The polysaccharides extracted from Claviclonium ovatum were studied by a combination of compositional assays, reductive partial hydrolysis, linkage analysis, Fourier Transform infrared (FTIR) spectroscopy, and C-13, H-1, and C-13/H-1 heteronuclear multiple quantum correlation (HMQC) two-dimensional nuclear magnetic resonance (NMR) spectroscopy. The chemical and spectroscopic data showed that the alkali-modified C. ovatum polysaccharides are composed of a nearly idealized repeating unit of 6'-O-methylcarrabiose 2,4'-disulfate (the repeating unit of 6-O-methylated iota-earrageenan), although some minor components were also present. The C. ovatum galactans are the most highly methylated carrageenans reported. (C) 2004 Elsevier Ltd. All rights reserved.

In this work it is proposed a simple and versatile undergraduate chemical experiment in polymer and environmental technology based on the process of polyethylene terephthalate (PET) hydrolysis. Polyethylene terephthalate from post-consume bottles is submitted to a controlled partial hydrolysis which allows the students to follow the reaction by a simple procedure. The students can explore the reaction kinetics, the effect of catalysts and the exposed polyethylene terephthalate surface area on the hydrolysis reaction. The second and innovative part of this experiment is the technological and environmental application of the hydrolyzed polyethylene terephthalate as a material with cation exchange properties. The surface hydrolyzed polyethylene terephthalate can be used as adsorbent for cationic contaminants.

Iron, aluminium and mixed iron aluminium pillared clays have been prepared by partial hydrolysis method and doped with IO% Mo, V and Cr. The samples have been characterised by XRD, FTIR and surface area and pore Volume measurements. The surface acid site distribution has been determined by temperature programmed desorption of ammonia. Vanadia incorporated systems show maximum acidity. Benzylation of o-xylene has been done as probe reaction to test catalytic activity. Benzyl chloride is a superior benzylating agent compared to benzyl alcohol in activity and selectivity. Cent percent selectivity towards monobenzylated product is obtained in all the cases. Fe pillared systems exhibit maximum activity. The catalytic activities of the systems can be correlated with the amount of strong add sites. The effects of various reaction variables on the reaction have been studied. Presence of moisture has a diminishing effect on the reaction rate.

Iron and mixed iron aluminium pillared montmorillonites prepared by partial hydrolysis method was subjected to room temperature exchange with transition metals of the first series. The materials exhibit good structural as well as thermal stability. Exchanged metals were found to be present inside the porous network, in the environs of the pillars. Mixed pillaring resulted in the intercalation of Al 13 like polymers in which Al is partially substituted by Fe. The acidic structure was followed by temperature programmed desorption of ammonia and cumene cracking test reaction. Weak and medium sites overshadow the strong sites in all systems. However, exchange with metals increases the number of strong sites. The prepared materials are efficient catalysts for gas phase MTBE synthesis. The catalytic activity can be well correlated with the total amount of weak and medium acid sites.

Iron and mixed iron aluminium pillared montrnorillonites prepared by partial hydrolysis method were subjected to room temperature exchange with transition metals of the first series. The resulting materials were characterised by different spectroscopic techniques and surface area measurements. About 1-3% transition metals were incorporated into the porous network. The structural stability of the porous network was not affected by exchange. XRD and AI NMR spectroscopy evidenced the presence of iron substituted Al13 like polymers in FeAl pillared systems. Acidity and basicity benefited much as a result of metal exchange. Acidity and basicity were quantified by model reactions, viz., cumene cracking and cyclohexanol decomposition respectively. The presence of basic sites in otherwise acidic pillared clays, though diminutive in amount can be of much importance in acid base catalysed reactions.

Yacon (Smallanthus sonchifolius [Poepp. & Endl.] H. Robinson) is an under-exploited native root crop of the Andes, which stores oligofructans (fructo-oligosaccharides, FOS) as its main component of dry matter (DM). FOS are of increasing economic interest because of their low caloric value in human diets and bifidogenic benefits on colon health. Two on-farm experiments were conducted to: (i) determine the effect of shaded, short-term storage at 1990 and 2930 m a.s.l. in the Andean highlands; and (ii) address the effects of a traditional sunlight exposure (‘sunning’) on the carbohydrate composition in the DM of tuberous yacon roots. After a 6-day shade storage FOS concentrations were smaller at the lower (36–48% of DM) than at the higher altitude (39–58% of DM). After 12 days FOS concentrations were nearly equal at both sites (27–39% of DM). The concentration of free sugars (fructose, glucose, sucrose) increased accordingly from 29–34 to 48–52%. During the 6-day sunning experiment FOS concentrations decreased from 50–62 to 29–44% and free sugars increased from 29–34 to 45–51%. The results indicate that partial hydrolysis of oligofructans starts shortly after harvest. Storage in highland environments should wherever possible exploit the cooler temperatures at higher altitudes. Sunning of yacon’s tuberous roots effectively reduces much of the roots’ water content, in this experiment 40%, and thus allows energy to be saved if yacon is processed into dehydrated products.

The polysaccharide (VSP) from the gum exudate of quaruba (Vochysia lehmannii) had two components of almost identical M. centred at 24,800, as shown by HSPEC-MALLS. The presence of aggregates was shown since carboxy-reduction gave VSP-RED, which contained low molecular weight components with M-w 19,000 > 5800 and polydispersity ratios dn/dc 0.160 and 0.149, respectively. VSP formed low viscosity aqueous solutions and acid hydrolysis gave Man (30%), Ara (16%), Gal (10%), and Glc (14%). The latter arose partly from GlcA (30%). Methylation analysis revealed mainly neutral units of 2-O- (60%) and 2,3-di-O-substituted Manp (5%), and those of nomeducing ends (8%), 2-O- (3%), and 4-O-substituted Arap and/or 5-O-substituted Araf units (6%). VSP-RED contained Glc (45%), Man (35%), and Ara (13%) and methylation analysis indicated mainly 4-O-substituted Glcp (31%) and 2-O- (51%) and 2,3-di-O-substituted Manp units (5%). A predominant alternating structure for VSP was shown by its C-13 NMR spectrum, which contained 10 main signals and a small one of C-6 of GlcpA. This was confirmed by formation, on partial hydrolysis of VSP, of a tetrasaccharide, which was characterised by NMR spectroscopy and ESI-MS as beta-GlcpA-(1 --> 2)-alpha-Manp-(1 --> 4)-beta-GlcpA-(1 --> 2)-Man, which arose from the main chain, thus confirming VSP to be a glycoglucuronomannan. Cassava bagasse, the solid residue of the starch industry, was studied as a potential ingredient in high-fibre dietary products. The bagasse contained 80% of starch, 11.5% fibre, 1.14% ash and 0.85% of protein, and it had a pH of 5.8. Glucose was the major sugar and the hemicellulose content was 22.31%. Microscopic analysis of the bagasse revealed a large number of starch granules inside the parenchyma cells and fibres, as well as outside the cells. The bagasse may be used in high-fibre products, and total or partial hydrolysis of its starch content might increase its commercial value.

The plant cell wall is composed mainly of polysaccharides some constituted of repeating units of a single sugar, as cellulose or by two or more sugars grouped in repeating oligosaccharide blocks as the galactomannans and xyloglucans. Variations in composition and fine structure of these cell wall polysaccharides have been used as taxonomic markers and in the comprehension of the evolutive process, particularly in the Leguminosae. Partial hydrolysis of these compounds give rise to oligomers, some of which are capable of eliciting the synthesis of defensive substances in plants named phytoalexins. Species which differ in respect to phytoalexin liberation also differ in cell wall composition, particularly in the pectic fraction of the wall. Pectinases (mainly endopolygalacturonases) present in fungi, have been shown to hydrolyze plant cell walls yielding phytoalexin-eliciting oligosaccharides which differ in composition and in eliciting capacity in different species. These differences can be associated with the capacity of a given species to produce phytoalexins. On the other hand, the phytoalexin induction in plants is being used as a method of producing novel bioactive secondary metabolites.

Metabolic studies are very important to improve quality of functional dairy products. For this purpose, the behaviors of pure cultures of Streptococcus thermophilus (St) and Lactobacillus rhamnosus (Lr) as well a co-culture of them (St-Lr) were investigated during skim milk fermentation, and the inulin effect as prebiotic was assessed. Lr was able to metabolize 6 g/100 g more galactose than St and St-Lr. Final lactic acid production by Lr was higher (9.8 g/L) compared to St (9.1 g/L) and St-Lr (9.1 g/L). Acetic acid concentration varied from 0.8 g/L (St-Lr) to 1.5 g/L (Lr) and that of ethanol from only 0.2 g/L (St-Lr) to 0.4 g/L (Lr), which suggests the occurrence in Lr of a NADH oxidase activity and citrate co-metabolization via pyruvate, both dissipating a part of the reducing power. Diacetyl and acetoin accumulated at the highest levels (18.4 and 0.8 mg/L, respectively) with St-Lr, which suggests possible synergistic interactions between these microorganisms as well as the Lr capability of co-metabolizing citrate in the presence of lactose. Inulin stimulated both biomass growth and levels of all end-products, as the likely result of fructose release from its partial hydrolysis and subsequent metabolization as an additional carbon and energy source. N-Vinylamidderivate sind eine toxikologische unbedenkliche Monomerklasse. Mit diesen Monomeren wurden verschiedene technische Anwendungsgebiete im Bereich der Kern-Schale-Partikel und der fließfähigen und vernetzten Hydrogele untersucht. Kern-Schale-Partikel Für die Synthese von Kern-Schale-Partikeln wurden die N-Vinylamidderivate als Schalenpolymere auf kommerziellen Poly(styrol-stat.-butadien)-Kernpartikeln aufpolymerisiert. Mit Hilfe verschiedener Untersuchungsmethoden (DLS, SEM, FFF, Ultrazentrifuge) wurde die Kern-Schale-Strukturbildung und die Effizienz der Pfropfungsreaktion untersucht und eine erfolgreiche Synthese der Kern-Schale-Partikel belegt. Durch die gezielte Modifizierung des Schalenpolymers wurde ein kationisches, organisches Mikropartikelsystem entwickelt, charakterisiert und auf die Eignung als „Duales Flockungsmittel“ untersucht. Diese Versuche belegten die Eignung der modifizierten Kern-Schale-Partikel als „Duales Flockungsmittel“ und bieten eine Alternative zu kommerziell verwendeten Retentionsmitteln. Außerdem wurden die filmbildenden Eigenschaften der Poly(N﷓vinylformamid)-Kern-Schale-Dispersionen untersucht. Nach der Verfilmung der Dispersionen wurden transparente und harte Filme erhalten. Die Auswirkungen auf die mechanischen Eigenschaften der Filme wurden durch die Variation verschiedener Parameter eingehend studiert. Auf der Basis dieser Partikel wurden selbstvernetzende Dispersionssysteme entwickelt. Das P﷓(VFA)-Schalenpolymer wurde teilweise hydrolysiert und die generierten freien Aminogruppen des Poly(N-vinylamins) durch eine Michael-Addition mit einem divinylfunktionalisierten Acrylat (Tetraethylenglykoldiacrylat) vernetzt. Untersuchungen zur mechanischen Beständigkeit der Filme zeigten bei geringen Vernetzungsgraden eine deutliche Optimierung der maximalen Zugbelastungen. Die Untersuchungen belegten, dass die Verwendung des selbstvernetzenden Dispersionssystems als Dispersion für eine Polymerbeschichtung möglich ist. Hydrogele Die Synthese von fließfähigen und quervernetzten Hydrogelen erfolgte auf der Basis verschiedener N﷓Vinylamide. Mit Hilfe geeigneter Vernetzer wurden feste Hydrogelplatten synthetisiert und für die Auftrennung von DNA-Sequenzen mit Hilfe der Gelelektrophorese verwendet. Scharfe und gute Auftrennung der verschiedenen „DNA-Ladder Standards“ wurden durch die Variation des Vernetzeranteils, der Polymerzusammensetzung, der angelegten Spannung und der Verweilzeit in der Gelelektrophoresekammer mit P﷓(MNVA)-Hydrogelplatten erreicht. Fließfähige und quervernetzte Elektrolytgele auf Poly-(N-vinylamid)-Basis wurden in wartungsfreien pH﷓Elektroden eingesetzt. Die Eignung dieser Hydrogele wurden in Bezug auf die Anwendung eingehend charakterisiert. Elektroden befüllt mit Poly(N-vinylamid)-Gelen wurden in Dauerbelastungsexperimenten, direkt mit kommerziellen pH﷓Elektroden verglichen. Es konnte gezeigt werden, dass die fließfähigen und quervernetzten Poly-(N-vinylamid)-Elektrolytgele in ihren Messeigenschaften gleichwertige bzw. bessere Potentialstabilitäten aufweisen als kommerzielle Referenzelektroden. Die Hydrogele auf Basis von Poly(N-vinylamidderivaten) boten für die beiden getesteten Anwendungen eine toxikologisch unbedenkliche Alternative zu Poly(acrylamid)-Gelen. In dieser Arbeit konnten die durchgeführten Untersuchungen belegen, dass N﷓Vinylamide eine attraktive Monomerklasse ist, die erfolgreich in vielen technischen Anwendungen einsetzbar ist.

Baeyer–Villiger oxidation of cyclic ketones, using H2O2 as the oxidising agent, was systematically studied using a range of metal chlorides in different solvents, and in neat chlorogallate(III) ionic liquids. The extremely high activity of GaCl3 in promoting oxidation with H2O2, irrespective of solvent, was reported for the first time. The activity of all other metal chlorides was strongly solvent-dependent. In particular, AlCl3 was very active in a protic solvent (ethanol), and tin chlorides, SnCl4 and SnCl2, were active in aprotic solvents (toluene and dioxane). In order to eliminate the need for volatile organic solvent, a Lewis acidic chlorogallate(III) ionic liquid was used in the place of GaCl3, which afforded typically 89–94% yields of lactones in 1–120 min, at ambient conditions. Raman and 71Ga NMR spectroscopic studies suggest that the active species, in both GaCl3 and chlorogallate(III) ionic liquid systems, are chlorohydroxygallate(III) anions, [GaCl3OH]−, which are the products of partial hydrolysis of GaCl3 and chlorogallate(III) anions; therefore, the presence of water is crucial.

An extracellular polygalacturonase (PG) produced from Paecilomyces variotii was purified to homogeneity through two chromatography steps using DEAE-Fractogel and Sephadex G-100. The molecular weight of P. variotii PG was 77,300 Da by gel filtration and SDS-PAGE. PG had isoelectric point of 4.37 and optimum pH 4.0. PG was very stable from pH 3.0 to 6.0. The extent of hydrolysis of different pectins by the purified enzyme was decreased with an increase in the degree of esterification. PG had no activity toward non-pectic polysaccharides. The apparent K (m) and V (max) values for hydrolyzing sodium polypectate were 1.84 mg/mL and 432 A mu mol/min/mg, respectively. PG was found to have temperature optimum at 65 A degrees C and was totally stable at 45 A degrees C for 90 min. Half-life at 55 A degrees C was 50.6 min. Almost all the examined metal cations showed partial inhibitory effects under enzymatic activity, except for Na(+1), K(+1), and Co(+2) (1 mM) and Cu(+2) (1 and 10 mM).

Treatment of Aspergillus niveus with 30 mu g tunicamycin/ml did not interfere with alpha-glucosidase production, secretion, or its catalytic properties. Fully- and under-glycosylated forms of the enzyme had similar molecular masses, similar to 56 kDa. Moreover, the absence of N-glycans did not affect either pH optimum (6.0) or temperature optimum (65A degrees C). The K(m) and V(max) values of under- and fully-glycosylated forms of alpha-glucosidase were similar when assessed for hydrolysis of starch (similar to 0.6 mg/ml, similar to 350 mu mol glucose per min per ml), maltose (similar to 0.54 mu mol, similar to 330 mu mol glucose per min per ml) and p-nitrophenyl-alpha-d-glucopyranoside (similar to 0.54 mu mol, similar to 8.28 mu mol p-nitrophenol per min per ml). However, the under-glycosylated form was sensitive to high temperatures probably because, in addition to stabilizing the protein conformation, glycosylation may also prevent unfolded or partially folded proteins from aggregating. Binding assays clearly showed that the under-glycosylated protein did not bind to concanavalin A but has conserve its jacalin-binding property, suggesting that only O-glycans might be intact on the tunicamycin treated form of the enzyme.
