Bacillus subtilis tem a capacidade de utilizar arabinoligossacáridos presentes na parede das células vegetais, através de um consórcios de enzimas envolvidas na hidrólise enzimática dos mesmos. A captação destes carbohidratos por parte do Bacillus subtilis depende de dois transportadores membranares AraE e AraNPQ-MsmX. Após a captação de L-arabinose e arabinoligossacáridos, estes substratos serão metabolizados pelos produtos enzimáticos codificados por genes pertencentes ao operão responsável pela metabolização de arabinose araABDLMNPQabfA. Neste operão, para além dos genes responsáveis pelo transportador AraNPQ, o gene araL determina a síntese de uma enzima com atividade de fosfatase, AraL, que tem putativamente como principal função a destoxificação de intermediários metabólicos fosforilados, em situações particulares. O objectivo deste trabalho consistiu em descobrir quais os determinantes moleculares envolvidos em ambos os processos de reconhecimento molecular, ora o reconhecimento de arabinoligossacáridos por parte da AraN, do ponto de vista dos carbohidratos ora de carbohidratos fosforilados do ponto de vista da AraL. Para elucidar estes processos de reconhecimento molecular proteína-carbohidrato foi utilizada a técnica de RMN, modelação computacional e mutagénese dirigida. No primeiro capitulo são introduzidos conceitos fundamentais para a percepção da ação de ambas as proteínas (AraN e AraL), no organismo Bacillus subtilis. O segundo capitulo, refere-se ao estudo dos mecanismos envolvidos no reconhecimento proteína-carbohidrato através da técnica de STD-RMN para estudar interações do ponto de vista do carbohidrato, bem como abordagens bioinformáticas tais como alinhamentos de sequencia primária e dockings moleculares, para identificar resíduos do ponto de vista da proteína passiveis de se encontrarem envolvidos no reconhecimento molecular, que numa última instância são mutados para se confirmar a sua relevância no processo de reconhecimento molecular do ponto de vista da proteína. Neste capitulo é demonstrado que a AraN é responsável pelo reconhecimento de arabinoligossacáridos e celoligossacáridos. No que diz respeito aos primeiros, a AraN reconhece preferencialmente arabinoligossacáridos com três subunidades, verificando-se a perda de saturação na subunidade não redutora da arabinotetraose. O processo de reconhecimento molecular apresenta uma constante de dissociação com uma ordem de grandeza na ordem dos μM. Do ponto de vista da proteína são identificados resíduos (W253, Y254 e Y54) putativos de se encontrarem também eles envolvidos no reconhecimento molecular do ponto de vista da AraN através de interações CHstacking. Finalmente no terceiro capitulo, é apresentada a optimização da sobrexpressão da AraL, dado que numa primeira fase a sua produção ocorria sob a forma de corpos de inclusão. Para tal são descritas metodologias de solubilização e renaturação da mesmas, com recurso a agentes caotrópicos (ureia e GmdCl) e detergentes (SDS).

NMR is a valuable screening tool for the binding of ligands to proteins providing structural information on both protein and ligands and is thus largely applied to drug-discovery. Among the recent NMR techniques to probe weak binding protein-ligand complexes we have critically evaluated the advantages and disadvantages of STD (Saturation Transfer Difference), WaterLOGSY (Water Ligand Observation with Gradient Spectroscopy), NOE pumping and DOSY-NOESY (Diffusion-Ordered NOESY) using a mixture of BSA (bovine serum albumin) plus salicylic acid, caffeine, citric acid, adipic acid and D-glucose.

This text describes an experiment on fractional precipitation of a polymer together with determination of average degree of polymerization by NMR. Commercial sodium polyphosphate was fractionated by precipitation from aqueous solution by adding increasing amounts of acetone. The polydisperse salt and nine fractions obtained from it were analyzed by 31P Nuclear Magnetic Resonance and the degree of polymerization of the salts and of the fractions were calculated. Long-chain sodium polyphosphate was also synthesized and analyzed. This experiment was tested in a PChem lab course but it can be used also to illustrate topics of inorganic polymers and analytical chemistry.

The [RuCl(bipy)(dppb)(4-pic)]PF6 complex was prepared and fully characterized. The X-ray crystal structure of this complex was determined in order to make an unambiguous distinction between the two possible positions of the 4-methylpyridine ligand (4-pic) in the compound: trans to phosphorus atom or trans to nitrogen atom. The [RuCl(bipy)(dppb)(4-pic)]PF6 complex exhibits an unusual temperature-dependent accidental degeneracy of the 31P chemical shifts in its solution NMR spectrum.

The Steady-State Free Precession (SSFP) sequence has been widely used in low-field and low-resolution imaging NMR experiments to increase the signal-to-noise ratio (s/n) of the signals. Here, we analyzed the Scrambled Steady State - SSS and Unscrambled Steady State - USS sequences to suppress phase anomalies and sidebands of the 13C NMR spectrum acquired in the SSFP regime. The results showed that the application of the USS sequence allowed a uniform distribution of the time interval between pulses (Tp), in the established time range, allowing a greater suppression of phase anomalies and sidebands, when compared with the SSS sequence.

Objective: To design, introduce, and evaluate STD syndrome packets containing recommended drugs for each syndrome, four condoms, a partner treatment card, and a patient information leaflet, with the goal of improving sexually transmitted disease (STD) case management. Methods: Packet design evolved around available packaging technology, informed by pilot testing with nurses working in primary care clinics, doctors in private medical practices, and patients with an STD, in Hlabisa, South Africa. Evaluation 1 year later included analysis of distribution records and interviews with 16 nurses and 61 patients. Results: A cheap packet (2 U, S, cents each, excluding contents) compatible with current legislation was designed and introduced to six public sector clinics and as a short pilot to five private medical practices, Four thousand eighty-five packets were distributed to the clinics, equivalent to approximately 115% of the STDs reported over that period. All 16 nurses reported using the packets, but only 63% did so all the time because of occasional supply problems, All believed the packets improved treatment by saving time (75%), improving supply of condoms and partner cards (44%), and making treatment easier (56%), Patients also responded positively, and most said they would buy a packet (up to $5) at a pharmacy (84%) or store (63%) if available. Conclusions: The STD syndrome packets have the potential to improve STD syndromic management by standardizing therapy and improving the supply of condoms, partner cards, and information leaflets. Packets are popular with practitioners and patients, but consistent supply is essential for maximal impact, There may be scope for social marketing of the packets, which could further increase use.

Objective: To determine whether coinfection with sexually transmitted diseases (STD) increases HIV shedding in genital-tract secretions, and whether STD treatment reduces this shedding. Design: Systematic review and data synthesis of cross-sectional and cohort studies meeting. predefined quality criteria. Main Outcome Measures: Proportion of patients with and without a STD who had detectable HIV in genital secretions, HIV toad in genital secretions, or change following STD treatment. Results: Of 48 identified studies, three cross-sectional and three cohort studies were included. HIV was detected significantly more frequently in participants infected with Neisseria gonorrhoeae (125 of 309 participants, 41%) than in those without N gonorrhoeae infection (311 of 988 participants, 32%; P = 0.004). HIV was not significantly more frequently detected in persons infected with Chlamydia trachomatis (28 of 67 participants, 42%) than in those without C trachomatis infection (375 of 1149 participants, 33%; P = 0.13). Median HIV load reported in only one study was greater in men with urethritis (12.4 x 10(4) versus 1.51 x 10(4) copies/ml; P = 0.04). In the only cohort study in which this could be fully assessed, treatment of women with any STD reduced the proportion of those with detectable HIV from 39% to 29% (P = 0.05), whereas this proportion remained stable among controls (15-17%), A second cohort study reported fully on HIV load; among men with urethritis, viral load fell from 12.4 to 4.12 x 10(4) copies/ml 2 weeks posttreatment, whereas viral load remained stable in those without urethritis. Conclusion: Few high-quality studies were found. HIV is detected moderately more frequently in genital secretions of men and women with a STD, and HIV load is substantially increased among men with urethritis, Successful STD treatment reduces both of these parameters, but not to control levels. More high-quality studies are needed to explore this important relationship further.

A cristalização e formação de depósitos parafínicos é um problema operacional crítico na indústria do petróleo em todo o mundo e provoca grandes perdas econômicas na recuperação do óleo. São muitos os estudos que têm sido realizados para desenvolver modelos termodinâmicos de predição da precipitação de parafinas, sendo que boa parte deles utilizam a ressonância magnética nuclear (RMN) em alto ou baixo campo magnético juntamente com outras técnicas como calorimetria exploratória diferencial (DSC), análise elementar e cromatografia gasosa para caracterizar a fase sólida formada a partir do petróleo. Este trabalho busca um maior entendimento da cristalização de ceras parafínicas por meio de experimentos de espectroscopia de RMN de 1H e 13C conduzidos em temperaturas variáveis, o que pode auxiliar na previsão e remediação dos problemas causados pela sua deposição nas linhas de escoamento da produção. Para isso, o estudo desse fenômeno foi conduzido inicialmente em amostras de parafina comercial e, posteriormente, em uma amostra de petróleo parafínico com variação de temperatura sem extrair a fase sólida, garantindo a não interferência de solventes que podem influenciar no processo de cristalização. A metodologia desenvolvida demonstrou ser útil para determinar a temperatura inicial de aparecimento de cristais (TIAC), sendo obtida uma boa concordância com os resultados de DSC para a parafina comercial e petróleo. O registro de espectros em diferentes temperaturas permitiu também a identificação das variações de intensidade e largura de linha dos sinais associados aos diferentes grupos químicos presentes nos materiais estudados.

Technology plays a double role in Education: it can act as a facilitator in the teaching/learning process and it can be the very subject of that process in Science & Engineering courses. This is especially true when students perform laboratory activities where they interact with equipment and objects under experimentation. In this context, technology can also play a facilitator role if it allows students to perform experiments in a remote fashion, through the Internet, in a so-called weblab or remote laboratory. No doubt, the Internet has been revolutionizing the educational process in many aspects, and it can be stated that remote laboratories are just an angle of that on-going revolution. As any other educational tool or resource, the i) pedagogical approach and the ii) technology used in the development of a remote laboratory can dictate its general success or its ephemeral existence. By pedagogical approach we consider the way remote experiments address the process by which students acquire experimental skills and link experimental results to theoretical concepts. In respect to technology, we discuss different specification and implementation alternatives, to show the case where the adoption of a family of standards would positively contribute to a larger acceptance and utilization of remote laboratories, and also to a wider collaboration in their development.

INTRODUCTION: The study aimed to assess the prevalence of Neisseria gonorrhoeae and Chlamydia trachomatis infections and identify demographic, behavioral and clinical factors correlated withsuch infections in men attending six sexually transmitted disease clinics in Brazil. METHODS: Multicentric, cross-sectional study performed among men attending STD clinics in Brazil. The study included STD clinics in six cities distributed throughout the five geographic regions of Brazil in 2005. Patients provided 20 ml of first catch urine for testing for NG and CT by DNA-PCR. RESULTS: A total of 767 (92.9%) men were included in the study. The mean age was 26.5 (SD 8.3) years-old. Prevalence of Chlamydia infection was 13.1% (95%CI 10.7%-15.5%) and gonorrhea was 18.4% (95%CI 15.7%-21.1%). Coinfection prevalence was 4.4% (95%CI 2.95%-5.85%) in men who sought attendance in STI clinics. Factors identified as associated with C. trachomatis were younger age (15-24) [OR=1.4 (95%CI 1.01-1.91)], present urethral discharge [OR=4.8 (95%CI 1.52-15.05)], genital warts [OR=3.0 (95%CI 1.49-5.92)] and previous history of urethral discharge [OR=2.4 (95%CI 1.11-5.18)]. Variables associated with gonorrhea were younger age (15 to 24) [OR=1.5 (95%CI 1.09-2.05)], presence of urethral discharge [OR=9.9 (95%CI 5.53-17.79)], genital warts [OR=18.3 (95%CI 8.03-41.60)] and ulcer present upon clinical examination [OR=4.9 (95%CI 1.06-22.73)]. CONCLUSIONS: These findings have important implications for education and prevention actions directed toward men at risk of HIV/STD. A venue-based approach to offer routine screening for young men in STD clinics should be stimulated.

Los polímeros son materiales que poseen una variedad muy grande de aplicaciones. Por esta razón, el estudio de sus propiedades físicas y químicas resulta de gran interés. Un polímero es una macromolécula cuyo peso molecular puede llegar a varios millones de umas. Mediante la selección de el o los monómeros y de su secuenciamiento en el proceso de polimerización (microestructura del polímero), se puede lograr que el material tenga propiedades predeterminadas. Es posible, entonces, encontrar o desarrollar polímeros para las más variadas aplicaciones: elásticos, rígidos, resistentes a la temperatura, conductores, aisladores, inertes, etc. (...) La Resonancia Magnética Nuclear (RMN) de alta resolución es una de las técnicas más poderosas para la caracterización de los polímeros, brindando información sobre la microestructura y la dinámica de estas macromoléculas, tanto cuando se encuentran en estado sólido como cuando están disueltas en soluciones líquidas. El entendimiento de la microestructura de un polímero es de interés porque ella está íntimamente relacionada con las propiedades macroscópicas del material. Por otra parte, la microestructura de un polímero depende del proceso de polimerización utilizado y en consecuencia, es posible obtener información sobre los mecanismos de reacción química que ocurren durante su síntesis, dentro de los reactores de polimerización. NMR permite, también, obtener información detallada sobre la dinámica de los polímeros. La gran longitud de los polímeros hace que su dinámica molecular sea sumamente compleja. Sin embargo, mediante el empleo de secuencias de pulsos particulares y mediciones de los tiempos de relajación característicos de los espines nucleares, se obtiene información sobre la dinámica de la macromolécula, de los segmentos que la componen y de los grupos colgantes que pueda poseer. Objetivos Generales y Específicos El trabajo a realizar en el período correspondiente al subsidio solicitado, es la continuación de las investigaciones comenzadas en septiembre de 1995. Se avanzará en el entendimiento de los elastómeros que se están estudiando actualmente. El estudio abarca los elastómeros sin tratamientos térmicos y con tratamientos térmicos (vulcanizados).

Purpose/Objective(s): RTwith TMZ is the standard for GBM. dd TMZ causes prolongedMGMTdepletion in mononuclear cells and possibly in tumor. The RTOG 0525 trial (ASCO 2011) did not show an advantage from dd TMZ for survival or progression free survival. We conducted exploratory, hypothesis-generating subset analyses to detect possible benefit from dd TMZ.Materials/Methods: Patients were randomized to std (150-200 mg/m2 x 5 d) or dd TMZ (75-100 mg/m2 x 21 d) q 4 weeks for 6- 12 cycles. Eligibility included age.18, KPS$ 60, and. 1 cm2 tissue for prospective MGMTanalysis for stratification. Furtheranalyses were performed for all randomized patients (''intent-to-treat'', ITT), and for all patients starting protocol therapy (SPT). Subset analyses were performed by RPA class (III, IV, V), KPS (90-100, = 50,\50), resection (partial, total), gender (female, male), and neurologic dysfunction (nf = none, minor, moderate).Results: No significant difference was seen for median OS (16.6 vs. 14.9 months), or PFS (5.5 vs. 6.7 months, p = 0.06). MGMT methylation was linked to improved OS (21.2 vs. 14 months, p\0.0001), and PFS (8.7 vs. 5.7 months, p\0.0001). For the ITT (n = 833), there was no OS benefit from dd TMZ in any subset. Two subsets showed a PFS benefit for dd TMZ: RPA class III (6.2 vs. 12.6 months, HR 0.69, p = 0.03) and nf = minor (HR 0.77, p = 0.01). For RPA III, dd dramatically delayed progression, but post-progression dd patients died more quickly than std. A similar pattern for nf = minor was observed. For the SPT group (n = 714) there was neither PFS nor OS benefit for dd TMZ, overall. For RPA class III and nf = minor, there was a PFS benefit for dd TMZ (HR 0.73, p = 0.08; HR 0.77, p = 0.02). For nf = moderate subset, both ITT and SPT, the std arm showed superior OS (14.4 vs. 10.9 months) compared to dd, without improved PFS (HR 1.46, p = 0.03; and HR 1.74, p = 0.01. In terms of methylation status within this subset, there were more methylated patients in the std arm of the ITT subset (n = 159; 32 vs. 24%). For the SPT subset (n = 124), methylation status was similar between arms.Conclusions: This study did not demonstrate improved OS for dd TMZ for any subgroup, but for 2 highly functional subgroups, PFS was significantly increased. These data generate the testable hypothesis that intensive treatment may selectively improve disease control in those most likely able to tolerate dd therapy.
