This paper has the objective of monitoring the biological activity of composting process of sewage sludge, sugarcane bagasse and ground coffee in a hermetic rotary reactor using the respirometric method in laboratory scale, in order to obtain parameters and system design for large scale projects. Another particularity of this study is the use of a hermetic reactor with gas purging cycles. Purging was performed when the percentage of oxygen reached less than 5%, thus eliminating the gaseous mixture (with elevated CO2 ratio) and the introduction of environmental air with around 21% of O2, successively until the compost was stabilized. The average purge intervals obtained were 29 h and 2 min with reactor rotation frequency of 15 min. The time of the compost stabilization was optimized in 60% if compared to the 90 days in the traditional method. The results obtained can be used to design the process in industrial scale using a simple O2 gas analyzer.

Este trabalho objetivou estudar o efeito da vinhaça na biodegradação em solo da borra oleosa proveniente da refinaria de petróleo Replan-Petrobras. Foi utilizado o método respirométrico de Bartha para verificar a eficiência de tratamentos constituídos de solo, borra oleosa nas concentrações 7 e 14 % (m/m) e ajuste da umidade do solo com e sem vinhaça (0,11 mL/g solo seco) durante 121 dias. Embora a adição da vinhaça tenha proporcionado um aumento da população microbiana nos tratamentos, esta não se mostrou adequada para aumentar a eficiência de biodegradação da borra oleosa em solo, uma vez que não houve diferença entre o CO2 produzido nos tratamentos com ou sem vinhaça após o consumo total da vinhaça. Assim, o uso da vinhaça como agente estimulante em processos de biodegradação mostrou-se ineficiente nas condições estudadas.

This work aimed to assess the aerobic biodegradation of butanol/gasoline, blends (5; 10; 15 and 20% v/v), being the latter compared to the ethanol/gasoline blend (20% v/v). Two experimental techniques were employed, namely the respirometric method and the redox indicator DCPIP test. in the former, experiments simulating the contamination of natural environments (addition of 50 mL of fuel kg(-1) of soil from a non-contaminated site and 20 mL of fuel L(-1) of water from a river) were carried out in biometer flasks (250 mL), used to measure the microbial CO(2) production. The DCPIP test assessed the capability of four inocula to biodegrade the blends of 20%. The addition of butanol at different concentrations enhanced the biodegradation of gasoline in soil. However, no practical gains were observed for concentrations of butanol above 10%. Ethanol showed to have a much faster biodegradation rate than butanol, particularly in water, and the following order of biodegradability was found: ethanol > butanol > gasoline. The addition of the alcohols to the gasoline resulted in positive synergic effects on the biodegradation of the fuels in soil and water matrices. Furthermore, results suggest that, in soil, butanol better enhanced the biodegradation of gasoline than ethanol. (C) 2009 Elsevier Ltd. All rights reserved

Para avaliar a possibilidade de biorremediação em áreas contaminadas, é necessário determinar a biodegradação dos poluentes no solo. Para esta determinação, emprega-se comumente no Brasil, o método respirométrico de Bartha, adaptado de uma norma holandesa. Porém, os solos tropicais possuem características bem diferentes dos solos de regiões de clima temperado. Neste trabalho, foi estudada a aplicabilidade de tal método para um latossolo, tipo de solo predominante no Estado de São Paulo. A partir dos resultados obtidos, foi possível verificar que reações abióticas geram gás carbônico em quantidades significativas. Constatou-se, também, a dificuldade de esterilização do solo em autoclave, o que impossibilita a avaliação da remoção dos poluentes por outros mecanismos ou a biodegradação dos mesmos por microrganismos exógenos somente. Portanto, não se recomenda a aplicação do teste respirométrico de Bartha para a determinação da biodegradabilidade de poluentes em latossolos.

The biodegradability of pure diesel and biodiesel and blends with different proportions of biodiesel (2% (commercial); 5% and 20%) was evaluated employing the respirometric method and the redox indicator 2,6-dichlorophenol indophenol (DCPIP) test. In the former, experiments simulating the contamination of natural environments (soil from a petrol station or water from a river) were carried out in Bartha biometer flasks (250 ml), and used to measure the microbial CO 2 production. With the DCPIP test, the capability of three inocula to biodegrade the blends was tested. Results show that although biodiesel is more easily and faster biodegraded than diesel oil, among the blends evaluated (2%, 5% and 20%), only the blend with higher concentration of biodiesel presented biodegradability significantly different from diesel and it was not verified an improvement on the biodegradation of the diesel by means of co-metabolism. © 2008 Academic Journals.

This study aimed to investigate the biodegradation of polypropylene/ poly(hydroxybutyrate-co-hydroxyvalerate) (PP/PHBV) blend (70/30, w/w) films in soil, monitoring the evolution of CO2 using the respirometric method. The polymeric films were incubated at 28°C ± 2°C for 180 days in biometer flasks, and the sequence of biodegradation percentage was PP/PHBV (70/30) > PP, that is, 15% and 0%, respectively. Fourier transform infrared spectroscopy and X-ray diffraction measurements showed that biodegradation occurs in the blend PP/PHBV interphases. Preferentially, the microbial action occurs in the fraction of the biodegradable polymer (PHBV), and it influences the PP fraction morphology, which showed some significant changes in the monomer unit sequences and the organization of the chains. © 2013 Wiley Periodicals, Inc.

The increase in the oil price and the current trend of using renewable raw materials for the production of chemicals renew the interest in the production of biobutanol that, produced by fermentation of agricultural raw materials, can be used as a component of gasoline and diesel. With the commercialization of new fuels, environmental damages due to spills can occur. Among other techniques, the clean-up of these contaminated areas can be achieved with bioremediation, a technique based on the action of microorganisms, which has the advantage of turning hazardous contaminants into non toxic substances such as CO2, water and biomass. Thus, bearing in mind the use of biobutanol in the near future as a gasoline extender and due to the lack of knowledge of the effects of butanol on the biodegradation of gasoline, this work aimed to assess the aerobic biodegradation of butanol/gasoline blends and butanol/diesel (20% v/v), being the latter compared to the ethanol/gasoline blend and biodiesel/diesel (20% v/v), respectively. The present paper describes a novel, simple and reliable differential pulse voltammetric method for determining amitriptyline (AMT) in pharmaceutical formulations. It has been described for many authors that this antidepressant is electrochemically inactive at carbon electrodes. However, the procedure proposed herein consisted in electrochemically oxidizing AMT at an unmodified carbon nanotube paste electrode in the presence of 0.1 mol L(-1) sulfuric acid used as electrolyte. At such concentration, the acid facilitated the AMT electroxidation through one-electron transfer at 1.33 V vs. Ag/AgCl, as observed by the augmentation of peak current. Concerning optimized conditions (modulation time 5 ms, scan rate 90 mV s(-1), and pulse amplitude 120 mV) a linear calibration curve was constructed in the range of 0.0-30.0 μmol L(-1), with a correlation coefficient of 0.9991 and a limit of detection of 1.61 μmol L(-1). The procedure was successfully validated for intra- and inter-day precision and accuracy. Moreover, its feasibility was assessed through analysis of commercial pharmaceutical formulations and it has been compared to the UV-vis spectrophotometric method used as standard analytical technique recommended by the Brazilian Pharmacopoeia.

The present work compared the local injection of mononuclear cells to the spinal cord lateral funiculus with the alternative approach of local delivery with fibrin sealant after ventral root avulsion (VRA) and reimplantation. For that, female adult Lewis rats were divided into the following groups: avulsion only, reimplantation with fibrin sealant; root repair with fibrin sealant associated with mononuclear cells; and repair with fibrin sealant and injected mononuclear cells. Cell therapy resulted in greater survival of spinal motoneurons up to four weeks post-surgery, especially when mononuclear cells were added to the fibrin glue. Injection of mononuclear cells to the lateral funiculus yield similar results to the reimplantation alone. Additionally, mononuclear cells added to the fibrin glue increased neurotrophic factor gene transcript levels in the spinal cord ventral horn. Regarding the motor recovery, evaluated by the functional peroneal index, as well as the paw print pressure, cell treated rats performed equally well as compared to reimplanted only animals, and significantly better than the avulsion only subjects. The results herein demonstrate that mononuclear cells therapy is neuroprotective by increasing levels of brain derived neurotrophic factor (BDNF) and glial derived neurotrophic factor (GDNF). Moreover, the use of fibrin sealant mononuclear cells delivery approach gave the best and more long lasting results.
