Este Trabalho Final de Mestrado consiste num estágio que se encontra reportado neste relatório e que traduz o acompanhamento por parte do discente de uma empreitada de remodelação e reconversão de um edifício em Lisboa. O estágio com duração de seis meses desenvolveu-se entre Fevereiro e Julho de 2012 na empresa Arlindo Correia e Filhos, SA. Consistiu no acompanhamento da obra das futuras instalações da ASAE (Autoridade de Segurança Alimentar e Económica) na freguesia de São Mamede, concelho de Lisboa, sendo o orientador do estágio por parte da empresa, o Eng.º Vítor Soares, Director de Produção em causa. Os objectivos do estágio curricular visam a consolidação das matérias e temas abordados na vertente académica, assim como o contacto com a realidade do mundo laboral. A integração na empresa Arlindo Correia e Filhos SA e o acompanhamento regular das frentes de trabalho e de questões relacionadas com a obra, contribuíram de forma decisiva para cumprir com sucesso esta missão.

Climate change has created the need for new strategies in conservation planning that account for the dynamics of factors threatening endangered species. Here we assessed climate change threat to the European otter, a flagship species for freshwater ecosystems, considering how current conservation areas will perform in preserving the species in a climatically changed future. We used an ensemble forecasting approach considering six modelling techniques applied to eleven subsets of otter occurrences across Europe. We performed a pseudo-independent and an internal evaluation of predictions. Future projections of species distribution were made considering the A2 and B2 scenarios for 2080 across three climate models: CCCMA-CGCM2, CSIRO-MK2 and HCCPR HAD-CM3. The current and the predicted otter distributions were used to identify priority areas for the conservation of the species, and overlapped to existing network of protected areas. Our projections show that climate change may profoundly reshuffle the otter's potential distribution in Europe, with important differences between the two scenarios we considered. Overall, the priority areas for conservation of the otter in Europe appear to be unevenly covered by the existing network of protected areas, with the current conservation efforts being insufficient in most cases. For a better conservation, the existing protected areas should be integrated within a more general conservation and management strategy incorporating climate change projections. Due to the important role that the otter plays for freshwater habitats, our study further highlights the potential sensitivity of freshwater habitats in Europe to climate change.

The Basic Law of Governance in Saudi Arabia stipulates that the king of Saudi Arabia has absolute power in the government of Saudi Arabia. However, after King Abdullah's accession to the throne in 2005, his political powers were limited because of the presence of the so-called Sudeiri Seven, the powerful royal group that consists of the seven sons of King Abdel-Aziz's purported favorite wife, Sheikha Hussa bin Ahmad Sudeiri. The death of the crown prince Sultan in 2011 followed by the death of the next crown prince Naif in 2012, both members of the Sudeiri Seven, weakened the power of the Sudeiri Seven. As a result, King Abdullah's power had increased greatly compared to that of the Sudeiri Seven. Also, the sons of King Abdullah, who occupied prominent governmental posts, were acquiring strong influence in the regime. The death of King Abdullah in January 2015 and Salman's accession to the throne caused changes to the ruling regime in Saudi Arabia. King Salman appointed Prince Muqrin as crown prince and deputy premier, and Prince Muhammad b. Naif as deputy crown prince. King Salman also appointed his son Muhammad b. Salman as defence minister and head of the royal court. Finally, King Salman issued a royal order on January 29 to reshuffle his cabinet and dismiss the governors of the Riyadh and Makka.

A method for determining the kinetic fate of structured disulfide species (i.e., whether they are preferentially oxidized or reshuffle back to an unstructured disulfide species) is introduced. The method relies on the sensitivity of unstructured disulfide species to low concentrations of reducing agents. Because a structured des species that preferentially reshuffles generally first rearranges to an unstructured species, a small concentration of reduced DTT (e.g., 260 μM) suffices to distinguish on-pathway intermediates from dead-end species. We apply this method to the oxidative folding of bovine pancreatic ribonuclease A (RNase A) and show that des[40–95] and des[65–72] are productive intermediates, whereas des[26–84] and des[58–110] are metastable dead-end species that preferentially reshuffle. The key factor in determining the kinetic fate of these des species is the relative accessibility of both their thiol groups and disulfide bonds. Productive intermediates tend to be disulfide-secure, meaning that their structural fluctuations preferentially expose their thiol groups, while keeping their disulfide bonds buried. By contrast, dead-end species tend to be disulfide-insecure, in that their structural fluctuations expose their disulfide bonds in concert with their thiol groups. This distinction leads to four generic types of oxidative folding pathways. We combine these results with those of earlier studies to suggest a general three-stage model of oxidative folding of RNase A and other single-domain proteins with multiple disulfide bonds.
