The low-lying doublet and quartet electronic states of the species SeF correlating with the first dissociation channel are investigated theoretically at a high-level of electronic correlation treatment, namely, the complete active space self-consistent field/multireference single and double excitations configuration interaction (CASSCF/MRSDCI) using a quintuple-zeta quality basis set including a relativistic effective core potential for the selenium atom. Potential energy curves for (Lambda+S) states and the corresponding spectroscopic properties are derived that allows for an unambiguous assignment of the only spectrum known experimentally as due to a spin-forbidden X (2)Pi-a (4)Sigma(-) transition, and not a A (2)Pi-X (2)Pi transition as assumed so far. For the bound excited doublets, yet unknown experimentally, this study is the first theoretical characterization of their spectroscopic properties. Also the spin-orbit coupling constant function for the X (2)Pi state is derived as well as the spin-orbit coupling matrix element between the X (2)Pi and a (4)Sigma(-) states. Dipole moment functions and vibrationally averaged dipole moments show SeF to be a very polar species. An overview of the lowest-lying spin-orbit (Omega) states completes this description. A economia portuguesa tem contado com o investimento estrangeiro como um factor relevante de crescimento, mas nos anos recentes este conheceu valores anormalmente baixos. As novas normas legais sobre o denominado Golden Visa, ao permitir a atribuição de vistos a investidores estrangeiros, com condições especiais e a possível atribuição de nacionalidade portuguesa, decorridos cinco anos, vem captar investimento estrangeiro de países não europeus, chamados de países terceiros. O Serviço de Estrangeiros e Fronteiras (SEF) é um dos serviços públicos de grande importância no desenvolvimento e aplicação de mecanismos, no que respeita à imigração em Portugal. No decorrer do tempo, cada vez mais esta instituição tem uma voz ativa nas questões relacionadas com a entrada e saída de cidadãos estrangeiros, em território nacional. Em Portugal existem várias políticas de integração dos imigrantes, bem como noutros países. Portanto, neste estudo pretendemos expor algumas políticas adotadas pelo Estado português, com base em três níveis evidentes: o social, o laboral e o educativo. A nível humanitário, o nosso estudo também demonstra extrema preocupação com as pessoas estrangeiras, uma vez que cada vez mais são aprovadas leis para a proteção dos cidadãos imigrantes no país, como: a discriminação, a inclusão social, as organizações não-governamentais, o asilo e a proteção internacional. Na Constituição da República Portuguesa, a discriminação é condenável e não é aceite, pois todos têm igual acesso aos direitos sociais, económicos e culturais por parte dos cidadãos de grupos desfavorecidos, como por exemplo os imigrantes e as minorias étnicas. Para a economia da nossa reflexão a inclusão social, importa realçar exemplos mais marcantes de programas e iniciativas públicos para a integração social, a nível das políticas sociais e de emprego, que em Portugal são tuteladas pelos Ministérios da Solidariedade, Emprego e Segurança Social, e da Educação. No caso português é grande o número de organizações nãogovernamentais a trabalhar com a imigração. A maior parte destas organizações situam-se nas zonas e bairros mais problemáticos, onde o número de cidadãos imigrantes é mais elevado, e trabalham os problemas concretos existentes nestas populações. Existem outras ONG que trabalham a nível nacional e que funcionam como grupos de pressão, promovendo os direitos dos imigrantes e lutando contra o racismo e a discriminação. No decorrer dos tempos, o avanço tecnológico veio também contribuir para um melhor controlo e funcionamento do hoje designado Serviço de Estrangeiros e Fronteiras (SEF) e, consequentemente, para um tratamento mais célere e eficiente dos serviços que tratam o fenómeno da imigração. A eficácia dos S.E.F. muito deve a uma progressiva dotação de equipamentos eletrónicos, para poder desempenhar melhor as suas funções em território nacional; mas sem nunca esquecer o potencial humano existente.

The involvement of type 1 fimbriae in colonisation of the rat gastrointestinal tract in vivo was investigated with Salmonella enterica serotype Enteritidis LA5 and a mutant of LA5 denoted EAV3 unable to elaborate type 1 fimbriae (SEF 21), Rats were given a single dose of LA5 or EAV3 or a 1:1 mixture of both, LA5 was found in higher numbers in the stomach and small intestine than EAV3 at 6 h after infection with a single strain, but not after 6 days, LA5 did not out-compete EAV3 when the strains were administered together. Indeed, after 6 and 21 days, EAV3 was found in the distal small intestine and large intestine in far higher numbers than LA5. These findings suggest that SEF 21 have an important role(s) in the early stages of infection in vivo, However, SEF 21 expression may disadvantage the pathogen in the longer term as indicated by EAV3 out-competing LA5 in the gut at 21 days.

Cytokine-induced transcription of the serum amyloid A3 (SAA3) gene promoter requires a transcriptional enhancer that contains three functional elements: two C/EBP-binding sites and a third site that interacts with a constitutively expressed transcription factor, SAA3 enhancer factor (SEF). Deletion or site-specific mutations in the SEF-binding site drastically reduced SAA3 promoter activity, strongly suggesting that SEF is important in SAA3 promoter function. To further elucidate its role in the regulation of the SAA3 gene, we purified SEF from HeLa cell nuclear extracts to near homogeneity by using conventional liquid chromatography and DNA-affinity chromatography. Ultraviolet cross-linking and Southwestern experiments indicated that SEF consisted of a single polypeptide with an apparent molecular mass of 65 kDa. Protein sequencing, oligonucleotide competition and antibody supershift experiments identified SEF as transcription factor LBP-1c/CP2/LSF. Cotransfection of SEF expression plasmid with SAA3-luciferase reporter resulted in 3- to 5-fold activation of SAA3 promoter. Interestingly, when SEF-transfected cells were treated with either conditioned medium (CM) or interleukin (IL) 1, the SAA3 promoter was synergistically activated in a dose-dependent manner. Furthermore, when SEF-binding site was mutated, the response of SAA3 promoter to IL-1 or CM stimulation was abolished or drastically decreased, suggesting that SEF may functionally cooperate with an IL-1-inducible transcription factor. Indeed, our functional studies showed that NFκB is a key transcription factor that mediates the IL-1-induced expression of SAA3 gene, and that SEF can synergize with NFκBp65 to activate SAA3 promoter. By coimmunoprecipitation experiments, we found that SEF could specifically interact with NFκBp65, and that the association of these two factors was enhanced upon IL-1 and CM stimulation. This suggests that the molecular basis for the functional synergy between SEF and NFκB may be due to the ability of SEF to physically interact with NPκB. In addition to its interaction with SEF, NFκB-dependent activation also requires the weak κB site in the C element and its interaction with C/EBP. Besides its role in regulating SAA3 gene expression, we provide evidence that SEF could also bind in a sequence-specific manner to the promoters of α2-macroglobulin, Aα fibrinogen, and 6–16 genes and to an intronic enhancer of the human Wilm's tumor 1 gene, suggesting a functional role in the regulation of these genes. By coimmunoprecipitation experiments, we determined that SEF could specifically associate with both Stat3 and Stat2 upon cytokine stimulation. To examine the functional roles of such interactions, we evaluated the effects of SEF on the transcriptional regulation of two reporter genes: Aα fibrinogen and 6–16, which are IL-6- and interferon-α-responsive, respectively. Our results showed that cotransfection of SEF expression plasmid can activate the expression of Aα fibrinogen gene and 6–16 gene. Moreover, SEF can dramatically enhance the interferon-α-induced expression of 6–16 gene and IL-6-induced expression of Aα fibrinogen gene, suggesting that SEF may functionally cooperate with ISGF3 and Stat3 to mediate interferon-α and IL-6 signaling. ^ Our findings that SEF can interact with multiple cytokine-inducible transcription factors to mediate the expression of target genes open a new avenue of investigation of cooperative transcriptional regulation of gene expression, and should further our understanding of differential gene expression in response to a specific stimulus. In summary, our data provide evidence that SEF can mediate the signaling of different cytokines by interacting with various cytokine-inducible transcription factors. ^
