Colleters are widely occurring in eudicots showing relevant taxonomic importance in several families. Nevertheless, there are few records in monocots, restricted to only one description of these glands in Orchidaceae. The genus Oncidium is polyphyletic, currently the subject of taxonomic studies. In this context, the secretory structures can be an important diagnostic character that may help in the delineation of this group. O. flexuosum Sims presents colleters in vegetative - leaf primordium of protocorms, apical and axillary buds in the mature rhizomes - and reproductive organs - at the base of bracts, bracteoles and sepals. All the colleters observed are finger-like trichomes, composed of two uniseriated cells, where the apical one is elongated and possesses dense cytoplasm. The exsudate accumulates in a subcuticular space. causing displacement of the cuticle. Histochemical tests indicate the presence of mucilage in association with lipophilic and proteinic compounds inside the secretory cell. Secretion is abundant, hyaline and slightly viscous. The localization of the trichomes and their exsudate indicate the involvement of these colleters with the protection of meristematic regions in vegetative and reproductive organs. These results can be useful in the taxonomy of the genus Oncidium and for future studies about colleters in monocots. (C) 2010 Elsevier GmbH. All rights reserved.

The aim of this study was to evaluate the in vitro growth of Cattleya loddigesii in alternative agents to agar with starch and physical matrix with acclimatization of regenerated plants. Protocorms with 90 days after sowing (0.5 cm of length) were subcultured in 1/2 MS culture medium among the treatments consisting of: agar 7 g L-1 (T1, which corresponds the control), agar 3,5 g L-1 with cassava starch 30 g L-1 (T2), cassava starch 60 g L-1 (T3), cotton fiber (T4) and chopped polyurethane foam (T5). Plantlets were retained in these treatments for over 150 days, and at the end of in vitro culture, were analyzed by their biometric data and acclimatized in a greenhouse during 120 days and evaluated the survival and relative growth rate (RGR). The substrate comprising of chopped polyurethane foam (T5) showed greater efficiency for growth in vitro and also increased survival rate, while substrate cassava starch (T3) provided delay for plantlet growth. Therefore, chopped polyurethane foam is recommended because of low cost and suitable characteristics for the propagation of Cattleya loddigesii.

Dendrobium is a large genus in the family Orchidaceae that exhibits vast diversity in floral characteristics, which is of considerable importance to orchid breeders, biotechnologists and collectors. Native species have high value as a result of their medicinal properties, while their hybrids are important as ornamental commodities, either as cut flowers or potted plants and are thus veritable industrial crops. Thus, preservation of Dendrobium germplasm is valuable for species conservation, breeding programs and the floriculture industry. Cryopreservation represents the only safe, efficient and cost-effective long-term storage option to facilitate the conservation of genetic resources of plant species. This review highlights 16 years of literature related to the preservation of Dendrobium germplasm and comprises the most comprehensive assessment of thorough studies performed to date, which shows reliable and reproducible results. Air-drying, encapsulation-dehydration, encapsulation-vitrification, vitrification and droplet-vitrification are the current cryopreservation methodologies that have been used to cryopreserve Dendrobium germplasm. Mature seeds, pollen, protoplasts, shoot primordia, protocorms and somatic embryos or protocorm-like bodies (PLBs) have been cryopreserved with different levels of success. Encapsulation-vitrification and encapsulation-dehydration are the most used protocol, while PLBs represent the main explant explored.
