 2 mit Hyp-Cl kann eine zweite Hypersilylgruppe in die 2-Position des Guajazulen-Gerüstes eingeführt werden. Das entstehende 2,6-bis(Hyp)-H2gua (14) kann anschließend mit nBuLi in das extrem luft- und feuchtigkeitsempfindliche Li[2,6-bis(Hyp)-Hgual] (15) überführt werden, dass wie 1 eine dimere Sandwich-Struktur aufweist. Durch Einführung des zweiten Hypersilylrestes werden die chemischen Eigenschaften des Azulenids dramatisch verändert. Während Verbindung 1 sich als guter Precursor für Metallocene erwies, gelang es uns bislang nicht, entsprechende Derivate der Verbindung 15 zu isolieren.

Poly(ethylene glycol) (PEG) is used in a broad range of applications due to its unique combination of properties and is approved use in formulations for body-care products, edibles and medicine. This thesis aims at the synthesis and characterization of novel heterofunctional PEG structures and the establishment of diethyl squarate as a suitable linker for the covalent attachment to proteins. Chapter 1 is an introduction on the properties and applications of PEG as well as the fascinating chemistry of squaric acid derivatives. In Chapter 1.1, the synthesis and properties of PEG are described, and the versatile applications of PEG derivatives in everyday products are emphasized with a focus on PEG-based pharmaceuticals and nonionic surfactants. This chapter is written in German, as it was published in the German Journal Chemie in unserer Zeit. Chapter 1.2 deals with PEGs major drawbacks, its non-biodegradability, which impedes parenteral administration of PEG conjugates with polyethers exceeding the renal excretion limit, although these would improve blood circulation times and passive tumor targeting. This section gives a comprehensive overview of the cleavable groups that have been implemented in the polyether backbone to tackle this issue as well as the synthetic strategies employed to accomplish this task. Chapter 1.3 briefly summarizes the chemical properties of alkyl squarates and the advantages in protein conjugation chemistry that can be taken from its use as a coupling agent. In Chapter 2, the application of diethyl squarate as a coupling agent in the PEGylation of proteins is illustrated. Chapter 2.1 describes the straightforward synthesis and characterization of squaric acid ethyl ester amido PEGs with terminal hydroxyl functions or methoxy groups. The reactivity and selectivity of theses activated PEGs are explored in kinetic studies on the reactions with different lysine and other amino acid derivatives, followed by 1H NMR spectroscopy. Further, the efficient attachment of the novel PEGs to a model protein, i.e., bovine serum albumin (BSA), demonstrates the usefulness of the new linker for the PEGylation with heterofunctional PEGs. In Chapter 2.3 initial studies on the biocompatibility of polyether/BSA conjugates synthesized by the squaric acid mediated PEGylation are presented. No cytotoxic effects on human umbilical vein endothelial cells exposed to various concentrations of the conjugates were observed in a WST-1 assay. A cell adhesion molecule - enzyme immunosorbent assay did not reveal the expression of E-selectin or ICAM-1, cell adhesion molecules involved in inflammation processes. The focus of Chapter 3 lies on the syntheses of novel heterofunctional PEG structures which are suitable candidates for the squaric acid mediated PEGylation and exhibit superior features compared to established PEGs applied in bioconjugation. Chapter 3.1 describes the synthetic route to well-defined, linear heterobifunctional PEGs carrying a single acid-sensitive moiety either at the initiation site or at a tunable position in the polyether backbone. A universal concept for the implementation of acetal moieties into initiators for the anionic ring-opening polymerization (AROP) of epoxides is presented and proven to grant access to the degradable PEG structures aimed at. The hydrolysis of the heterofunctional PEG with the acetal moiety at the initiating site is followed by 1H NMR spectroscopy in deuterium oxide at different pH. In an exploratory study, the same polymer is attached to BSA via the squarate acid coupling and subsequently cleaved from the conjugate under acidic conditions. Furthermore, the concept for the generation of acetal-modified AROP initiators is demonstrated to be suitable for cholesterol, and the respective amphiphilic cholesteryl-PEG is cleaved at lowered pH. In Chapter 3.2, the straightforward synthesis of α-amino ω2-dihydroxyl star-shaped three-arm PEGs is described. To assure a symmetric length of the hydroxyl-terminated PEG arms, a novel AROP initiator is presented, who’s primary and secondary hydroxyl groups are separated by an acetal moiety. Upon polymerization of ethylene oxide for these functionalities and subsequent cleavage of the acid-labile unit no difference in the degree of polymerization is seen for both polyether fragments.

During my PhD we focused on different research projects concerning the synthesis and characterization of new rhodium carbonyl clusters. More specifically, we studied the reactivity between Rh4(CO)12 and different bidentate phosphines, obtaining seven different species: Rh4(CO)10(dppe), Rh4(CO)8(dppe)2, Rh4(CO)10(dppf), {Rh4(CO)10(dpp-hexane)}2, {Rh4(CO)10(t-dppe)}2, Rh2(CO)2(dppf)2 and Rh4(CO)9(μ2-dppe)(μ1-dppeO). The reactivity of [Rh7(CO)16]3- with [AuCl4]- and Au(Et2S)Cl led to the formation of seven bimetallic clusters, of which four new ones, namely [Rh16Au6(CO)36]6-, [Rh10Au(CO)26]3-, [Rh16Au6(CO)36]4-, [Rh16Au6(CO)36]5-, [Rh22Au3(CO)47]5-, [Rh19Au5(CO)40]4- and [Rh20Au7(CO)45]5-. The reactivity of [Rh16Au6(CO)36]6- and [Rh10Au(CO)26]3- was studied as well. The reactivity of [Rh7(CO)16]3- with AgBF4, AgNO3 and with Pt(Et2S)2Cl2 was investigated, yielding only to the already known [Rh6N(CO)15]-, [PtRh5(CO)15]- and [PtRh4(CO)14]2- compounds. [Rh7(CO)16]3- war reacted with SnCl2·2H2O in acetone obtaining [Rh7Sn4Cl10(CO)14]5-, and [Rh12Sn(CO)23Cl2]4- was reacted with H+ obtaining [Rh18Sn3Cl2(CO)44]4-. Reactivity of [Rh7(CO)16]3- with InCl3 resulted in the isolation of [Rh12In(CO)28]3- and [Rh11In3(CO)25Cl2]3-, already known in our research lab, and the new [HRh11In(CO)26]3-. Moreover, a more straightforward synthesis for [Rh6InCl3(CO)15]2- was found, and this also led to the isolation of the [Rh6InCl2(DMF)(CO)15]-. The recover or rhodium as valuable carbonyl compound was also studied, and starting from a mixture of by-products it was possible to obtain the starting material [Rh7(CO)16]3-.

A homoenolate generated by tellurium/lithium exchange reaction was employed in a straightforward enantioselective synthesis of (+)-endo-brevicomin in 70% yield and 84.4% e.e.

The synthesis of selenium derivatives of naturally occurring chiral molecules is becoming increasingly important in recent years. In this context, we describe herein an easy, straightforward synthetic route for the preparation of a series of chiral seleno-furanosides, starting from the readily available carbohydrate D-xylose. In addition, selected compounds were screened as inhibitors of the delta-aminolevulinate dehyclratase (delta-ALA-D) enzyme. Diselenide 4 was found to reduce significantly the enzymatic activity, while seleno-furanoside la increased delta-ALA-D activity. (C) 2010 Elsevier Ltd. All rights reserved.
