Yhteiskunnan globaalistumisen myötä johtamiselle on kasautunut huomattava määrä paineita. Kiireen ja kovan kilpailun keskellä työntekijöitä on motivoitava mahdollisimman tehokkaaseen työskentelyyn. Johtamisessa on siirrytty ihmisten johtamisen suuntaan. Syväjohtaminen perustuu transformationaaliseen johtamiseen, jossa johtajuus perustuu esimiehen ja alaisen väliseen vuorovaikutukseen. Tutkimuksen tavoitteena on tutkia, kuinka Porin Prikaatissa työskentelevät naissiviilityöntekijät kokevat johtajuuden puolustusvoimissa. Toiseksi työ tarkastelee sitä, kuinka syväjohtaminen näkyy esimiesten johtamiskäyttäytymisessä. Kolmanneksi työ tarkastelee sitä, miten johtajuus vaikuttaa työyhteisön työilmapiiriin ja miten sen avulla työilmapiiriä voidaan kehittää. Käyttämäni tutkimusote on fenomenografinen. Tutkimusmenetelmänä käytän puolistrukturoitua teemahaastattelua, jota analysoin diskurssianalyysin menetelmin, tuloksia analysoin kvalitatiivisesti. Tutkimusotteeni on holistisen hermeneuttinen. Kohderyhmänä ovat Porin Prikaatin siviiliviroissa työskentelevät naiset. Tutkimuksen tuloksena voidaan todeta, että johtajat käyttävät vaihtelevasti niin transformationaalista kuin transaktionaalistakin johtamista. Siviilityöntekijänaisten työnkuva on erittäin kaavamainen, jolloin se rajoittaa esimiehen mahdollisuutta käyttää tehokkaasti syväjohtamisen kulmakiviä. Tutkimuksen perusteella, keskeisimmät syväjohtamisen kulmakivet ovat luottamuksen rakentaminen ja ihmisen yksilöllinen kohtaaminen. Tärkeimmät keinot työilmapiirin kehittämiseen ovat tiedonkulun parantaminen ja tasa-arvoisuus työyhteisön sisällä.

Aims: In Escherichia coli, increased expression of efflux pumps and/or decreased expression of porins can confer multiple antibiotic resistance (MAR), causing resistance to at least three unrelated classes of antibiotics, detergents and dyes. It was hypothesized that in Campylobacter jejuni, the efflux systems CmeABC, CmeDEF and the major outer membrane porin protein, MOMP (encoded by porA) could confer MAR. Methods: The expression of cmeB, cmeF and porA in 32 MAR C. jejuni isolated from humans or poultry was determined by comparative (C)-reverse transcriptase (RT)-PCR and denaturing DHPLC. A further 13 ethidium bromide-resistant isolates and three control strains were also investigated. Accumulation of ciprofloxacin carbonyl cyanide-m-chlorophenyl hydrazone (CCCP) was also determined for all strains. Results: Although resistance to ethidium bromide has been associated with MAR, expression of all three genes was similar in the ethidium bromide-resistant isolates. These data indicate that CmeB, CmeF and MOMP play no role in resistance to this agent in C. jejuni. Six MAR isolates over-expressed cmeB, 3/32 over-expressed cmeB and cmeF. No isolates over-expressed cmeF alone. Expression of porA was similar in all isolates. All nine isolates that over-expressed cmeB contained a mutation in cmeR, substituting glycine 86 with alanine. All cmeB over-expressing isolates also accumulated low concentrations of ciprofloxacin, which were restored to wild-type levels in the presence of CCCP. Conclusions: These data indicate that over-expression of cmeB is associated with MAR in isolates of C. jejuni. However, as cmeB was over-expressed by only one-third (9/32) of MAR isolates, these data also indicate other mechanisms of MAR in C. jejuni.

The outer membrane protein M35 of Moraxella catarrhalis is an antigenically conserved porin. Knocking out M35 significantly increases the MICs of aminopenicillins. The aim of this study was to determine the biological mechanism of this potentially new antimicrobial resistance mechanism of M. catarrhalis and the behaviour of M35 in general stress situations.

All mitochondria have integral outer membrane proteins with beta-barrel structures including the conserved metabolite transporter VDAC (voltage dependent anion channel) and the conserved protein import channel Tom40. Bioinformatic searches of the Trypanosoma brucei genome for either VDAC or Tom40 identified a single open reading frame, with sequence analysis suggesting that VDACs and Tom40s are ancestrally related and should be grouped into the same protein family: the mitochondrial porins. The single T. brucei mitochondrial porin is essential only under growth conditions that depend on oxidative phosphorylation. Mitochondria isolated from homozygous knockout cells did not produce adenosine-triphosphate (ATP) in response to added substrates, but ATP production was restored by physical disruption of the outer membrane. These results demonstrate that the mitochondrial porin identified in T. brucei is the main metabolite channel in the outer membrane and therefore the functional orthologue of VDAC. No distinct Tom40 was identified in T. brucei. In addition to mitochondrial proteins, T. brucei imports all mitochondrial tRNAs from the cytosol. Isolated mitochondria from the VDAC knockout cells import tRNA as efficiently as wild-type. Thus, unlike the scenario in plants, VDAC is not required for mitochondrial tRNA import in T. brucei.

BACKGROUND: The outer membrane protein M35 is a conserved porin of type 1 strains of the respiratory pathogen Moraxella catarrhalis. It was previously shown that M35 is involved in the uptake of essential nutrients required for bacterial growth and for nasal colonization in mice. The aim of this study was (i) to characterize the potential roles of M35 in the host-pathogen interactions considering the known multifunctionality of porins and (ii) to characterize the degree of conservation in the phylogenetic older subpopulation (type 2) of M. catarrhalis. RESULTS: Isogenic m35 mutants of the type 1 strains O35E, 300 and 415 were tested for their antimicrobial susceptibility against 15 different agents. Differences in the MIC (Minimum Inhibitory Concentration) between wild-type and mutant strains were found for eight antibiotics. For ampicillin and amoxicillin, we observed a statistically significant 2.5 to 2.9-fold MIC increase (p < 0.03) in the m35 mutants. Immunoblot analysis demonstrated that human saliva contains anti-M35 IgA. Wild-type strains and their respective m35 mutants were indistinguishable with respect to the phenotypes of autoagglutination, serum resistance, iron acquisition from human lactoferrin, adherence to and invasion of respiratory tract epithelial cells, and proinflammatory stimulation of human monocytes. DNA sequencing of m35 from the phylogenetic subpopulation type 2 strain 287 revealed 94.2% and 92.8% identity on the DNA and amino acid levels, respectively, in comparison with type 1 strains. CONCLUSION: The increase in MIC for ampicillin and amoxicillin, respectively, in the M35-deficient mutants indicates that this porin affects the outer membrane permeability for aminopenicillins in a clinically relevant manner. The presence of IgA antibodies in healthy human donors indicates that M35 is expressed in vivo and recognized as a mucosal antigen by the human host. However, immunoblot analysis of human saliva suggests the possibility of antigenic variation of immunoreactive epitopes, which warrants further analysis before M35 can be considered a potential vaccine candidate.

Multiple isoforms of type 1 hexokinase (HK1) are transcribed during spermatogenesis in the mouse, including at least three that are presumably germ cell specific: HK1-sa, HK1-sb, and HK1-sc. Each of these predicted proteins contains a common, germ cell-specific sequence that replaces the porin-binding domain found in somatic HK1. Although HK1 protein is present in mature sperm and is tyrosine phosphorylated, it is not known whether the various potential isoforms are differentially translated and localized within the developing germ cells and mature sperm. Using antipeptide antisera against unique regions of HK1-sa and HK1-sb, it was demonstrated that these isoforms were not found in pachytene spermatocytes, round spermatids, condensing spermatids, or sperm, suggesting that HK1-sa and HK1-sb are not translated during spermatogenesis. Immunoreactivity was detected in protein from round spermatids, condensing spermatids, and mature sperm using an antipeptide antiserum against the common, germ cell-specific region, suggesting that HK1-sc was the only germ cell-specific isoform present in these cells. Two-dimensional SDS-PAGE suggested that all of the sperm HK1-sc was tyrosine phosphorylated, and that the somatic HK1 isoform was not present. Immunoelectron microscopy revealed that HK1-sc was associated with the mitochondria and with the fibrous sheath of the flagellum and was found in discrete clusters in the region of the membranes of the sperm head. The unusual distribution of HK1-sc in sperm suggests novel functions, such as extramitochondrial energy production, and also demonstrates that a hexokinase without a classical porin-binding domain can localize to mitochondria.

The Baltic Sea was studied with respect to selected organic contaminants and their ecotoxicology. The research consisted of analyses of total hydrocarbons, polycyclic aromatic hydrocarbons, bile metabolites, hepatic ethoxyresorufin-O-deethylase (EROD) activity, polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs). The contaminants were measured from various matrices, such as seawater, sediment and biota. The methods of analysis were evaluated and refined to comparability of the results. Polyaromatic hydrocarbons, originating from petroleum, are known to be among the most harmful substances to the marine environment. In Baltic subsurface water, seasonal dependence of the total hydrocarbon concentrations (THCs) was seen. Although concentrations of parent polycyclic aromatic hydrocarbons (PAHs) in sediment surface varied between 64 and 5161 ug kg-1 (dw), concentrations above 860 ug kg-1 (dw) were found in all the studied sub-basins of the Baltic Sea. Concentrations commonly considered to substantially increase the risk of liver disease and reproductive impairment in fish, as well as potential effects on growth (above 1000 ug kg-1 dw), were found in all the studied sub-basins of the Baltic Sea except Kattegat. Thus, considerable pollution in sediments was indicated. In bivalves, the sums of 12 PAHs varied on a wet weight basis between 44 and 298 ug kg-1 (ww). The predominant PAHs were high molecular weight and the PAH profiles of M. balthica differed from those found in sediment from the same area. The PAHs were both pyrolytic and petrogenic in origin, and a contribution from diesel engines was found, which indicates pollution of the Baltic Sea, most likely caused by the steadily increasing shipping in the area. The HPLC methods developed for hepatic EROD activity and bile metabolite measurements proved to be fast and suitable for the study of biological effects. A mixed function oxygenase enzyme system in Baltic Sea perch collected from the Gulf of Finland was induced slightly: EROD activity in perch varied from 0.30 14 pmol min-1 mg-1 protein. This range can be considered to be comparable to background values. Recent PAH exposure was also indicated by enhanced levels (213 and 1149 ug kg-1) of the bile metabolite 1-hydroxypyrene. No correlation was indicated between hepatic EROD activity and concentration of 1-hydroxypyrene in bile. PCBs and OCPs were observed in Baltic Sea sediment, bivalves and herring. Sums of seven CBs in surface sediment (0 5 cm) ranged from 0.04 to 6.2 ug kg-1 (dw) and sums of three DDTs from 0.13 to 5.0 ug kg-1 (dw). The highest levels of contaminants were found in the most eastern area of the Gulf of Finland where the highest total carbon and nitrogen content was found and where the lowest percentage proportion of p,p -DDT was found. The highest concentrations of CBs and the lowest concentration of DDTs were found in M. balthica from the Gulf of Finland. The highest levels of DDTs were found in M. balthica from the Hanö Bight, which is the outer part of the Bornholm Basin close to the Swedish mainland. In bivalves, the sums of seven CBs were 72 108 ug kg-1 (lw) and the sums of three DDTs were 66 139 ug kg-1 (lw). Results from temporal trend monitoring showed, that during the period 1985 2002, the concentrations of seven CBs in two-year-old female Baltic herring were clearly decreased, from 9 16 to 2 6 ug kg-1 (ww) in the northern Baltic Sea. At the same time, concentrations of three DDTs declined from 8 15 to 1 5 ug kg-1 (ww). The total concentration of the fat-soluble CBs and DDTs in Baltic herring muscle was shown to be age-dependent; the average concentrations in ten-year-old Baltic herring were three to five-fold higher than in two-year-old herring. In Baltic herring and bivalves, as well as in surface sediments, CB 138 and CB153 were predominant among CBs, whereas among DDTs p,p'-DDD predominated in sediment and p,p'-DDE in bivalves and Baltic herring muscle. Baltic Sea sediments are potential sources of contaminants that may become available for bioaccumulation. Based on ecotoxicological assessment criteria, cause for concern regarding CBs in sediments was indicated for the Gulf of Finland and the northern Baltic Proper, and for the northern Baltic Sea regarding CBs in Baltic herring more than two years old. Statistical classification of selected organic contaminants indicated high-level contamination for p,p'-DDT, p,p'-DDD, p,p'-DDE, total DDTs, HCB, CB118 and CB153 in muscle of Baltic herring in age groups two to ten years; in contrast, concentrations of a-HCH and g-HCH were found to be moderate. The concentrations of DDTs and CBs in bivalves is sufficient to cause biological effects, and demonstrates that long-term biological effects are still possible in the case of DDTs in the Hanö Bight.

Tutkimuksen tavoitteena oli selvittää vuoden 2008 lopulla alkaneen laman näkymistä Suomen metalliteollisuudessa. Tehtävänä oli arvioida missä määrin toimialan tuotanto ja investoinnit ovat muuttuneen laman myötä. Lisäksi tuli tarkastella näiden muutosten aluetaloudellisia vaikutuksia. Tutkimus kohdennettiin yhdeksään seutukuntaan: Helsinki, Jyväskylä, Kemi-Tornio, Pori, Raahe, Rauma, Tampere, Turku ja Vaasa. Näillä alueilla toimii merkittäviä alan yrityksiä ja metalliteollisuudella on huomattava vaikutus. Lähtötiedot kerättiin eLomake-kyselyllä, haastatteluin ja hyödyntämällä ETLA:n maakunnallista tuotantoennustetta. Laskelmat tehtiin seutukuntatasoisina vuosille 2009–2013 käyttäen Ruralia-instituutissa kehitettyä yleisen tasapainon RegFinDyn-aluemallia. Tutkimuksen on rahoittanut Työ- ja elinkeinoministeriö. Kyselyyn vastanneiden ja ETLA:n ennusteen mukainen seutukuntien metalliteollisuuden lamanäkemys ovat lähellä toisiaan ja tuottavat saman johtopäätöksen. Alan tuotannon menetyksillä on huomattava negatiivinen vaikutus seutukuntien talouskasvuun ja työllisyyteen. Alueellisen BKT:n muutos on lamavuosina välillä -1,5% (Helsinki) ja -18,0% (Raahe) normaalikehitykseen verrattuna kun kerroinvaikutuksetkin huomioidaan. Vaikutukset ovat suhteellisesti suurimmat seutukunnissa, jotka ovat pieniä ja joissa toimiala on erittäin merkittävä. Tällaisia seutukuntia ovat Kemi-Tornio ja Raahe. Negatiivinen vaikutus kokonaistuotantoon on merkittävä myös Jyväskylän, Rauman, Tampereen ja Turun seutukunnissa. Kokonaistuotannon lasku on pienin Helsingin ja Porin seutukunnissa. Vaasan seutukunnan metalliteollisuuden kehitystä voi taloustilanteeseen nähden pitää hyvänä. Metalliteollisuuden lähiajan vaikutus alueen talouskasvuun ja työllisyyteen on neutraali. Tuotannon menetykset veisivät yhdeksän seutukunnan metalliteollisuudesta yhteensä 3500 henkilötyövuotta kun muun talouden kompensoiva vaikutuskin huomioidaan. Eniten henkilötyövuosia, yli 700 menettäisi Raahen seutukunta. Myös Helsingin, Tampereen ja Turun seutukuntien työpaikkojen menetykset olisivat tuntuvat, 400–600. Tulokset heijastelevat seutukunnan vahvimpien metalliteollisuuden alatoimialojen tilannetta. Raahen ja Kemi-Tornion vahvin metalliteollisuuden toimiala on perusmetallien valmistus, jonka vientiä lama on tiputtanut erityisen rajusti. Vaasan seutukunnan muista parempaa tilannetta selittää ainakin osaksi seudun voimakas energiaklusteri. Kyselyn ja ETLA:n ennusteiden mukaiset tuotannon muutokset eivät tutkimuksen mukaan pitäisi johtaa niin suureen toimialan investointien laskuun kuin mitä kyselyn vastaukset ennakoivat. Kemi-Tornion ja Tampereen seutukunnissa investointien ennakoitiin laskevan 70%:lla lamavuosien aikana. Syynä äkkijarrutukseen lienee maailmantalouden suuri epävarmuus, rahoituksen saamisen kiristyminen sekä yritysten pyrkimys investointeja leikkaamalla vähentää riskejään ja parantaa taseitaan. Tulosten mukaan tehokas yritys olisi leikannut investointejaan paljon vähemmän, 5–10%:lla. Tulosten mukaan hintojen lasku eli deflaatio on seutukuntatasolla selvästi suurempaa kuin koko kansantalouden tasolla on laman aikana tähän mennessä koettu. Metalliteollisuudesta riippuvaisissa seutukunnissa kuluttajahinnat laskivat välillä 1,1% (Helsinki) ja 5,8% (Raahe). Deflaatio on tulosten mukaan kuitenkin vain tilapäinen ilmiö, mutta auttaa laman aikana ylläpitämään yksityistä kulutusta. Teknologiateollisuus ry:n äskettäin esitetty lamanäkemys on synkkä, metalliteollisuus toipuisi vain hitaasti lamasta. Kyselyyn vastanneet suurten yritysten edustajat olivat sitä vastoin suhteellisen toiveikkaita. Heidän arvionsa oli, että lama voisi taittua jo vuoden 2010 keväästä lähtien.

Identifying the structures of membrane bound proteins is critical to understanding their function in healthy and diseased states. We introduce a surface enhanced Raman spectroscopy technique which can determine the conformation of membrane-bound proteins, at low micromolar concentrations, and also in the presence of a substantial membrane-free fraction. Unlike conventional surface enhanced Raman spectroscopy, our approach does not require immobilization of molecules, as it uses spontaneous binding of proteins to lipid bilayer-encapsulated Ag nanoparticles. We apply this technique to probe membrane-attached oligomers of Amyloid-beta(40) (A beta(40)), whose conformation is keenly sought in the context of Alzheimer's disease. Isotope-shifts in the Raman spectra help us obtain secondary structure information at the level of individual residues. Our results show the presence of a beta-turn, flanked by two beta-sheet regions. We use solid-state NMR data to confirm the presence of the beta-sheets in these regions. In the membrane-attached oligomer, we find a strongly contrasting and near-orthogonal orientation of the backbone H-bonds compared to what is found in the mature, less-toxic A beta fibrils. Significantly, this allows a ``porin'' like beta-barrel structure, providing a structural basis for proposed mechanisms of A beta oligomer toxicity.

果实为开花植物所特有的发育器官,在种子的成熟和传播过程中发挥着重要作用。同时,肉质果实中含有丰富的营养物质,包括纤维素、维生素、抗氧化剂等,成为人们饮食的重要组成部分。由于果实的成熟衰老和抗病性直接影响果品的质量和市场价值,因此,研究果实成熟衰老和抗病性的调控机制具有重要的理论意义和应用前景。本文主要利用蛋白质组学的方法,探讨外源化学物质抑制果实成熟衰老和诱导抗病性的调控机制。 1. 硅对果实的抗病性诱导:用硅酸钠(1%)处理采后的甜樱桃果实,再接种褐腐病原菌(Molinilia fracticola),置于20C下,观测贮藏期间果实的发病率,并分析硅处理后诱导的主要蛋白质及调控机制。研究结果表明:硅酸钠处理可显著抑制贮藏期间褐腐病的发生,其抑病机理与硅诱导PR-蛋白的表达,提高果实的抗氧化水平,减轻由病原菌侵染造成的氧化胁迫相关。同时,硅处理还能保护细胞骨架结构,有利于增强果实对病原菌入侵的抵抗力。 2. 水杨酸对果实的抗病性诱导:用水杨酸(SA,2mM)在果园处理三种成熟度的甜樱桃果实,然后接种青霉病原菌(Penicillium expansum)观察其发病情况,并取样分析参与抗病性应答的主要蛋白质及调控机制。试验结果表明:SA处理能显著降低青霉病的发病率和抑制病斑扩展,而且SA对低成熟度甜樱桃果实的抗性诱导效果更好。在八成熟的果实中,有5个热激蛋白和4个脱氢酶蛋白被SA诱导,这些蛋白参与了糖酵解和三羧酸循环。抗氧化蛋白和PR蛋白主要参与较低成熟度果实的抗性应答,而热激蛋白和脱氢酶在较高成熟度果实的抗性应答中更明显,SA诱导的抗性与代谢途径相关。 3. 草酸对果实的抗性诱导:用5mM的草酸处理冬枣果实后,接种青霉菌(P. expansum),观察果实发病情况,测定果实相关的生理指标,分析参与果实抗性应答的主要蛋白质及调控机制。结果表明:草酸能明显延缓冬枣果实的衰老,提高果实对青霉菌的抗性。草酸处理能抑制果实乙烯的释放量和呼吸强度,延缓叶绿素的降解,减少乙醇积累。利用蛋白质组学的研究方法证实了在25个参与了草酸处理应答的蛋白中,胱硫醚-β-合酶结构域包含蛋白(CBB domain-containing protein)和3个与光合作用相关蛋白[二磷酸核酮糖羧化酶/加氧酶(Ribulose bisphosphate carboxylase/oxygenase activase, chloroplast precursor),二磷酸核酮糖羧化酶/加氧酶大亚基结合蛋白(RuBisCO large subunit-binding protein subunit beta, chloroplast precursor),植物光系统Ⅱ放氧复合蛋白2(PSII oxygen-evolving complex protein 2)]的表达量上调,乙醇脱氢酶的表达量出现下调。草酸处理还提高了与乙烯合成前体相关蛋白的表达,抑制了ACC合成酶的活性。草酸提高果实抗病的机制与延缓果实成熟衰老和保持果实抗性有关。 4. 果实衰老的调控机制:采用高氧(100%)和低氧(2-3%)处理苹果果实,观察果实衰老的进程,并基于蛋白质组学的研究方法,探讨苹果果实衰老与线粒体蛋白质组的关系。结果表明,在苹果衰老过程中有22个蛋白的表达量发生变化,这些蛋白主要参与了三羧酸循环,电子传递,碳代谢和胁迫应答。高氧处理能诱导氧化胁迫,加速了果实的衰老。质谱鉴定结果证明:在高氧胁迫下,超氧化物歧化酶(manganese superoxide dismutase,MnSOD)和线粒体外膜通道蛋白(porin) 的表达量降低,MnSOD的活性受到抑制,由此提高了线粒体中超氧阴离子的含量,增加了蛋白质的氧化损伤。 此外,高氧处理改变了porin的功能,导致了线粒体膜的透势发生变化,从而引起外膜损伤。由此阐明了活性氧在果实的成熟衰老调控中的重要作用。

Voltage-dependent anion channel (VDAC, also known as mitochondrial porin) is acknowledged to play an important role in stress-induced mammalian apoptosis. In this study, Paralichthys olivaceus VDAC (PoVDAC) gene was identified as a virally induced gene from Scophthalmus Maximus Rhabdovirus (SMRV)-infected flounder embryonic cells (FEC). The full length of PoVDAC cDNA is 1380 bp with an open reading frame of 852 bp encoding a 283 amino acid protein. The deduced PoVDAC contains one alpha-helix, 13 transmembrane beta-strands and one eukaryotic mitochondrial porin signature motif. Constitutive expression of PoVDAC was confirmed in all tested tissues by real-time PCR. Further expression analysis revealed PoVDAC mRNA was upregulated by viral infection. We prepared fish antiserum against recombinant VDAC proteins and detected the PoVDAC in heart lysates from flounder as a 32 kDa band on western blot. Overexpression of PoVDAC in fish cells induced apoptosis. Immunofluoresence localization indicated that the significant distribution changes of PoVDAC have occurred in virus-induced apoptotic cells. This is the first report on the inductive expression of VDAC by viral infection, suggesting that PoVDAC might be mediated flounder antiviral immune response through induction of apoptosis. RarA is an AraC-type regulator in Klebsiella pneumoniae, which, when overexpressed, confers a low-level multidrug-resistant (MDR) phenotype linked to the upregulation of both the acrAB and oqxAB efflux genes. Increased rarA expression has also been shown to be integral in the development of tigecycline resistance in the absence of ramA in K. pneumoniae. Given its phenotypic role in MDR, microarray analyses were performed to determine the RarA regulon. Transcriptome analysis was undertaken using strains Ecl8?rarA/pACrarA-2 (rarA-expressing construct) and Ecl8?rarA/pACYC184 (vector-only control) using bespoke microarray slides consisting of probes derived from the genomic sequences of K. pneumoniae MGH 78578 (NC_009648.1) and Kp342 (NC_011283.1). Our results show that rarA overexpression resulted in the differential expression of 66 genes (42 upregulated and 24 downregulated). Under the COG (clusters of orthologous groups) functional classification, the majority of affected genes belonged to the category of cell envelope biogenesis and posttranslational modification, along with genes encoding the previously uncharacterized transport proteins (e.g., KPN_03141, sdaCB, and leuE) and the porin OmpF. However, genes associated with energy production and conversion and amino acid transport/metabolism (e.g., nuoA, narJ, and proWX) were found to be downregulated. Biolog phenotype analyses demonstrated that rarA overexpression confers enhanced growth of the overexpresser in the presence of several antibiotic classes (i.e., beta-lactams and fluoroquinolones), the antifungal/antiprotozoal compound clioquinol, disinfectants (8-hydroxyquinoline), protein synthesis inhibitors (i.e., minocycline and puromycin), membrane biogenesis agents (polymyxin B and amitriptyline), DNA synthesis (furaltadone), and the cytokinesis inhibitor (sanguinarine). Both our transcriptome and phenotypic microarray data support and extend the role of RarA in the MDR phenotype of K. pneumoniae.

Abstract The emergence of multi and extensively drug resistant tuberculosis (MDRTB and XDRTB) has increased the concern of public health authorities around the world. The World Health Organization has defined MDRTB as tuberculosis (TB) caused by organisms resistant to at least isoniazid and rifampicin, the main first-line drugs used in TB therapy, whereas XDRTB refers to TB resistant not only to isoniazid and rifampicin, but also to a fluoroquinolone and to at least one of the three injectable second-line drugs, kanamycin, amikacin and capreomycin. Resistance in Mycobacterium tuberculosis is mainly due to the occurrence of spontaneous mutations and followed by selection of mutants by subsequent treatment. However, some resistant clinical isolates do not present mutations in any genes associated with resistance to a given antibiotic, which suggests that other mechanism(s) are involved in the development of drug resistance, namely the presence of efflux pump systems that extrude the drug to the exterior of the cell, preventing access to its target. Increased efflux activity can occur in response to prolonged exposure to subinhibitory concentrations of anti-TB drugs, a situation that may result from inadequate TB therapy. The inhibition of efflux activity with a non-antibiotic inhibitor may restore activity of an antibiotic subject to efflux and thus provide a way to enhance the activity of current anti-TB drugs. The work described in this thesis foccus on the study of efflux mechanisms in the development of multidrug resistance in M. tuberculosis and how phenotypic resistance, mediated by efflux pumps, correlates with genetic resistance. In order to accomplish this goal, several experimental protocols were developed using biological models such as Escherichia coli, the fast growing mycobacteria Mycobacterium smegmatis, and Mycobacterium avium, before their application to M. tuberculosis. This approach allowed the study of the mechanisms that result in the physiological adaptation of E. coli to subinhibitory concentrations of tetracycline (Chapter II), the development of a fluorometric method that allows the detection and quantification of efflux of ethidium bromide (Chapter III), the characterization of the ethidium bromide transport in M. smegmatis (Chapter IV) and the contribution of efflux activity to macrolide resistance in Mycobacterium avium complex (Chapter V). Finally, the methods developed allowed the study of the role of efflux pumps in M. tuberculosis strains induced to isoniazid resistance (Chapter VI). By this manner, in Chapter II it was possible to observe that the physiological adaptation of E. coli to tetracycline results from an interplay between events at the genetic level and protein folding that decrease permeability of the cell envelope and increase efflux pump activity. Furthermore, Chapter III describes the development of a semi-automated fluorometric method that allowed the correlation of this efflux activity with the transport kinetics of ethidium bromide (a known efflux pump substrate) in E. coli and the identification of efflux inhibitors. Concerning M. smegmatis, we have compared the wild-type M. smegmatis mc2155 with knockout mutants for LfrA and MspA for their ability to transport ethidium bromide. The results presented in Chapter IV showed that MspA, the major porin in M. smegmatis, plays an important role in the entrance of ethidium bromide and antibiotics into the cell and that efflux via the LfrA pump is involved in low-level resistance to these compounds in M. smegmatis. Chapter V describes the study of the contribution of efflux pumps to macrolide resistance in clinical M. avium complex isolates. It was demonstrated that resistance to clarithromycin was significantly reduced in the presence of efflux inhibitors such as thioridazine, chlorpromazine and verapamil. These same inhibitors decreased efflux of ethidium bromide and increased the retention of [14C]-erythromycin in these isolates. Finaly, the methods developed with the experimental models mentioned above allowed the study of the role of efflux pumps on M. tuberculosis strains induced to isoniazid resistance. This is described in Chapter VI of this Thesis, where it is demonstrated that induced resistance to isoniazid does not involve mutations in any of the genes known to be associated with isoniazid resistance, but an efflux system that is sensitive to efflux inhibitors. These inhibitors decreased the efflux of ethidium bromide and also reduced the minimum inhibitory concentration of isoniazid in these strains. Moreover, expression analysis showed overexpression of genes that code for efflux pumps in the induced strains relatively to the non-induced parental strains. In conclusion, the work described in this thesis demonstrates that efflux pumps play an important role in the development of drug resistance, namely in mycobacteria. A strategy to overcome efflux-mediated resistance may consist on the use of compounds that inhibit efflux activity, restoring the activity of antimicrobials that are efflux pump substrates, a useful approach particularly in TB where the most effective treatment regimens are becoming uneffective due to the increase of MDRTB/XDRTB.

Viimeisen vuosikymmenen aikana Suomi on osallistunut toistaiseksi vaarallisimpaan kriisinhallintaoperaatioon koko rauhanturvahistoriansa aikana. Tämä on johtanut henkisten ongelmien kasvuun kriisinhallintaveteraanien keskuudessa. Tästä johtuen Suomen puolustusvoimat aloitti vuonna 2008 kriisinhallintaveteraaneille suunnatun psykososiaalisen tukijärjestelmän. Tämän tutkimuksen tavoitteena on kartoittaa kriisinhallintaoperaatioista palaavien sotilaiden psykososiaalista tukea Puolustusvoimissa. Tutkimus pyrkii vastaamaan minkälaisen prosessin rauhanturvaajat käyvät läpi kotiutumisen yhteydessä. Tutkimuksen tulokset on kerätty vuosina 2013 ja 2014 pääasiassa kahdella asiantuntijahaastattelulla. Haastateltavat asiantuntijat olivat Porin prikaatin henkilökuntaa, jotka vastasivat psykososiaalisen tuen järjestämisestä. Keskeisimpänä tuloksena ja johtopäätöksenä voidaan todeta Puolustusvoimien järjestämän psykososiaalisen tukijärjestelmän olevan tarpeeseen ja resursseihin suhteutettuna tarkoituksenmukaisesti toteutettu. Rauhanturvaajat käyvät kotiutumisen jälkeen läpi tehokkaan prosessin, jonka päätarkoituksena on kouluttaa heille vertaistukea. Tämän lisäksi järjestelmä pyrkii seulomaan mahdollisesti tuen tarpeessa olevia henkilöitä ja saattamaan heidät tarvittavan jatkohoidon piiriin.
