We investigated the condensation of calf thymus DNA by amphiphilic polystyrene(m)-b-poly(l-lysine)(n) block copolymers (PSm-b- PLys(n), m, n = degree of polymerization), using small-angle X-ray scattering, polarized optical microscopy and laser scanning confocal microscopy. Microscopy studies showed that the DNA condenses in the form of fibrillar precipitates, with an irregular structure, due to electrostatic interactions between PLys and DNA. This is not modified by the presence of hydrophobic PS block. Scattering experiments show that the structure of the polyplexes corresponds to a local order of DNA rods which becomes more compact upon increasing n. It can be concluded that for DNA/ PSm-b- PLys(n) polyplexes, the balance between the PLys block length and the excess charge in the system plays an essential role in the formation of a liquid crystalline phase.
Nonspherical assemblies generated from polystyrene-b-poly(L-lysine) polyelectrolyte block copolymers

This report describes the aqueous solution self-assembly of a series of polystyrene(m)-b-poly(L-lysine)n block copolymers (m = 8-10; n = 10-70). The polymers are prepared by ring-opening polymerization of epsilon-benzyloxycarbonyl-L-lysine N-carboxyanhydride using amine terminated polystyrene macroinitiators, followed by removal of the benzyloxycarbonyl side chain protecting groups. The critical micelle concentration of the block copolymers determined using the pyrene probe technique shows a parabolic dependence on peptide block length exhibiting a maximum at n = approximately 20 (m = 8) or n = approximately 60 (m = 10). The shape and size of the aggregates has been studied by dynamic and static light scattering, small-angle neutron scattering (SANS), and analytical ultracentrifugation (AUC). Surprisingly, Holtzer and Kratky analysis of the static light scattering results indicates the presence of nonspherical, presumably cylindrical objects independent of the poly(L-lysine)n block length. This is supported by SANS data, which can be fitted well by assuming cylindrical scattering objects. AUC analysis allows the molecular weight of the aggregates to be estimated as several million g/mol, corresponding to aggregation numbers of several 10s to 100s. These aggregation numbers agree with those that can be estimated from the length and diameter of the cylinders obtained from the scattering results.

The microphase structure of a series of polystyrene-b-polyethylene oxide-b-polystyrene (SEOS) triblock copolymers with different compositions and molecular weights has been studied by solid-state NMR, DSC, wide and small angle X-ray scattering (WAXS and SAXS). WAXS and DSC measurements were used to detect the presence of crystalline domains of polyethyleneoxide (PEO) blocks at room temperature as a function of the copolymer chemical composition. Furthermore, DSC experiments allowed the determination of the melting temperatures of the crystalline part of the PEO blocks. SAXS measurements, performed above and below the melting temperature of the PEO blocks, revealed the formation of periodic structures, but the absence or the weakness of high order reflections peaks did not allow a clear assessment of the morphological structure of the copolymers. This information was inferred by combining the results obtained by SAXS and (1)H NMR spin diffusion experiments, which also provided an estimation of the size of the dispersed phases of the nanostructured copolymers. The microphase separation of block copolymer (BCP) thin films can afford a simple and cost-effective means to studying nanopattern surfaces, and especially the fabrication of nanocircuitry. However, because of complex interface effects and other complications, their 3D morphology, which is often critical for application, can be more complex than first thought. Here, we describe how emerging microscopic methods may be used to study complex BCP patterns and reveal their rich detail. These methods include helium ion microscopy (HIM) and high resolution x-section transmission electron microscopy (XTEM), and complement conventional secondary electron and atomic force microscopies (SEM and TEM). These techniques reveal that these structures are quite different to what might be expected. We illustrate the advances in the understanding of BCP thin film morphology in several systems, which result from this characterization. The systems described include symmetric, lamellar forming polystyrene-b-polymethylmethacrylate (PS-b-PMMA), cylinder forming polystyrene-b-polydimethylsiloxane (PS-b-PDMS), as well as lamellar and cylinder forming patterns of polystyrene-b-polyethylene oxide (PS-b-PEO) and polystyrene-b-poly-4-vinylpyridine (PS-b-P4VP). Each of these systems exhibits more complex arrangements than might be first thought. Finding and developing techniques whereby complex morphologies, particularly at very small dimensions, can be determined is critical to the practical use of these materials in many applications. The importance of quantifying these complex morphologies has implications for their use in integrated circuit manufacture, where they are being explored as alternative pattern forming methods to conventional UV lithography.

Le protocole pour le greffage irréversible du copolymère amphiphile polystyrène-b-poly (acrylate de sodium) PS-b-PANa, sur un substrat de mica et de silice hydrophobe a été développé, en utilisant la méthode de greffage à partir de solution. Les propriétés de surface du bloc chargé ont été évaluées. L’effet de la force ionique sur le gonflement des chaînes a été investigué par ellipsométrie. Les forces d’interaction entre les surfaces recouvertes du copolymère ont été évaluées par la technique SFA. Les profils de force ont démontré être stables et nettement répulsifs en compression et décompression, montrant l’irréversibilité du greffage. Les forces de frottement entre les brosses de PANa sont élevées, mais aucune évidence d’endommagement de la surface n’a été observée. La comparaison entre le comportement à la surface des chaînes de l’acide polyacrylique PAA et celles du PANa, obtenues par deux méthodes de greffage différentes, est également investiguée.

L'auto-assemblage des copolymères à bloc (CPBs) attire beaucoup d'intérêt grâce à leur capacité de générer spontanément des matériaux ordonnés avec des propriétés uniques. Les techniques Langmuir-Blodgett (LB) et Langmuir-Schaefer (LS) sont couramment utilisées pour produire des monocouches ou des films ultraminces à l'interface air/eau suivi de transfert aux substrats solides. Les films LB/LS de CPBs amphiphiles s'auto-assemblent dans des morphologies variables dépendamment de la composition du CPB ainsi que d'autres facteurs. Dans notre travail, nous avons étudié les films LB/LS de polystyrène-b-poly(4-vinyl pyridine) (PS-P4VP) et leurs complexes supramoléculaires avec le naphtol (NOH), l'acide naphtoïque (NCOOH) et le 3-n-pentadécylphenol (PDP). La première partie de ce mémoire est consacré à l'investigation du PS-P4VP complexé avec le NOH et le NCOOH, en comparaison avec le PS-P4VP seul. Il a été démontré qu'un plateau dans l'isotherme de Langmuir, indicatif d'une transition de premier ordre, est absent à des concentrations élevées des solutions d'étalement des complexes. Cela a été corrélé avec l'absence de morphologie en nodules avec un ordre 2D hexagonal à basse pression de surface. L'ordre au-delà de la pression de cette transition, lorsque présente, change à un ordre 2D carré pour tout les systèmes. La deuxième partie du la mémoire considère à nouveau le système PS-P4VP/ PDP, pour lequel on a démontré antérieurement que la transition dans l'isotherme correspond a une transition 2D d'un ordre hexagonal à un ordre carré. Cela est confirmé par microscopie à force atomique, et, ensuite, on a procédé à une étude par ATR-IR des films LB pour mieux comprendre les changements au niveau moléculaire qui accompagnent cette transition. Il a été constaté que, contrairement à une étude antérieure dans la littérature sur un autre système, il n'y a aucun changement dans l'orientation des chaînes alkyles. Au lieu de cela, on a découvert que, aux pressions au-delà de celle de la transition, le groupe pyridine, qui est orienté à basse pression, devient isotrope et qu'il y a une augmentation des liaisons hydrogènes phénol-pyridine. Ces observations sont rationalisées par un collapse partiel à la pression de transition de la monocouche P4VP, qui à basse pression est ordonné au niveau moléculaire. Cette étude a mené à une meilleure compréhension des mécanismes moléculaires qui se produisent à l'interface air/eau, ce qui fournit une meilleure base pour la poursuite des applications possibles des films LB/LS dans les domaines de nanotechnologie.

Clay-containing nanocomposites of polystyrene-b-poly(ethylene-co-butylene)-b-polystyrene (SEBS) copolymers having cylindrical domains were obtained by melt extrusion using a tape die. One type of sample (SEBS-MA) had maleic anhydride attached to the middle block. Two types of organoclays were added, namely Cloisite 20A and Cloisite 30B. Small angle X-ray scattering and transmission electron microscopy (TEM) analyses showed that the addition of 20A clay to SEBS and SEBS-MA resulted in nanocomposites with intercalated and partially exfoliated structures, respectively. The addition of 30B clay to SEBS and SEBS-MA promoted the formation of composites containing relatively large micron-sized and partially exfoliated clay particles, respectively. Our TEM analysis revealed that clay particles embedded in SEBS are preferably in contact with the polystyrene cylindrical domains, while in SEBS-MA they are in contact with the maleated matrix. The extrusion processing promoted alignment of the axes of the polystyrene cylinders along the extrusion direction in all samples, and the basal planes of the clay particles were mostly parallel to the main external surfaces of the extruded tapes. © 2013 Society of Chemical Industry.

Im Rahmen dieser Dissertation wurden die Synthese und die Charakterisierung verschiedener, zum Teil neuartiger Blockcopolymere beschrieben, wobei die Einbeziehung radikalischer Polymeri-sationsmechanismen den konzeptionellen Kern ausmachte. Mit einer auf die jeweilige Kombination von Monomeren zugeschnittenen Syntheseroute gelang die Verknüpfung von Segmenten, die allein mittels der herkömmlich zur Synthese von Blockcopolymeren genutzten, ionischen Mechanismen nur mit hohem Aufwand oder gar nicht zu verbinden sind. Auf materieller Seite stand die Herstellung amphiphiler Strukturen im Vordergrund. Diese wurden entweder direkt beim Aufbau der Blockcopo-lymere oder nach anschließender polymeranaloger Umsetzung eines ihrer Segmente erhalten. Solche amphiphilen Substanzen besitzen aufgrund ihrer Grenzflächenaktivität Anwendungspotential z. B. als Stabilisatoren in der Dispersionspolymerisation oder als Flokkulantien. Es wurden drei Verfahren zum Aufbau von Blockcopolymeren untersucht:1. Die Transformation von anionischer zu freier radikalischer Polymerisation für die Synthese von Polystyrol-b-poly(N-vinylformamid) (PS-b-P(VFA)).2. Die Transformation von anionischer zu kontrollierter radikalischer Polymerisation (ATRP) für den Aufbau von Blockcopolymeren aus Poly(dimethylsiloxan) PDMS und Segmenten von t-Butylacrylat (t-BuA) bzw. (2-(Trimethylsiloxy)ethyl)methacrylat (TMS-HEMA).3. Die kontrollierte radikalische Polymerisation unter Einsatz von Triazolinyl als Gegenradikal zur Synthese von Poly[(2-(trimethylsiloxy)ethyl)methacrylat]-b-polystyrol (P(TMS-HEMA)-b-PS) als alternative Route zur anionischen Polymerisation.

In der vorliegenden Arbeit werden drei Polyelektrolyt-Architekturen zunehmender Verzweigung auf der Basis von L-Lysin vorgestellt. Zunächst wird auf das Aggregationsverhalten des linearen Blockpolyelektrolyten Polystyrol-b-Poly(L-Lysin) eingegangen. Dabei wird der Einfluss der Lysinblocklänge (NLys = 10…70) bei gleich bleibendem, sehr kurzem hydrophobem Polystyrolsegment untersucht. Wie sich der Polystyrolblock auf die Helixbildung auswirkt, kann mit Hilfe von Zirkulardichroismus nachgewiesen werden. Nach Bestimmung der kritischen Mizellenkonzentration über Fluoreszenzspektroskopie wird mittels statischer Streumethoden (SLS, SANS) eine zylinderförmige Mizelle mit einem Kernradius von 4,4 nm charakterisiert. Im zweiten Abschnitt werden die optischen Eigenschaften von sternförmigen, rot fluoreszierenden Perylendiimid-Poly(L-lysin)-Konjugaten mit variierender Armzahl (n = 4, 8, 16) und Kettenlänge (NLys = 10, 50, 100) beschrieben. Die guten Absorptionseigenschaften und schlechten Fluoreszenzeigenschaften zeigen weder eine Abhängigkeit von der Sekundärstruktur der Poly(L-lysin)-Arme noch von deren Zahl oder Kettenlänge. Der dritte Teil der Arbeit handelt von amin- (Fmoc-, TFAA- oder Z-) geschützten L-Lysindendrone bis zur dritten Generation, welche durch Verknüpfung der Carboxylfunktion der Dendrone mit der Amingruppe von Vinylbenzylamin in Makromonomere überführt werden. Das Polymerisationsverhalten der Makromonomere wird in Abhängigkeit der Dendrongeneration und der Monomerkonzentration zu Beginn der Polymerisation untersucht. Anhand von AFM-Aufnahmen kann nachgewiesen werden, dass das Polystyrolrückgrat der dendronisierten Polymere der ersten Z-geschützten Generation eine Streckung erfährt.

Die dieser Arbeit zugrundeliegenden Nanopartikel wurden mittels der Makromonomer-Strategie aus polymerisierbaren Polystyrol-b-Poly(2-vinylpyridin) Oligomeren dargestellt. Die Bürstenpolymere besitzen eine polare PS-Schale und einen polaren Kern (P2VP), dessen Polarität durch Quaternisierung deutlich erhöht werden kann. Die Bürstenpolymere weisen bei Molmassen um 400 - 800 kg/mol einen Teilchendurchmesser von ca. 15 - 20 nm auf. Die Nanopartikel eignen sich dazu, hydrophile Farbstoffe in unpolaren Lösungsmitteln zu solubilisieren. Durch spektroskopische Untersuchungen wurden in Abhängigkeit der chemischen Struktur und der Bürstenpolymere Beladungsgrade von über 1 g Farbstoff pro Gramm Polymer ermittelt. Die Beladung der Nanopartikel folgt hierbei einer nichttrivialen Kinetik, was möglicherweise durch eine wasserinduzierte Überstrukturbildung während der Beladung bedingt ist. Mittels isothermer Titrationskalorimetrie konnten die Wechselwirkungen zwischen polymeren Substrat und niedermolekularen Liganden genauer charakterisiert werden. Teilweise werden hierbei zweistufige Titrationsverläufe und "überstöchiometrische" Beladung der Bürstenpolymere beobachtet. Den Hauptbeitrag zur Wechselwirkung liefert hierbei die exotherme Wechselwirkung zwischen basischen Polymer und saurem Farbstoff. Die hohe Farbstoffbeladung führt zur deutlichen Vergrößerung der einzelnen Nanopartikel, was sowohl in Lösung durch Lichtstreu-Techniken als auch auf Oberflächen mit Hilfe des AFM zu beobachten ist. Durch Untersuchungen mit der analytischen Ultrazentrifuge konnte nachgewiesen werden, dass sich der eingelagerte Farbstoff in einem Polaritäts-abhängigen Gleichgewicht mit der Umgebung steht, er somit auch wieder aus den Nanopartikeln freigesetzt werden kann. Darüberhinaus wurden im Rahmen der Arbeit erste Erfolge bei der Synthese von wasserlöslichen Nanopartikeln mit Poly(2-vinylpyridin)-Kern erzielt. Als hierfür geeignet stellte sich eine Synthesestrategie heraus, bei der zunächst ein Bürstenpolymer mit P2VP-Seitenketten dargestellt und dieses anschließend mit geeignet funktionalisierten Polyethylenoxid-Ketten zum Kern-Schale Teilchen umgesetzt wurde. Neben Untersuchungen zum Mizellisierungsverhalten von PEO-b-P2VP Makromonomeren wurden deren Aggregate in Wasser hinsichtlich ihrer Zelltoxizität durch in-vitro Experimente an C26-Mäusekarzinom-Zellen charakterisiert. Die extrem geringe Toxizität macht das PEO-P2VP System zu einem potentiellen Kandidaten für drug-delivery Anwendungen. Besonders die pH-abhängige Löslichkeitsänderung des Poly(2-vinylpyridin) erscheint hierbei besonders interessant.

Integrin-mediated cell adhesion to extracellular matrices provides signals essential for cell cycle progression and differentiation. We demonstrate that substrate-dependent changes in the conformation of adsorbed fibronectin (Fn) modulated integrin binding and controlled switching between proliferation and differentiation. Adsorption of Fn onto bacterial polystyrene (B), tissue culture polystyrene (T), and collagen (C) resulted in differences in Fn conformation as indicated by antibody binding. Using a biochemical method to quantify bound integrins in cultured cells, we found that differences in Fn conformation altered the quantity of bound α5 and β1 integrin subunits but not αv or β3. C2C12 myoblasts grown on these Fn-coated substrates proliferated to different levels (B > T > C). Immunostaining for muscle-specific myosin revealed minimal differentiation on B, significant levels on T, and extensive differentiation on C. Differentiation required binding to the RGD cell binding site in Fn and was blocked by antibodies specific for this site. Switching between proliferation and differentiation was controlled by the levels of α5β1 integrin bound to Fn, and differentiation was inhibited by anti-α5, but not anti-αv, antibodies, suggesting distinct integrin-mediated signaling pathways. Control of cell proliferation and differentiation through conformational changes in extracellular matrix proteins represents a versatile mechanism to elicit specific cellular responses for biological and biotechnological applications.

Background and objective The time course of cardiopulmonary alterations after pulmonary embolism has not been clearly demonstrated and nor has the role of systemic inflammation on the pathogenesis of the disease. This study aimed to evaluate over 12 h the effects of pulmonary embolism caused by polystyrene microspheres on the haemodynamics, lung mechanics and gas exchange and on interleukin-6 production. Methods Ten large white pigs (weight 35-42 kg) had arterial and pulmonary catheters inserted and pulmonary embolism was induced in five pigs by injection of polystyrene microspheres (diameter similar to 300 mu mol l(-1)) until a value of pulmonary mean arterial pressure of twice the baseline was obtained. Five other animals received only saline. Haemodynamic and respiratory data and pressure-volume curves of the respiratory system were collected. A bronchoscopy was performed before and 12 h after embolism, when the animals were euthanized. Results The embolism group developed hypoxaemia that was not corrected with high oxygen fractions, as well as higher values of dead space, airway resistance and lower respiratory compliance levels. Acute haemodynamic alterations included pulmonary arterial hypertension with preserved systemic arterial pressure and cardiac index. These derangements persisted until the end of the experiments. The plasma interleukin-6 concentrations were similar in both groups; however, an increase in core temperature and a nonsignificant higher concentration of bronchoalveolar lavage proteins were found in the embolism group. Conclusion Acute pulmonary embolism induced by polystyrene microspheres in pigs produces a 12-h lasting hypoxaemia and a high dead space associated with high airway resistance and low compliance. There were no plasma systemic markers of inflammation, but a higher central temperature and a trend towards higher bronchoalveolar lavage proteins were found. The process of adhesion of three different strains of Trichomonas vaginalis to a polystyrene substrate was analysed. The process of adhesion was dependent on the time of incubation and the pH of the phosphate-buffered solution (PBS) in which the parasites were suspended. The highest indices of adhesion were observed after an incubation time of 60 min at pH 6.6. The adhesion index increased when the parasites were incubated in the presence of culture media or when Ca++ or Mg++ was added to the PBS solution, whereas cytochalasin B, trypsin or neuraminidase reduced adhesion. Incubation of the parasites in the presence of poly-L-lysine facilitated the process of adhesion. Incubation of the parasites or polystyrene beads in the presence of poly-L-lysine led to important changes in their surface charge.

The present work focuses on the modification of the commonly used thermoplastics, polypropylene and polystyrene using nanosilica preparcd from a cheap source of sodium silicate. Melt compounding technique has been used for nanocomposite preparation as it is simple and suited to injection moulding. Nanosilica in a polymer matrix provide significant enhancement in strength, stiffness and impact strength. Incorporation of silica particles in a polymer also improves its thennal stability. To achieve better dispersion of fillers in polymer matrices the mixing was done at different shear rates. The enhancement in material properties indicates that at higher shear rates there is greater interaction between particles and the matrix and it depends on filler concentration and type of polymer used. N anosilica is a useful filler in thennoplastic polymers and has been applied in automotive applications, electronic appliances and consumer goods.This thesis is divided into six chapters. General introduction to the topic is described in chapter 1. Salient features of polymer nanocomposites, their synthesis, properties and applications are presented. A review of relevant literature and the scope and objectives are also mentioned in this chapter.The materials used and the vanous experimental method and techniques employed in the study are described in chapter 2. Preparation of nanocomposites by melt blending using Thenno Haake Rheocord, preparation of samples, evaluation of mechanical and thennal properties using UTM, Impact testing and characterization using DMA, TGA and DSC and morphology by SEM are described.The preparation of nanosilica from a laboratory scale to a pilot plant scale is described in chapter 3. Generation of surface modified silica, evaluation of kinetic parameters of the synthesis reaction, scale up of the reactor and modeling of the reactor are also dealt with in this chapter.The modification of the commodity thennoplastic, Polypropylene using nanosilica is described in chapter 4. Preparation of PP/silica nanocomposites, evaluation of mechanical properties, thermal and crystallization characteristics, water absorption and ageing resistance studies are also presented.The modification of Polystyrene using synthesized nanosilica IS described in chapter 5. The method of preparation of PS/silica nanocomposites, evaluation of mechanical properties (static and dynamic), thermal properties melt flow characteristics using Haake Rheocord, water absorption and ageing resistance of these nanocomposites are studied.

Vibrational overtone spectra of styrene (liquid) and polystyrene (solid), studied by the laser-induced thermal lens (for ΔV=6) and the conventional near infrared absorption (for ΔV=3–5) techniques, are reported. For polystyrene, the overtone energy-bond length correlation predicts that the aryl CH bonds are ∼0.0005 Å longer than that in benzene, while no such conclusions could be drawn on styrene. Thesp 3 CH overtones in polystyrene are observed on the low energy side of the aryl CH overtones.
