This work investigated the purification of phosphoric acid using a suitable organic solvent, followed by re-extraction of the acid from the solvent using water. The work consisted of practical batch and continuous studies and the economics and design of a full scale plant, based on the experimental data. A comprehensive literature survey on the purification of wet process phosphoric acid by organic solvents is presented and the literature describing the design and operation of mixer-settlers has also been reviewed. In batch studies, the equilibrium and distribution curves for the systems water-phosphoric acid-solvent for Benzaldehyde, Cyclohexanol and Methylisobutylketone (MIBK) were determined together with hydrodynamic characteristics for both pure and impure systems. The settling time increased with acid concentration, but power input had no effect. Drop size was found to reduce with acid concentration and power input. For the continuous studies a novel horizontal mixer~settler cascade was designed, constructed and operated using pure and impure acid with MIBK as the solvent. The cascade incorporates three air turbine agitated, cylindrical 900 ml mixers, and three cylindrical 200 ml settlers with air-lift solvent interstage transfer. Mean drop size in the fully baffled mixer was correlated. Drop size distributions were log-normal and size decreased with acid concentration and power input and increased with dispersed phase hold-up. Phase inversion studies showed that the width of the ambivalent region depended upon rotor speed, hold-up and acid concentration. Settler characteristics were investigated by measuring wedge length. Distribution coefficients of impurities and acid were also investigated. The following optimum extraction conditions were found: initial acid concentration 63%, phase ratio of solvent to acid 1:1 (v/v), impeller speed recommended 900 r.p.m. In the washing step the maximum phase ratio of solvent to water was 8:1 (v/v). Work on phosphoric acid concentration involved constructing distillation equipment consisting of a 10& spherical still. A 100 T/d scale detailed process design including capital cost, operating cost and profitability was also completed. A profit model for phosphoric acid extraction was developed and maximised. Recommendations are made for both the application of the results to a practical design and for extensions of the study.

The tanning industries are those which transform animal hide or skin into leather. Due to the complexity of the transformation process, greater quantities of chemicals are being used which results in the generation of effluents with residual solids. The chromium in the residual waters generated by tanning tend to be a serious problem to the environment, therefore the recovery of this metal could result in the reduction of manufacturing costs. This metal is usually found in a trivalent form which can be converted into a hexavalent compound under acidic conditions and in the presence of organic matter. The present study was carried out with the objective to recover chromium through an extraction/re-extraction process using micro emulsions. Micro emulsions are transparent and thermodynamically stable system composed of two immiscible liquids, one forming the continuous phase and the other dispersed into micro bubbles, established by an interfacial membrane formed by surface active and co-surface active molecules. The process of recovering the chromium was carried out in two stages. The first, an extraction process, where the chromium was extracted in the micro emulsion phase and the aqueous phase in excess was separated. In the second stage, a concentrated acid was added to the micro emulsion phase rich in chromium in order to obtain a Winsor II system, where the water that formed in the micro emulsion phase separates into a new micro emulsion phase with a higher concentration of chromium, due to the lowering of the hydrophiles as well as the ionisation of the system. During the experimental procedure, a study was initiated with a synthetic solution of chromium sulphate passing onto the effluent. A Morris extractor was used in the extraction process. Tests were carried out according to the plan and the results were analysed by statistical methods in order to optimise the main parameters that influence the process: the total rate of flow (Q), stirring speed (w) and solvent rate (r). The results, after optimization, demonstrated that the best percentuals in relation to the chromium extraction (99 %) were obtained in the following operational conditions: Q= 2,0 l/h, w= 425 rpm and r= 0,375. The re-extraction was carried out at room temperature (28 °C), 40 °C and 50°C using hydrochloric acid (8 and 10 M) and sulphuric acid (8 M) as re-extracting agents. The results obtained demonstrate that the process was efficient enough in relation to the chromium extraction, reaching to re-extraction percentage higher than 95 %.

The efficiency of methods for the determination of hexachlorobenzene (HCB) and pentachlorophenol (PCP) in soil samples was evaluated. An on-line method was applied for HCB determination. Soil samples were transferred to chromatographic columns prepacked with alumina. The HCB elution was processed with n-hexane. The PCP was extracted from soil samples with n-hexane-acetone in an ultrasonic bath. After re-extraction with K2CO3 solution PCP was acetylated with acetic anhydride. The pentachlorophenyl acetate derivative was then extracted with n-hexane. The HCB and PCP derivative were analyzed by gas chromatography with electron capture detection (GC-ECD). Mean recoveries obtained from soil samples fortified at levels of 0.5; 4 and 20 ng g(-1) ranged from 91 to 100% for HCB, and for PCP, at levels of 10; 40 and 200 ng g(-1), ranged from 88 to 101%. Parahancornia fasciculata (Poir.) Benoist (Apocynaceae), também conhecida como Parahancornia amapa (Hub.) Ducke, é uma espécie vegetal empregada popularmente no tratamento da malária, infecções no útero, gastrite, anemia, problemas respiratórios, entre outros. Os objetivos do presente trabalho foram realizar o estudo fitoquímico, avaliar a toxicidade oral aguda e a atividade antimalárica in vitro e in vivo de extratos, frações e substância isolada obtidas a partir de cascas do caule de P. fasciculata. Foram realizados dois tipos de extrações com o pó das cascas de P. fasciculata, por maceração / percolação, com etanol 96°GL e diclorometano, esta última tendo sido realizada a com o pó das cascas alcalinizado com hidróxido de amônio, obtendo-se os extratos secos EEPF e EDAPF, respectivamente. Uma terceira extração foi realizada a partir do EEPF por aquecimento sob refluxo, sucessivamente, com Hex:DCM (1:1), AcOEt:DCM (1:1) e AcOEt. EEPF foi, também, submetido a fracionamento por extrações ácido-base resultando nas frações de neutros (EEPFN) e de alcalóides (EEPFA). A prospecção fitoquímica realizada com o EEPF foi desenvolvida por CCD em cromatoplacas de sílica gel tendo sido detectada a presença de triterpenos, esteróides, heterosídeos flavônicos, saponinas, polifenóis, taninos, heterosídeos antracênicos e heterosídeos cardiotônicos. EDAPF foi submetido à cromatografia em coluna de sílica gel. Foram recolhidas 30 frações sendo que as frações Fr1-3, Fr4, Fr5-7 e Fr11 concentraram a maior parte da massa do extrato cromatografado. Da Fr5-7 foi isolada uma mistura de ésteres do lupeol que representam os componentes majoritários do EDAPF. Esta fração passou por um processo de hidrólise alcalina e o produto obtido (Fr5-7Hid) foi analisado por espectrometrias no IV, RMN de 1H e 13C e foi identificado como o triterpeno lupeol. A fração insolúvel em AcOEt obtida a partir do EEPF, por aquecimento sob refluxo, apresentou resultado positivo para o teste de proantocianidinas e foi submetido a doseamento desta classe de metabólitos. Os resultados foram expressos em porcentagem dos teores para a amostra não diluída (10,46±0,3419%), amostra diluída a 1:10 (9,94± 0,1598%) e amostra diluída a 1:100 (10,55± 0,9299%). A avaliação da atividade antiplasmódica in vitro em culturas de cepas W2 de Plasmodium falciparum foi realizada pelo teste da Proteína II Rica em Histidina (HRP-II) tendo sido testados EEPF, EEPFN, EEPFA, Fr1-3, Fr4, Fr5-7(ésteres do lupeol), Fr11 e o Fr5-7Hid (lupeol). Os melhores resultados obtidos foram para EEPF, EEPFA E EEPFN (CI50= ~ 50 μg/mL) sendo considerados moderadamente ativos. As demais amostras apresentaram CI50 > 50 μg/mL e foram consideradas inativas. Realizou-se também a avaliação da atividade antimalárica in vivo em camundongos fêmeas suíços infectados com cepas ANKA de P. berghei com o EEPF e o EEPF-HEX:DCM (1:1) em concentrações de 500, 250 e 125mg/kg de peso. EEPF foi parcialmente ativo, somente no 8° dia, em todas as concentrações. Já EEPF-HEX:DCM (1:1) foi parcialmente ativo na dose de 500mg/kg de peso e nas demais doses foi inativo. O teste de toxicidade oral aguda foi realizado em camundongos fêmeas suíços, pelo método da dose fixa (5.000mg/kg), com EEPF e não apresentou nenhum sinal de toxicidade evidente, o que foi confirmado pela ausência de alterações nos exames anátomohistopatológicos realizados.

O presente trabalho descreve a análise fitoquímica e avaliação da atividade antiplasmódica em Plasmodium falciparum (W2) de Himatanthus articulatus (Vahl) Woodson. O extrato etanólico (EEHS) obtido por percolação do pó das cascas, após concentração, forneceu um precipitado (EEHSP) e um resíduo pastoso que foi submetido a liofilização (EEHS). Fracionou-se este por: re-extração sob refluxo, partição ácido-base e coluna cromatográfica de sílica gel. Além disso, submeteu-se o pó das cascas a extração com ácido clorídrico 1 N para separação de alcalóides. A prospecção fitoquímica, em CCD, foi realizada com EEHS e EEHSP, enquanto que EEHS, frações (FrDCM EEHS, FrAcOET EEHS e FrMeOH EEHS) e FAHS2 DCM foram analisadas por cromatografia líquida de alta eficiência acoplada a DAD (arranjo de diiodo-CLAE-DAD). FrAcOET EEHS foi fracioanda por coluna cromatográfica fornecendo uma substância isolada (S1). Analisou-se o EEHS, a fração majoritária obtida na coluna cromatográfica (F71), FAHS2 DCM e S1 por cromatografia liquida acoplada a espectrometria de massas (CL-EM). O espectro no infravermelho foi obtido para F71 e S1. A estrutura química de S1 foi definida como sendo o plumierídeo por ressonância magnética nuclear. A avaliação de atividade antiplasmódica foi realizada com EEHS, EEHSP, FrDCM EEHS, FrAcOET EEHS, FrMeOH EEHS, FAHS1 DCM, FAHS2 DCM, FAEEHS, FNEEHS, FNHS DCM e S1 pelo método da lactato desidrogenase parasitária (pLDH). Na prospecção fitoquímica, EEHS e EEHSP apresentaram resultados positivos para polifenóis e taninos, saponinas, triterpenos e esteroides, alcaloides e geninas flavônicas. EEHS e frações analisadas por CLAE-DAD mostraram os picos mais intensos sugestivos de iridoides, comprovados por CL-EM de EEHS que mostrou como substância majoritária o iridóide plumierídeo, coerente com o espectro no IV de F71 e S1 que revelou absorções de grupos funcionais presentes em iridóides. Nas análises destas por CL-EM, observou-se um pico a m/z 471 (M+H), atribuído ao íon pseudomolecular do plumierídeo e/ou isoplumierídeo. S1 foi identificada como plumierídeo. Até o presente momento não foi relatada a presença de alcaloides para a espécie, porém nas frações alcaloídicas (FAHS1 DCM, FAHS2 DCM, FAEEHS) e de neutros (FNEEHS e FNHS DCM) em análise por CCD revelada com reagente Dragendorff observaram-se manchas sugestivas de alcalóides, coerente com análise por CLAE-DAD de FAHS2 DCM, a qual mostrou pico majoritário com cromóforo sugestivo de alcaloide β-carbolínico. Em análises por CL-EM de FAHS2 DCM observou-se pico majoritário sugestivo do alcalóide 10-hidroxi-antirina-N-óxido com massa molecular 328 u. Os ensaios de atividade antiplasmódica foram negativos para EEHS, EEHSP, FrDCM EEHS, FrAcOET EEHS, FrMeOH EEHS, S1, FAEEHS, FAHS1 DCM, FNEEHS, FNHS DCM e moderadamente ativo (CI50 22,89 μg/mL) para FAHS2 DCM. Estes resultados indicam que a atividade antiplasmódica da planta pode se atribuída aos alcaloides, cuja presença em H. articulatus é descrita pela primeira vez.

Generalizing the notion of an eigenvector, invariant subspaces are frequently used in the context of linear eigenvalue problems, leading to conceptually elegant and numerically stable formulations in applications that require the computation of several eigenvalues and/or eigenvectors. Similar benefits can be expected for polynomial eigenvalue problems, for which the concept of an invariant subspace needs to be replaced by the concept of an invariant pair. Little has been known so far about numerical aspects of such invariant pairs. The aim of this paper is to fill this gap. The behavior of invariant pairs under perturbations of the matrix polynomial is studied and a first-order perturbation expansion is given. From a computational point of view, we investigate how to best extract invariant pairs from a linearization of the matrix polynomial. Moreover, we describe efficient refinement procedures directly based on the polynomial formulation. Numerical experiments with matrix polynomials from a number of applications demonstrate the effectiveness of our extraction and refinement procedures.

BACKGROUND: Tumor levels of steroid hormone receptors, a factor used to select adjuvant treatment for early-stage breast cancer, are currently determined with immunohistochemical assays. These assays have a discordance of 10%-30% with previously used extraction assays. We assessed the concordance and predictive value of hormone receptor status as determined by immunohistochemical and extraction assays on specimens from International Breast Cancer Study Group Trials VIII and IX. These trials predominantly used extraction assays and compared adjuvant chemoendocrine therapy with endocrine therapy alone among pre- and postmenopausal patients with lymph node-negative breast cancer. Trial conclusions were that combination therapy provided a benefit to pre- and postmenopausal patients with estrogen receptor (ER)-negative tumors but not to ER-positive postmenopausal patients. ER-positive premenopausal patients required further study. METHODS: Tumor specimens from 571 premenopausal and 976 postmenopausal patients on which extraction assays had determined ER and progesterone receptor (PgR) levels before randomization from October 1, 1988, through October 1, 1999, were re-evaluated with an immunohistochemical assay in a central pathology laboratory. The endpoint was disease-free survival. Hazard ratios of recurrence or death for treatment comparisons were estimated with Cox proportional hazards regression models, and discriminatory ability was evaluated with the c index. All statistical tests were two-sided. RESULTS: Concordance of hormone receptor status determined by both assays ranged from 74% (kappa = 0.48) for PgR among postmenopausal patients to 88% (kappa = 0.66) for ER in postmenopausal patients. Hazard ratio estimates were similar for the association between disease-free survival and ER status (among all patients) or PgR status (among postmenopausal patients) as determined by the two methods. However, among premenopausal patients treated with endocrine therapy alone, the discriminatory ability of PgR status as determined by immunohistochemical assay was statistically significantly better (c index = 0.60 versus 0.51; P = .003) than that determined by extraction assay, and so immunohistochemically determined PgR status could predict disease-free survival. CONCLUSIONS: Trial conclusions in which ER status (for all patients) or PgR status (for postmenopausal patients) was determined by immunohistochemical assay supported those determined by extraction assays. However, among premenopausal patients, trial conclusions drawn from PgR status differed--immunohistochemically determined PgR status could predict response to endocrine therapy, unlike that determined by the extraction assay.

AIM: To assess dimensional ridge alterations following immediate implant placement in molar extraction sites. MATERIAL AND METHODS: Twelve subjects received 12 immediate transmucosal implants in molar extraction sites. Peri-implant defects were treated according to the principles of Guided Bone Regeneration by means of a deproteinized bone substitute and a bioresorbable collagen membrane. Changes in vertical (IS-BD, CREST-BD) and horizontal distances (EC-I, IC-I) of alveolar bony walls to the bottom of the defects (BD) and to the implant surfaces (I) were compared between implant placement and surgical re-entry at 6 months. RESULTS: The implant survival rate at 6 months was 100%. Statistically significant differences (P<0.01) were observed in the mean changes in vertical distances IS-BD and CREST-BD between baseline and re-entry. At re-entry, all peri-implant marginal defects assessed from the internal socket wall to the implant surface (IC-I) were healed. The residual combined thickness of the buccal wall with the newly formed peri-implant bone at sites with an initial thickness of 1 mm was statistically significantly smaller (P<0.05) compared with that of sites with an initial buccal thickness of 2 mm (2.50 +/- 0.76 vs. 4+/-0 mm). CONCLUSIONS: The marginal defects around immediate implants placed in molar extraction sites were completely filled after 6 months of healing through de novo bone formation. Bone resorption was observed from the external aspects of the buccal and oral socket walls. Dimensional changes of the external socket walls were mostly pronounced at the buccal aspects.

In this study we demonstrate a new in-fermenter chemical extraction procedure that degrades the cell wall of Escherichia coli and releases inclusion bodies (IBs) into the fermentation medium. We then prove that cross-flow microfiltration can be used to remove 91% of soluble contaminants from the released IBs. The extraction protocol, based on a combination of Triton X-100, EDTA, and intracellular T7 lysozyme, effectively released most of the intracellular soluble content without solubilising the IBs. Cross-flow microfiltration using a 0.2 mum ceramic membrane successfully recovered the granulocyte macrophagecolony stimulating factor (GM-CSF) IBs with removal of 91% of the soluble contaminants and virtually no loss of IBs to the permeate. The filtration efficiency, in terms of both flux and transmission, was significantly enhanced by infermenter Benzonase(R) digestion of nucleic acids following chemical extraction. Both the extraction and filtration methods exerted their efficacy directly on a crude fermentation broth, eliminating the need for cell recovery and re-suspension in buffer. The processes demonstrated here can all be performed using just a fermenter and a single cross-flow filtration unit, demonstrating a high level of process intensification. Furthermore, there is considerable scope to also use the microfiltration system to subsequently solubilise the IBs, to separate the denatured protein from cell debris, and to refold the protein using diafiltration. In this way refolded protein can potentially be obtained, in a relatively pure state, using only two unit operations. (C) 2004 Wiley Periodicals Inc.

Linear unmixing decomposes an hyperspectral image into a collection of re ectance spectra, called endmember signatures, and a set corresponding abundance fractions from the respective spatial coverage. This paper introduces vertex component analysis, an unsupervised algorithm to unmix linear mixtures of hyperpsectral data. VCA exploits the fact that endmembers occupy vertices of a simplex, and assumes the presence of pure pixels in data. VCA performance is illustrated using simulated and real data. VCA competes with state-of-the-art methods with much lower computational complexity.

In recent years a set of production paradigms were proposed in order to capacitate manufacturers to meet the new market requirements, such as the shift in demand for highly customized products resulting in a shorter product life cycle, rather than the traditional mass production standardized consumables. These new paradigms advocate solutions capable of facing these requirements, empowering manufacturing systems with a high capacity to adapt along with elevated flexibility and robustness in order to deal with disturbances, like unexpected orders or malfunctions. Evolvable Production Systems propose a solution based on the usage of modularity and self-organization with a fine granularity level, supporting pluggability and in this way allowing companies to add and/or remove components during execution without any extra re-programming effort. However, current monitoring software was not designed to fully support these characteristics, being commonly based on centralized SCADA systems, incapable of re-adapting during execution to the unexpected plugging/unplugging of devices nor changes in the entire system’s topology. Considering these aspects, the work developed for this thesis encompasses a fully distributed agent-based architecture, capable of performing knowledge extraction at different levels of abstraction without sacrificing the capacity to add and/or remove monitoring entities, responsible for data extraction and analysis, during runtime.

In this paper we present a description of the role of definitional verbal patterns for the extraction of semantic relations. Several studies show that semantic relations can be extracted from analytic definitions contained in machine-readable dictionaries (MRDs). In addition, definitions found in specialised texts are a good starting point to search for different types of definitions where other semantic relations occur. The extraction of definitional knowledge from specialised corpora represents another interesting approach for the extraction of semantic relations. Here, we present a descriptive analysis of definitional verbal patterns in Spanish and the first steps towards the development of a system for the automatic extraction of definitional knowledge.

Factors affecting the detennination of PAHs by capillary GC/MS were studied. The effect of the initial column temperature and the injection solvent on the peak areas and heights of sixteen PAHs, considered as priority pollutants, USillg crosslinked methyl silicone (DB!) and 5% diphenyl, 94% dimethyl, 1% vinyl polysiloxane (DBS) columns was examined. The possibility of using high boiling point alcohols especially butanol, pentanol, cyclopentanol, and hexanol as injection solvents was investigated. Studies were carried out to optimize the initial column temperature for each of the alcohols. It was found that the optimum initial column temperature is dependent on the solvent employed. The peak areas and heights of the PAHs are enhanced when the initial column temperature is 10-20 c above the boiling point of the solvent using DB5 column, and the same or 10 C above the boiling point of the solvent using DB1 column. Comparing the peak signals of the PAHs using the alcohols, p-xylene, n-octane, and nonane as injection solvents, hexanol gave the greatest peak areas and heights of the PAHs particularly the late-eluted peaks. The detection limits were at low pg levels, ranging from 6.0 pg for fluorene t9 83.6 pg for benzo(a)pyrene. The effect of the initial column temperature on the peak shape and the separation efficiency of the PARs was also studied using DB1 and DB5 columns. Fronting or splitting of the peaks was obseIVed at very low initial column temperature. When high initial column temperature was used, tailing of the peaks appeared. Great difference between DB! and.DB5 columns in the range of the initial column temperature in which symmetrical.peaks of PAHs can be obtained is observed. Wider ranges were shown using DB5 column. Resolution of the closely-eluted PAHs was also affected by the initial column temperature depending on the stationary phase employed. In the case of DB5, only the earlyeluted PAHs were affected; whereas, with DB1, all PAHs were affected. An analytical procedure utilizing solid phase extraction with bonded phase silica (C8) cartridges combined with GC/MS was developed to analyze PAHs in water as an alternative method to those based on the extraction with organic solvent. This simple procedure involved passing a 50 ml of spiked water sample through C8 bonded phase silica cartridges at 10 ml/min, dried by passing a gentle flow of nitrogen at 20 ml/min for 30 sec, and eluting the trapped PAHs with 500 Jll of p-xylene at 0.3 ml/min. The recoveries of PAHs were greater than 80%, with less than 10% relative standard deviations of nine determinations. No major contaminants were present that could interfere with the recognition of PAHs. It was also found that these bonded phase silica cartridges can be re-used for the extraction of PAHs from water.
