É do conhecimento geral que a população portuguesa está cada vez mais preocupada com a sua saúde e qualidade de vida, bem como em mudar alguns dos seus hábitos alimentares. Consequentemente, a procura por alimentos que proporcionam, para além de boas propriedades organolépticas e nutritivas, benefícios à saúde do consumidor, aumentou significativamente nas últimas décadas, onde os produtos lácteos fermentados surgem em grande destaque. Vários são os microrganismos empregues no desenvolvimento de produtos lácteos fermentados. Na produção de iogurtes, são utilizadas culturas lácteas tradicionais (Lactobacillus delbrueckii subsp. bulgaricus e Streptococcus thermophilus), também denominadas por flora específica do iogurte, e/ou bactérias probióticas, como as bifidobactérias, devido às suas boas características tecnológicas, terapêuticas e sensoriais. O trabalho descrito nesta dissertação de mestrado foca-se na validação e implementação de metodologias que permitem a contagem da flora específica do iogurte e de bifidobactérias, em iogurtes, na empresa SGS Portugal, S.A. A enumeração da flora específica do iogurte teve como base a norma ISO 7889:2003, por meio da técnica de sementeira por incorporação no meio de cultura apropriado e posterior contagem em placa dos microrganismos presentes. Foram analisadas 20 amostras de iogurtes, obtendo-se valores de contagens da flora específica do iogurte na ordem de 108 UFC/g de produto, indo ao encontro dos valores estabelecidos pela legislação portuguesa em vigor. A enumeração de bifidobactérias teve como base a norma ISO 29981:2010, por meio da técnica de sementeira por incorporação no meio de cultura apropriado e posterior contagem em placa dos microrganismos presentes. Das 20 amostras de iogurtes probióticos analisadas, apenas 15 obtiveram resultados positivos, apresentando valores de contagens de bifidobactérias superiores a 106 UFC/g de produto. Estas amostras, para além de apresentarem valores de acordo com os valores estabelecidos pela legislação portuguesa em vigor, são terapeuticamente eficazes, durante todo o prazo de validade do produto, uma vez que apresentam valores de contagem de bactérias probióticas superiores a 106 UFC/g, sendo este o padrão mínimo necessário para que um iogurte seja considerado como probiótico. Com base nos resultados obtidos e no cumprimento dos parâmetros preconizados nos procedimentos de validação de métodos microbiológicos, foi possível a validação destas metodologias e as suas implementações na empresa SGS Portugal, S.A.

As alergias alimentares são um grande problema de saúde pública a nível mundial, que ocorre em todas as faixas etárias, tendo maior incidência em crianças. Mediada pelo sistema imunitário, a alergia alimentar é uma reação adversa que ocorre devido à presença de alergénios alimentares, que são identificados erroneamente como sendo um perigo ao organismo. Como é uma patologia que não possui cura, a prevenção é a melhor forma de evitar que ocorram reações alérgicas em indivíduos sensíveis, sendo a rotulagem um componente indispensável para a identificação dos alergénios presentes nos alimentos. Dentro dos métodos utilizados para a deteção de alergénios está a Reação em Cadeia da Polimerase em tempo real (PCR em tempo real), que é capaz de identificar o gene que codifica a proteína alergénica. Este método possui elevada sensibilidade e especificidade, sendo atualmente utilizado na área alimentar para a deteção de organismos genéticamente modificados e alergénios. Devido a uma grande procura de clientes e consumidores, para identificação de alergénios em alimentos, o objetivo do presente trabalho foi implementar e validar, a partir de análises de sensibilidade, precisão e robustez, o método de PCR em tempo real. Dessa forma, foi testada a deteção qualitativa de oito alergénios em alimentos, sendo estes o aipo, amendoim, avelã, caju, noz, pistáchio, sésamo e soja, no Laboratório da SGS - Société Générale de Surveillance. Destes alergénios, foi possível a validação de sete, demonstrando que o método de PCR em tempo real, para os presentes alergénios, possui elevada precisão e sensibilidade nos resultados obtidos.

Sub-grid scale (SGS) models are required in order to model the influence of the unresolved small scales on the resolved scales in large-eddy simulations (LES), the flow at the smallest scales of turbulence. In the following work two SGS models are presented and deeply analyzed in terms of accuracy through several LESs with different spatial resolutions, i.e. grid spacings. The first part of this thesis focuses on the basic theory of turbulence, the governing equations of fluid dynamics and their adaptation to LES. Furthermore, two important SGS models are presented: one is the Dynamic eddy-viscosity model (DEVM), developed by \cite{germano1991dynamic}, while the other is the Explicit Algebraic SGS model (EASSM), by \cite{marstorp2009explicit}. In addition, some details about the implementation of the EASSM in a Pseudo-Spectral Navier-Stokes code \cite{chevalier2007simson} are presented. The performance of the two aforementioned models will be investigated in the following chapters, by means of LES of a channel flow, with friction Reynolds numbers $Re_\tau=590$ up to $Re_\tau=5200$, with relatively coarse resolutions. Data from each simulation will be compared to baseline DNS data. Results have shown that, in contrast to the DEVM, the EASSM has promising potentials for flow predictions at high friction Reynolds numbers: the higher the friction Reynolds number is the better the EASSM will behave and the worse the performances of the DEVM will be. The better performance of the EASSM is contributed to the ability to capture flow anisotropy at the small scales through a correct formulation for the SGS stresses. Moreover, a considerable reduction in the required computational resources can be achieved using the EASSM compared to DEVM. Therefore, the EASSM combines accuracy and computational efficiency, implying that it has a clear potential for industrial CFD usage.

To compare time and risk to biochemical recurrence (BR) after radical prostatectomy of two chronologically different groups of patients using the standard and the modified Gleason system (MGS). Cohort 1 comprised biopsies of 197 patients graded according to the standard Gleason system (SGS) in the period 1997/2004, and cohort 2, 176 biopsies graded according to the modified system in the period 2005/2011. Time to BR was analyzed with the Kaplan-Meier product-limit analysis and prediction of shorter time to recurrence using univariate and multivariate Cox proportional hazards model. Patients in cohort 2 reflected time-related changes: striking increase in clinical stage T1c, systematic use of extended biopsies, and lower percentage of total length of cancer in millimeter in all cores. The MGS used in cohort 2 showed fewer biopsies with Gleason score ≤ 6 and more biopsies of the intermediate Gleason score 7. Time to BR using the Kaplan-Meier curves showed statistical significance using the MGS in cohort 2, but not the SGS in cohort 1. Only the MGS predicted shorter time to BR on univariate analysis and on multivariate analysis was an independent predictor. The results favor that the 2005 International Society of Urological Pathology modified system is a refinement of the Gleason grading and valuable for contemporary clinical practice.

Background: Despite governmental and private efforts on providing malaria control, this disease continues to be a major health threat. Thus, innovative strategies are needed to reduce disease burden. The malaria vectors, through the injection of saliva into the host skin, play important role on disease transmission and may influence malaria morbidity. This study describes the humoral immune response against Anopheles (An.) darlingi saliva in volunteers from the Brazilian Amazon and addresses the association between levels of specific antibodies and clinical presentation of Plasmodium (P.) vivax infection. Methods: Adult volunteers from communities in the Rondonia State, Brazil, were screened in order to assess the presence of P. vivax infection by light microscopy and nested PCR. Non-infected volunteers and individuals with symptomatic or symptomless infection were randomly selected and plasma collected. An. darlingi salivary gland sonicates (SGS) were prepared and used to measure anti-saliva antibody levels. Plasma interleukin (IL)-10 and interferon (IFN)-gamma levels were also estimated and correlated to anti-SGS levels. Results: Individuals infected with P. vivax presented higher levels of anti-SGS than non-infected individuals and antibody levels could discriminate infection. Furthermore, anti-saliva antibody measurement was also useful to distinguish asymptomatic infection from non-infection, with a high likelihood ratio. Interestingly, individuals with asymptomatic parasitaemia presented higher titers of anti-SGS and lower IFN-gamma/IL-10 ratio than symptomatic ones. In P. vivax-infected asymptomatic individuals, the IFN-gamma/IL-10 ratio was inversely correlated to anti-SGS titers, although not for while in symptomatic volunteers. Conclusion: The estimation of anti-An. darlingi antibody levels can indicate the probable P. vivax infection status and also could serve as a marker of disease severity in this region of Brazilian Amazon.

The use of chloroplast DNA markers (cpDNA) helps to elucidate questions related to ecology, evolution and genetic structure. The knowledge of inter-and intra-population genetic structure allows to design effective conservation and management strategies for tropical tree species. With the aim to help the conservation of Hymenaea stigonocarpa of the Cerrado (Brazilian savanna) in Sao Paulo State, an analysis of the spatial genetic structure (SGS) was conducted in two populations using five universal chloroplast microsatellite loci (cpSSR). The population of 68 trees of H. stigonocarpa in the Ecological Station of Itirapina (ESI) had a single haplotype, indicating a strong founder effect. In turn, the population of 47 trees of H. stigonocarpa in a contiguous area that includes the Ecological Station of Assis and the Assis State Forest (ESA), showed six haplotypes ((n) over cap (h) = 6) with a moderate haplotype diversity ((h) over cap = 0667 + 0094), revealing that it was founded by a small number of maternal lineages. The SGS analysis for the population ESA/ASF, using Moran`s I index, indicated limited seed dispersal. Considering SGS, for ex situ conservation strategies in the population ESA/ASF, seed harvesting should require a minimum distance of 750 m among seed-trees.

Understanding the mating patterns of populations of tree species is a key component of ex situ genetic conservation. In this study, we analysed the genetic diversity, spatial genetic structure (SGS) and mating system at the hierarchical levels of fruits and individuals as well as pollen dispersal patterns in a continuous population of Theobroma cacao in Para State, Brazil. A total of 156 individuals in a 0.56 ha plot were mapped and genotyped for nine microsatellite loci. For the mating system analyses, 50 seeds were collected from nine seed trees by sampling five fruits per tree (10 seeds per fruit). Among the 156 individuals, 127 had unique multilocus genotypes, and the remaining were clones. The population was spatially aggregated; it demonstrated a significant SGS up to 15m that could be attributed primarily to the presence of clones. However, the short seed dispersal distance also contributed to this pattern. Population matings occurred mainly via outcrossing, but selfing was observed in some seed trees, which indicated the presence of individual variation for self-incompatibility. The matings were also correlated, especially within ((r) over cap (p(m)) = 0.607) rather than among the fruits ((r) over cap (p(m)) = 0.099), which suggested that a small number of pollen donors fertilised each fruit. The paternity analysis suggested a high proportion of pollen migration (61.3%), although within the plot, most of the pollen dispersal encompassed short distances (28m). The determination of these novel parameters provides the fundamental information required to establish long-term ex situ conservation strategies for this important tropical species. BACKGROUND: The development of newer diagnostic technologies has reduced the need for invasive electroencephalographic (EEG) studies in identifying the epileptogenic zone, especially in adult patients with mesial temporal lobe epilepsy and hippocampal sclerosis (MTLE-HS). OBJECTIVE: To evaluate ictal single photon emission computed tomography (SPECT) in the evaluation and treatment of patients with MTLE-HS. METHODS: MTLE patients were randomly assigned to those with (SPECT, n = 124) and without ictal SPECT (non-SPECT, n = 116) in an intent-to-treat protocol. Primary end points were the proportion of patients with invasive EEG studies, and those offered surgery. Secondary end points were the length of hospital stay and the proportion of patients with secondarily generalized seizures (SGS) during video-EEG, postsurgical seizure outcome, and hospital cost. RESULTS: The proportion of patients offered surgery was similar in the SPECT (85%) and non-SPECT groups (81%), as well as the proportion that had invasive EEG studies (27% vs 23%). The mean duration of hospital stay was 1 day longer for the SPECT group (P < 0.001). SGS occurred in 51% of the SPECT and 26% of the non-SPECT group (P < 0.001). The cost of the presurgical evaluation was 35% higher for the SPECT compared with the non-SPECT group (P < 0.001). The proportion of patients seizure-free after surgery was similar in the SPECT (59%) compared with non-SPECT group (54%). CONCLUSION: Ictal-SPECT did not add localizing value beyond what was provided by EEG-video telemetry and structural MRI that altered the surgical decision and outcome for MTLE-HS patients. Ictal-SPECT increased hospital stay was associated with increased costs and a higher chance of SGS during video-EEG monitoring. These findings support the notion that a protocol including ictal SPECT is equivalent to one without SPECT in the presurgical evaluation of adult patients with MTLE-HS.
