Indagamos, a partir de los manuscritos inéditos de Das Ich und das Es, los puntos eminentes instaurados por la lectura de sus tres versiones: borrador, copia en limpio y texto publicado, en el momento en que surge una disimetría entre lo reprimido-icc y un Icc no-todo reprimido. Estos documentos revelan una transcripción 'casi' directa de las formulaciones freudianas en un estado naciente que llevan la marca de pensamientos urgidos por lo real del psicoanálisis, cuando todavía no está presente el tiempo de hacerse comprender en el contexto de su obra. Y es, justamente, en esta condición de inicial que el texto manuscrito ofrece una novedosa redefinición del Icc. Tal como señala en la Introducción las cuestiones que interroga en El yo y el ello retoman los pensamientos iniciados en su escrito de 1920 pero sin seguir la elaboración teórica que llama 'especulación analítica', aunque a último momento incluya sus conclusiones. Esta relectura interroga el encuentro de la pulsión de muerte con la segunda tópica, es decir, con ese material Icc que permanece no-reconocido. Y, a partir de 1924, establece de que manera el supuesto de la pulsión de muerte ilumina la discontinuidad existente ente icc e Icc [ello]. El cruce entre la hipótesis especulativa y un masoquismo erógeno primario produce un giro y da entrada a la cura misma como satisfacción sustitutiva. En síntesis, la lectura de los textos de El yo y el ello contando, por primera vez, con las tres versiones y sus dos anexos, introducen, con la disimilitud icc e Icc, un cambio de pregunta; anticipan la hendidura o Spaltung del sujeto, con ese objeto [el yo-cuerpo] que Freud no termina de construir; y anuncian que el Icc es lo que se funda de la huella de lo no-reconocido, que clama por ese mismo campo heterogéneo que obligaba en 1920 a tomar en consideración un más allá del principio de placer que divide el espacio euclidiano dejando asomar también su carácter disímil, asimétrico. Así, en ciertos momentos privilegiados de un análisis, vía trabajo del sueño, se produce la activación de los restos de lo visto y de lo oído, es decir, de un saber hecho de sedimentos, de los excedentes traumáticos del tesoro de palabras, para cada cual, de su lengua materna, cuando dicha lengua se separa del lenguaje produciendo un Icc no-todo reprimido que bordea el agujero de lo no-reconocido, es decir, de su punto de fracaso mismo: la imposibilidad con que el sexo se inscribe en el inconsciente

Background: Cancer shows a great diversity in its clinical behavior which cannot be easily predicted using the currently available clinical or pathological markers. The identification of pathways associated with lymph node metastasis (N+) and recurrent head and neck squamous cell carcinoma (HNSCC) may increase our understanding of the complex biology of this disease. Methods: Tumor samples were obtained from untreated HNSCC patients undergoing surgery. Patients were classified according to pathologic lymph node status (positive or negative) or tumor recurrence (recurrent or non-recurrent tumor) after treatment (surgery with neck dissection followed by radiotherapy). Using microarray gene expression, we screened tumor samples according to modules comprised by genes in the same pathway or functional category. Results: The most frequent alterations were the repression of modules in negative lymph node (N0) and in non-recurrent tumors rather than induction of modules in N+ or in recurrent tumors. N0 tumors showed repression of modules that contain cell survival genes and in non-recurrent tumors cell-cell signaling and extracellular region modules were repressed. Conclusions: The repression of modules that contain cell survival genes in N0 tumors reinforces the important role that apoptosis plays in the regulation of metastasis. In addition, because tumor samples used here were not microdissected, tumor gene expression data are represented together with the stroma, which may reveal signaling between the microenvironment and tumor cells. For instance, in non-recurrent tumors, extracellular region module was repressed, indicating that the stroma and tumor cells may have fewer interactions, which disable metastasis development. Finally, the genes highlighted in our analysis can be implicated in more than one pathway or characteristic, suggesting that therapeutic approaches to prevent tumor progression should target more than one gene or pathway, specially apoptosis and interactions between tumor cells and the stroma.

A synthetic version of the metal-regulated gene A (mrgA) promoter from Bacillus subtilis, which in this Gram-positive bacterium is negatively regulated by manganese, iron, cobalt, or copper turned out to promote high level of basal gene expression that is further enhanced by Co(II), Cd(II), Mn(II), Zn(II), Cu(II), or Ni(II), when cloned in the Gram-negative bacterium Cupriavidus metallidurans. Promoter activity was monitored by expression of the reporter gene coding for the enhanced green fluorescent protein (EGFP), and cellular intensity fluorescence was quantified by flow cytometry. Expression levels in C. metallidurans driven by the heterologous promoter, here called pan, ranged from 20- to 53-fold the expression level driven by the Escherichia coli lac promoter (which is constitutively expressed in C. metallidurans), whether in the absence or presence of metal ions, respectively. The pan promoter did also function in E. coli in a constitutive pattern, regardless of the presence of Mn(II) or Fe(II). In conclusion, the pan promoter proved to be a powerful tool to express heterologous proteins in Gram-negative bacteria, especially in C. metallidurans grown upon high levels of toxic metals, with potential applications in bioremediation. The opportunistic pathogen Pseudomonas aeruginosa PA14 possesses four fimbrial cup clusters, which may confer the ability to adapt to different environments. cupD lies in the pathogenicity island PAPI-1 next to genes coding for a putative phosphorelay system composed of the hybrid histidine kinase RcsC and the response regulator RcsB. The main focus of this work was the regulation of cupD at the mRNA level. It was found that the HN-S-like protein MvaT does not exert a strong influence on cupD transcript levels, as it does for cupA. cupD transcription is higher in cultures grown at 28 degrees C, which agrees with a cupD mutant presenting attenuated virulence only in a plant model, but not in a mouse model of infection. Whereas an rcsC in-frame deletion mutant presented higher levels of cupD mRNA, rcsB deletion had the opposite effect. Accordingly, overexpression of RcsB increased the levels of cupD transcription, and promoted biofilm formation and the appearance of fimbriae. A single transcription start site was determined for cupD and transcription from this site was induced by RcsB. A motif similar to the enterobacterial RcsB/RcsA-binding site was detected adjacent to the -35 region, suggesting that this could be the RcsB-binding site. Comparison of P. aeruginosa and Escherichia coli Rcs may provide insights into how similar systems can be used by different bacteria to control gene expression and to adapt to various environmental conditions.

Este trabalho tem o objetivo de desenvolver e avaliar um Protótipo de um EIS (Executive Information System) que integre os níveis hierárquicos operacional, tático e estratégico, a fim de suprir e dar suporte, quanto a informações, a uma das Pró- Reitorias da Universidade de Caxias do Sul - UCS. Quer-se demonstrar não só a importância mas também a contribuição da integração, através do acesso e compartilhamento das informações, dos níveis hierárquicos, proporcionada pelo Protótipo, para a obtenção da eficiência e eficácia das atividades desenvolvidas na Pró-Reitoria em estudo. Através de revisão bibliográfica e de levantamentos efetuados por meio de questionários, elaborou-se o Protótipo EIS -UCS, o qual posteriormente foi avaliado através de dois protocolos, aplicados em momentos distintos, e também através da grade de análise do modelo conceitual de EIS. Os resultados obtidos demonstram o cumprimento do objetivo de desenvolver e avaliar o protótipo EIS-UCS. Os resultados apresentam o obtido em relação ao processo, ao sistema, ao conteúdo, a integração dos níveis hierárquicos e o aprendizado da organização e do pesquisador. Concluiu-se que o Protótipo EIS-UCS integra, através do acesso e compartilhamento das informações, os níveis hierárquicos e também supre e dá suporte, quanto ao nível de informações à Pró-Reitoria de Extensão e Relações Universitárias da Universidade de Caxias do Sul.

This study analyzed the perception of ethics on the part of students of Post-graduation in Tourism, to develop their academic research. It is characterized as exploratory-descriptive, being considered quantitative with some elements analyzed qualitatively. Data were collected through an electronic questionnaire, originated in google docs and sent via e-mail to 54 students in three graduate programs in Tourism: The Tourism Master of the Federal University of Rio Grande do Norte (UFRN), the Master in Tourism and Hospitality of the University of Vale do Itajai (UNIVALI) and Master of Tourism at the University of Caxias do Sul (UCS), resulting in 22 responses. The study presents a profile of the dissertation research of post-graduate students, their perception about research ethics, as well as the difficulties encountered in the research process. Notes that research of dissertation are geared mostly for the historical and cultural aspects of tourism and the choice of topics for the essays came in first place, due to the influence of the guiding lines of research and, secondly, due to social relevance. The methods used in post-graduate research are descriptive, the literature and exploratory. It also notes that most students graduate in tourism (81%) attaches great importance to ethics in carrying out their dissertation research, students claim that ethics brings credibility and seriousness to research. Furthermore, it was realized that they have no knowledge about the practice of misconduct by researchers in the field of Tourism. Demonstrate that the major ethical challenge encountered by students is in relationship with the research subject, what happens during data collection. It also notes that none of the respondents sent any project to be evaluated by an Ethics Committee (CEP), moreover, most do not know the resolution 196/96, which has the main guidelines on research ethics at national level. Concludes that the students post-graduate in tourism have little knowledge about the standards and guidelines on research ethics. They demonstrate ignore the benefits of research ethics in regarding the protection and preservation of the participant, as well as the benefits it generates for society

The current concern with environmental issues by the scientific community, governmental and civil society also brings out the need to recover, preserve and conserve the environment. With this understanding, this work was prepared, which had as one of the main objectives of the historical process of land use and occupancy of the Pontal, since the nineteenth century to the present day. From this, the changes were evident in the types of land use, which rose from native forest to plant crops (permanent and temporary crops) and later as pasture. Through this process of occupation, exploitation of forests and replacement even three, Reserves being brought from the Far West Paulista, through the state government of Sao Paulo, they were almost decimated. Such areas decreed by law to be preserved and conserved, with the rampant occupations, were suffocated by the fakes and illegal occupation of land tenure contracts, leaving the region currently only a few forest fragments and protected areas of State Park Morro do Diabo, PRNP Boa Vista and Ecological Station Black Lion Tamarin. Furthermore, the article emphasizes the main legislation being developed by federal and state levels aimed at protecting natural resources and the effectiveness they had with the passage of time, demonstrating that the conservation of the environment, has several contributions in the law, however, and they are so vast that hinder its application.

The ferric uptake regulator protein Fur regulates iron-dependent gene expression in bacteria. In the human pathogen Helicobacter pylori, Fur has been shown to regulate iron-induced and iron-repressed genes. Herein we investigate the molecular mechanisms that control this differential iron-responsive Fur regulation. Hydroxyl radical footprinting showed that Fur has different binding architectures, which characterize distinct operator typologies. On operators recognized with higher affinity by holo-Fur, the protein binds to a continuous AT-rich stretch of about 20 bp, displaying an extended protection pattern. This is indicative of protein wrapping around the DNA helix. DNA binding interference assays with the minor groove binding drug distamycin A, point out that the recognition of the holo-operators occurs through the minor groove of the DNA. By contrast, on the apo-operators, Fur binds primarily to thymine dimers within a newly identified TCATTn10TT consensus element, indicative of Fur binding to one side of the DNA, in the major groove of the double helix. Reconstitution of the TCATTn10TT motif within a holo-operator results in a feature binding swap from an holo-Fur- to an apo-Fur-recognized operator, affecting both affinity and binding architecture of Fur, and conferring apo-Fur repression features in vivo. Size exclusion chromatography indicated that Fur is a dimer in solution. However, in the presence of divalent metal ions the protein is able to multimerize. Accordingly, apo-Fur binds DNA as a dimer in gel shift assays, while in presence of iron, higher order complexes are formed. Stoichiometric Ferguson analysis indicates that these complexes correspond to one or two Fur tetramers, each bound to an operator element. Together these data suggest that the apo- and holo-Fur repression mechanisms apparently rely on two distinctive modes of operator-recognition, involving respectively the readout of a specific nucleotide consensus motif in the major groove for apo-operators, and the recognition of AT-rich stretches in the minor groove for holo-operators, whereas the iron-responsive binding affinity is controlled through metal-dependent shaping of the protein structure in order to match preferentially the major or the minor groove.

Eosinophils are white blood cells that function in innate immunity and participate in the pathogenesis of various inflammatory and neoplastic disorders. Their secretory granules contain four cytotoxic proteins, including the eosinophil major basic protein (MBP-1). How MBP-1 toxicity is controlled within the eosinophil itself and activated upon extracellular release is unknown. Here we show how intragranular MBP-1 nanocrystals restrain toxicity, enabling its safe storage, and characterize them with an X-ray-free electron laser. Following eosinophil activation, MBP-1 toxicity is triggered by granule acidification, followed by extracellular aggregation, which mediates the damage to pathogens and host cells. Larger non-toxic amyloid plaques are also present in tissues of eosinophilic patients in a feedback mechanism that likely limits tissue damage under pathological conditions of MBP-1 oversecretion. Our results suggest that MBP-1 aggregation is important for innate immunity and immunopathology mediated by eosinophils and clarify how its polymorphic self-association pathways regulate toxicity intra- and extracellularly.

The RNA binding proteins RBM binding motif protein 38 (RBM38) and DEAD END 1 (DND1) selectively stabilize mRNAs by attenuating RNAse activity or protecting them from micro(mi)RNA-mediated cleavage. Furthermore, both proteins can efficiently stabilize the mRNA of the cell cycle inhibitor p21(CIP1). Since acute myeloid leukemia (AML) differentiation requires cell cycle arrest and RBM38 as well as DND1 have antiproliferative functions, we hypothesized that decreased RBM38 and DND1 expression may contribute to the differentiation block seen in this disease. We first quantified RBM38 and DND1 mRNA expression in clinical AML patient samples and CD34(+) progenitor cells and mature granulocytes from healthy donors. We found significantly lower RBM38 and DND1 mRNA levels in AML blasts and CD34(+) progenitor cells as compared to mature neutrophils from healthy donors. Furthermore, the lowest expression of both RBM38 and DND1 mRNA correlated with t(8;21). In addition, neutrophil differentiation of CD34(+) cells in vitro with G-CSF (granulocyte colony stimulating factor) resulted in a significant increase of RBM38 and DND1 mRNA levels. Similarly, neutrophil differentiation of NB4 acute promyelocytic leukemia (APL) cells was associated with a significant induction of RBM38 and DND1 expression. To address the function of RBM38 and DND1 in neutrophil differentiation, we generated two independent NB4RBM38 as well as DND1 knockdown cell lines. Inhibition of both RBM38 and DND1 mRNA significantly attenuated NB4 differentiation and resulted in decreased p21(CIP1) mRNA expression. Our results clearly indicate that expression of the RNA binding proteins RBM38 and DND1 is repressed in primary AML patients, that neutrophil differentiation is dependent on increased expression of both proteins, and that these proteins have a critical role in regulating p21(CIP1) expression during APL differentiation.
