Trypanosoma cruzi. The aim of this work was to analyze histologically and histometrically the sublingual gland of mice infected with the RAL strain of T cruzi, according to the sex. Swiss mice (Mus musculus) were inoculated with 2 x 10(4) blood trypomastigotes of the RAL strain of T cruzi. In the peak of the parasitemia (12th day) the mice were sacrificed, and the sublingual glands were fixed in ALFAC. HE-stained histological sections were evaluated histometrically. The parasitemia was higher in females. Histopatologically, acini of the infected animals were smaller, with scanty production of secretion, and smaller striated ducts. The nuclei of the demilunes were smaller and showed amastigote nests in the cytoplasm. Karyometrically, nuclei of the acini, demilunes and striated ducts were smaller in the infected mice. Stereologically, it was observed that relative volumes of acini and ducts were smaller and, inversely, relative volumen were greater for the conjunctive tissue in the infected males. The surface densities of acini and ducts were bigger and the diameter and thickness of the wall were smaller in this group. On the other hand, relative volume of acini was smaller and those of the ducts and conjunctive tissue were bigger in the infected females. The diameter and thickness of the wall of acini were smaller, and those of the striated ducts were bigger in this group. The RAL strain of T cruzi caused general atrophy in the sublingual gland, with numerous nests of parasites in the glandular parenchyma.

L’exposition du fœtus à l’éthanol est reconnue comme étant la principale cause de maladies évitables lors du développement. Une forte exposition à l’alcool durant la gestation peut occasionner des dysmorphies cranio-faciales et des retards mentaux, ainsi que des troubles d’apprentissages et du comportement. Le développement du système visuel est également perturbé chez une grande majorité d’enfants qui ont été exposés à l’alcool. Lorsque les doses prises sont élevées, le système visuel peut présenter une panoplie de symptômes comme une augmentation de la tortuosité des vaisseaux rétiniens, de la myopie, de l’hypermétropie, du strabisme et une hypoplasie du nerf optique. Cependant, très peu d’études se sont penchées sur les effets de plus faibles doses sur le développement du système visuel du primate. Le singe est un excellent modèle pour étudier le système visuel car il possède plusieurs similitudes avec l’humain tant au niveau développemental qu’au niveau structurel. De plus, le singe utilisé, le Chlrocebus aethiops sabeus, possède l’avantage que des individus de cette espèce ont une consommation naturelle et volontaire à l’alcool. Une étude (Clarren et al., 1990) a suggéré qu’une faible exposition à l’alcool du fœtus du primate non humain occasionnait une diminution du nombre de cellules ganglionnaires de la rétine (CGRs). Étant donné que le corps genouillé latéral dorsal (CGLd) reçoit la plupart de ses intrants de la rétine, il est raisonnable d’assumer que les couches rétino-récipientes du CGLd devraient être aussi affectées. Nous avons alors émis l’hypothèse que le CGLd devrait également subir une diminution du nombre de neurones. Pour la première fois, nous avons utilisé une méthode stéréologique pour quantifier le nombre de cellules dans les couches parvo- (P) et magnocellulaires (M) du CGLd. Contrairement à notre hypothèse de départ, nous n’avons pas observé de diminution dans le nombre global de neurones dans le CGLd des animaux exposés à l’alcool par rapport à des sujets contrôles, ni une diminution de son volume. Nous avons toutefois observé une diminution de la taille du corps cellulaire seulement dans la population M du CGLd. Ces résultats suggèrent que le système visuel est affecté par une faible exposition à l’alcool durant son développement qui devrait se traduire sur le comportement par des déficits dans les fonctions de la voie M.

ABSTRACT The aim of the present study was to analyze the influence of enriched environment on the distribution of perineuronal nets (PNNs) using a stereogically based unbiased protocol and visual acuity in adult Swiss albino mice that underwent monocular deprivation during the critical period of postnatal development. Eight female Swiss albino mice were monocular deprived were removed and cut at 70 µm thickness in a vibratome and processed for lectin histochemical staining with Wisteria floribunda agglutinin (WFA). Architectonic limits of area 17 were conspicuously defined by WFA histochemical staining, and the optical fractionator stereological method was applied to estimate the total number of PNNs in the supragranular, granular, and infragranular layers. All groups were compared using Student's t-test at a 95% confidence level. Comparative analysis of the average PNN estimations revealed that the EE group had higher PNNs in the supragranular layer (2726.33 ± 405.416, mean ± standard deviation) compared with the SE group (1543.535 ± 260.686; Student's t-test, p = .0495). No differences were found in the other layers. Visual acuity was significantly lower in the SE group (0.55 cycles/degree) than in the EE group (1.06 cycles/degree). Our results suggest that the integrity of the specialized extracellular matrix PNNs of the supragranular layer may be essential for normal visual acuity development.

Poucos estudos tem se dedicado a investigar em detalhe possíveis relações entre o declínio cognitivo associado ao envelhecimento e a plasticidade astroglial no hipocampo. No presente trabalho investigamos possíveis relações entre o desempenho em testes de memória de reconhecimento de objeto e o numero e a distribuição laminar dos astrocitos em CA3 em modelo murino. Para isso empregamos camundongos fêmeas adultas da linhagem C57Bl6 de 6 (n = 7) e 20 meses (n = 5) de idade, mantidos em gaiolas padrão desde o nascimento,comparando seus desempenhos em tarefas hipocampo-dependentes para reconhecimento da forma, do lugar e do momento em que os objetos selecionados lhes foram expostos. Apos os testes comportamentais todos os animais foram per fundidos com fixadores aldeidicos e tiveram seus cérebros removidos e processados para imunomarcação empregando anticorpo seletivo para detecção da proteína acida fibrilar dos astrocitos (GFAP). Para evitar possível viés amostral empregamos o fracionador óptico, um método estereológico que não e afetado pelo processamento histologico. Os resultados nos testes comportamentais isolados e integrados (memória episódica) revelaram que o envelhecimento compromete significativamente (teste T bi-caudal, p<0.05) o reconhecimento da forma, do lugar e do momento em que objetos selecionados são apresentados aos sujeitos. As análises estereologicas das estimativas do numero de astrocítos revelaram que o envelhecimento afetou a distribuicao laminar com aumento na proporção relativa daqueles na camada piramidal de CA3 dorsal e ventral e reducao no lacunoso molecular de CA3 dorsal. O número total de astrocitos em consequência apresentou significativa reorganização na distribuição laminar em função da idade com os animais senis mostrando redução no percentual daqueles localizados na camada oriens. Nenhuma diferenca significativa foi encontrada entre os volumes das camadas de CA3 sugerindo que as mudanças induzidas pelo envelhecimento alteram diretamente a plasticidade astroglial em CA3. Finalmente os estudos de correlação linear entre as estimativas do numero dos astrocitos da camada piramidal e os testes comportamentais demonstraram correlação inversa com os piores desempenhos estando associados a um maior número de astrocitos naquela camada. Evidencias diretas adicionais dessa correlação com os astrocitos alterados em CA3 e possíveis mecanismos moleculares para explicar o declinio cognitivo associado ao envelhecimento permanecem por ser investigados.

The quantification of the structural properties of snow is traditionally based on model-based stereology. Model-based stereology requires assumptions about the shape of the investigated structure. Here, we show how the density, specific surface area, and grain boundary area can be measured using a design-based method, where no assumptions about structural properties are necessary. The stereological results were also compared to X-ray tomography to control the accuracy of the method. The specific surface area calculated with the stereological method was 19.8 ± 12.3% smaller than with X-ray tomography. For the density, the stereological method gave results that were 11.7 ± 12.1% larger than X-ray tomography. The statistical analysis of the estimates confirmed that the stereological method and the sampling used are accurate. This stereological method was successfully tested on artificially produced ice beads but also on several snow types. La mineralogía de procesos se ha convertido en los últimos años en una herramienta indispensable dentro del ámbito minero-metalúrgico debido fundamentalmente a la emergencia de la Geometalurgia. Esta disciplina en auge, a través de la integración de datos geológicos, mineros y metalúrgicos, proporciona la información necesaria para que el circuito de concentración mineral pueda responder de manera rápida y eficaz a la variabilidad mineralógica inherente a la geología del yacimiento. Para la generación del modelo geometalúrgico, la mineralogía de procesos debe aportar datos cuantitativos sobre los rasgos mineralógicos influyentes en el comportamiento de los minerales y para ello se apoya en el uso de sistemas de análisis mineralógico automatizado. Estos sistemas son capaces de proporcionar gran cantidad de datos mineralógicos de manera rápida y precisa. Sin embargo, cuando se trata de la caracterización de la textura, el mineralogista debe recurrir a descripciones cualitativas basadas en la observación, ya que los sistemas actuales no ofrecen información textural automatizada. Esta tesis doctoral surge precisamente para proporcionar de manera sistemática información textural relevante para los procesos de concentración mineral. La tesis tiene como objetivo principal la identificación y caracterización del tipo de intercrecimiento que un determinado mineral presenta en las partículas minerales, e inicialmente se han tenido en cuenta los siete tipos de intercrecimiento considerados como los más relevantes bajo el punto de vista del comportamiento de las partículas minerales durante flotación, lixiviación y molienda. Para alcanzar este objetivo se ha desarrollado una metodología basada en el diseño y cálculo de una serie de índices numéricos, a los que se ha llamado índices mineralúrgicos, que cumplen una doble función: por un lado, cada índice aporta información relevante para caracterizar los principales rasgos mineralógicos que gobiernan el comportamiento de las partículas minerales a lo largo de los procesos de concentración y por otro lado, estos índices sirven como variables discriminantes para identificar el tipo de intercrecimiento mineral mediante la aplicación de Análisis Discriminante. Dentro del conjunto de índices propuestos en este trabajo, se han considerado algunos índices propuestos por otros autores para su aplicación tanto en el ámbito de la mineralogía como en otros ámbitos de la ciencia de materiales. Se trata del Índice de Contigüidad (Gurland, 1958), Índice de Intercrecimiento (Amstutz y Giger, 1972) e Índice de Coordinación (Jeulin, 1981), adaptados en este caso para el análisis de partículas minerales. El diseño de los índices se ha basado en los principios básicos de la Estereología y el análisis digital de imagen, y su cálculo se ha llevado a cabo aplicando el método de interceptos lineales mediante la programación en MATLAB de varias rutinas. Este método estereológico permite recoger una serie de medidas a partir de las que es posible calcular varios parámetros, tanto estereológicos como geométricos, que han servido de base para calcular los índices mineralúrgicos. Para evaluar la capacidad discriminatoria de los índices mineralúrgicos se han seleccionado 200 casos en los que se puede reconocer de manera clara alguno de los siete tipos de intercrecimiento considerados inicialmente en este trabajo. Para cada uno de estos casos se han calculado los índices mineralúrgicos y se ha aplicado Análisis Discriminante, obteniendo un porcentaje de acierto en la clasificación del 95%. Esta cifra indica que los índices propuestos son discriminadores fiables del tipo de intercrecimiento. Una vez probada la capacidad discriminatoria de los índices, la metodología desarrollada ha sido aplicada para caracterizar una muestra de un concentrado de cobre procedente de la mina Kansanshi (Zambia). Esta caracterización se ha llevado a cabo para obtener la distribución de calcopirita según su tipo de intercrecimiento. La utilidad de esta distribución ha sido analizada bajo diferentes puntos de vista y en todos ellos los índices mineralúrgicos aportan información valiosa para caracterizar el comportamiento mineralúrgico de las partículas minerales. Los resultados derivados tanto del Análisis Discriminante como de la caracterización del concentrado de Kansanshi muestran la fiabilidad, utilidad y versatilidad de la metodología desarrollada, por lo que su integración como herramienta rutinaria en los sistemas actuales de análisis mineralógico pondría a disposición del mineralurgista gran cantidad de información textural complementaria a la información ofrecida por las técnicas actuales de caracterización mineralógica. ABSTRACT Process mineralogy has become in the last decades an essential tool in the mining and metallurgical sphere, especially driven by the emergence of Geometallurgy. This emergent discipline provides required information to efficiently tailor the circuit performance to the mineralogical variability inherent to ore deposits. To contribute to the Geometallurgical model, process mineralogy must provide quantitative data about the main mineralogical features implied in the minerallurgical behaviour of minerals. To address this characterisation, process mineralogy relies on automated systems. These systems are capable of providing a large amount of data quickly and accurately. However, when it comes to the characterisation of texture, mineralogists need to turn to qualitative descriptions based on observation, due to the fact that current systems can not offer quantitative textural information in a routine way. Aiming at the automated characterisation of textural information, this doctoral thesis arises to provide textural information relevant for concentration processes in a systematic way. The main objective of the thesis is the automated identification and characterisation of intergrowth types in mineral particles. Initially, the seven intergrowth types most relevant for flotation, leaching and grinding are considered. To achieve this goal, a methodology has been developed based on the computation of a set of numerical indices, which have been called minerallurgical indices. These indices have been designed with two main purposes: on the one hand, each index provides information to characterise the main mineralogical features which determine particle behaviour during concentration processes and, on the other hand, these indices are used as discriminant variables for identifying the intergrowth type by Discriminant Analysis. Along with the indices developed in this work, three indices proposed by other authors belonging to different fields of materials science have been also considered after being adapted to the analysis of mineral particles. These indices are Contiguity Index (Gurland, 1958), Intergrowth Index (Amstutz and Giger, 1972) and Coordination Index (Jeulin, 1981). The design of minerallurgical indices is based on the fundamental principles of Stereology and Digital Image Analysis. Their computation has been carried out using the linear intercepts method, implemented by means of MATLAB programming. This stereological method provides a set of measurements to obtain several parameters, both stereological and geometric. Based on these parameters, minerallurgical indices have been computed. For the assessment of the discriminant capacity of the developed indices, 200 cases have been selected according to their internal structure, so that one of the seven intergrowth types initially considered in this work can be easily recognised in any of their constituents. Minerallurgical indices have been computed for each case and used as discriminant variables. After applying discriminant analysis, 95% of the cases were correctly classified. This result shows that the proposed indices are reliable identifiers of intergrowth type. Once the discriminant power of the indices has been assessed, the developed methodology has been applied to characterise a copper concentrate sample from the Kansanshi copper mine (Zambia). This characterisation has been carried out to quantify the distribution of chalcopyrite with respect to intergrowth types. Different examples of the application of this distribution have been given to test the usefulness of the method. In all of them, the proposed indices provide valuable information to characterise the minerallurgical behaviour of mineral particles. Results derived from both Discriminant Analysis and the characterisation of the Kansanshi concentrate show the reliability, usefulness and versatility of the developed methodology. Therefore, its integration as a routine tool in current systems of automated mineralogical analysis should make available for minerallurgists a great deal of complementary information to treat the ore more efficiently.

The physical disector is a method of choice for estimating unbiased neuron numbers; nevertheless, calibration is needed to evaluate each counting method. The validity of this method can be assessed by comparing the estimated cell number with the true number determined by a direct counting method in serial sections. We reconstructed a 1/5 of rat lumbar dorsal root ganglia taken from two experimental conditions. From each ganglion, images of 200 adjacent semi-thin sections were used to reconstruct a volumetric dataset (stack of voxels). On these stacks the number of sensory neurons was estimated and counted respectively by physical disector and direct counting methods. Also, using the coordinates of nuclei from the direct counting, we simulate, by a Matlab program, disector pairs separated by increasing distances in a ganglion model. The comparison between the results of these approaches clearly demonstrates that the physical disector method provides a valid and reliable estimate of the number of sensory neurons only when the distance between the consecutive disector pairs is 60 microm or smaller. In these conditions the size of error between the results of physical disector and direct counting does not exceed 6%. In contrast when the distance between two pairs is larger than 60 microm (70-200 microm) the size of error increases rapidly to 27%. We conclude that the physical dissector method provides a reliable estimate of the number of rat sensory neurons only when the separating distance between the consecutive dissector pairs is no larger than 60 microm.

The etoposide is an anticancer drug that interacts with topoisomerase II. Thirty-day-old rats received intraperitonially 2mg/kg of etoposide for 30 consecutive days. Their testes were analyzed in the adult phase under light microscopy according to histomorphometric and stereological parameters. Random 3mum-thick-paraplast sections of testis were stained with periodic acid-Schiff reaction and Harris' hematoxylin method. Serum testosterone level and reproductive performance were also investigated. The results showed an accentuated decrease in the frequency of germinal lineage cell types and differentiated spermatogonia were the most affected cell types. Morphometric and stereological testicular parameters exhibited highly, significant reductions in adult etoposide-treated rats. Their reproductive performance diminished but their serum testosterone level was not significantly altered. The mortality frequency of the progenies was 100%.

Stereology is an essential method for quantitative analysis of lung structure. Adequate fixation is a prerequisite for stereological analysis to avoid bias in pulmonary tissue, dimensions and structural details. We present a technique for in situ fixation of large animal lungs for stereological analysis, based on closed loop perfusion fixation.

The small trees of gas-exchanging pulmonary airways which are fed by the most distal purely conducting airways are called acini and represent the functional gas-exchanging units. The three-dimensional architecture of the acini has a strong influence on ventilation and particle deposition. Due to the difficulty to identify individual acini on microscopic lung sections the knowledge about the number of acini and their biological parameters like volume, surface area, and number of alveoli per acinus are limited. We developed a method to extract individual acini from lungs imaged by high-resolution synchrotron radiation based X-ray tomographic microscopy and estimated their volume, surface area and number of alveoli. Rat acini were isolated by semiautomatically closing the airways at the transition from conducting to gas-exchanging airways. We estimated a mean internal acinar volume of 1.148mm(3), a mean acinar surface area of 73.9mm(2), and a mean of 8470 alveoli per acinus. Assuming that the acini are similarly sized throughout different regions of the lung, we calculated that a rat lung contains 5470±833 acini. We conclude that our novel approach is well suited for the fast and reliable characterization of a large number of individual acini in healthy, diseased, or transgenic lungs of different species including humans.

Stochastic models for three-dimensional particles have many applications in applied sciences. Lévy–based particle models are a flexible approach to particle modelling. The structure of the random particles is given by a kernel smoothing of a Lévy basis. The models are easy to simulate but statistical inference procedures have not yet received much attention in the literature. The kernel is not always identifiable and we suggest one approach to remedy this problem. We propose a method to draw inference about the kernel from data often used in local stereology and study the performance of our approach in a simulation study.

The present paper describes a novel, simple and reliable differential pulse voltammetric method for determining amitriptyline (AMT) in pharmaceutical formulations. It has been described for many authors that this antidepressant is electrochemically inactive at carbon electrodes. However, the procedure proposed herein consisted in electrochemically oxidizing AMT at an unmodified carbon nanotube paste electrode in the presence of 0.1 mol L(-1) sulfuric acid used as electrolyte. At such concentration, the acid facilitated the AMT electroxidation through one-electron transfer at 1.33 V vs. Ag/AgCl, as observed by the augmentation of peak current. Concerning optimized conditions (modulation time 5 ms, scan rate 90 mV s(-1), and pulse amplitude 120 mV) a linear calibration curve was constructed in the range of 0.0-30.0 μmol L(-1), with a correlation coefficient of 0.9991 and a limit of detection of 1.61 μmol L(-1). The procedure was successfully validated for intra- and inter-day precision and accuracy. Moreover, its feasibility was assessed through analysis of commercial pharmaceutical formulations and it has been compared to the UV-vis spectrophotometric method used as standard analytical technique recommended by the Brazilian Pharmacopoeia.

The present work compared the local injection of mononuclear cells to the spinal cord lateral funiculus with the alternative approach of local delivery with fibrin sealant after ventral root avulsion (VRA) and reimplantation. For that, female adult Lewis rats were divided into the following groups: avulsion only, reimplantation with fibrin sealant; root repair with fibrin sealant associated with mononuclear cells; and repair with fibrin sealant and injected mononuclear cells. Cell therapy resulted in greater survival of spinal motoneurons up to four weeks post-surgery, especially when mononuclear cells were added to the fibrin glue. Injection of mononuclear cells to the lateral funiculus yield similar results to the reimplantation alone. Additionally, mononuclear cells added to the fibrin glue increased neurotrophic factor gene transcript levels in the spinal cord ventral horn. Regarding the motor recovery, evaluated by the functional peroneal index, as well as the paw print pressure, cell treated rats performed equally well as compared to reimplanted only animals, and significantly better than the avulsion only subjects. The results herein demonstrate that mononuclear cells therapy is neuroprotective by increasing levels of brain derived neurotrophic factor (BDNF) and glial derived neurotrophic factor (GDNF). Moreover, the use of fibrin sealant mononuclear cells delivery approach gave the best and more long lasting results.
