A implementação de programas de LTPP – Long-Term Pavement Performance (avaliação do desempenho de pavimentos a longo prazo), ao criar um registo histórico sobre as características e a evolução do estado dos pavimentos da rede, revela-se uma ferramenta de valor acrescido no auxílio à tomada de decisões, no âmbito da conservação e da reabilitação de pavimentos, por parte das administrações das infraestruturas rodoviárias. O LTPP americano, dentre os existentes no mundo, é o projeto pioneiro e um dos que tem a maior quantidade de informação armazenada. Por isso, foi o escolhido no âmbito desta dissertação para estudar a evolução do estado dos pavimentos em várias secções classificadas no programa como SPS-5, incluídas no estudo de reabilitação de pavimentos betuminosos. A avaliação efetuada considera cerca de 15 anos de registos de dados, na Califórnia, no Texas e no Mississippi. Após uma breve apresentação de vários projetos LTPP existentes no mundo, descreve-se o modo como são recolhidos e estruturados os dados no LTPP americano, referindo os módulos, as tabelas e os campos considerados para estudo que se faz nesta dissertação. Os estados americanos escolhidos apresentam características climáticas que não reproduzem de forma muito próxima o que acontece em Portugal. Mesmo assim, procurou estabelecer-se um paralelo entre aquelas zonas, com base na temperatura do ar e na precipitação, de modo a poder inferir, de forma aproximada, tendências de evolução do estado dos pavimentos para Portugal Continental. A correspondência climática entre os estados americanos e as regiões portuguesas, resultou nos pares Califórnia/Beja, Texas/Lisboa e Mississippi/Porto. No que se refere ao desenvolvimento de patologias nos pavimentos, analisou-se a evolução do fendilhamento longitudinal, transversal, em bloco (ou malha) e por fadiga (ou pele de crocodilo), da desagregação superficial, do Índice de Irregularidade Internacional (IRI) e das rodeiras. O fendilhamento em bloco (ou malha) e a desagregação superficial não foram objeto de estudos estatísticos para determinação de modelos de previsão. A evolução do estado dos pavimentos, traduzida por indicadores relacionados com as patologias analisadas, foi apreciada em função de diversas variáveis associadas às técnicas de reabilitação, tais como a preparação da superfície de trabalho (realização de atividades de fresagem antes da nova camada), a espessura da camada de reforço (50 ou 125 mm) e o tipo de mistura utilizada (nova ou contendo cerca de 30% de material reciclado). Para as situações em que os dados existentes eram coerentes e em quantidade suficiente, apresentam-se análises de regressão multivariada, de modo a prever a evolução no tempo do estado dos pavimentos, tendo em conta um conjunto de variáveis explicativas relacionadas com as técnicas de reabilitação aplicadas e com o tráfego que solicitou os pavimentos. A análise agregada dos dados para cada um dos estados permitiu concluir que a preparação da superfície por fresagem, o aumento da espessura do reforço, e a utilização de misturas betuminosas novas ou com material reciclado, interferem na evolução das degradações. A influência daqueles fatores é variável em função dos diferentes tipos de patologias em análise.

This research evaluated the concrete strength of two mixes which were used in the Polk County project NHS-500-1(3)--10-77 and were developed to meet a contract requirement of 900 psi third-point 28-day flexural strength. Two concrete mixes, the Proposed Mix and the Enhanced Mix, were tested for strength. Based on the experimental results, it was found that the addition of 50 lb of cementitious materials did not significantly increase concrete strength. The requirement of 900 psi 28-day third-point flexural strength (MOR-TPL) was not achieved by this amount of addition of cementitious materials.

Tungsten carbide has a wide range of applications, mainly cemented carbides made of WC and Co, as wear resistant materials. However, the high cost of WC-Co powders encourages the use of a substrate to manufacture a functionally graded material (FGM) tool made of WC-Co and a tool steel. These materials join the high wear resistance of the cemented carbide and the toughness of the steel. This work deals with the study interaction of the WC-Co and H13 steel to design a functionally graded material by means of spark plasma sintering (SPS). The SPS, a novel sintering technique reaching the consolidation of the powders at relatively low temperatures and short dwell times, is a promising technique in processing materials. In this study, WC, H13 steel, WC-Co, WC-H13 steel and WC-Co-H13 steel bulk samples were investigated using scanning electron microscopy and X-ray diffraction techniques to evaluate the phase transformations involved during SPS consolidation process. The W(2)C and W(3)Fe(3)C precipitation were identified after the SPS consolidation of the WC and WC-H13 steel samples, respectively. The precipitation Of W(4)Co(2)C was also identified in the WC-Co and WC-Co-H13 steel samples. The WC-H 13 steel and WC-Co-H13 steel were also evaluated after heat treatments at 1100 degrees C for 9 h, which enhanced the chemical interaction and the precipitation of W(3)Fe(3)C and W(4)Co(2)C, respectively. The lithium salt of the anionic SPS pincer ligand composed of a central hypervalent lambda(4)-phosphinine ring bearing two ortho-positioned diphenylphosphine sulfide side arms reacts with [Mn(CO)(5)Br] to give fac-[Mn(SPS)(CO)(3)], This isomer can be converted photochemicaily to mer-[Mn(SPS)(CO)(3)], with a very high quantum yield (0.80 +/- 0.05). The thermal backreaction is slow (taking ca. 8 h at room temperature), in contrast to rapid electrodecatalyzed mer-to-fac isomerization triggered by electrochemical reduction of mer-[Mn(SPS)(CO)(3)]. Both geometric isomers of [Mn(SPS)(CO)(3)] have been characterized by X-ray crystallography. Both isomers show luminescence from a low-lying (IL)-I-3 (SPS-based) excited state. The light emission of fac-[Mn(SPS)(CO)(3)] is largely quenched by the efficient photoisomerization occurring probably from a low-lying Mn-CO dissociative excited state. Density functional theory (DFT) and time-dependent DFT calculations describe the highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) of fac- and mer-[Mn(CO)(3)(SPS)] as ligand-centered orbitals, largely localized on the phosphinine ring of the SPS pincer ligand. In line with the ligand nature of its frontier orbitals, fac-[Mn(SPS)(CO)(3)] is electrochemically reversibly oxidized and reduced to the corresponding radical cation and anion, respectively. The spectroscopic (electron paramagnetic resonance, IR, and UV-vis) characterization of the radical species provides other evidence for the localization of the redox steps on the SIPS ligand. The smaller HOMO-LUMO energy difference in the case of mer-[Mn(CO)(3)(SPS)], reflected in the electronic absorption and emission spectra, corresponds with its lower oxidation potential compared to that of the fac isomer. The thermodynamic instability of mer-[Mn(CO)(3)(SPS)], confirmed by the DFT calculations, increases upon one-electron reduction and oxidation of the complex.

Fragestellung / Zielsetzung Rollenspiel von standardisierten Patienten (SPs) bei hochstehenden Prüfungen unterliegt hohen Qualitätsansprüchen, um die Reliabilität und Validität der Prüfung zu gewährleisten. Die Literatur über die Qualitätskontrolle bei SPs konzentriert sich vor allen Dingen auf das Feedback (1,2) oder das Ausfüllen der Checklisten (3,4). Bei Qualitätskontrollen bezüglich des Rollenspiels sind Instrumente mit Bezug auf spezifische Rollen publiziert(5). In den meisten Fakultäten beobachten die SP Trainer die von ihnen trainierten SPs während der OSCEs und bilden sich so ein Urteil über die Qualität der Performance. Die Machbarkeit / Notwendigkeit einer systematischen Qualitätskontrolle des Rollenspiels von SPs in OSCEs über diese Praxis hinaus ist unter SP Trainern umstritten. Das Ziel des hier beschriebenen Projektes ist es, die Anwendungen einer Liste von rollenunabhängigen Qualitätsmerkmalen der Spiels von SPs im Rahmen von OSCEs in der Praxis zu testen. Materialien und Methoden Im Rahmen einer Konsensbildung von Experten auf nationaler Ebene wurde eine Liste von Qualitätsmerkmalen zum Rollenspiel von SPs bei OSCEs erstellt. Diese Liste wurde seitdem systematisch zur Vorbereitung, Durchführung und zum Debriefing von OSCEs eingesetzt. Ergebnisse 1. Bei der Erarbeitung der Liste wurde klar, dass verschiedene SP Trainer unterschiedliche Vorstellungen bezüglich von Qualitätsmerkmalen des Rollenspiels beim OSCE hatten. Diese Vorstellungen konnten im Rahmen Erarbeitung der Liste vereinheitlicht werden. 2. Die Liste wird eingesetzt, um den SPs die in sie gesetzten Erwartungen bezüglich eines guten Rollenspiels beim OSCE gezielt zu vermitteln. 3. Die Liste wird zur Qualitätssicherung des Schauspiels in OSCE Prüfungen eingesetzt. Dabei zeigte sich, dass die SPs in der Qualität ihres Spiels in über 90% der Beobachtungen den in sie gesetzten Erwartungen der SP Trainer entsprechen. 4. Anhand der Liste wird den SPs ein formatives Feedbacks zu ihrer Leistung gegeben. Die SPs in unserem Programm haben uns immer wieder zurückgemeldet, dass sie diese Rückmeldungen zu ihrer Performance sehr schätzen. 5. Die Dokumentation der Performance in den vergangenen Prüfungen kann im Training zur nächsten Rolle benutzt werden, um gezielt etwaige Schwächen zu trainieren und die Performance dadurch zu verbessern. 6. In seltenen Fällen, in denen ein SP nicht mehr im Programm behalten werden kann, liefert die Dokumentation der Performance eine gute Gesprächsgrundlage zur Begründung der Differenzen bezüglich der Erwartung und der aktuellen Leistung. Diskussion In unseren Händen ist der Einsatz der Liste von Qualitätsmerkmalen zur Beurteilung der Leistung von Schauspielpatienten mit hoher Konzentration verbunden. Die gewonnene Klarheit in Bezug auf die Erwartungen und die gebrachten Leistungen bringt eine Struktur und Transparenz in unser Programm, die wir und entsprechend ihrer Rückmeldungen auch unsere SPs sehr schätzen.

Pyrimidine-5'-nucleotidase type I (P5'NI) deficiency is an autosomal recessive condition that causes nonspherocytic hemolytic anemia, characterized by marked basophilic stippling and pyrimidine nucleotide accumulation in erythrocytes. We herein present two African descendant patients, father and daughter, with P5'N deficiency, both born from first cousins. Investigation of the promoter polymorphism of the uridine diphospho glucuronosyl transferase 1A (UGT1A) gene revealed that the father was homozygous for the allele (TA7) and the daughter heterozygous (TA6/TA7). P5'NI gene (NT5C3) gene sequencing revealed a further change in homozygosity at amino acid position 56 (p.R56G), located in a highly conserved region. Both patients developed gallstones; however the father, who had undergone surgery for the removal of stones, had extremely severe intrahepatic cholestasis and, liver biopsy revealed fibrosis and siderosis grade III, leading us to believe that the homozygosity of the UGT1A polymorphism was responsible for the more severe clinical features in the father. Moreover, our results show how the clinical expression of hemolytic anemia is influenced by epistatic factors and we describe a new mutation in the P5'N gene associated with enzyme deficiency, iron overload, and severe gallstone formation. To our knowledge, this is the first description of P5'N deficiency in South Americans.

We describe herein a general method for the controlled Heck arylation of allylated malonates. Both electron-rich and electron-poor aryldiazonium salts were readily employed as the aryl-transfer agents in good yields and in high chemo-, regio-, and stereoselectivity without formation of decarboxylated byproducts. Reaction monitoring via ESI-MS was used to support the formation of chelated Pd species through the catalytic cycle. Additionally, some Heck adducts were successfully used in the total synthesis of pharmacologically active γ-lactones.

Excessive occlusal surface wear can result in occlusal disharmony, functional and esthetic impairment. As a therapeutic approach, conventional single crowns have been proposed, but this kind of treatment is complex, highly invasive and expensive. This case report describes the clinical outcomes of an alternative minimally invasive treatment based on direct adhesive-pin retained restorations. A 64-year-old woman with severely worn dentition, eating problems related to missing teeth and generalized tooth hypersensitivity was referred for treatment. Proper treatment planning based on the diagnostic wax-up simulation was used to guide the reconstruction of maxillary anterior teeth with direct composite resin over self-threading dentin pins. As the mandibular remaining teeth were extremely worn, a tooth-supported overdenture was installed. A stabilization splint was also used to protect the restorations. This treatment was a less expensive alternative to full-mouth rehabilitation with positive esthetic and functional outcomes after 1.5 years of follow-up.

A series of novel 1-(substituted phenyl)-3-(2-oxo-1,3,4-oxadiazol-5-yl) β-carbolines (4a-e) and the corresponding Mannich bases 5-9(a-c) were synthesized and evaluated for their in vitro antitumor activity against seven human cancer cell lines. Compounds of 4a-e series showed a broad spectrum of antitumor activity, with GI50 values lower than 15μM for five cell lines. The derivative 4b, having the N,N-dimethylaminophenyl group at C-1, displayed the highest activity with GI50 in the range of 0.67-3.20μM. A high selectivity and potent activity were observed for some Mannich bases, particularly towards resistant ovarian (NCI-ADR/RES) cell lines (5a, 5b, 6a, 6c and 9b), and ovarian (OVCAR-03) cell lines (5b, 6a, 6c, 9a, 9b and 9c). In addition, the interaction of compound 4b with DNA was investigated by using UV and fluorescence spectroscopic analysis. These studies indicated that 4b interact with ctDNA by intercalation binding.

Mucositis induced by anti-neoplastic drugs is an important, dose-limiting and costly side-effect of cancer therapy. To evaluate the effect of the topical application of S-nitrosoglutathione (GSNO), a nitric oxide donor, on 5-fluorouracil (5-FU)-induced oral mucositis in hamsters. Oral mucositis was induced in male hamsters by two intraperitoneal administrations of 5-FU on the first and second days of the experiment (60 and 40 mg/kg, respectively) followed by mechanical trauma on the fourth day. Animals received saline, HPMC or HPMC/GSNO (0.1, 0.5 or 2.0 mM) 1 h prior to the 5-FU injection and twice a day for 10 or 14 days. Samples of cheek pouches were harvested for: histopathological analysis, TNF-α and IL-1β levels, immunohistochemical staining for iNOS, TNF-α, IL-1β, Ki67 and TGF-β RII and a TUNEL assay. The presence and levels of 39 bacterial taxa were analyzed using the Checkerboard DNA-DNA hybridization method. The profiles of NO released from the HPMC/GSNO formulations were characterized using chemiluminescence. The HPMC/GSNO formulations were found to provide sustained release of NO for more than 4 h at concentration-dependent rates of 14 to 80 nmol/mL/h. Treatment with HPMC/GSNO (0.5 mM) significantly reduced mucosal damage, inflammatory alterations and cell death associated with 5-FU-induced oral mucositis on day 14 but not on day 10. HPMC/GSNO administration also reversed the inhibitory effect of 5-FU on cell proliferation on day 14. In addition, we observed that the chemotherapy significantly increased the levels and/or prevalence of several bacterial species. Topical HPMC/GSNO accelerates mucosal recovery, reduces inflammatory parameters, speeds up re-epithelization and decreases levels of periodontopathic species in mucosal ulcers.

Resistant hypertension (RHTN) is a multifactorial disease characterized by blood pressure (BP) levels above goal (140/90 mmHg) in spite of the concurrent use of three or more antihypertensive drugs of different classes. Moreover, it is well known that RHTN subjects have high prevalence of left ventricular diastolic dysfunction (LVDD), which leads to increased risk of heart failure progression. This review gathers data from studies evaluating the effects of phosphodiesterase-5 (PDE-5) inhibitors (administration of acute sildenafil and short-term tadalafil) on diastolic function, biochemical and hemodynamic parameters in patients with RHTN. Acute study with sildenafil treatment found that inhibition of PDE-5 improved hemodynamic parameters and diastolic relaxation. In addition, short-term study with the use of tadalafil demonstrated improvement of LVDD, cGMP and BNP-32 levels, regardless of BP reduction. No endothelial function changes were observed in the studies. The findings of acute and short-term studies revealed potential therapeutic effects of IPDE-5 drugs on LVDD in RHTN patients.A Hipertensão arterial resistente (HAR) é uma doença multifatorial caracterizada por níveis pressóricos acima das metas (140/90 mmHg), a despeito de tratamento farmacológico otimizado de 3 ou mais fármacos anti-hipertensivos de diferentes classes. Pacientes diagnosticados como hipertensos resistentes apresentam alta prevalência de disfunção diastólica do ventrículo esquerdo (DDVE) que proporciona risco aumentado para insuficiência cardíaca. Esta revisão reúne dados de estudos prévios avaliando os efeitos dos inibidores de fosfodiesterase-5 (PDE-5) (administração aguda de sildenafil e de curto prazo de tadalafil) na função diastólica e nos parâmetros bioquímicos e hemodinâmicos em pacientes com HAR. O estudo agudo com sildenafil demonstrou que a inibição da PDE-5 melhorou os parâmetros hemodinâmicos e de relaxamento diastólico. Além disso, o estudo curto prazo com o uso de tadalafil revelou melhora da DDVE e dos níveis de GMPc e BNP-32, independente de redução de pressão arterial. A função endotelial não apresentou alteração com ambos os tratamentos. Os resultados dos estudos agudo e de curto prazo sugerem efeitos terapêuticos potenciais dos fármacos inibidores da PDE-5 na disfunção diastólica em pacientes com HAR.

High levels of substrate-based 1,5-stereoinduction are obtained in the boron-mediated aldol reactions of beta-oxygenated methyl ketones with achiral and chiral aldehydes. Remote induction from the boron enolates gives the 1,5-anti adducts, with the enolate pi-facial selectivity critically dependent upon the nature of the beta-alkoxy protecting group. This 1,5-anti aldol methodology has been strategically employed in the total synthesis of several natural products. At present, the origin of the high level of 1,5-anti induction obtained with the boron enolates is unclear, although a model based on a hydrogen bonding between the alkoxy oxygen and the formyl hydrogen has been recently proposed.

An efficient synthesis of the marine metabolite 3-bromoverongiaquinol (1) and the first total synthesis of 5-monobromocavernicolin (2), both isolated from the marine sponge Aplysina cavernicola, have been described based on the 1,2 addition of the lithium enolate of N,O-bistrimethylsilylacetamide (BSA, 4) to 1,4-benzoquinone (3). Bromination and purification of the crude product on silica gel chromatography provided 3-bromoverongiaquinol (1) in 50% overall yield. Under alkaline conditions, the crude product of the bromination reaction was converted to 5-monobromocavernicolin (2) in 20% yield which was also obtained in 13% yield (25% yield based on recovered starting material) from 3-bromoverongiaquinol (1).

The objective of this work was to compare the soybean crop mapping in the western of Parana State by MODIS/Terra and TM/Landsat 5 images. Firstly, it was generated a soybean crop mask using six TM images covering the crop season, which was used as a reference. The images were submitted to Parallelepiped and Maximum Likelihood digital classification algorithms, followed by visual inspection. Four MODIS images, covering the vegetative peak, were classified using the Parallelepiped method. The quality assessment of MODIS and TM classification was carried out through an Error Matrix, considering 100 sample points between soybean or not soybean, randomly allocated in each of the eight municipalities within the study area. The results showed that both the Overall Classification (OC) and the Kappa Index (KI) have produced values ranging from 0.55 to 0.80, considered good to very good performances, either in TM or MODIS images. When OC and KI, from both sensors were compared, it wasn't found no statistical difference between them. The soybean mapping, using MODIS, has produced 70% of reliance in terms of users. The main conclusion is that the mapping of soybean by MODIS is feasible, with the advantage to have better temporal resolution than Landsat, and to be available on the internet, free of charge.

The objective of this study was to analyze changes in the spectral behavior of the soybean crop through spectral profiles of the vegetation indexes NDVI and GVI, expressed by different physical values such as apparent bi-directional reflectance factor (BRF), surface BRF, and normalized BRF derived from images of the Landsat 5/TM. A soybean area located in Cascavel, Paraná, was monitored by using five images of Landsat 5/TM during the 2004/2005 harvesting season. The images were submitted to radiometric transformation, atmospheric correction and normalization, determining physical values of apparent BRF, surface BRF and normalized BRF. NDVI and GVI images were generated in order to distinguish the soybean biomass spectral response. The treatments showed different results for apparent, surface and normalized BRF. Through the profiles of average NDVI and GVI, it was possible to monitor the entire soybean cycle, characterizing its development. It was also observed that the data from normalized BRF negatively affected the spectral curve of soybean crop, mainly, during the phase of vegetative growth, in the 12-9-2004 image.
